Presence of low level infected iridovirus in sea perch Lateolabrax sp. imported to Korea

  • We have identified an iridovirus CH-1 from sea perch Lateolabrax sp. healthy externally and imported from China to Korea. In a comparison of the nucleotide sequences of the five different genomic regions, the CH-1 appears to be closely related to the ISKNV, IVS-1 and Ehime-1 strains detected in China, Korea and Japan respectively. In quantitative comparison of the viral DNA, level of CH-1 in tissue of imported fish was 10,000 times lower than that of IVS-1 strain presented in the infected rock bream Oplegnathus fasciatus of moribund stage. It allowed us to speculate the possibility of the asymtomatic iridovirus infection in the culturing sea perch. Such possibility of asymptomatic infection was supported by result of no appearance of dead fish with typical symptoms of iridoviral disease in keeping experiment of the imported sea pearch in laboratory for more than three weeks. Such asymptomatic infections with iridovirus were also found in spleen of the culturing and externally healthy sea perch of Korea by the presence of the iridoviral DNA in nested PCR.

  • Chromosome analysis using mitosis, meiosis and bicolor FISH were carried out in Bupleurum latissimum Nakai, which is one of the endemic plants in Ulleung island of korea. The somatic methaphase chromosomes number of this plant was 2n = 2x = 16 and the chromosome complements consisted of six pairs of metacentrics and two pairs of submetacentrics. The size of chromosomes ranged 2.40${\sim}$4.20 ${\mu}$m and NOR (nucleolus organizer region) chromosome did not observed using conventional staining. In meiosis chromosomes, metaphase-I and anaphase-I were observed. Metaphase-I anaphase-I showed 8 bivalents and chromosomes migration to make two daughter cells. Using bicolor FISH, one pair of 5S and 45S rDNA signals were detected on the centromeric region of chromosome 3 and the end of short of chromosome 2,respectively. We also observed the NOR using 45S rDNA probe.

Effect of Testosterone on the mRNA Levels of Gonadotropin Subunits in the Immature Rainbow Trout Pituitary

  • In order to clarify the role of gonadal sex steroids in the synthesis of gonadotropin (GTH) subunits in immature rainbow trout, we examined in vitro and in vivo effects of testosterone (T) on the pituitary mRNA levels of GTH I $\beta$, GTH II$\beta$ and a subunits by Northern blot analysis and on the pituitary content levels of GTH I$\beta$ and GTH II$\beta$by radioimmunoassay (RIA). The mRNA levels of the a subunit in T-treated fish were not changed more dramatically than those in control fish both in vivo and in vitro. Interestingly, the mRNA levels of GTH I$\beta$ in T-treated fish were shown to be slightly lower than those in the control fish under these experimental conditions, but no differences were observed in pituitary GTH I$\beta$ contents. In contrast, the mRNA levels and pituitary contents of GTH II$\beta$ subunit were strongly increased by T both in vivo and in vitro. These results demonstrate that the expressions of GTH I$\beta$ and II$\beta$ subunit genes in immatue rainbow trout pituitary are subjected to differential regulation by T.

Isopod Parasite Induced Secondary Microbial Infection in Marine Food Fishes

  • Isopods are parasitic crustaceans that pose serious threat to fisheries. Several studies have tried to explore the host-pathogen relationship between marine fishes and isopods. The present study aims to understanding the secondary infections in marine fishes pertaining to isopods. To assess the secondary infection in infected fishes, parasite infested and healthy tissues of fishes were collected. The samples were subjected to standard microbiological procedure to identify the presence of pathogenic bacteria and fungi. Our results showed the branchial region had the higher microbial load of non-sporulating cenocytic fungi in infected fishes. Moreover, fungal strains isolated from the parasitic lesion confirmed that the parasitation and body lesion facilitates the entry of several pathogenic microbes at the damaged host tissue. More over the immune regulation of fish fights back by producing minute cysts, trying to encapsulate the growing fungus. But this may eventually lead to systemic infestation and death of the fish.

  • Two wooden fish excavated from Wiryeseong Fortress on Seonggeosan Mountain in Cheonan-si, Chungcheongnam-do Province were frail due to their deterioration within the burial environment. To prevent further deformation, conservation treatment was conducted on them to strengthen the wooden materials and stabilize the dimensions. Prior to the conservation treatment, the species of wood was determined to be pine and infrared Irradiation on the patterns remaining on the wooden fish revealed that they were depictions of carp. For conservation treatment, the wooden fish were impregnated with PEG #3, 350 in a water solution with a 40% concentration, followed by freeze-drying.

  • Morphological development of eggs, larvae and juvenile of the sunrise sculpin, Pseudoblennius cottoides were studed in the field and laboratory at the coastal Dolsan-do, Yeosu-shi from October, 2000 to April, 2001 Egg mass of Pseudoblennius cottoides in peribranchial cavity of Halocynthia hilgendorfi, were observed during late fall to winter in the study area. Fertiliged eggs were spherical in shape, demersal, adhesive, transparent and greenwish yellow color, measuring 1.84 mm (1.83-1.87 mm) in diameter. There were numerous and 17 (15-20) various-sized oil globules accounted in the yolk. Granular materials formed a mass in the yolk. Fertiliged eggs hatched at 301 hr 20 min after morula stage. Newly hatched larvae 6.31 mm (6.24-6.37 mm) in total length (TL), had a large yolk. At 3 days after hatching, the larvae, 6.77 mm (6.69-7.14 mm) in TL came out through the excurrent siphon of Halocynthia hilgendorfi. At 13 days after hatching, the larvae 12.59 mm (12.42-12.63 mm) in TL transformed to postlarval stage. At 32 days after hatching, postlarvae of TL 19.18 mm (19.01-19.46 mm) have reached the juvenile stage.

Bactericidal Efficacies of an Aquatic Disinfectant Tablet Composed to Calcium Hypochlorite Against Vibrio anguillarum and Streptococcus iniae

  • Vibrio spp. and Streptococcus spp. have caused a considerable disease of farmed fish and economic loss in fish farming and seafood industry. In this study, the efficacy of an aquatic disinfectant tablet composed to calcium hypochlorite was evaluated against V. anguillarum and S. iniae. A bactericidal efficacy test by broth dilution method was used to determine the lowest effective dilution of the disinfectant following exposure to test bacteria for 30 min at $4^{\circ}C$. An aquatic disinfectant tablet and test bacteria were diluted with distilled water (DW), hard water (HW) or organic matter suspension (OM) according to treatment condition. V. anguillarum on the DW, HW and OM condition was completely inactivated with 16,000 15,000 and 13,000 fold dilutions of the disinfectant, respectively. On the DW, HW and OM condition, S. iniae was absolutely inactivated with 17,000 16,000 and 14,000 fold dilutions of the disinfectant, respectively. As an aquatic disinfectant tablet possesses bactericidal efficacy against fish pathogenic bacteria such as V. anguillarum and S. iniae this disinfectant solution can be used to control the spread of fish infective bacterial diseases.
