Characterization of the Infectious Pancreatic Necrosis Virus (IPNV) isolated from Pan-Cultured Rainbow Trout in Korea (한국산 양식송어에서 분리된 전염성 췌장괴저 바이러스의 특성)

  • Infectious Pancreatic Necrosis Virus (IPNV) is one of the most important pathogens for inland fish farming and had been first reported in Korea from returning adult chum salmon (Oncorhynchus keta) at hatcheries on the east coast. During the past years, several viruses identified as IPNV were isolated not only from chum salmon, but also from gold fish (Carassius auratus), eel(Anguilla japonica), and rainbow trout (Salmo gairdneri). An isolate, coded DRT, from fingerlings of pan-cultured rainbow trout in Daechung Dam showed different serotype from three known reference serogroups of IPNV such as VR-299, Sp, and Ab. Antisera to three of these serotypes, however, partially neutralized the infectivity of this isolate. Anti-Sp type was rather effective than either anti-VR-299 or anti-Ab, implying DRT could be more closely related to Sp. DRT has been purified and its RNA genome segments were compated showing that the isolate does not belong to any of known serogroups even with some common antigenicity.

A Study on the Crevice Corrosion for Ferritic Stainless Steel (페라이트 스테인리스강의 틈부식에 대한 연구)

  • In recently days, the breed fish farm is increased in the beach side for farming fish. In such a farm, the heater is requested for preventing freezing in cold season. The heating material are requested high corrosion resistance and strength for endurance high corrosive salt and pressure. In case of low corrosion resistance and/or strength, the heating element shall be broke down and eventually make spillage or leaking contaminated salt. In the most cases, crevice corrosion is localized form of corrosion usually associated with a stagnant solution on the micro-environmental level. In this study, the crevice corrosion of Ferritic type 430 stainless steel is investigated. The size of specimen is $15{\times}20{\times}3mmt$. Test solution is 1N H2SO4 + 0.05N NaCl. The artificial crevice gap size is $0.24{\times}3{\times}15mmL$. Crevice corrosion is measured under applied voltage 300mV(SCE) to the external surface. the result of this study showed that 1) the induced time for initiation of crevice is 750seconds, 2) potential is dropped in the crevice from the top of gap opening from -320 to -399mV. The result confirmed that the potential drop(IR mechanism) in the crevice is one of mechanism for crevice corrosion.

  • Fisheries policy change from catching to farming requires more intensive consideration for aquaculture industry. The oceanic farm is a desirable cost effective aquaculture method. However. in odor to gather fish in the oceanic farm, eating sound or any attracting sound should be radiated through underwater loudspeaker. In this Paper, it has been found in literature that the frequency range responding to fish is about 16Hz to 13kHz but sensitive frequency range is about 150Hz to 2kHz and sound pressure level is about 100dB to 150dB reference $1{\mu}Pa$. Therefore, frequency range and output sound level of designed underwater loudspeaker has been specified as 150Hz to 3kHz and 100dB to 145dB reference $1{\mu}bar$. respectively To verify the stability and the endurance to the pressure of 40m water depth, manufactured underwater loudspeaker was examined before sea trial in manufactured water pressure tank which gives a maximum of 10 atmospheric Pressure. We experimented on acoustic characteristic with manufactured underwater loudspeaker under water depth of 10m.

A Study on Information Asymmetry and the Country of Origin Labeling in Live Small Fish Trade for Aquaculture and Naturalization (이식용수산물 교역의 정보비대칭과 원산지표시제도 영향에 관한 연구)

  • The main objective of this study is to analyze problems of the Country of Origin Labeling(COOL) in small live fish(SLF) trade by applying the asymmetric information theory which is called ${\ulcorner}$Lemon Market Theory${\lrcorner}$. The purpose of importing SLF is to enhance fishing household income and import substitution effect as well. SLF importation is quite different in its nature from importing general fish and fish products for final/direct consumption. SLF are imported for the purpose of domestic aquaculture and naturalization where domestic production/supply of SLF is less than domestic demand for or such species are not native to Korean waters. Information asymmetric problems arise even in SLF trade as the same way in general goods and services. However, the information asymmetry issues in SLF trade are much more complex than non-living goods because SLF are traded in the live state. To alleviate such problems the Korean government initiated the general COOL scheme to imported SLF. However, many experts argues that such policy on SLF would not be appropriate because of SLF's very nature. Applying the lemon market theory, we can analyze how information discovery schemes are able to signal correct information to SLF trading parties and to result in more symmetric information in SLF trade markets. This research carried out a case study about small live eel(SLF) trade and its farming. The results showed that applying the COOL to small live eels just right upon coming into fish farms tends to increase substantially fish farmer's income and at the same time to lower unnecessary transaction costs. In particular, such transaction costs by imposing simply the general COOL on SLF may easily outweigh its benefits. For instance, to resolve the problems, the Korean ministry of agriculture and forestry(KMAF) has developed a quite different COOL rules from the general ones and has applied them to imported live cattle and medium/small livestocks. The KMAF's differentiated COOL policy on some imported livestocks may be a good case which can be applied to imported SLF. In addition to the differentiated COOL on SLF, Sanitary and Phytosanitary (SPS) and traceability system would play crucial complementary roles in alleviating information asymmetry problems in SLF trade. Advanced fisheries tend to strengthen their SPS system rather than to adopt the general comprehensive COOL schemes into imported SLF trades and domestic market exchanges.

Antibacterial Effects of Pseudomonas aeruginosa MB I-3 against Listonella anguillarum (어류 병원세균, Listonella anguillarum에 대한 Pseudomonas aeruginosa MB I-3의 항균 효과)

  • To study the possible use of probiotics in fish farming, The in vitro and in vivo antibacterial effects of Pseudomonas aeruginosa MB I-3 (MB I-3) against the fish pathogenic bacterium Listonella anguillarum were evaluated. The inhibitory effects of MB I-3 against vibrios were investigated by the double layer method and the co-culture. The results showed that MB I-3 inhibited the growth of pathogenic vibrios including Listonella anguillarum, Vibrio alginolyticus, Vibrio cholerae, Vibrio fluvialis, Vibrio furnissii, Vibrio harveyi, Vibrio parahaemolyticus and Vibrio vulnificus. Extracellular substances obtained from the cultural supernatant of MB I-3 by ethyl acetate extraction showed inhibitory effects on L. anguillarum. The antibacterial substance of MB I-3 was evaluated to destroy the cell membrane of L. anguillarum in electron micrographs. The probiotic effects of MB I-3 was tested by exposing olive flounder (Paralichthys olivaceus) fry to L. anguillarum with or without MB I-3. The cumulative mortality of olive flounder fry infected with L. anguillarum was 24% in the group with MB I-3, while it was 46% in the control group without MB I-3. These results indicate that MB I-3 has potential applications as a probiotic for the control of fish pathogenic vibrios in fish rearing system.

Anti-scuticociliate effects of a combined treatment with formalin and blue LED (포르말린과 청색 LED 병용처리에 의한 항스쿠티카 효과)

  • Scuticociliatosis caused by Miamiensis avidus is a very important parasitic disease in olive flounder farming industry. The aim of this study was to determine effect of combined treatment with blue LED (light-emitting diode) illumination and formalin on olive flounder (Paralichthys olivaceus) infected with M. avidus. Different intensity of 405 nm LED (20, 40, and 60 μmol·m-2·s-1) was illuminated on 2.2×104 cells/well of M. avidus in a 24 well microplate for 24 h. Also, 2.4×104 cells/well of M. avidus were exposed to varying combinations of 60 μmol·m-2·s-1 of 405 nm LED and serial 10-fold dilutions of formalin (from 10 to 100 ppm) for 15, 30, 45, and 60 min. Surviving M. avidus were counted using a hemocytometer. For in vivo test, flounder acclimatized at 11-12 practical salinity unit (psu) were challenged with 2×106 cells/ml of M. avidus by immersion method for 1 h. Then, fish were moved and divided into four groups; "F" group, treated with formalin at 50 ppm; "L" group, treated with 60 μmol·m-2·s-1 of 405 nm LED; "C" group, treated with combination of the two methods; and the control group. After treatment for 30 min, fish were transferred to new tanks (salinity = 11-12 psu) and observed for 3 weeks. As a result, illumination of 405 nm LED at 60 μmol·m-2·s-1 killed 100% of M. avidus after 12 h, while 67% and 90% of the scuticociliate died at 20 and 40 μmol·m-2·s-1, respectively, after 24 h exposure. One hundred percent of M. avidus was killed at 90, 80, 80 and 70 ppm after exposure to formalin for 15, 30, 45 and 60 min, respectively. However, combined method (e.g., 60 μmol·m-2·s-1 of 405 nm-LED plus 50 ppm formalin) killed the parasite within 30 min. From in vivo test, similarly, survival rates of fish challenged with M. avidus were 100%, 43%, 29% and 0% in the C, F, L, and control groups, respectively. Results obtained in this study demonstrates that the combined treatment method has clear synergistic effect on scuticociliatosis in fish.

Chemical Fluxes at the Sediment-Water Interface Below Marine Fish Cages on the Coastal Waters off Tong-Young, South Coast of Korea (남해안 통영지역 가두리양식장 해수-퇴적물 경계면에서의 chemical fluxes)

  • Benthic respiration and chemical fluxes were measured at the sediment-water interface underlying the marine fish cages floating on the open coastal waters off Tong-Young, the South Coast of Korea. The effects of cage farming on coastal benthic environment and on mass balance of organic carbon in the benthic boundary layer under the marine fish cages are addressed. In a growing season of caged fishes of June, 1995, benthic chambers and sediment traps were deployed on the sediment-water interfaces of the two sites chosen for this study: 1) Cage Site, directly underlying the fish cages of the farm at 18 m water depth, and 2) Control Site, about 100 m away from the farm at 32 m water depth. Benthic respiration rates and chemical fluxes were calculated from the evolution of dissolved oxygen and chemicals in the chamber water, and mass balance of organic carbon in the benthic boundary layer was constructed based on the vertical flux of particulate organic matter (POM) and chemical fluxes out of the sediment. High organic dumping (6400 mg C $m^{-2}d^{-1}$) and high benthic respiration (230 mmol $O_2\;m^{-2}d^{-1}$) were observed at the Cage Site. Equivalent to 40% of vertical flux of organic carbon into the Cage Site seemed to be decomposed concurrently and released back to overlying waters (2400 mg C $m^{-2}d^{-1}$). Consequently, up to 4000 mg C $m^{-2}d^{-1}$ of organic carbon could be buried into the farm sediment (equivalent to 60% of organic carbon flux into the Cage Site). At the Control Site, relatively less input of organic carbon (4000 mg C $m^{-2}d^{-1}$) and low benthic respiration rate (75 mmol $O_2\;m^{-2}d^{-1}$) were observed despite short distance away from the cages. The influence of cage farming on benthic chemical fluxes might be restricted and concentrated in the sea bottom just below the fish cages in spite of massive organic dumping and high current regime around the fish cage farm.

  • The family of lemnacae colloquially known as duckweed contains the world' smallest species of flowering plants (macrophytes). Aquatic and free-floating, their most striking qualities are a capacity for explosive reproduction and an almost complete lack of fibrous material. They are widely used for reducing chemical loading in facultative sewage lagoons, but their greatest potential lies in their ability to produce large quantities of protein rich biomass, suitable for feeding to a wide range of animals, including fish, poultry and cattle. Despite these qualities there are numerous impediments to these plants being incorporated into western farming systems. Large genetically determined variations in growth in response to nutrients and climate, apparent anti-nutritional factors, concerns about sequestration of heavy metals and possible transference of pathogens raise questions about the safety and usefulness of these plants. A clear understanding of how to address and overcome these impediments needs to be developed before duckweed is widely accepted for nutrient reclamation and as a source of animal feed.

  • Marine chlorella (Nannochloropsis oculata) is cultivated at the fish farming association of experimental fishery laboratory. In the culture of the chlorella, it is necessary to count the number of chlorella in order to understand the condition of increase. For that purpose, we develop automatic chlorella counter using the image processing and the neural network. Its effectiveness is confirmed through the experiment.

Identification of intestinal microflora in rainbow trout

  • Although trout farming is well established in Korea, very little information is available on the composition of intestinal microflora in rainbow trout (Salmo gairdnerii). In 1994, from October through November, we investigated the composition and succession of intestinal bacteria. As fish grew, total viable counts increased dramatically until 45 days after fertilization when anaerobes started to appear on the media. After this time, they increased steadily. Ten aerobic generic were identified and Gram negative bacteria constituted 85% of total isolates. Among these, Pseudomonas, Eikenella, and Alcaligenes were the three major genera. Six anaerobic genera were isolated and identified. The ratio of anaerobes to aerobes was about 1 : 1 in adult trout and the composition of genera was changed under different conditions.
