Preparation and Characterization of Polyurethane Nanofibers Containing Dendropanax morbiferus Extracts (황칠나무 추출물을 함유한 폴리우레탄 나노섬유의 제조 및 특성)

  • In this study, we first analyzed the total polyphenol and flavonoid contents and the antioxidant activity of Dendropanax morbiferus extracts obtained from different parts using different solvents. We then investigated the possibility of producing Dendropanax morbiferus leaf extract loaded polyurethane (PU) nanofibers via electrospinning. The total polyphenol and flavonoid content and the antioxidant activity of the Dendropanax morbiferus leaf extracts obtained distilled water were found to be higher than those of the extracts obtained under other conditions, and it was therefore used to prepare the extracts/PU nanofibers by varying the concentrations of both the Dendropanax morbiferus leaf extract and PU. The most appropriate fiber morphology were when 1.5% and 1.0% of the Dendropanax morbiferus leaf extracts were added to 12% and 13% PU, respectively. Fourier-tranform infrared (FT-IR) spectroscopy and X-ray diffraction (XRD) analyse reveal the successful incorporation of the Dendropanax morbiferus leaf extracts into the PU matrix. In addition, the nanofibers containing this leaf extract were confirmed to exhibit antibacterial properties against Staphylococcus aureus, suggesting the potential usefulness of nanofacial masks containing the Dendropanax morbiferus leaf extract.

Antioxidant Activity and Properties Characteristics of Pound Cakes Prepare using Freeze Dried Maquiberry (Aristotelia chilensis [Mol.]) Powder (냉동 동결 건조 머큐베리 분말 첨가 파운드케이크의 항산화 활성과 품질 특성)

  • A study was undertaken to examine the effect of adding freeze dried maquiberry powder on the quality of pound cakes. Freeze dried maquiberry was added to the flour at a ratio of 0.5, 1, 1.5 and 2.5%. The antioxidant activity was estimated by DPPH free radical scavenging activity, the total phenolic compounds content, and flavonoid content in freeze dried maquiberry powder of pounds cakes. Antioxidative activity was highly correlated with the total phenolic and total flavonoids contents of freeze dried maquiberry powder of pound cakes (r=0.9126, p<0.001, r=0.8366, p<0.001, respectively). The quality characteristics of freeze dried maquiberry powder of poundcakes were estimated. The specific volume decreased significantly with increased substitution level of freeze dried maquiberry powder (p<0.01). The lightness significantly decreased with increased freeze-dried maquiberry powder of pound cake crust and crumb (p<0.01, p<0.01). The hardness and adhesiveness increased, while gumminess tended to reduce with increased maquiberry powder. The consumer acceptability score of 0.5~2.5% freeze dried maquiberry powder of pound cakes ranked significantly (p<0.01) higher than those of the other groups in overall preference. These results showed that freeze dried maquiberry powder is a good ingredient to increase consumer acceptability and health.

Effects of Citrus Flavonoid, Hesperidin and Naringin on Lipid Metabolism in HepG2 Cells (간배양 HepG2 세포의 지질대사에 미치는 Hesperidin 및 Naringin의 영향)

  • The effects of citrus flavonoids, hesperidin and naringin, on the lipid metabolism were investigated in cultured human hepatocyte HePG2 cells. HepG2 cells were cultured for 6 h and 24 h to the control medium or the media containing hespridin and narigin, which concentrations were 0.5 and 5.0 mg/$m\ell$. There were no significant effects on cell proliferation and cellular protein content, except for increased in these parameters by adding both citrus flavonoids (0.5 mg/$m\ell$). The cellular content of triacylglycerol after 6 h incubation with 0.5 mg/$m\ell$ hesperidin and naringin was markedly increased, and after 24 h incubation that was decreased in both citrus flavonoids supplementation. The supplementation of 5.0 mg/$m\ell$ hesperidin caused a marked decrease in the cellular cholesterol content following 6 h incubation, and that was also reduced markdly, in a dose-dependent manner, during incubation for 24 h. However, there was no significant difference in the cellular cholesterol content in medium supplemented with naringin. The effect of hesperidin and naringin on acyl-CoA: cholesterol acyltransferase (ACAT) activity was studied in vivo and in vitro. The data confirmed that hesperidin inhibit ACAT activity in vivo and in vitro, whereas naringin had no such effect on ACAT activity in vivo but not in vitro. The present study suggests that hesperidin reduces the cellular triacyglycerol and cholesterol contents in human hepatocyte HepG2 cells.

Marker compound contents and antioxidant capacities of the taproot and lateral root of Danshen (Salvia miltiorrhiza Radix)

  • In this study, the marker compound contents of both the taproot and lateral root of Danshen (Salvia miltiorrhiza Radix), which is cultivated in Korea, were investigated. The salvianolic acid B content was the highest in the taproot (5.17-6.75%) and lateral root (3.99-5.69%). The cryptotanshinone, tanshinone I, and tanshinone IIA contents were higher in the lateral root than in the taproot of Danshen (p <0.05). Principal component analysis results revealed that the taproot was correlated to the salvianic acid A, rosmarinic acid, salvianolic acid B, and salvianolic acid A contents, whereas the lateral root was correlated to the cryptotanshinone, tanshinone I, and tanshinone IIA contents. The total phenolic content and total flavonoid content of the taproot were higher than those of the lateral root (p <0.05); however, the antioxidant activities of the taproot and lateral root of Danshen were similar. The salvianolic acid B content was correlated to the TPC of the taproot (r=0.748) and the 2,2-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid radical scavenging activity of the lateral root (r=0.847). This study could provide useful information for the classification of Danshen as a herbal medicinal product.

Development and Characterization of Peach Powder-added Chocolate and Chocolate-covered Freeze-dried Peach Snack (복숭아 분말 첨가 초콜릿과 초콜릿 코팅 복숭아 스낵의 제조 및 품질 특성)

  • In this study, peach powder-added chocolate and chocolate-covered freeze-dried peach snack were developed, and their physicochemical and bioactive properties were characterized. To make the chocolate, 0~15% of peach powders made of Fantasia cultivar were used. To make freeze-dried peach snack, three types of peach cultivars were tested: Mihwang, Kanoiwa Hakuto, andFantasia. In the case of chocolate, sugar content increased and pH decreased as peach powder content increased. Upon 15% peach powder addition, chocolate showed significant increase in flavonoid content and reducing power. In the case of freeze-dried peach snack, phenol content and antioxidant activity were significantly high in the Mihwang cultivar among the three types of peach cultivars tested. These results can be applied to the development of new food products using peach while maintaining its original physicochemical and bioactive properties.

Polyphenol Contents of Rumex crispus Root Extract with Hot Water and its Antioxidative Effect (소리쟁이 뿌리 열수 추출물의 폴리페놀 함량 및 추출물의 항산화 효과)

  • This study was conducted to investigate the extraction characteristics including total polyphenol compound content (TPC) and the antioxidative effect of Rumex crispus root extract on commercial corn oil. Extraction yield was increased with extraction temperature but decreased with extraction ratio. No significant differences in aromatics content were found among the extracts prepared with various extraction conditions; extraction ratio, extraction temperature, extraction pH and composition of extractant. Total flavonoid content of the extract was increased with extraction temperature and extraction pH, and highest value of it was found when extractant composition of ethanol in water was 75%. Total polyphenol compound content (TPC) of the extract with 10 fold extraction ratio showed the highest value, but no significant difference in TPC was found with extraction temperature. Reducing power and DPPH (${\alpha}$,${\alpha}$-diphenyl-${\beta}$-picrylhydrazyl) radical scavenging ability (RSA) of the extract in spectrophotometric absorbance were decreased with extraction ratio but increased with extraction temperature and showed 63.1%~98.4% and 67.6%~86.7% of those of ascorbic acid in reducing power and DPPH RSA, respectively. The antioxidative effects of the extract on corn oil were 84.8~93.0% of that of commercial butylated hydroxytoluene, the antioxidant index value was highest when the ethanol composition to water in extractant was equal ratio.

Functional Properties and Biological Activity of Breeding Lines, Parts, and Various solvents from Acanthopanax (오갈피나무의 계통별, 부위별, 추출용매별 생리활성물질 분석 및 기능적 특성)

  • This study was evaluated to biological activity of breeding lines from Acantopanax( A. sessiliflorus: ASF, A. koreanum: AKN, A. chiicanensis: ACS, A. senticosus: AST) and parts(root, stem, fruit, and leaf) and various extracted solvents( 100% water, 100% EtOH, 50% EtOH). Total polyphenol content of AKN root in 100% water extracts was high detected 464.46 mg/100 g. Total flavonoid content in the leaf was significantly higher than in other parts. The content of total sugars was high in the 50% EtOH extracts and fruit. The major free amino acids were arginine in all extracts. The content of arginine was detected in the root of AKN(1.807 mg/100 mg). Contents of eleutheroside B, E were high detected in 100% water extracts. Antioxidative capacity in the leaves of AKN was the higher than other extracts($EC_{50}=84.8{\mu}g/mL$). The results would be useful for understanding of the physiological properties of AKN extracts.

Quality Characteristics and Antioxidant Activity of Cookies with Black Ginseng Powder (흑삼 분말을 첨가한 쿠키의 품질 특성 및 항산화 활성)

  • This study evaluated the quality characteristics and antioxidant activities of cookies after adding black ginseng powder at ratios of 0%, 1%, 3%, 5%, and 7%, and the potential of black ginseng as a functional food. The moisture content increased with an increase in the black ginseng powder content while the pH decreased. The L value decreased with increase in the black ginseng powder content while the a and b values increased. The hardness decreased with increase in the black ginseng powder content. Notably, antioxidant activities, such as 2,2-diphenyl-1-picryl-hydrazyl (DPPH), 2-2'-azinobis(3-ethylbenzothiazoline-6-sulfonic acid)diammonium salt (ABTS) radical scavenging showed the highest increases in cookies containing 7% black ginseng powder. The total phenol and flavonoid contents significantly increased according to the amount of black ginseng powder added. These results suggest that black ginseng powder can be applied to cookies to achieve high quality and antioxidant activity.

Selective Allele Stacking of a Novel Quantitative Trait Locus Facilitates the Enhancement of Seed Epicatechin Contents in Soybean (Glycine max (L.) Merr.)

  • (-)-Epicatechin (EC), a primary form of flavan-3ol and a building block of proanthocyanidins, has health benefits as it is a potent antioxidant. So far, no quantitative trait loci (QTLs) associated with EC have yet been identified in soybean. In this study, QTLs for EC and hilum color were identified in recombinant inbred lines (RILs) derived from the varieties Jinpung and IT109098 using high-resolution single nucleotide polymorphism linkage mapping. This revealed two major QTLs for EC content, qEC06 and qEC08. qEC06 spanned the T Locus encoding flavonoid 3'-hydroxylase. qEC08, located near the I locus on Chr08, was also a major QTL for hilum color; however, allelic stacking of qEC08 and I revealed no relationship between I and EC content. RILs with IT 109098 alleles at both qEC06 and qEC08 had higher EC content than other lines. These results will enable the production of soybean varieties with high EC content via marker-assisted selection.

Determination of Total Phenol Content and Selected Phenolic Metabolites Analysis of Rice (Oryza sativa L.) Genetic Resources

  • The study of total phenol (TP) content from 700 rice varieties was evaluated using the UV-Vis spectrophotometric method. The calibration curve of serial diluted gallic acid as a standard of this study showed the acceptable performances (R2 = 0.999, mean accuracy 90%) and the mean of % relative standard deviation (%RSD: 0.07%) within the range of 7.8 to 1000 ppm concentrations. The mean value of total phenol content from 700 rice varieties was 2723.15 ㎍/g ranged from 55.48 ㎍/g to 9922.23 ㎍/g and the mean %RSD was 2.5%. Furthermore, this study aim was to analyze and profile individual phenolic compounds in the rice genetic resources to construct an integrative database for development of new rice variety with high functionality for health and understanding of phenolics characteristics in the rice grain. Herein, we analyzed selected 100 rice varieties based on high TP content and identified total 15 phenolic compounds by LC-ESI-MS/MS. Among selected 100 rice genetic resources, the phenolic metabolites consisted of higher amount of flavonoid (catechin) and phenolic acid mainly protocatechuic acid. Further research of more selected rice genetic resources would be continued to provide for an integrative phenolics profile of different rice genetic resources.
