Visual census and hydro-acoustic survey of demersal fish aggregations in Ulju small scale marine ranching area (MRA), Korea (수중촬영조사법과 음향자원조사법을 활용한 울주군 연안 소규모 바다목장 해역의 어류 군집 조사)

  • Visual census and hydro-acoustic survey was carried out at Ulju small scale marine ranching area (MRA) to estimate demersal fish aggregations on September and November 2013. In this hydro-acoustic survey, the authors combined an image sonar with a scientific echo sounder to monitor an underwater situation and compare two acoustic data. Consequently, visual census survey was useful to estimate fish species composition for hydro-acoustic survey, because it is easy to identify aggregated fish species and overcome limits on a fishing depth and ability of an conventional fishing gear like a bottom gill-net or a fish trap at marine ranching area. Mean fish density was estimated as $0.757g/m^2$ on September and $0.219g/m^2$ on November and Fish abundance was finally calculated as 1.51ton (coefficient of variation, CV=13.1%) on September and 0.44ton (CV=47.7%) on November, respectively. Hydro-acoustic survey combined with the image sonar was useful to monitor fish aggregations and estimate fish stocks around artificial reefs at shallow coastal MRA. We were able to easily identify the underwater structures like an artificial reef and a fishing rope as well as fish aggregations from image sonar data. Therefore, the method was effective to separate unwanted echo signals in acoustic data of scientific echo sounder.

Fishing capacity assessment of the octopus coastal trap fishery using data envelopment analysis(DEA) (DEA 기법을 이용한 낙지통발어업의 어획능력 측정)

  • Estimating fishing capacity is one of current hot issues in the international fisheries. It is because that increased fishing capacity has caused not only fish stocks to be reduced, but also additional fishing costs to be incurred without additional incomes, which resulted in decrease of economically viability of fisheries. In order to solve this problem, FAO adopted 'the International Plan of Action for the Management of Fishing Capacity' in 1999 and recommended that member countries to measure fishing capacity and to implement the domestic action plan to reduce excess fishing capacity. This study is aimed at assessing fishing capacity of the octopus coastal trap fishery(OCTF) using data envelopment analysis(DEA) which is a method recommended by FAO. The DEA results on 10 individual OCTF vessels showed that the capacity utilization(CU) was a 0.93 on average, indicating some differences in CU among vessels(0.79-1.0). In addition, results of the sensitivity analysis revealed that under the current level of catch, the gross tonnage, horse power, days fished, and traps per trip could be reduced by 35%, 33%, 16%, and 18% on average, respectively.

Developing a Gene-trapping Approach for Gene Identification Using Nuclear Transfer in Zebrafish (지브라물고기 복제방법에 의한 유전자 동정 및 유전자트랩법 개발)

  • This involves identifying and cloning trapped genes from cultured cells carrying the gene-trap constructs and generating cloned zebrafish using these cells for functional study. Gene-trapping studies in gene-trapped cells were carried out in initial and cloned zebrafish carrying gene-trap events were successfully produced based on the nuclear transplantation technique. Two kind of retroviral gene-trap constructs were adopted. The first one(SA/GFP-TP), constructed in my laboratory, carries a GFP reporter gene containing a splicing acceptor and an internal neo gene. The second one(Neo-TP), obtained from Dr. Hicks (Hicks et al., 1997), contains a promoter-less neo gene located in the LTR sequence of a retroviral vector. The infected cells were subjected to drug selection(neomycin treatment) because the two constructs carry the neomycin resistant gene. All those cells survived the neomycin treatment should carry the proviral insertions. For Neo-TP, Isolated DNA from the neomycin-resistant fibroblast cells infected by Neo-TP, was digested with EcoR1 restriction enzyme and transformed into bacteria after ligation. This procedure led to the isolation of seven clones carrying flanking cellular DNA with a typical retroviral integration signature sequence. These clones contained genomic DNA ranging from 1kb to 7kb and sequences of 300-600 bp were obtained from each of the rescued plasmids. Database searching showed that all of them share high homology to zebrafish sequences. For fish cloning using tagged cells, initially, nucleus donors directly selected from a mixture of cells(Neo-TP cells) were used. A total of 44 embryos(3.7%) out of 1179 transplants were reached blastula stage; 8 of these embryos(0.8%) hatched and 3(0.3%) of them survived to adulthood. One out of three lived cloned zebrafish has an amplified fragment and was labeled with 32P.

Telomeric Dynamics and Telomerase Activity in Early Bovine Embryos (소의 초기 배 발생단계별 Telomeric DNA 함량 및 Telomerase Activity 분석)

  • Objective: This study was carried out to analyze the amount of telomeric DNA and telomerase activity in early bovine embryos. Methods: The amount of telomeric DNA in early bovine embryos at the 8 cell, morula and blastocyst stages was analyzed by Quantitative Fluorescence In Situ Hybridization (Q-FISH) technique using a bovine telomeric DNA probe. Telomerase activity was analyzed by Telomeric Repeat Amplification Protocol (TRAP assay). Results: The relative amount of telomeric DNA in early bovine embryos was gradually increased from 8 cell to blastocyst stage. It was not significantly associated with the grade of embryo quality. While telomerase activity was detected in the early bovine embryos at these stages, it significantly increased at morula stage and showed maximum activity at the blastocyst stage. Conclusion: The amount of telomeric DNA and the telomerase activity of bovine embryos increase during the progression of early embryogenesis, suggesting a positive correlation between telomeric DNA and telomerase activity. The telomerase activity seems to increase to maintain the levels of telomeric DNA through embryo development which are required for extensive cell division.

  • Telomeres consist of TTAGGG tandem repeated DNA sequences with specific proteins and locate at chromosome ends. Telomeres are essential for chromosome stability and are related with cell senescence, apoptosis and cancer. Telomerase is a ribonucleoprotein which has a template for the synthesis of telomeric DNA. This study was carried out to analyze the amount of telomeric DNA and telomerase activity in cattle cells. Analysis of the quantity of telomere in lymphocytes was done at different ages, sex and among Korean cattle and Holstein breeds. The telomerase activity was also analyzed in liver, brain, heart, kidney, and testis tissues of fetal calf and of 18 month old cattle. The amount of telomeres in lymphocytes and other tissue cells was analyzed by Quantitative-Fluorescence in situ Hybridization (Q-FISH) technique using a telomeric DNA probe. Telomerase activity was analyzed by Telomeric Repeat Amplification Protocol assay (TRAP). The amount of telomeric DNA on the lymphocytes during the whole life span was decreased along with age. Quantity of telomeres in Korean cattle was significantly higher than that in Holstein breed. The amount of telomeric DNA in males was significantly higher than that in females. Telomerase activity was up-regulated in most bovine tissues during fetal stage, but was down-regulated in most tissues at mature 18 month age except the testis cells. This study indicates that the amount of telomeres and telomerase activity of cells can be used as an age marker or/and a physiological marker of cattle.

Efficiency Analysis of the Ice Harbor Type Fishway Installed at the Gongju Weir on the Geum River using Traps (Trap을 이용한 공주보 아이스하버식 어도의 효과분석)

  • To overcome the stream fragmentation and blockage of migration of fishes by dams and weirs, fishways are commonly installed. However, limited studies were conducted for effectiveness and suitability of fishways installed in Korean streams. In this study, we investigated fish usages (by time periods, locations and months) of the Ice Harbor type fishway installed in the Gongju weir using traps. The monitoring were monthly conducted from June to October, 2012. The number of individuals which used fishway in September and October decreased than in June to August. Although no statistical significance was identified, many numbers of species and individuals were occurred at the trap installed at the left end of fishway than others. Fishes of more diverse size classes occurred at this trap as well. The number of collected individuals and water level of weir were positively correlated though they showed low correlation coefficient. Conversely, occurrence rate of fishes smaller than 70 mm of total length decreased with increasing water level of weir. fishway usage time of fish were different depends on ecological characteristics of each species. Various sizes of fishes can use fishway for their upstream migration. These results are useful for establishing management and evaluation plans of Ice Harbor type fishway in S. Korea.

Seasonal Fluctuations in Abundance and Species Composition of Fishes in Cheonsu Bay Using Trap Net Catches (小型定置網資料에 의한 淺水灣 魚類의 水質에 따른 種組成 및 量的 變動)

  • Fish samples were collected by a trap net during 1981∼82 at the mouth of Cheonsu Bay to determine seasonal fluctuations in abundance and species composition. Examination of a series of trap net catches indicated that trap net data could be used, at least, for a qualitative analyses of a fish community and for a quantitative study of some pelagic fishes of Cheonsu Bay. Of the 64 species collected, a few temporal components predominated in abundance because of great seasonal fluctuations of water temperature. The early spring catch showed a low abundance value, and two cold weather seasonals, juvenile Enedrias fangi and Ammodytes personatus predominated in number and in weight. The number of species, abundance value in number and in biomass showed a peak in late spring when fishes move into bay for spawning and feeding after over-winterinig in the sea. Three pelagic species or warm weather seasonals, Harengula zunasi , Ammodytes personatus and Konosirus punctatus, predominated in abundance values. In this period night catch was significantly larger than day catch in terms of number of species, abundance in number and in weight Summer data showed a marked reduction of the number of species and abundance due to the loss of spawners probably by mortality or wide dipersion after spawning. A slight increase in abundance was observed in autumn by recruitment of the young-of-the-years. This catch was predominated by three juvenile fishes, H. zunasi, E. japonica and K. punctatus, which were the same species as in spring. The autumn catch showed no significant difference between day and night compositions.

Summer Patterns and Diel Variations of Fish Movements Using Fish Trap Sampling Technique in the Juksan Weir (죽산보의 어도에서 트랩 샘플링 기법을 이용한 하절기, 일주기별 어류 이동성 평가)

  • The objective of this study was to evaluate seasonal patterns and diel variations of fish movements in the Juksan Weir which was constructed in 2010 at the down-stream of Yeongsan-River watershed. For this study, we monitored day-and-night movement(24 Hr cycle) and seasonal fish migration(June ~ August) patterns in 2011 along with species compositions and abundances depending on the locations of the traps within the fishway. Total number of species sampled was 14 and the total number of individuals was 1,263 with only the size-fractions(as total length) of the fish < 20 cm during the study. Seasonal analysis of fish movement in the fish way showed that highest frequency in the movement occurred in June - July, which is closely associated with a spawning peak season. The most dominant species using the fishway was Squalidus chankaensis tsuchigae, and this species turned out to be 26.9% of the total in the use rate of fishway. Daily monitoring of fish movements showed that most frequent movements occurred between 18:00 PM and 21:00 PM when the fish have a feeding time generally. The migratory fish were not found in the fishway during the study. Mean current velocity during the study $0.42{\pm}0.02ms^{-1}$(n = 42), and there were no significant statistical differences(p > 0.05) among the daily and monthly velocities in the fishway. The use rate of fish passage, in terms of fish species, was 48%, compared with total sampling of fish species(29 species) at the down-river regions during the same period, indicating a low use rate. Further continuous long-term monitoring should be conducted to evaluate the impacts of the weir construction in the river.

마우스 수정란의 발생단계별 telomeric DNA의 분포양상과 telomerase activity 분석

  • Telomere란 진핵세포에 존재하는 DNA-protein 복합체로서 염색체의 말단부에 tandem repeated DNA 서열 (TTAGGG)과 특정 단백질로 구성되어 있으며 세포 분열이 진행함에 따라 이의 길이가 짧아지게 되고 일정 길이 이하가 되면 세포의 사망이 유발된다. 반면 telomerase는 ribonucleoprotein으로서 telomeric DNA의 합성에 관여하는 것으로 염색체의 말단에 telomeric DNA의 소실을 보충하는 역할로 알려져 있다. 최근 암, 노화 등과 관련하여 telomere 및 telomerase의 연구들이 활발히 진행되고 있으며, 다양한 세포들에 있어 이들의 존재와 역할에 대해서도 많은 연구들이 수행되고 있다. 포유동물의 초기 배자에 있어 telomere의 분포 양상과 telomerase의 activity의 분석은 배 발생의 기작과 배자의 세포적 특성을 구명하는데 매우 중요한 과제라 사료된다. 따라서 본 연구에서는 마우스의 초기 배 발생 단계별 수정란의 telomeric DNA의 분포 양상과 각 단계별 배자들의 telomerase activity를 제시하고자 하였다. 시험에 공시된 마우스는 4-6주령된 ICR계통으로 이들을 과배란 처리 후 자연 교배시켜 얻은 2-, 4-, 8-세포기배, 상실배 및 배반포배를 대상으로 하였다. Telomeric DNA의 양적 분석은 각 발생단계별 수정란의 표본을 제작하고 human telomere repeat probe를 이용하여 FISH (fluorescence in situ hybridization)를 시행하였으며, 분리된 할구들을 형광현미경으로 관찰 후 상을 포착하고 image analyser program (MataMorph, UIC, USA)을 이용하여 한 개의 세포내 telomere의 상대적 함량을 분석하였다. 발생 단계별 배자의 telomerase activity의 분석은 TRAP (telomeric repeat amplofication protocol) assay로 분석한 바 각 발생 단계별 30개의 수정란으로부터 핵 단백질을 추출하여 telomerase를 신장시키고 PCR을 시행한 후 15% PAGE gel loading하여 이의 activity를 확인하였다. 분석 결과, telomeric DNA의 함유율은 발생단계별 다소의 차이를 나타내었으며 telomerase activity는 모든 발생단계의 수정란에서 확인할 수 있었고, 특히 상실배부터 높게 나타남을 확인하였다.

  • Telomeres are the end of chromosomes and consist of a tandem repeat sequence of (TTAGGG)n and associated proteins. Telomerase is a ribonucleoprotein which act as a template for the synthesis of telomeric DNA. Telomeres are essential for chromosome stability and are related with cell senescence, apoptosis and cancer. Even though telomeres and telomerase have been studied extensively, very little is known about telomere dynamics in embryonic cells. This study was carried out to analyze the telomeres distribution and telomerase activity of chicken cells during embryonic and developmental stages. The target cells for analysing were sperms, ovulated ova, early embryonic cells and the cells from brain, heart, liver, kidney and germinal tissue in fetus. Telomeres distribution on target cells was analyzed by Q-FISH (Quantitation-Fluorescence in situ Hybridization) techniques using a chicken telomere repeat probe. Telomerase activity was performed by TRAP assay (Telomeric repeat Amplification Protocol) with target DNA. In results, the telomeres of chicken were found at the ends of all chromosomes. In addition, chicken had interstitial telomeres on chromosomes 1, 2 and 3. Telomerase activity was highly detectable in early embryonic cells, germinal tissues and kidney cells. Whereas telomerase activity was gradually down-regulated when the organs, including brain, heart, and liver, were developed from embryos. In the distribution of telomeric DNA on the embryonic and developmental stages, most of the cells was gradually decreased in telomere quantity during ontogenesis.
