Domestic Smart Aqua-farming Technology (국내 스마트양식 기술 동향)

  • A stable protein source is required to support the rapidly increasing global population, and fishery products are a particularly important part of the required protein supply. However, due to continued overfishing, fishery resources are depleted, and the number of fish caught by fishing boats has stagnated. Consequently, the aquaculture industry is becoming increasingly important. Internationally, smart aquaculture technology that minimizes labor and environmental pollution has been established through technological developments supported by large investments in automation and water treatment technology over the last several decades. In the case of Korea, the aquaculture industry has not yet emerged as a labor-intensive primary industry. However, in recent years various attempts have been made to apply ICT technology to aquaculture to overcome these problems. In this study, domestic and foreign technologies and patent trends for smart aquaculture are analyzed. In addition, the current status of the smart aquaculture cluster business that the Ministry of Oceans and Fisheries has been promoting since 2019 to utilize ICT technology in aquaculture is introduced.

Long-tenn Evaluation of Extruded Pellet Diets Compared to Raw Fish Moist Pellet Diet for Growing Flounder, Paralichthys olivaceus (넙치 미성어 건조 배합사료 및 습사료의 장기사육 평가)

  • This experiment was conducted to compare the effects of extruded pellets and raw fish-based pellet on olive flounder Paralichthys. olivaceus. Six diets were prepared for this study: two formulated extruded pellets (FEP1 & FEP2), three commercially available extruded pellets (CEP1, CEP2 & CEP3) and moist pellet (MP). Weight gain offish fed FEP1 and CEP3 were significantly higher (P<0.05) than those of fish fed FEP2, CEP1, CEP2 and MP, while that of fish fed MP was not significantly different (f<0.05) from those of fish fed the FEP2, CEP1 and CEP2. Feed efficiency of fish fed CEP2 was significantly lower (P<0.05) than those of fish fed FEP1, FEP2, CEP1, CEP3 and MP. There was no significant difference in protein efficiency ratio and hepatosomatic index between fish fed FEP1 and CEP3, and among fish fed FEP2, CEP1 and CEP2. There was no significant difference in condition factor among fish fed the FEP1 and CEP3, and between fish fed FEP2, CTP1 and MP. However, fish fed MP had a lower survival rate than fish fed the other five EP These results suggest that diet FEPl could be developed to replace MP for the owing stage of flounder without adverse effects on growth performance.

Evaluation of Nutritional Content in Convenience Store Lunchboxes by Meal Type, Price, and Store Brand (편의점 도시락의 식단형태별, 가격대별, 편의점 브랜드별 영양성분 함량 평가)

  • Objectives: This study investigated the menu and nutritional contents of convenience store lunchboxes, and evaluated the nutritional content by meal type, price, and store brand. Methods: In September 2019, 93 convenience store lunchboxes from the top five franchise stores were purchased. Relevant information on price, food weight, food ingredients, cooking methods, and nutrition labeling were subsequently collected. Nutritional content was evaluated based on the daily value (DV) and Index of Nutritional Quality (INQ), and energy contribution of carbohydrate, protein, fat, saturated fat and sugar was compared with the recommended range. Results: Most lunchboxes included the food groups; grains/starches, meats/fish/eggs/legumes, and vegetables. However, none provided fruits, and only a few lunchboxes provided milk/milk products. Stir-frying, deep-frying, and pan-frying were the most frequent methods of cooking. The average energy content of the lunchboxes was 736.6 kcal, whereas the average contents of protein, fat and saturated fat were higher than 40% of the DV, and sodium content was 66.8% of the DV. The contents of most nutrients in traditional type lunchboxes were higher, as compared to nutrients in onedish type lunchboxes. Considering pricing of lunchboxes, protein and sodium contents were higher in the higher-priced lunchboxes as compared to lower-priced lunchboxes, but there were no differences in the INQs. The contents of energy, protein, fat, saturated fat, and cholesterol significantly differed by brand. Conclusions: Our results indicate that convenience store lunchboxes contain high levels of protein, fat, saturated fat, and sodium. The nutritional contents differed by meal type, price, and store brand, and higher price did not imply higher nutritional quality. We propose the need to educate consumers to check nutrition labels and purchase appropriate lunchboxes. Manufacturers also need to make efforts to reduce the amounts of fat, saturated fat, and sodium, and not provide protein in excess.

  • In order to develop a new type of food source for the effective utilization of fish protein, plastein reaction was applied to improve the functional properties of sardine protein. Conditions necessary for optimal plastein productivity from sardine protein using pepsin, ${\alpha}-chymotrypsin$, protease(from Aspergillus saitoi) and papain were established. Sardine protein concentrate was hydrolyzed with pepsin yielding an approximate degree of hydrolysis of 78.4%. Enzyme induced plastein was optimized at : pH 4 for pepsin, pH 7 for ${\alpha}-chymotrypsin$, pH 5 for pretense and pH 6 for papain : Substrate concentrate 40% for pepsin and ${\alpha}-chymotrypsin$, 50% for pretense and papain : the time of incubation, 24hr : enzyme/substrate ratio, 1 : 100(w/v) incubation temperature, $50^{\circ}C$.

Study on the Preparation and Utilization of Sardine Protein (정어리 단백질 제조와 이용에 관한 연구)

  • To utilize sardine protein more effectively, fish meat paste products mixing sardine protein concentrate with pollack frozen meat paste at the ratio 0%, 15%, 20% and 25% were produced, and the change of firmness, sensory evaluation and the properties of amino and fatty acid were investigated. The quantity of sardine protein and it was almost gushed out around one hour at 100$\^{C}$. The firmness of the meat paste product was found as 0.54% and was better when the concentrated sardine protein was added at the ratio 15% and it was much higher than just that of pollack meat paste. In that case, total amino acid was the highest as 90.701 mg/g from the point of view of the amino acid composition. In terms of the fatty acid composition, unsaturated fatty acid of raw and boiled sardine was 61,8634% and 61.9384% each. We could find out that the high value of C$\_$20:5/ and C$\_$22:6/ of raw sardine was 7.2931% and 27.7843%, respectively.

Milk Proteins and Allergy (우유 단백질과 알레르기)

  • Food allergy is defined as adverse reactions toward food mediated by aberrant immune mechanisms. Therefore, an allergic response to a food antigen can be thought of as an aberrant mucosal immune response. Food allergy most often begins in the first 1~2 years of life with the process of sensitization by which the immune system responds to specific food proteins, most often with the development of allergen-specific immunoglobulin E (IgE). Over time, most food allergeies are lost, although allergy to some foods is often long lived. The most important allergen sources involved in early food allergy are milk, eggs, peanut, soybean, meat, fish and cereals. Milk allergy seem to be associated with casein and whey protein. Important features of proteins as allergenicity are size, abundance and stability. Strategies for the prevention of milk allergy is breast-feeding, partially hydrolysised infant formula, using of probiotics, immune components in milk, preparation of low allergenicity milk protein and allergy therapy (immune therapy).

A Study on the Dietary Fiber Intake and Iron Metabolism in Korean Female College Students (일부 여대생의 식이섬유 섭취와 철분대사에 관한 연구)

  • This study was conducted to evaluate dietary fiber intake, iron status, and their correlations in 50 female college students living in Seoul. The nutritional status was analyzed using 3-day dietary record, duplicated diet collection, and venous blood sampling. The mean values for age, height, weight, BMI, and blood pressure of the subjects were 23.2 years, 160.2cm, 53.9kg, 21.0kg/$m^2$, and 110.1/68.4mmHg, respectively. Daily intakes of enery, protein, fiber, crude fiber, iron, and heme iron were 1635.5㎉, 54.3g, 22.5g, 6.8g, 16.2mg, and 0.2mg, respectively. Fiver intake was positively correlate with energy, protein, carbohydrate, vitamin C, iron, and crude fiber intakes. Also, iron intake was positively correlated with energy, protein, lipid, carbohydrate, and vitamin C intakes. There was a significant correlation between heme iron and MFP(meat, fish, poultry) intakes. To exame the iron balance, iron intake and excretion were measured. Iron intake and excretions through urine and feces were 19.5mg, 8.5mg, and 1.6mg, respectively. Based on these iron retention and iron apparent absorbability were calculated as 9.4mg and 52.4%, respectively. There was no significant relationship between dietary fiber intake and iron status. However, there were significant positive correlation between fiber intake and urinary iron excretion.

Effect of Feeding Date Pits on Milk Production, Composition and Blood Parameters of Lactating Ardi Goats

  • Twenty Ardi lactating goats were used to investigate the effect of substituting 10%, 15%, and 20% of concentrate feed with date pits on milk production, composition, and blood parameters. Four isocaloric and isonitrogenous dietary treatments were used. Four levels (0% [control], 10%, 15%, and 20%) of date pits were used to replace concentrate feed. The forages to concentrate ratio used was 60 to 40. Dry matter intake (DMI) of goats fed diets containing 10% and 15% date pits was significantly (p<0.05) higher than those fed diets containing 0% and 20%. However, goats fed a diet containing 20% date pits were significantly (p<0.05) lower in DMI compared to those fed control diet. The protein percent was significantly higher for goats fed control diet compared to the other dietary treatments. Total solids percent was significantly the lowest for goats fed diet supplemented with 10% date pits. Goats fed a diet containing 20% date pits was significantly (p<0.05) higher in the total protein compared to those fed a diet containing 10%. In addition, goats fed a diet containing 15% date pits exhibited no significant differences in the total protein percent compared to those fed a diet containing 20% date pits. Triglyceride was significantly higher for goats fed diets containing 10% and 20% date pits compared to those fed 15% date pits. Results obtained in the present study suggest that date pits can be added up to 20% of the concentrate feeds into lactating Ardi goat diets without negative effects on their productive performance.

Molecular Cloning and Characterization of Alkaliphilic Phospholipase B (VFP58) from Vibrio fluvialis

  • Vibrio fluvialis, an enteropathogenic bacterium, produces a phospholipase which is thought to be an important factor in the pathogenesis of disease. In this study, the phospholipase gene (vfp) was identified from V fluvialis (KCTC 2473) and its sequence was determined. The entire open reading frame was composed of 1,689 nuc1eotides and 563 amino acids. The phospholipase gene (vfp) was overexpressed in Escherichia coli as a his-tag fused protein. This recombinant protein (rVFP58) was solubilized with 6 M urea and purified by Ni-NTA affinity chromatography. The action mode of rVFP58 was determined by TLC and GC-MS, and it showed phospholipase B activity, which had both phospholipase A and lysophospholipase activities. The rVFP58 showed a maximum activity at pH around 9- 10 and temperature of about 40OC, and it was stable under alkaline condition over pH 9. The cytotoxicity of rVFP58 was evaluated, using a fish cell line, CHSE-2l4, and was found to cause significant cell death after 14 h of exposure to 250 $\mu$g of the protein.

In situ Hybridization of a Megalocytivirus Using Nucleic Acid Probes against ATPase and the Major Capsid Protein of Rock Bream Iridovirus

  • Systemic infections of maricultured fishes by Megalocytivirus species have occurred over a broad area in South Korea, causing extensive economic loss. We developed digoxigenin-labeled nucleic acid probes against the 230-bp ATPase and 311-bp major capsid protein (MCP) of rock bream Oplegnathus fasciatus iridovirus (RBIV) using polymerase chain reaction, and an in situ hybridization (ISH) method to detect Megalocytivirus in formalin-fixed tissues of mariculture species (rock bream, sea bass, and olive flounder). ISH-positive cells were abundant in the hematopoietic and connective tissues of various organs, while brain tissue showed little or no signal. The ISH procedure can become an important diagnostic tool in complement with histopathological methods, and advances epidemiological studies on the origin and distribution of Megalocytivirus in mariculture.
