Early Life History Characteristics of an Induced Hybrid Between Rhodeus ocellatus and Rhodeus notatus (흰줄납줄개(Rhodeus ocellatus)와 떡납줄갱이(Rhodeus notatus) 잡종의 초기생활사 특징)

  • This study aimed to investigate taxonomic differences in the early life history characteristics of Rhodeus ocellatus and Rhodeus notatus through an interspecific hybridization experiment. The hatching rates of the cross-bred eggs were 50% for the ON cross (R. ocellatus ♀×R. notatus♂) and 67% for the NO cross (R. notatus♀×R. ocellatus♂). Interestingly, the hatching time of the hybrids was longer than that of their parents. The newly hatched larvae exhibited distinct differences in their physical characteristics. The head of the adult fish protruded forward more than that of the ON cross. The pterygoid process was 45° for the ON cross and 90° for the NO cross. Furthermore, the tail shape of the ON and NO crosses closely resembled that of R. notatus.

  • The authors analyzed the daily catch data which were obtained from two different regions at the Southern Sea in Korea, Neungpo, Geoje island and Yangwhagum, Namhae island, during three or four years in 1978, 1981-1985, in order to know the fishing characteristics of the set net fisheries. The favorable fishing season was summer in the southern sea. The total catches during one year were greatly affected by those several day when the dominant species of fish were caught in large quantities. The dominant species of fish at Neungpo were little horse mackerel, sand lance, sardine, hair tail, spanish mackerel and common mackerel, and those at Yangwhagun were anchovy, little horse mackerel, sardine, spanish mackerel and gizzard shad, in order of catch. Especially, the little horse mackerel and the sardine appeared to make up very big schools at both Neungpo and Yangwhagum. The occurrence seasons of the dominant species were different by the region, but generally those were earlier at western part than at eastern part of the Southern Sea in Korea.

  • The spermatogenesis of Siamese fighting fish, Betta splendens, belongs to Osphronemidae was investigated by light and electron microscopic observations. In primary spermatocyte stage, the nucleus was comparatively large ellipsoidal, and mitochondria showed a marked development in cytoplasm. In secondary spermatocyte stage, the germ cells were smaller than that of primary spermatocytes. The nucleus was a spherical shape and intercellular space was formed between germ cells. In spermatid stage, the early spermatids were not much different from a secondary spermatocyte. But, the chromatin condensation was occurred from the outside to the inside. The nucleus was more condensed. Intracellular space was larger than early spermatid. The mitochondria were rearranged in a middle piece, and occupied about half of the head part in early sperm. In sperm stage, the head of mature sperm was a spherical shape and had no acrosome. The flagellum was showed the typical 9+2 array of microtubules. Also, the tail of sperm had no lateral fins and outer coarse fibers. These ultrastructural characteristics can be used in classification of species.

Embryonic Developmen Larvae and Juveniles of the Small Yellow Croaker (Larimichthys polyactis) Reared in Aquarium (실내수조에서 사육한 참조기 배발생 및 자치어의 형태)

  • On 11 June 1991, eggs from the brood stock of small yellow croaker (Larimichthys polyactis) were artificially fertilized using the standard dry method and were hatched. Each of the fertilized eggs (1.1-1.2 mm in diameter) had an oil globule and was transparent and buoyant. The fertilized eggs hatched in a range of water temperatures $(17.5-20.3^{\circ}C)$ 44 hrs after fertilization. The total lengths of the newly hatched larvae were 3.1-3.3 mm, and these hatchlings had 31 myotomes (10+21). Melanophores and yellow-brown chromatophores were concentrated on the head, at the ventral part of the yolk, and in the middle of the tail. Four days after hatching, the larvae completely absorbed the yolk and became flexions of 5.1-5.5 mm in total length. Fifteen days after hatching, one spine (the anterior tip of the maxillary) appeared in the upper jaw and three spines developed at the upper parts of the eyes and on the posterior part of the head. At this stage, the larvae were approximately 8.3 mm long. Thirty-nine days after hatching, juveniles (1.9-3.4 mm in total length) had a pointed tail fin. By 66 days after hatching, the juvenile fish (about 4.0-6.5 mm in total length) were similar to adult fish in body shape. The larvae of L. polyactis could be distinguished from those of L. croacea by two distinct characteristics: the large number of vertebrae (28-29), and a relatively small bony ridge on the occipital region of the head.

  • To keep relics excavated in the era of the Japanese Colonist's rule in the Korean Peninsula in order, the National Museum of Korea selected some of horse strap pendants (Haengyeob) that were excavated from Seobongchong, Sikrichong and Geumryeongchong, and it investigated conservation and production technique. The horse strap pendants selected were classified into three, that is to say, Oval and fish-tail shape, heart shape and atypical: At the results of microscopic observation, XRF and SEM-EDS analysis, base plate was made of iron, and top plate as well as pattern plate was made of iron, copper and silver. Both Oval and fish-tail shape and heart shape horse strap pendants had horse strap pendants having same production technique and sizes at all of three ancient tombs were made of iron and copper. The gold layer of horse strap pendants except the atypical one having no gold layer were made by amalgam gilding.

  • The ultrastructures of spermatogenesis and sperm in Xiphophorus maculatus, ovoviviparous fish were investigated by electronmicroscopy The testis of Xiphophorus maculatus contained numerous testicular sacs, and spermatogenesis was synchronized in these testicular sac. In the case of spermatogonium, the nucleus was comparatively large ellipsoidal, and the nucleolus and mitochondria showed a marked development. The size of primary spermatocyte was smaller than that of spermatogonia, and that of secondary spermatocyte was smaller than that of primary spermatocyte. The chromatin of spermatocyte was highly condensed according to their development. The nucleus with electron-dense was round shape. In spermiogenesis, flagella started to be formed and chromatin was more condensed. The mitochondria were rearranged along the tail. The sperm was formed by loss of cytoplasm. The head of mature sperm was long cone shape and had not acrosome. The microtubules of flagella were arranged 9+2 structure. Also, the sperm has a loop-like structure at the end of a tail.

Life History and Morphological Responses of Daphnia similis against Chemical Compounds Exuded by Various Cohabiting Animals and Triops longicaudatus in the Rice Paddy Ecosystem (논 생태계에 서식하는 생물과 긴꼬리투구새우가 분비한 화학물질이 물벼룩의 생활사와 형태에 미치는 영향)

  • The aim of this study was to assess the interaction between Daphnia similis and various organisms related to the rice paddy ecosystem. We selected several organisms that are likely to prey on D. similis and evaluate predation rate as well as responses of D. similis to the chemical compounds exuded by these organisms. As a result of predation experiment, larval dragonfly (Anax parthenope) and Triops longicaudatus were clearly shown decreasing abundances of D. similis. Especially, Triops longicaudatus was observed higher feeding rates on D. similis than larval dragonfly. Chemical compounds from the vertebrates such as fish (Misgurnus anguillicaudatus, Pseudorasbora parva, Micropterus salmoides) and tadpole of frog (Rana nigromaculata) did not affect the life history of Daphnia. However, a potential predatory fish P. parva induced significantly longer tail spine in Daphnia. In addition, among the invertebrates (T. longicaudatus, A. Parthenope, Micronecta spp., Palaemon paucidens), chemical compounds exuded by T. longicaudatus induced shorter body and significantly longer tail spine in D. similis.

Effects of Phosphate Complex on the Functional Properties of Fish Meat paste (혼합 인산염의 첨가가 어류 연육의 기능적 성질에 미치는 영향)

  • Effects of four kinds of phosphate complex on the water holding capacity(W.H.C) and protein solubility of yellow-corvenia(Pseudosciance manchurica) and hair tail(Trichurus lepturns) meat paste were investigated. The formulations of four kinds of phosphate complex employed to this experiment were made by mixing several phosphates such as sodium polyphosphate, sodium pyro-phosphate, sodium acid pyro-phosphate, potassium pyro-phosphate, sodium tetra meta-phosphate, sodium ultra meta-phosphate and sodium hexa meta-phosphate, and monoglyceride at different mixture ratios. Among the four kinds of phosphate complex, phosphate B complex which was formulated by mixing sodium poly-phosphate 50%, sodium pyro-phosphate 20%, sodium tetra meta-phosphate 20%, sodium acid pyrophosphate 5% and sodium ultra meta-phosphate 5% was most effective on enhancing the W.H.C and protein solubility of yellow corvenia meat paste, and in case of hair tail meat paste, phosphate C complex which was formulated by mixing sodium poly-phosphate 40%, sodium pyro-phosphate 30%, potassium pyro-phosphate 15%, sodium tetra meta-phosphate 10%, and sodium hexa meta-phosphate 5% was more effective than other phosphate complex, and their optimum addition level was 0.4% respectively in weight of fish meat paste. Texture characteristics such as hardness, cohesiveness, and springiness value of Kamaboko(fish meat paste product) were evaluated as best when 0.3% of phosphate B complex was added. The optimum cooking condition of Kamaboko to get good texture was heating for 45 minutes at 85$^{\circ}C$.

Concatemer-Associated Transgene Expression Patterns in Transgenic Marine Medaka Oryzias dancena Strains

  • To examine the interrelationship between transgenic insertion patterns and transgene expression profiles in established transgenic fish lines, four stable transgenic marine medaka Oryzias dancena germlines harboring ${\beta}$-actin regulator-driven RFP reporter constructs were selected. The established transgenic strains were characterized with regard to their transgenic genotypes (insertion pattern, concatemer formation, and transgene copy number based on genomic Southern blot hybridization and qPCR assay) and expression characteristics at the mRNA (qRT-PCR), protein (western blot), and phenotypic (fluorescent appearance) levels. From comparative examinations, it was found that transgenic expression at both the transcription and translation levels could be significantly downregulated in transgenic strains, potentially through methylation-mediated transgene silencing that was particularly associated with the formation of a long tail-to-head tandem concatemer in the chromosomal integration site(s). When this occurred, an inverse relationship between the transgene copy number and fluorescence intensity was observed in the resultant transgenic fish. However, with the other transgenic genotype, transgenic individuals with an identical Southern blot hybridization pattern, containing a tandem concatemer(s), had very different expression levels (highly robust vs. low expression strengths), which was possibly related to the differential epigenetic modifications and/or degrees of methylation. The concatemer-dependent downregulation of transgene activity could be induced in transgenic fish, but the overall pattern was strain-specific. Our data suggest that neither a low (or single) transgene copy number nor tandem transgene concatemerization is indicative of strong or silenced transgene expression in transgenic fish carrying a ubiquitous transgene. Hence, a sufficient number of transgenic lineages, with different genotypes, should be considered to ensure the establishment of the best-performance transgenic line(s) for practical applications.

Morphological characterization of Vibrio alginolyticus specific bacteriophage isolated from fish farms on west coast of Korea (서해안 양식장에서 분리한 Vibrio alginolyticus의 특이 bacteriophage에 대한 구조적 특성)

  • Vibrio alginolyticus (V. alginolyticus), which is one of bacterial pathogens evoking severe infection in fish and shellfish as well as in human has been found at high frequency around all coast areas in Korea. Both V. alginolyticus and V. alginolyticus specific bacteriophage were isolated from sea water and various fishes from fish farms on west coast in Korea. In a morphological study based on electron microscope, the purified phage appeared to be composed of hexagon head of 60 nm and short tail of 20 nm. In the denatured SDS-PAGE analysis, the structural proteins of the phage were found to be 7 different protein fractions ranging from 37.8 to 198 kda. The kind of nucleic acid of the phage was ascertained to a double stranded DNA.
