Study of Grid Dependency of Sheet Atomization Model of a Pressure-Swirl Atomizer (스월형 분사기 분무 예측 모델에서의 격자 의존성 연구)

  • An improved spray model of a pressure-swirl atomizer was developed and the grid dependency of the model was investigated. Since the Lagrangian-Eulerian approach was adopted for tracking droplets, very small grids could not be used. However, in order to detect swirl flow accurately, small grids were needed because of the consideration of swirl injection. In order to overcome these limitations, numerical studies were performed by using various grids with cell sizes ranging from 10.0 $\times$ 10 mm to 0.625 $\times$ 0.625 mm. From these calculated results, it was observed that the most efficient grid cell size was 1.25 $\times$ 1.25 mm.

  • Aerosol slurry samples were collected in 60-min interval using Korean Semi-continuous Elements in Aerosol Sampler (KSEAS) between May 19 and June 6, 2010 at an urban site of Gwangju. The $PM_{2.5}$ samples were collected with a flow rate of 16.7 L/min and particles are grown by condensation of water vapor in a condenser maintained at ${\sim}5^{\circ}C$ after saturation by direct injection of steam. The resulting droplets are collected in a liquid slurry with a airdroplet separator. Concentrations of 16 elements (Al, Fe, Mn, Ca, K, Cu, Zn, Pb, Cd, Cr, Ti, V, Ni, Co, As, Se) in the collected slurry samples were determined off-line by ICP-MS. KSEAS sample analysis encompassed the sampling periods for which 24-hr average elemental species concentrations were calculated for comparison with those derived from 24-hr integrated filter samples. Relationship between elemental species measured by two methods indicated high correlation coefficients (r), mostly greater than r of 0.80. However, we note that concentrations of Al, K, Ca, Mn, and Fe, which are often associated with crustal elemental particles, in the KSEAS samples, were substantially lower (1.4~11 times) than those found in the typical filter-based samples. This discrepancy is probably due to difficulties in transferring insoluble dust particles to the collection vials in the KSEAS. Temporal profiles of elemental concentrations indicate that some transient events in their concentrations are observed over the sampling periods. For the elemental species studied, atmospheric concentrations during the transient events increased by factors of 4 in Mn~80 in Zn, compared to their background levels. Principle component analyses were applied to the hourly KSEAS data sets to identify sources affecting the concentrations of the metal constituents observed. In this study, we conclude that hourly measurements for particle-bound elemental constituents were extremely useful for revealing the short-term variability in their concentrations and developing insights into their sources.

  • Automatic addition of glucose and ammonium to an alcohol distille깨 wastewater and the control of them at low $\infty$ncentration were tried to efficiently increase the cell concentration of a baker's yeast c비tivated in that wastewater. Added g glucose was indirectly controlled to less than 116 mg/L by a method which used DO as control parameter. Ammonium was a automatically measured and controlled within the range of 7.0~27.7 mM by a homemade on-line system which adopted FIA a as measurement method. Maximum specific growth rate and biomass yield to glucose were $0.21 hr^{-1}$ and about 0.78 g/g, w which were significantly increased values in contrast to those of an experiment without ammonium control. Biomass yield to a ammonium was 11.3 gIg. Cell cone엉ntration could be increased from 2.6 g/L to 18.5 g/L by the add ion of glucose and a ammonium.

  • Hydraulic fracturing is used as a method for promoting the fluid flow in the rock and, in the energy field such as geothermal development and the development of sales gas, many studies has been actively conducted. In many cases, hydraulic fracturing is not performed in isotropic rock and especially in the case of sedimentary rocks, hydraulic fracturing is conducted in the transverse isotropic rock. The direction of the crack growth on hydraulic fracturing does not necessarily coincides with the direction of maximum principal stress in the transverse isotropic rock. Therefore, in this study, bonded particle model with hydro-mechanical coupling analysis was adopted for analyzing the characteristics of hydraulic fracturing in transverse isotropic rock. In addition, experiments of hydraulic fracturing were conducted in laboratory-scale to verify the validity of numerical analysis. In this study, the crack growth and crack patterns showed significant differences depending on the viscosity of injection fluid, the angle of bedding plane and the influence of anisotropy. In the case of transverse isotropic model, the shear crack growth due to hydraulic fracturing appeared prominently.

  • Open-loop groundwater heat pump (GWHP) system generally has benefits such as a higher coefficient of performance (COP), lower initial cost, and flexible system size. The hydrogeological conditions in Korea have the potential to facilitate the use of the GWHP system because a large number of monitoring wells show stable groundwater temperatures, shallow water levels, and high well yields. However, few studies have been performed in Korea regarding the GWHP system and the most studies among them dealt with Standing Column Well (SCW). Because the properties of the aquifer have an influence on designing open-loop systems, it is necessary to perform studies on various hydrogeological settings. In this study, the hydrogeological and thermal properties were estimated through various tests in the riverside alluvial layer where a GWHP system was installed. Under different groundwater flow velocities and pumping and injection rates, a sensitivity analysis was performed to evaluate the effect of such properties on the design of open-loop systems. The results showed that hydraulic conductivity and thermal dispersivity of the aquifer are the most sensitive parameters in terms of performance and environmental aspects, and sensitivities of the properties depend on conditions.

Studies on Voice Changes Associated with Alcohol Intake (음주에 따른 음성 변화에 대한 고찰)

  • Background and Objectives : It has been pointed out that alcohol intake in human beings induces changes in voice register and maximum phonation time. These changes supposedly result from injection of the vibratory vocal folds. The purpose of this study was to clarify the voice changes associated with alcohol intake and the changes of laryngeal mucosa. Materials and Methods : The subjects included 29 volunteers, including 20 men and 9 women ranging in age from 22 to 31 years. Alcohol intake was accomplished by oral administration of 23% soju 1 bottle (255cc). Serum alcohol concentration levels were evaluated hourly for 3 h after ingestion of alcohol. Seven measurements were performed at pre-alcohol intake and post-alcohol intake hourly : fundamental frequency, jitter, shimmer, noise to harmonic ratio as the acoustic analysis, maximal phonation time, mean flow rate, and subglottal pressure as the aerodynamic analysis. The changes of laryngeal mucosa were evaluated by flexible laryngoscope at each measurement. Results : By comparing the acoustic and aerodynamic data and laryngeal mucosa before and after alcohol intake, there were not remarkable changes (p>0.05). Conclusion : The voice and laryngeal mucosa have not remarkably changed according to alcohol concentration in this study. Furthermore studies on the voice change induced by multiple alcohol concentrations are required.

Quantitative Analysis of the Eleven Marker Components in Traditional Korean Formula, Jakyakgamcho-Tang Decoction Using an Ultra-Performance Liquid Chromatography Coupled to Electrospray Ionization Tandem Mass Spectrometry (UPLC-MS/MS를 이용한 작약감초탕 물 추출물 중 11종 성분의 함량분석)

  • Jakyakgamcho-tang is a well-known traditional herbal medicine and has been used for the treatment of mainly pains in oriental medicine. In this study, analytical method for the quantitative determination of the eleven marker components, gallic acid (1), oxypaeoniflorin (2), paeoniflorin (3), albiflorin (4), liquiritin (5), isoliquiritin (6), ononin (7), liquiritigenin (8), benzoylpaeoniflorin (9), paeonol (10), and glycyrrhizin (11) in Jakyakgamcho-tang decoction was performed using an ultra-performance liquid chromatography-electrospray ionization-mass spectrometer. The analytical column for separation of the compounds 1~11 was used an UPLC BEH $C_{18}$ ($100{\times}2.1mm$, $1.7{\mu}m$) column and column oven temperature was maintained at $45^{\circ}C$. The mobile phase consisted of 0.1% (v/v) aqueous formic acid (A) and acetonitrile (B) by gradient elution. The flow rate was 0.3 ml/min and injection volume was $2.0{\mu}l$. Correlation coefficient in the calibration curves of the compounds 1~11 were showed a good linearity with more than 0.99. The limit of detection and limit of quantification values of the compounds 1~13 were detected in the ranges 0.06~18.43 ng/ml and 0.18~58.29 ng/ml, respectively. Among the compounds 1~11, the compounds 10 were not detected in this sample, while the ten compounds, 1~9 and 11, were detected $44.05{\sim}19,289.05{\mu}g/g$ in Jakyakgamcho-tang extract.

Quantitative Analysis of Citrate in Foods Using a Potentiometric Enzyme Biosensor (전위차법 효소 바이오센서를 이용한 식품의 구연산 정량분석)

  • Potentiometric biosensor using flow injection analysis system was developed to determine citrate concentration in foods. Biosensor system consisted of sample injector, peristaltic pump, enzyme reactor, carbonate ion selective solid-state electrode, reference electrode, detector, and recorder. Enzyme reactor was prepared with immobilized citrate lyase and oxaloacetate decarboxylase. Carbonate ions produced through enzyme reactions of citrate were potentiometrically detected by ion selective electrode. Optimum conditions for biosensor system were investigated. Interference effect of major sugars and organic acids was less than 5% on citrate biosensor system. Citrate concentrations in fruit juices were determined by biosensor and gas chromatography. No significant difference was observed between two analytical methods. Results indicate citrate biosensor is useful in determining citrate concentration in foods.

Effect of Relaxin Expressing Adenovirus for Rat Skin Flap Viability (Relaxin을 분비하는 아데노바이러스가 피판의 생존에 미치는 영향)

  • Purpose: Of various effects of relaxin, we assumed that anti-fibrotic effects, neovascularization effects and vasodilatation effects of relaxin might enhance the survival rate of skin flap. In the current study, we used adenovirus expressing relaxin genes to examine whether these genes could enhance the survival rate of a skin flap. Methods: A total of 30 Sprangue-Dawley rats were divided into three groups: RLX group (10; relaxin virus injected group), CTR group (10; no gene coded virus injection group), and PBS group (10; PBS injected group). Each group was intradermally injected with the virus ($10^7$ PFU) and PBS 48 hours before and immediately before the flap elevation. A distally based flap $3{\times}9\;cm$ in size was elevated on the dorsal aspect of each rat. Following this, a flap was placed in the original location and then sutured using a #4-0 Nylon. A surviving area of the flap was measured and then compared on postoperative days 3, 7 and 10. Using a laser Doppler, the amount of blood flow was measured. On postoperative day 10, tissues were harvested for histologic examination and the number of blood vessels was counted. Results: There was a significant increase in the area of the flap survival in the RLX group on postoperative days 3 and 7. The Doppler measurement also showed significantly increased blood flow immediately after the operation and on postoperative days 7 and 10. The number of blood vessels was significantly greater in the RLX group in the tissue harvested on postoperative day 10. The VEGF concentration was significantly higher in the RLX group than others in the tissues harvested on postoperative day 10. Conclusion: Following an analysis of the effects of relaxin-secreting adenovirus on the survival of a flap, the surviving area of the flap and the blood flow also increased. A histopathology also showed an increase in the number of blood vessels and the concentration of VEGF.

Development and Validation of an HPLC Method for the Pharmacokinetic Study of Fenoprofen in Human (페노프로펜 체내동태 연구를 위한 혈청 중 페노프로펜의 HPLC 정량법 개발 및 검증)

  • A selective and sensitive reversed-phase HPLC method for the determination of fenoprofen in human serum was developed, validated, and applied to the pharmacokinetic study of fenoprofen calcium. Fenoprofen and internal standard, ketoprofen, were extracted from human serum by liquid-liquid extraction with diethyl ether and analyzed on a Luna C18(2) column with the mobile phase of acetonitrile-3 mM potassium dihydrogen phosphate (32:68, v/v, adjusted to pH 6.6 with phosphoric acid). Detection wavelength of 272 nm and flow rate of 0.25 mL/min were fixed for the study. The assay robustness for the changes of mobile phase pH, organic solvent content, and flow rate was confirmed by $3^{3}$ factorial design using a fixed fenoprofen concentration $(2\;{\mu}g/mL)$ with respect to its peak area and retention time. And also, the ruggedness of this method was investigated at three different laboratories using same quality control (QC) samples. This method showed linear response over the concentration range of $0.05-100\;{\mu}g/mL$ with correlation coefficients greater than 0.999. The lower limit of quantification using 1 mL of serum was $0.05\;{\mu}g/mL$, which was sensitive enough for pharmacokinetic studies. The overall accuracy of the quality control samples ranged from 92.27 to 109.20% for fenoprofen with overall precision (% C.V.) being 5.51-11.71 %. The relative mean recovery of fenoprofen for human serum was 81.7%. Stability (freeze-thaw, short and long-term) studies showed that fenoprofen was not stable during storage. But, extracted serum sample and stock solution were allowed to stand at ambient temperature for 12 hr prior to injection without affecting the quantification. The peak area and retention time of fenoprofen were not significantly affected by the changes of mobile phase pH, organic solvent content, and flow rate under the conditions studied. This method showed good ruggedness (within 15% C.V.) and was successfully used for the analysis of fenoprofen in human serum samples for the pharmacokinetic studies of orally administered Fenopron tablet (600 mg as fenoprofen) at three different laboratories, demonstrating the suitability of the method.
