Bioinformatics in Fish: its Present Status and Perspectives with Particular Emphasis on Expressed Sequence Tags

  • Characterization of a single pass of cDNA sequence, an expressed sequence tag (EST) has been a fast growing activity in fish genomics. Despite its relatively short history, fish EST databases (dbESTs) have already begun to play a significant role in bridging the gaps in our knowledge on the gene expression in fish genome. This review provides a brief description of the technology for establishing fish dbESTs, its current status, and implication of the ESTs to aquaculture and fisheries science with particular emphasis on the discovery of novel genes for transgenic application, the use of polymorphic EST markers in genetic linkage mapping and the evaluation of signal-responsive gene expression.

Fish Activity State based an Intelligent Automatic Fish Feeding Model Using Fuzzy Inference (퍼지추론을 이용한 어류 활동상태 기반의 지능형 자동급이 모델)

  • The automated fish feed system currently used in Korea supplies a certain amounts of feed to water tanks at a certain time. This automated system can reduce the labor cost of managing aqua farms, but it is very difficult to control intelligently and appropriately the amount of expensive feed that is critical to aqua farm productivity. In this paper, we propose the FIIFF Inference Model( Fuzzy Inference-based Intelligent Fish Feeding Model) that can solves the problems of these existing automatic fish feeding devices and maximizes the efficiency of feed supply while properly maintaining the growth rate of fish in aqua farms. The proposed FIIFF inference model has the advantage of being able to control feed amounts appropriately since it computes the amount of feed using the current water environments and fish activity state of the aqua farms. The result of the feed amount yield experiment with the proposed FIIFF Inference Model represents the effect of saving 14.8% over the eight months of actual feed amount in the aqua farm.

Zinc status and dietary quality of type 2 diabetic patients: implication of physical activity level

  • The purpose of this study was to analyze the relationships among zinc status, diet quality, glycemic control and self-rated physical activity level of type 2 diabetic patients. Dietary intakes for two non-consecutive days were measured by 24-hour recall method for seventy-six diabetic patients. Fasting blood glucose and HbA1c were measured for the assessment of glycemic control. We evaluated the extent of dietary adequacy by the percentage of subjects with a dietary intake of a nutrient less than the estimated average requirement(EAR), the dietary diversity score(DDS) and the dietary variety score(DVS). Zinc status was assessed from serum levels and urinary excretion. Dietary inadequacy was serious for five nutrients: riboflavin, calcium, thiamin, zinc and vitamin C. Dietary intakes from the meat, fish, and egg food groups and the milk food group were below the recommended level. Effects of Dietary Inclusion of Astaxanthin on Growth, Muscle Pigmentation and Antioxidant Capacity of Juvenile Rainbow Trout (Oncorhynchus mykiss)

  • This study was designed to investigate the effects of dietary astaxanthin levels on growth performance, feed utilization, muscle pigmentation, and antioxidant capacity in juvenile rainbow trout. Four experimental diets were formulated to contain 0, 50, 75, and 100 mg/kg astaxanthin (designed as AX0, AX50, AX75, and AX100). Each diet was fed to triplicate groups of fish (18.5 g/fish) for 10 weeks. Growth performance and muscle composition of fish were not affected by dietary astaxanthin levels. Total carotenoid concentration in the muscle of fish fed the AX50 diet was higher than that of fish fed the AX0 diet, but no significant differences were observed between these fish and those fed the AX75 and AX100 diets. Muscle astaxanthin content increased with increased astaxanthin in the diet. Deposition of astaxanthin in the flesh resulted in a decrease in lightness and an increase in redness and yellowness. The fillets from trout fed the AX75 diet had significantly lower lightness than trout fed the AX50 and AX100 diets. Fish fed the AX50 and AX75 diets showed significantly lower catalase activity than those fed the control diet. Total antioxidant status increased significantly in all astaxanthin supplemented groups when compared to the control group. Superoxide dismutase activity was significantly decreased in fish fed the AX50 diet compared to fish fed the AX0 diet. These findings suggest that while fillet pigmentation increased with increasing dietary astaxanthin concentration, indices of fish antioxidant capacity may not be affected in a dose dependent manner.

Effects of 4-tert-octylphenol on glutathione-related antioxidant status in olive flounder Paralichthys olivaceus

  • Effects on glutathione-related antioxidant parameters were examined after a chronic exposure of olive flounder, Paralichthys olivaceus to dietary 4-tert-octylphenol (4-tert-OP). Fish were fed diets containing 4-tert-OP at 0, 1, 5 and 10 mg/kg diet for 6 weeks. Antioxidant parameters examined were reduced glutathione (GSH) contents and enzyme activities of glutathione reductase (GR), glutathione S-transferase (GST) and glutathione peroxidase (GPx) in tissue homogenates of the liver, kidney and gill. It was observed that all parameters examined increased although there were some differences in dose responses and temporal patterns in the increase. GSH contents increased after exposure to 4-tert-OP in the three organs examined. However, the GSH increase was evident only after 4 weeks in the liver whereas it was elevated after 2 weeks in the kidney and gill. GR activity exhibited a significant increase in response to 4-tert-OP at 1 mg/kg in all three organs, however, its activity returned to control levels when exposed to 5 and 10 mg/kg. Hepatic GST activity showed an earlier increase at week 2 in contrast to the kidney and gill where they increased after 4 weeks of 4-tert-OP exposure. Temporal patterns in GPx activity changes to 4-tert-OP exposure were dissimilar among the organs: hepatic activity increased from week 2 through week 6; renal activity increased transiently at week 2; gill levels were higher through weeks 4 - 6. The results suggest that elevation of several GSH-related antioxidant parameters can be considered as evaluation criteria for 4-tert-OP-induced oxidative stress in a fish.

Induction of oxidative stress in Clarias gariepinus from Eleyele River in Nigeria

  • This study evaluated some markers of oxidative stress in the organs of African Catfish, Clarias gariepinus from Eleyele River in Oyo State, Nigeria. Clarias gariepinus (250 g-400 g) were collected from Eleyele River (a suspected polluted River) and Clarias gariepinus from a clean fish farm (Durantee fisheries) were used as the control. Levels of Malondialdehyde (index of lipid peroxidation), Glutathione (GSH) and activities of antioxidant enzymes- Superoxide dismutase (SOD), Catalase and Glutathione-S-Transferase (GST) were evaluated in the liver, kidney and gills of the fish. From the results, there were significant (p<0.001) increases in malondialdehyde and GSH levels in the liver, kidney and gills of Clarias gariepinus from Eleyele River compared with control. The activity of GST increased significantly (p<0.05; p<0.001) in the liver and kidney of fish from Eleyele River compared with control. There was a significant decrease (p<0.05; p<0.001) in SOD activity in all the organs of Clarias gariepinus from Eleyele River compared with conrol and also a significant (p<0.001) decrease in catalase activity in the gills and kidney of the fish but catalase activity increased in the liver. Increase in lipid peroxidation and alterations in antioxidant status in Clarias gariepinus from Eleyele River show that the fish were under oxidative stress. These suggest that the River is polluted probably as a result of various wastes frequently discharged into the River. This could pose serious health risks to consumers of water and aquatic organisms from the River.

Nutritional Status and Cognitive Status of the Elderly Using Public Health Center in Ulsan (울산지역 보건소 이용 노인들의 영양상태와 인지상태)

  • This study was designed to obtain the information concerning food intake, dietary habits, functional status, health condition and cognitive status of the elderly using public health center in Ulsan area. The subjects of this study consisted 154 elderly persons aged 60~82 years. Interviews were conducted using the health habits and food frequency questionnaires to provide basic information for nutrition education program. We evaluated the current food intake, dietary cholesterol intake (cholesterol index), functional status, cognitive function and blood analysis of the subjects. The results of this study were as follows : Mean age of the subjects was 68.7 $\pm$ 6.7years. The average cognitive status score of the subjects was 7.9 $\pm$ 2.0 (full score was 10.0). Male had a higher cognitive status score than female. There was significant difference between cognitive status score and age, education level, pocket money, physical activity and living condition. The subjects who had a higher cognitive status score ate more fish and meats group and milk and milk products than the subjects had a lower cognitive status score. And hemoglobin level, serum triglyceride, HDL-cholesterol and atherogenic index affected to cognitive status while fasting blood glucose and LDL-cholesterol did not any effect on cognitive status. These results have demonstrated that various socioeconomic variables and food intake pattern and nutritional status affect on cognitive status with aging and suggest that proper nutrition education and adequate nutrient intake in quality and quantity are essential in maintaining cognitive status in later life.

Relationship of Socioeconomic Status and Food Intake to Cognitive Status of the Older Population

  • This study was done to investigate the relationship of socioeconomic status and food intake to cognitive status of the older population. The subjects of this study consisted of 214 older persons aged 60 - 84 years. Interviews were conducted using the health-related habits and food frequency questionnaires to provide basic information for nutrition education program. We evaluated the current food consumption-pattern and cognitive status of the subjects. The results of this study were as follows: Mean age of the subjects was 69.7${\pm}$7.4 years. The average cognitive function score of the subjects was 7.9${\pm}$2.0 (full score was 10.0). Male had a higher cognitive status score than female. There was significant difference between cognitive status score and age, education level, pocket money, physical activity and family type. The subjects who had a higher cognitive status score ate more fish and meats group and milk and milk products than the subjects that had a lower cognitive status score. These results have demonstrated that various socioeconomic variables and food intake pattern affect on cognitive status with aging and suggest that proper nutrition education and adequate nutrient intake in quality and quantity are essential in maintaining cognitive status in later life.

Effects of Diet Supplementation with Onion Allium cepa L. Juice Processing By-products on Juvenile Black Rockfish Sebastes schlegelii Growth, Feed Utilization and Antioxidant Enzymes Activity (배합사료내 양파(Allium cepa L.)즙 부산물 첨가 함량에 따른 조피볼락(Sebastes schlegelii) 치어의 성장, 사료이용성 및 항산화효소 활성에 미치는 영향)

  • An eight-week feeding trial was conducted to evaluate the effects of a diet consisting of different onion juice processing by-product (OJPB) levels on juvenile black rockfish Sebastes schlegelii growth, feed utilization, and, lysozyme and antioxidant enzymes activities. Juvenile rockfish (2.2 g) were randomly distributed into 15 flow-through tanks (30 fish/tank). Five experimental diets were prepared in triplicate. The experimental diets were supplemented with OJPB at different levels of 0 (control), 0.25, 0.5, 0.75 and 1% (designated as OJPB0, OJPB0.25, OJPB0.5, OJPB0.75 and OJPB1, respectively). At the end of the feeding trial, the results revealed that the fish that were fed the OJPB0.75 and OJPB1 diets showed enhancement in growth (weight gain and specific growth rate) and feed utilization (feed efficiency and protein efficiency ratio) compared with the fish that were fed other diets. Plasma lysozyme, glutathione concentration, and superoxide dismutase and catalase activities significantly increased in the fish that were fed the OJPB0.75 and OJPB1 diets. In conclusion, dietary supplementation of 0.75-1% OJPB in juvenile rockfish diet improved the growth performance and antioxidant status.

Effect of Dietary Selenium and Fish oil on Lipid Peroxidation and Fatty Acid Profile in the Rat (식이 셀레늄 수준과 식이 지방산 조성이 쥐의 지질과 산화 상태와 조직의 지방산 조성에 미치는 영향)

  • The influence of selenuium deficiency and fish oil on lipid peoxidation status and fatty acid composition of tissues(plasma aorta and liver) was studied. Male Sprague Dawley rats were fed for eight weeks semipurified diets containing 7% corn oil(by weight) or 5, 5% fish oil(MaxEPA) plus 1.5% corn oil with oil with or without selenium status (glutathione peroxidase activity and selenium levels) were significantly lower in the rats given inadequate selenium in plasma aorta (p<0.02 and p<0.001 respectively) gut not that in plasma Selenium supplementation decreased hepatic MDA levels(p<0.02) Increases in the levels of 20:5(n-3) 22:5(n-3), 22:6(n-3) 20:3(n-6) and a decrease in the level of 20:4(n-6) were observed in plasma total lipids and aortic and hepatic phospholipids when fish oil was fed. Though selenium supplementation increased the level of n-3 fatty acids(such as 22:6(n-3)) in plsama and the aorta is overall effect was smaller than the effect of fish oil feeding. These data suggest that selenium may play a significant but minor role in protecting against lipid peroxide-tion even when vitamin E intakes are in excess of current recommendations in both corn oil and fish oil diets.
