중국에서 수입된 잉어 및 붕어의 잉어부종바이러스병 및 허피스바이러스성 조혈기괴사증 모니터링 (Monitoring of carp edemavirus disease and herpesviral hematopoietic necrosis in Cyprinus carpio and Carassius auratus imported from China)

  • In this study, carp edemavirus disease and herpesviral hematopoietic necrosis were examined in carp (Cyprinus carpio) and crucian carp (Carassius auratus) imported from China. From August 2016 to May 2017, 100 carp and 300 crucian carp were examined. There were no clinical symptoms in all the tested fish, and no internal or external parasitic infections were identified except for the detection of Argulus sp. in one leather carp. Carp edema virus disease was examined by PCR in C. carpio, but all were negative. In C. auratus, herpesviral hematopoietic necrosis was tested by PCR and 4 out of 300 were positive. The results of this study are expected to be useful as basic data for conducting risk analysis on the relevant organisms and counterpart countries or for designating new pathogens for quarantine.

  • We identified a microsatellite marker, Poli121TUF, which appears to be significantly linked (P<0.001) with a lymphocystis disease virus (LCDV)-resistance gene in the olive flounder, Paralichthys olivaceus. The olive flounder is an economically important food fish, that is widely cultured in Korea, Japan, and China. Lymphocystis disease has spread in these countries and has seriously reduced the economic value of the fish. LCDV causes lymphocystis cells (LC) to form on the body surface, fins, gills, mouth, and intestine. Fish with LC lose commercial value due to their deformed appearance. The identified micro satellite marker can be used as a candidate locus for marker-assisted selection (MAS) in order to enhance the efficiency of selection for LCDV resistance in the olive flounder.

  • Infection with salmonid alphavirus (SAV) is a serious disease that mainly affects rainbow trout (Oncorhynchus mykiss) and Atlantic salmon (Salmo salar L.) reared in seawater or freshwater. SAV is prevalent in European countries including Norway that exports rainbow trout and Atlantic salmon to Korea. Consequently, SAV was listed as a disease notifiable to the OIE and many salmonid-producing countries either designate SAV as their notifiable disease or do research on the development of diagnosis and epidemiology to reduce the possibility of SAV infection. Unlike other salmonid-producing countries, SAV is not listed as a notifiable disease in Korea, thereby arousing concern that SAV will get into the country through the importation of live salmonids. Under the circumstance, Korea needs to have a legal basis to take much stricter follow-up measures, including listing SAV as a notifiable disease, establishing surveillance system based on OIE standards to declare Korea free from SAV, killing infected fish and conducting fallow system against affected farms.

Larval Anisakid Infections in Marine Fish from Three Sea Areas of the Republic of Korea

  • The present study was performed to determine the infection status of anisakid larvae in marine fish collected from 3 sea areas of the Republic of Korea. Total 86 marine fish (8 species) collected from the East Sea (Goseong-gun, Gangwon-do), 171 fish (10 species) from the South Sea (Sacheon-si, Gyeongsangnam-do), and 92 fish (7 species) from the Yellow Sea (Incheon Metropolitan City) were examined by both naked eyes and artificial digestion method. Among the total of 349 fish examined, 213 (61.0%) were infected with 8 species of anisakid larvae, i.e., Anisakis simplex, 6 types of Contracaecum spp., and Raphidascaris sp., and the mean larval density was 13.8 per infected fish. Anisakid larvae were detected in 45 fish (52.3%) from the East Sea, 131 fish (76.6%) from the South Sea, and 37 fish (40.2%) from the Yellow Sea. The average numbers of larvae detected were 4.0, 16.6, and 15.9, respectively. Anisakis simplex larvae were detected in 149 fish (42.7%), and the mean larval density was 9.0 per infected fish. They were found in 26 fish (30.2%) collected from the East Sea, 96 fish (56.1%) from the South Sea, and 27 fish (29.3%) from the Yellow Sea. The average numbers of larvae detected were 2.9, 10.3, and 10.5, respectively. Conclusively, the present study suggests that the infection rate and density of anisakid larvae are more or less higher in the fish from the South Sea than those from the East Sea or the Yellow Sea.

국민건강영양조사를 활용한 담수어패류 섭취량 산정: 위해성 평가를 위한 파라메타 도출 (Estimating Freshwater Fish Intake for Human Health Risk Assessment Using Korea National Health and Nutrition Examination Survey)

  • Freshwater fish intake is a critical parameter for deriving water quality criteria and water quality standards for protection of human health based on human health risk assessment. Although the freshwater fish intake parameter should be accurate and representative of Korean fish consumption for the water quality criteria to be reliable, data are limited in Korea and have low reliability. In this study, Korean National Health and Nutrition Examination Survey data from 2008-2010 were analyzed to reevaluate freshwater fish consumption. Based on these results, an average consumption rate of 3.0 g/day per person, a $90^{th}$ percentile consumption rate of 0.0 g/day per person, an average consumption rate of 65.7 g/day per fish consumer, and a $90^{th}$ percentile consumption rate of 153.4 g/day per fish consumer were proposed for derivation of water quality criteria using a conservative approach and various exposure scenarios.

  • 자연산어류의 질병 감염 실태를 파악하기 위하여 2006년 6월에 남해안 해역에서 트롤에 의하여 어획된 개체 중 13 종 176 개체의 어류와 1 종 15개체의 새우류 등 총 191 개체의 자연산어류에 대한 바이러스성 및 세균성 질병에 대한 모니터링을 실시하였다. 조사 결과 우리나라 양식 어류의 주요 질병과 관련한 바이러스성 병원체의 존재는 확인되지 않았다. 시료 6 개체에서 세균이 분리되어 확인한 결과 Pseudomonas속 세균 5 균주와 Proteus속 세균 1균주가 확인되었으나 양식 어류의 폐사와 관련한 세균군은 아닌 것으로 판단되며, 양식산어류의 질병 발생에 대한 관련성을 밝히기 위하여 자연산어류에 대한 더 많은 조사가 필요할 것으로 사료된다.

Susceptibility of Koi, Koi×Red Common Carp, and Red Common Carp×Koi to Koi Herpesvirus (KHV)

  • The disease-causing koi herpes virus (KHV), also known as cyprinid herpesvirus-3 (CyHV3), causes mass mortality of koi and carp. Koi (Cyprinus carpio) is a host for KHV, one of 12 virus species in the Alloherpesviridae family. We examined the effects of KHV disease koi (KK), and on koi×red common carp (KR) and red common carp×koi (RK) cross, using a virus challenge test. The infected fish had clinical signs that included gill necrosis and skin lesions. The RK and KR were highly more resistant (cumulative mortality: RK; 6% and KR; 8%) to KHV infection than KK fish (cumulative mortality: 28%). KHV DNA was confirmed in the tissues of all dead fish in groups by use of polymerase chain reaction (PCR), and the presence of the KHV protein in kidney was confirmed by immunohistochemistry. Histological analysis showed severe gill lesions and fusion of the lamellae in KK fish, but less severe damage in RK fish. In immunohistochemistry analysis, the KHV protein localized in the cytoplasm of infected kidney cells of KK, but the cross groups had lower levels of KHV antigen. Our data indicate that the cross groups had increased resistance to KHV disease.

Identification of potential molecular markers for disease resistance in giant gourami through major histocompatibility complex (MHC) II gene analysis

  • Research to obtain molecular markers related to the major histocompatibility complex (MHC) gene in both strains of gourami is essential to increase the success of the selection program of disease resistance traits. Using a completely randomized design (CRD), the challenge test consists of four treatments and seven replications. The treatment was Jambi gourami injected with PBS (KJ), Kalimantan gourami injected with PBS (KK), Jambi strain injected with Aeromonas hydrophila (GJ), and Kalimantan strain injected with A. hydrophila (GK). The GJ population was more resistant to A. hydrophila than the GK population. The MHC II gene was detected in both test strains (GJ and GK), both resistant and susceptible fish. However, there were differences in the results of amplifying the MHC II gene in susceptible and resistant fish. Two DNA fragments approximately 400 and 585 bp were detected in the genome of susceptible fish, while in the genome of susceptible fish, only one DNA fragment was detected (400 bp). Therefore, the MHC II gene fragment with a size of about 585 bp can be used as a potential candidate for specific molecular markers to obtain resistance to A. hydrophila bacteria in the giant gourami.

배합사료 및 습사료 투여가 양식 넙치, Paralichthys olivaceus의 건강도에 미치는 영향 (Effects of extruded pellet and moist pellet feed on health conditions of olive flounder Paralichthys olivaceus)

  • 2008년 7월부터 12월까지 양식장 2개소에서 사육중인 넙치를 대상으로 상업용 배합사료 (extruded pellet, EP) 및 습사료 (raw fish moist pellet, MP)를 급이하고, 시험 사료별로 병원체 검출률과 혈액학적 성상의 차이를 조사하였다. 그 결과, EP 사료를 먹인 시험구 (EP 그룹)가 MP 사료를 먹인 시험구 (MP 그룹)에 비해 혈청 GOT, GPT 및 glucose 성분이 높게 나타났다. 시험기간 동안의 평균 생존율과 병원체 검출률에서는 뚜렷한 차이가 없는 것으로 나타났으나, 계절적으로 수온이 높은 7월부터 10월까지 EP 그룹에 비해 MP 그룹에서 병원체 검출률이 높게 나타났다. EP 그룹에서는 Dactylogyrus sp., E. tarda 및 red seabream iridovirus (RSIV)가 주로 검출되었으며, MP 그룹에서는 Trichodina sp., Streptococcus spp. 및 viral nervous necrosis virus (VNNV)가 우점적으로 검출되었다.

High Endemicity with Clonorchis sinensis Metacercariae in Fish from Yongjeon-cheon (Stream) in Cheongsong-gun, Gyeongsangbuk-do, Korea

  • The infection status with Clonorchis sinensis metacercariae (CsMc) was examined in freshwater fishes from Yongjeon-cheon (a branch of Nakdong-gang) located in Cheongsong-gun, Gyeongsangbuk-do, the Republic of Korea (Korea). A total of 750 fishes in 19 species were examined by the artificial digestion method for 2 years (2019 and 2020). CsMc were detected in 378 (51.4%) out of 735 fishes in 14 species (73.7%), and the infection intensity was 666 per fish infected. In 2019, CsMc were found in 172 (68.0%) out of 253 fishes in 10 species, and the infection intensity was 565 per fish infected. In 2020, CsMc were detected in 206 (62.2%) out of 331 fishes in 10 species, and the infection intensity was 751 per fish infected. The other zoonotic trematode, ie. Metagonimus spp., Centrocestus armatus, Echinostoma spp. and Clinostomum complanatum, metacercariae were also detected in fishes from the survey streams, but their endemicities were relatively low. Conclusively, it was first confirmed that CsMc are highly endemic in fishes from Yongjeon-cheon in Cheongsong-gun, Gyeongsangbuk-do, Korea.
