Identification and Expression Patterns of fvexpl1, an Expansin-Like Protein-Encoding Gene, Suggest an Auxiliary Role in the Stipe Morphogenesis of Flammulina velutipes

  • Expansins are cell wall proteins that mediate cell wall loosening and promote specific tissue and organ morphogenesis in plants and in some microorganisms. Unlike plant expansins, the biological functions of fungal expansin-like proteins have rarely been discussed. In the present study, an expansin-like protein-encoding fvexpl1 gene, was identified from Flammulina velutipes by using local BLAST. It consisted of five exons with a total length of 822 bp. The deduced protein FVEXPL1 contained 274 amino acids with a predicted molecular mass and isoelectric point of 28,589 Da and pH 4.93, respectively. The first 19 amino acids from the N terminal are the signal peptide. Phylogenetic analysis and multiple protein alignment indicated FVEXPL1 was an expansin-like protein. The expression level of fvexpl1 gene in the stipe was significantly higher than that in the mycelia, primordia, and cap. However, the expression level of fvexpl1 gene was significantly higher in the fast elongation region of the stipe as compared with the slow elongation region. Expression analysis indicated that fvexpl1 gene might have an auxiliary role in the stipe morphogenesis of F. velutipes.

Development of a New Strain and Mating Type Analysis by Inbreeding Method in Flammulina velutipes (근연간 교배방법에 의한 팽이 탄핵균주의 교배형분석 및 균주 육성)

  • To breed new superior strains, collected strains were characterized and then several white strains were selected as parents. Monokaryons from the parents were isolated and studied. All tested white strains showed same mating genotype. Growth rate of monokaryons were lower than collected dikaryons. New dikaryotic strains were derived from inbreeding method, which means mating between monokaryotic isolates from different white strains having same mating genotype. Some of them showed higher yields of fruitbody than their parents. Specially Fv 4-1 strain showed the best productivity. Furthermore some mating combination showed cytoplasmic effect, when they mated reciprocally.

Medium composition for Flammulina velutipes bottle cultivation utilizing cassava stem chips (카사바줄기칩을 활용한 팽이버섯 병재배용 배지의 조성)

  • This study was conducted to establish replacement the corncob used in winter mushroom bottle cultivation. Corncob is unstable quality in moisture content or total nitrogen(T-N) content. Fruit body yields according to the ratio of cassava stem chips mixing were compared. After treatment-1 and treatment-2, fruit body yields increased by 8.8% and 5.4% and raw material cost decreased by 7% and 19%. The results showed that cassava stem chips could replace 33% to 67% of corncob for winter mushroom bottle cultivation.

  • This experiment was undertaken to investigate proper conditions for protoplast isolation from strains of Agaricus bisporus, Flammulina velutipes, Lentinus edodes, Pleurotus florida, Pleurotus ostreatus, Pleurotus sajor-caju and Volvariella volvacea. Combination of ${\beta}-D-glucanase$, novozym 234 and snail enzyme with 0.6M $MgSO_4$ in 0.05M Na-maleate buffer, pH 5.8 was the most effective for isolation of protoplasts. High yields of protoplasts were obtained from $4{\sim}5$ days old mycelia of P. florida and P. ostreatus on mushroom complete agar medium. Protoplasts were also obtained in good yield from other fungi but A. bisporus and V. volvacea. In the latter two cases protoplast release was observed; however, the yield was much lower than those of the other fungi.

  • Super water absorbent (CPAM-AS-hyd-1) was prepared by polymerization of acrylamide and allyl sulfonate salt with N,N'-methylene-bis-acrylamide as crosslinking agent, followed by alkaline hydrolysis and the effect on mycelial growth and sporophore production of edible mushrooms in the artificial cultivation was examined. The mycelial growth of edible mushrooms did not depend on the addition of super water absorbent upto 200 g of hydrated polymer gel per 100 cc medium. The proper hydrated polymer gel concentration for sporophore production of Pleurotus sajor-caju and Hericium erinaceus were 200 g and $200{\sim}250\;g$ per 100 cc medium, respectively. The proper hydrated polymer gel and puffed rice hull concentration for sporophore production of Flammulina velutipes was 200 g per 100 mm medium and 10% (v/v), respectively.

Identification and Characterization of Two New S-Adenosylmethionine-Dependent Methyltransferase Encoding Genes Suggested Their Involvement in Stipe Elongation of Flammulina velutipes

  • Two new SAM-dependent methyltransferase encoding genes (fvsmt1 and fvsmt2) were identified from the genome of Flammulina velutipes. In order to make a comprehensive characterization of both genes, we performed in silico analysis of both genes and used qRT-PCR to reveal their expression patterns during the development of F. velutipes. There are 4 and 6 exons with total length of 693 and 978 bp in fvsmt2 and fvsmt1, respectively. The deduced proteins, i.e., FVSMT1 and FVSMT2 contained 325 and 230 amino acids with molecular weight 36297 and 24894 Da, respectively. Both proteins contained a SAM-dependent catalytic domain with signature motifs (I, p-I, II, and III) defining the SAM fold. SAM-dependent catalytic domain is located either in the middle or at the N-terminal of FVSMT2 and FVSMT1, respectively. Alignment and phylogenic analysis showed that FVSMT1 is a homolog to a protein-arginine omega-N-methyltransferase, while FVSMT2 is of cinnamoyl CoA O-methyltransferase type and predicted subcellular locations of these proteins are mitochondria and cytoplasm, respectively. qRT-PCR showed that fvsmt1 and fvsmt2 expression was regulated in different developmental stages. The maximum expression levels of fvsmt1 and fvsmt2 were observed in stipe elongation, while no difference was found in mycelium and pileus. These results positively demonstrate that both the methyltransferase encoding genes are involved in the stipe elongation of F. velutipes.

Effect of Enoki Mushroom (Flammulina velutipes) Addition on the Quality of Kimchi during Fermentation (팽이버섯 첨가가 김치의 숙성 중 품질에 미치는 영향)

  • Enoki mushroom (Flammulina velutipes) was added in preparation of kimchi, and the quality was evaluated during fermentation at $10^{\circ}C$. Kimchi showed a rapid decrease in pH until 11 days, but revealed a retarded decrease thereafter. Titratable acidity increased sharply until 15 days, but increased slowly thereafter. Treatment with 5% enoki mushroom showed a lower pH and titratable acidity than control. Total microbial load and lactic acid bacterial count were maximum at 5 days, and were about $2.0{\times}10^{8}\;CFU/ml$. Radical scavenging activity was greater in enoki mushroom added kimchi after 11 days, and it was mainly due to the materials soluble in methanol and ethanol. Kimchi with 5% enoki mushroom showed higher scores than others in color and taste for sensory evaluation.

The change on characterizations of fruiting body according to the growing temperature in Flammulina velutipes (팽이버섯 생육온도에 따른 자실체의 특성 변화)

  • Total eight different winter mushroom (Flammulina velutipes) strains were characterized by comparisons of fruiting body morphology, hardness and color after growing at different temperature. Although variations depending on the strains were observed, in general, both stipe length and pilus diameter were shorter while pilus thickness was bigger as growing temperatures were decreased. No significant differences in stipe thickness were observed. The ASI4065 and 4151 had the tallest stipe and the ASI4021 had the shortest stipe. The ASI4103 strain had the biggest pilus diameter. The ASI4153 had most thick pilus, while the ASI 4021 had least thick pilus. Also, the ASI4166 had the most thick stipe and the ASI4065 had the most thin stipe. The hardness of white mushrooms were increased by decreasing growth temperatures, while no relationships between the hardness of brown mushrooms. In most strains, stipe were much stronger than pilei and white mushrooms were harder than brown ones. The strain having most strong stipe was ASI4021. Both ASI 4021 and ASI4166 had the most strong pilus. The color of pilus has much lighter than that of stipe. White mushrooms had high brightness value ranging from 70-90, while brown mushrooms showed low brightness value from 30-70. Saturation value was higher in brown mushrooms. Based on our study, it was shown that environmental conditions had some effects on winter mushrooms, but genetic differences among strains were more important factors to determine most characteristics of winter mushrooms.

Effect of Microbes and Formic acid on Storage of Spent Mushroom (Flammulina velutipes) Substrates (미생물과 개미산 첨가가 팽이버섯 수확후배지의 저장성에 미치는 영향)

  • This study was conducted to suggest effective storage methods for the use spent mushroom (Flammulina velutipes) substrates (SMS) as animal feed. SMS was storage by deep stacking, composting and vinyl mulching after treated with 1% microbes and 0.5% formic acid. During the storage periods, all of the treatment was covered with hyphae of mushroom. The external change of SMS was shown that the storage of SMS can be improved by anaerobic condition and 1% microbial treatment. The comparatively high temperature, pH and total bacterial number was shown in the SMS treated without microbes and formic acid by composting.

Electrophoretic karyotype of Flammulina velutipes and its variation among cultivars (팽이버섯의 핵형분석과 균주 사이의 핵형 다양성)

  • The karyotype of F. velutipes Korean cultivar, Fv 3-6, was compared with those of Japanese cultivars, Fv 0-1, Fv 1-5, Fv 11-1, by CHEF gel electrophoresis. The Korean cultivar, Fv 3-6, showed the difference from the three Japanese cultivars in number and size of chromosomes; the Fv 3-6 had two and one more chromosomes then Fv 0-1 and Fv 11-4, and Fv 1-5 had, respectively. The karyotyping by CHEF gel electrophoresis is quite suitable to define new Korean cultivars against Japanese cultivars.
