As a part of investigation for quality improvement of surimi gel from fish with a red muscle by addition of emulsion curd, we investigated the processing conditions of emulsion curd contained succinylated gelatin from conger eel skin as an emulsifier and emulsion curd-added surimi gel. Activity and stability of emulsion curd on standing at room temperature, chilled temperature and vibration were remarkably improved by the addition of 15 tunes of soybean oil and 5 times of water to succinylated gelatin from conger eel skin. The proximate composition of the emulsion curd was moisture 18%, protein 5%, lipid 76% and ash 0.5% and its appearance was white. Peroxide value and fatty acid composition of emulsion curd contained succinylated gelatin as an emulsifier were similar to these of soybean oil. By the addition of 6% of emulsion curd to mackerel surimi, gel strength, appearance and texture of the resulting surimi gel were improved, while its peroxide value and brown pigment revealed minor change. From the results of volatile basic nitrogen, viable cell counts and histamine content, the emulsion curd-added mackerel surimi gel can be safe In the sense of food sanitation.
This experiment was performed to compare the techniques of the metabolizable energy determination and to, examine the problems associated with metabolizable energy determination of poultry feedingstuffs. The feedingstuffs evaluated herein, were corn, sorghum, barley. fish meal and soybean meal The results obtained were as followings : 1. When comparing AME and AMEn values of five feedingstuffs(measured) by Total Collection ana Index Methods there was no significant difference (P>.05) between two methods except fish meal. 2. The AME$_{F}$ values of feedingstuffs were not different (P>.05) from AME values. which implied that bioassay of AME$_{F}$ be applicable to rapid measure AME value of feedingstuffs for poultry and the problems connecting to AME$_{F}$ determination were discussed. 3. The AME$_{FC}$ and TME values of corn, sorghum and barley were significantly higher (P<.05) than AME and AME$_{F}$ values but those of fish meal and soybean meal were similar or lower than AME and AME$_{F}$ values. This fact was indicative that the techniques measuring AME$_{FC}$ and TME has some depressing factors when fish meal and soybean meal were determined. 4. Comparing of TME, AME$_{FC}$ and T-AME values tile TME values of corn sorghum and barley were significantly higher(P<.05) than AME$_{FC}$ and T- AME values. But that of fish meal and soybean meal was much lower(P< .05)comparing with AME$_{FC}$ and T-AME values. According]y, it night be concluded that the relationship among the values of TME. AME$_{FC}$ and T-AME was shown great variation according to the feedingstuffs, especially in the protein diets.
In order to investigate the immune response induced by supplementation of herb medical stuff in diet on the nile tilapia. Oreochromis niloticus, experiments were performed with feeding of four different experimental diets supplemented with 2% ginseng. Panax ginseng, 3% Kugija. Lycium chinense, 3% Hasuo, Polygonum multiflorum, 2% Omija, Schizandra chinensis, respectively, for 84 days. The non-specific immune responses changed during the feeding period were investigated at 2, 4, 6, 8, 10, 12 weeks in each group. Average body weight of the nile tilapia with supplemented diets was heavier than control group. The fish fed on diet supplemented with 3% Kugija showed the better growth than the other tested groups. Complement activity such as complete hemolytic activity ($CH_{50}$) and bactericidal activity against Escherichia coli tended to be increased by the supplementation of herb medical stuff. The lysozyme activity of serum and adherent phagocyte activity showed higher in the fish fec on diet supplemented with 3% kugija than the other tested groups. In respect to the RPS against experimental Edwardsiella tarda infection, all of the group fed on the herb medical stuff in diet appeared higher response compared with control group. From these results, herb medical stuff (Panax ginseng, Lycium chinense. Polygonum multiflorum, Schizandra chinensis) might be used a additives of diet for the increasing of non-specific immune response or resistance against bacterial fish diseases.
This study was designed to investigate the effects of various sources of dietary fats on the blood and liver cholesterol(CHOL) levels in young Single Comb White Leghorn male chicks, In experiment 1, corn oil, palm gil, tallow and fish oil were added individually at a level of 4% to semipurified type diets composed of isolated soyprotein and glucose as major components. The diets were fed ad libitum for a period of 15 days. In experiment 2, various fats such as corn oil, soybean oil, repeseed oil, palm oil, tallow, fish oil and hydrogenated fish oil(HFO) were added individually at a level of 11.4% to practical type diets primarily based on corn and soybean meal. Control diet contained 3% of corn oil. All these diets were formulated to contain equivalent amount of nutrients such as protein, vitamins and minerals on a basis of unit kcal of metabolizable energy. The third Experiment was to compare the effects of different levels of calorie/protein(C/P ratio) of diets on the performances and various biological parameters in the chicks. Control diet was the same as in experiment 2. Another diet was added with 11.14% corn oil(C/P ratio=146) and the other diet with 10% corn oil(C/P ratio=164), The diets in experiment 2 and 3 were fed ad libitum for 26 days. In the first experiment, the chicks fed the diet containing vegetable oils tended to grow faster and show better feed efficiency without significance than those fed diets added with animal fats. However, this tendency was not observed in the experiment 2. Birds consumed the diets added with fish oil appeared to have heavier liver weight and higher liver CHOL than the others(p〈0.05), No significant differences in the levels of blood CHOL and triacylglycerol(TG) were observed among the chicks of various dietary groups(Exp. 1). Weights of liver or heart were significantly heavier in the chicks consumed the diets added with HFO or fish oil, respectively(Exp. 2). However, chicks ingested diet containing fish oil appeared to have significantly lower plasma CHOL. No significant differences were observed in the levels of liver CHOL and plasma TG among the dietary groups. Birds consumed the diet with a wider C/P ratio resulted in higher liver TG levels in experiment 3(p〈0.05). Although no statistical differences were observed among the various dietary groups, chicks fed the diet with a wider C/P ratio tended to show higher levels of plasma CHOL, TG, liver CHOL and total liver lipids compared to those of the control group.

Surimi yields from acid and alkaline processing of 5 fishes were compared to those from conventional processing Effect of sarcoplasmic protein and NaCl on heating gel from acid and alkaline surimi were also investigated by punch test and color. Yield of alkaline surimi was higher than that of conventional surimi. However, the breaking force, deformation and whiteness of heating gel from alkaline surimi were lower than those of heating gel from conventional surimi. The sarcoplasmic protein increased a breaking force and a deformation of gel. A breaking force was decreased, but deformation not significantly with NaCl concentration. Myosin heavy chain (MHC) and actin were greatly degraded in acid processing. Alkaline process for surimi is a valuable way of increasing the utilization of frozen and pelagic fishes, and making kamaboko-type products.
Antifreeze proteins (AFPs) have ice binding affinity, depress freezing temperature and inhibit ice recystallization which protect cellular membranes in polar organisms. Recent structural studies of antifreeze proteins have significantly expanded our understanding of the structure-function relationship and ice crystal growth inhibition. Although AFPs (Type I-IV AFP from fish, insect AFP and Plant AFP) have completely different fold and no sequence homology, they share a common feature of their surface area for ice binding property. The conserved ice-binding sites are relatively flat and hydrophobic. For example, Type I AFP has an amphipathic, single ${\alpha}$-helix and has regularly spaced Thr-Ala residues which make direct interaction with oxygen atoms of ice crystals. Unlike Type I AFP, Type II and III AFP are compact globular proteins that contain a flat ice-binding patch on the surface. Type II and Type III AFP show a remarkable structural similarity with the sugar binding lectin protein and C-terminal domain of sialic acid synthase, respectively. Type IV is assumed to form a four-helix bundle which has sequence similarity with apolipoprotein. The results of our modeling suggest an ice-binding induced structural change of Type IV AFP. Insect AFP has ${\beta}$-helical structure with a regular array of Thr-X-Thr motif. Threonine residues of each Thr-X-Thr motif fit well into the ice crystal lattice and provide a good surface-surface complementarity. This review focuses on the structural characteristics and details of the ice-binding mechanism of antifreeze proteins.
The aim of this study was to characterize the hemolytic Aeromonas sp. MH-8 exposed to green tea catechin, epigallocatechin gallate (EGCG). Initially, the hemolytic Aeromonas sp. MH-8 was enriched and isolated from stale fish. Bactericidal effects of MH-8 exposed to EGCG ranging from 1 mg/mL to 4 mg/mL were monitored, and complete bactericidal effects were achieved within 3 h at 3 mg/mL and higher concentrations. SDS-PAGE with silver staining revealed that the amount of lipopolysaccharides increased or decreased in the strain MH-8 treated to different concentrations and exposing periods of EGCG in exponentially growing cultures. The stress shock proteins (70-kDa DnaK and 60-kDa GroEL), which might contribute to enhancing the cellular resistance to the cytotoxic effect of EGCG, were induced at different concentrations of EGCG exposed to cell culture of MH-8. Scanning electron microscopic analysis demonstrated the presence of irregular rod shapes with umbilicated surfaces for cells treated with EGCG. 2-DE of soluble protein fractions from MH-8 cultures showed 18 protein spots changed by EGCG exposure. These proteins involved in chaperons (e.g., DnaK, GroEL and trigger factor), enterotoxins (e.g., aerolysin and phospholipase C precursor), LPS synthesis (e.g., LPS biosynthesis protein and outer membrane protein A precursor), and various biosynthesis and energy metabolism were identified by peptide mass fingerprinting using MALDI-TOF. In consequence, EGCG was found to have substantial antibacterial effects against food-poisoning causing bacterium, hemolytic Aeromonas sp. MH-8. Also the results provide clues for understanding the mechanism of EGCG-induced stress and cytotoxicity on Aeromonas sp. MH-8.
To study the correlation with increasing body fat mass for the Standard Guideline for Korean Dietary Life, a nationwide, cross-sectional survey was performed with 178 male college students in december 2005. Among the student subjects, 76 were analyzed for body protein mass, body mineral mass, body fat mass, percent body fat, waist-hip ratio(WHR), body mass index(BMI) and obesity degree using Inbody 3.0(Biospace Co, Seoul, Korea). Body protein mass was at a high level of 92.1%, body mineral mass was at a low level of 94.7%. Body fat mass was at a high level of 28.9%, percent body fat was at a high level of 37.3%, WHR was at a high level of 31.6%, and obesity degree was at a high level of 38.2%, BMI was at a high level of 24.3% and BMR was observed at a high level of 41.8%. WHR was correlated with '7. Prepare food proper amount sanitarily.' and obesity degree, while BMl was were correlated with '4. Increase movement and eat proper amount.' positively. BMR was correlated with '6. Enjoy 3 meal a day regularly.' ArmCircle was correlated with '4. Increase movement and eat proper amount.' and fitness score was corrected with '4. Increase movement and eat proper amount', and '8. Enjoy Korean rice food style.' positively. Body protein mass and body mineral mass were negatively correlated with fruit, ArmCircle was positively correlated with fish, obesity degree was positively correlated with soup and BMI and ArmCircle were positively correlated with sweet. '8. Enjoy Korean rice food style.' was negatively significantly correlated with overweight, over percentage body fat, obesity degree > 120 and BMI. Male students were not practicing the Standard Guideline for Korean Dietary Life well with an average score of 37.7%. However, male students who are within the normal range of body composition analysis value have to notice that over range of weight, protein, percentage body fat, WHI and obesity degree was observed above average score of 30%.
We developed predictive growth models for Staphylococcus aureus and Bacillus cereus on various food matrices consisting primarily of ready-to-eat (RTE) foods. A cocktail of three S. aureus strains, producing enterotoxins A, C, and D, or a B. cereus strain, were inoculated on sliced bread, cooked rice, boiled Chinese noodles, boiled bean sprouts, tofu, baked fish, smoked chicken, and baked hamburger patties at an initial concentration of 3 log CFU/g and stored at 8, 10, 13, 17, 24, and $30^{\circ}C$. Growth kinetic parameters were determined by the Gompertz equation. The square-root and Davey models were used to determine specific growth rate and lag time values, respectively, as a function of temperature. Model performance was evaluated based on bias and accuracy factors. S. aureus and B. cereus growth were most delayed on sliced bread. Overall, S. aureus growth was significantly (p<0.05) more rapid on animal protein foods than carbohydrate-based foods and vegetable protein foods. The fastest growth of S. aureus was observed on smoked chicken. B. cereus growth was not observed at 8 and $10^{\circ}C$. B. cereus growth was significantly (p<0.05) more rapid on vegetable protein foods than on carbohydrate-based foods. The secondary models developed in this study showed suitable performance for predicting the growth of S. aureus and B. cereus on various food matrices consisting of RTE foods.

Lathyrus sativus, locally known as Khesari, is a leguminous pulse crop grown in many parts of the world for food (used by poor people) and animal feed/fodder. Its seeds are rich in protein and energy but contains anti-nutritional factors prominent among which is ${\beta}$-N-Oxalylamino-L-Alanine (BOAA), a neurotoxin causing lathyrism in humans due to prolong consumption. Keeping in view the chemical characteristics of this toxin and literary facts on L. sativus feeding in terrestrial animals, it was hypothesized that aquatic species may better utilise this ingredient in mixed extruded diets. Diets were prepared with varying levels (0, 10, 18, 26 and 34%) of L. sativus seeds and fed for 60 days to study growth, body composition and digestibility of nutrients. Final body weight, specific growth rate and feed and protein conversion ratio did not differ (p>0.05) between treatments. Crude protein digestibility was reduced (p<0.01) beyond 26% inclusion level of L. sativus. Final carcass composition with regard to protein, lipid and ash did not differ (p>0.05) among the treatments. Comparable craniosomatic, viscerosomatic, renosomatic and hepatic indices and no mortality implies no apparent adverse effect on the vital organs and fish health. It was concluded that L. sativus can be a promising feed ingredient that can be used up to 34% or possibly higher level in fingerlings diet. To our knowledge, it appears to be the first report of its kind under laboratory conditions.
