난황 콜레스테롤과 지방산 조성에 관한 서로 다른 지방의 첨가 효과 (Effects of Dietary Fats on the Cholesterol Content and Fatty Acid Composition of Egg Yolk)

  • 계란 품질, 난황 콜레스테롤 함량 및 지방산 조성의 변화에 관한 산란계 사료 내 서로 다른지방의 첨가효과를 조사하였다. 36주령의 Hyline brown 산란계 총 200수(4처리구${\times}$50수/반복)를 공시하여 우지, 옥수수유, 아마인유 및 어유를 각각 5.0% 함유하는 실험사료를 10주간 급여하였다. Haugh unit, 파란 강도, 난각 두께 및 난황색에 대한 처리구 간 유의적 차이는 나타나지 않았다. 첨가지방은 계란 콜레스테롤 함량과 고도 불포화지방산 조성을 현저하게 변화시켰다. 난황의 n-6지방산 함량은 옥수수유 첨가구가 31.61%로써 가장 높았고 우지, 아마인유 및 어유 첨가구순으로 낮았으며, n-3 지방산 함량은 아마인유와 어유 첨가구가 각각 9.74%, 5.16%로써 유의적 증가 경향을 나타냈다(p<0.05). 난황지방 산 조성의 n-3지방산 증가는 계란 콜레스테롤 함량 감소를 초래하였다. 난황 콜레스테롤(mg/g yolk) 및 계란 콜레스테롤(mg/60g egg) 범위는 각각 $15.98{\sim}18.37mg$$227{\sim}261mg$으로써 우지 첨가구가 가장 높았고 어유, 아마인유 및 옥수수유 첨가구 순으로 낮아지는 경향을 보였다(p<0.05). 난황 콜레스테롤의 감소율은 우지 첨가구와 비교하였을 때 어유 13.01%, 아마인유 11.49% 및 옥수수유 6.91%로써 유의적인 차이를 나타냈다(p<0.05). 이상의 결과는 산란계 사료 내 첨가되는 지방급원을 조절해 줌으로써 계란 콜레스테롤을 낮출 수 있으며 n-6와 n-3 지방산을 난황으로 축적하는 것이 가능함을 시사해준다.

수유기에 식이와 함께 섭취한 DHA가 산모의 혈액과 모유의 지질조성에 미치는 영향 (The Influence of DHA Supplementation in Maternal Diets on Fatty Acid Compositions of Plasma Lipids and Human Milk)

  • The most abundant long-chain polyunsaturated fatty acid in brain lipids is docosahexaenoic acid(C22 : 6 N-3, DHA). It is incorporated into nerve tissues mostly in utero and during the first year of life. DHA in brain is derived from either pre-formed DHA in human milk or by infant hepatic synthesis from linolenic acid in milk. This study was designed to investigate the effects of DHA supplementation on fatty acid profiles in maternal plasma lipid and breast milk. Twenty lactating women participated in the study. Seven women took 3g of fish oil per day and vitamin E for 28 days starting from the day of giving birth. Five women consumed 1.5g of fish oil as well as tivamin E, and the rest took vitamin E supplements for the same period of time. Dietary questionnaires and 3 consecutive 24-h recalls were collected to evaluate theri nutritional status and food habits. Finding that DHA intake from fish was not significantly different among three experimental groups, the partcipants were instructed to continue eating their usual home diets. Milk samples were taken on the day of giving birth, as well as the 7th, 14th and 28th day being the supplement phase, and finally 2 weeks after the cessating of DHA supplements. The amounts of the fish oil supplements produced significant dose-dependent increased in the DHA content of milk and plasma, but to a lesser degree. Base-line for 28 days raised the level to 2.05$\pm$0.43% and 1.5g/day supplement produced DHA levels of 1.02$\pm$0.19%. The results of this study indicated that relatively small amount of dietary DHA supplementation significantly elevats DHA content in milk. This would clearly elevate the infant's DHA intake which in turn may have implications for the infant's brain development.

Effects of Phospholipid Extract from Squid Viscera on Lipid Oxidation of Fish Oil

  • Phospholipid(PL), phosphatidylcholine(PC) and phosphatidylcholine free PL(PCF) were extracted from squid viscera and the antioxidant effects of each fraction on the oxidation of refined fish oil were evaluated. Polyunsaturated fatty acid contents were the highest in PC(46.7%) followed by PL(44.8%) and PCF(40.9%). The effects of each phospholipid fraction on stabilizing fish oil were compared by incubating at 40$^{\circ}C$ for 10 days. At the initial period(2 days), changes in peroxide value did not show any significant difference ; however, as incubation time was extended, PC fraction showed the strongest antioxidant activity. PL and PCF added to fish oils also resulted in increased stability against oxidation. Antioxidative effect of PC at the 5% level was equivalent to 0.05% BHT, 1% catechin and 1% tocopherol.

반응표면분석법에 의한 정제어유 미세캡슐화 공정의 최적화 및 미세캡슐 저장안정성 분석 (Optimization of Fish Oil Microencapsulation by Response Surface Methodology and Its Storage Stability)

  • ${\omega}3$계 고도불포화 지방산의 한 종류인 DHA(docosahexaenoic acid, $C_{22:6},\;{\omega}3$)를 함유하는 정제어유를 중심물질로 하고 agar와 waxy corn starch를 피복물질로하여 미세캡슐화 공정을 수행할 때 반응표면분석법(response surface methodology, RSM)을 이용하여 최적 조건을 확립하고자 하였다. 이때 정제어유 미세캡슐화의 수율을 정량화하기 위하여 5% cupric acetatepyridine 용액에 발색정도가 뛰어난 oleic acid를 정제 어유내에 20%(w/w) 농도로 첨가하여 중심물질로 사용하였다. 반응표면분석결과 최적화된 미세캡슐화 조건은 [중심물질, Cm]:[피복물질, Wm]의 비율 =4.9 : 5.1(w/w), 유화제(sorbitan monolaurate, H.L.B. 16.7)의 농도 = 0.48%(w/w), 분산매의 온도 = $19.4^{\circ}C$이었고, 실제 이 조건에서 99.9%의 수율을 보였다. 또한 최적 조건하에서 제조된 미세캡슐물질 저장을 위한 최적 온도 및 pH는 각각 $25{\circ}C$와 pH 7.0인 것으로 나타났으며, $25^{\circ}C$, pH 7.0의 분산매내에서 7일간 저장한 후에도 99%이상의 미세캡슐이 안정한 상태로 존재함을 확인하였다.

N-6와 n-3 지방산이 풍부한 식이가 뇌졸중 유발 모델에서 뇌경색 크기 및 항산화 효소계에 미치는 영향 (Neuroprotective & antioxidant effects of diets high in n-6 and n-3 fatty acids in rat focal brain ischemia model)

  • This study was undertaken to investigate the effects of n-6(corn oil) & n-3(fish oil) fatty acids on infarction size and the cerebral activities of antioxidant enzyme in rat focal brain ischemia model. Weaning Sprague-Dawley rats were fed with either corn oil supplemented diet(COD, 14% corn oil) or fish oil supplemented diet(FOD, 14% menhaden oil) for 6 weeks. The right middle cerebral artery was occluded for 2 hours with a silicon rubber coated nylon surgical thread. After 24 hours of recirculation, the rats were sacrificed and brain sections were photographed using CCD camera after staining with 2, 3, 5-triphenyltetrazolium chloride for 60 minutes in room temperature. The infarcted area was measured and the volume of infarction was calculated. Catalase(CAT), superoxide dismutase(SOD) activities, and fatty acid composition in the brain were also measured. The total and corrected infarction volumes were not significantly different between FOD and COD group. The docosagexaenoic acid(DHA) and DHA content/arachidonic acid(AA) ratio of the cerebral cortex, an index of defense against lipid oxidation, were significantly increased in FOD group compared to those of COD group(p<0.05). In the left cortex(non-infarction side) as well as the right cortex(infarction side) of FOD group, CAT and Cu/Zn SOD activities were higher than those of the COD group(p<0.05). However, CAT and Cu/Zn SOD activities were not significantly different between the left cortex(non-infarction side) and the right cortex(infarction side) of both FOD and COD group. GPx activities were also not significantly different between two groups. Our results demonstrate that the brain infarction size in FOD and COD were not significantly different. However, cerebral lipid composition and antioxidant enzyme activities in FOD and COD group were different. Fish oil, a source of n-3 polyunsaturated fatty acid(PUFA) and corn oil, that of n-6(PUFA) may have a protective effect against oxidative stress induced via different mechanisms.

고당질 식이시 n6 와 n3 불포화 지방산이 쥐의 혈장지질 저하기전에 미치는 영향 (Plasma Lipid-Lowering Effect of n6 and n3 Polyunsaturated Fatty Acids in Rats Fed High Carbohydrate Diet)

  • To compare the hypolipidemic effects of n6 linoleic acid. n3 $\alpha$-linolenic acid and n3 eicosapentaenoic acid in rats fed high carbohydrate(70% Cal) diet. male Sprague Dawley rats were fed different experimental diets for 6 weeks. which were different only in fatty acid composition. The dietary fats were beer tallow(BT) as a source of saturated fatty acid (SFA), corn oil(CO) for n6 linoleic acid(LA), perilla oil(PO) for n3 $\alpha$-linolenic acid(LL) and fish oil(FO) for n3 eicosapentaenoic acid(EPA) and docosahexaenoic acid (DHA) Plasma total cholesterol(Chol) level was increased by n6 LA but decreased by n3 LL and n3 EPA and most effectively reduced by n3 EPA. HDL-Chol level was raised by n6 LA, but there was no significant change in HDL-Chol levels by n3 LL and was lowered by n3 EPA. Plasma TG level was reduced by n6 LA, but lipogenesis in liver was not affected by n6 LA. However, plasma TG level was lowered by n3 LL and EPA. Both lipogenic enzyme activity and liver TC level were also decreased by n3 PUFA. The relative proportions of TG in VLDL was significantly lowered by n3 EPA. but the proportions of Apo B in VLDL was not changed by n3 EPA. Overall. the hypolipidemic effect was in the order of EPA+ DHA(n3) >LL(n3) >LA(n6) and fish oil and perilla oil rich in n3 PUFA may have important nutritional applications in the prevention and treatment of hypertriglyceridemia.

Effects of Dietary Cimetidine, a Cytochrome P450 Inhibitor, on the Benzo[a]pyrene-induced Lipid Peroxidation of Liver in Olive Flounder, Paralichthys olivaceus

  • Effects of cimetidine, a cytochrome P450 inhibitor, on the benzo[a]pyrene (BaP)-mediated cytochrome P450 induction and lipid peroxidation of liver in olive flounder, Paralichthys olivaceus, were investigated. Fish were fed either a cimetidine-supplemented diet or a cimetidine-free control diet once daily to satiation for 3 days. After 6 hrs of last feeding, the fish received intraperitoneal (i.p.) injection of BaP (20 mg/kg of body weight) dissolved in sterile corn oil $(100 \mu L)$ or received only a corn oil i.p. injection. At 1, 2, 3, and 7 days after the injection, hepatic cytochrome P450 and thiobarbituric acid reactive substances (TBARS), an indicator of lipid peroxidation, were analyzed. BaP injection resulted in an increase of hepatic cytochrome P450, and the fish fed the cimetidine-supplemented diet before injection of BaP showed delayed increase of hepatic cytochrome P450 compared to the fish fed a cimetidine-free diet and BaP injected. Injection of BaP clearly induced hepatic lipid peroxidation, and consistently higher TBAR values were shown in the fish fed a cimetidine­supplemented diet before injection of BaP than the fish injected with BaP alone.

지방공급원이 병아리의 혈액 및 간 Cholesterol 함량에 미치는 영향 (A Study on the Effects of Dietary Fat Sources on the Plasma and Liver Cholesterol Levels in Young Chicks)

  • 본 실험에서는 식물성이면서 포화도가 높은 팜유와 동물성이면서 불포화도가 높은 어유를 병아리에 섭취시켜 혈액과 간내의 cholesterol 및 TG 함량에 미치는 영향을 조사하는데 그 목적을 두었다. 그 결과 에너지 섭취량은 각 첨가 지방의 에너지가가 g당 9kcal로 가정하고 다른 원료 사료들은 각각의 대사에너지 함량을(Scott, 1976) 기준해서 계산하였는데 그 결과 지방 첨가 수준이나 첨가 지방의 종류에 관계없이 에너지 섭취량이 전반적으로 비슷하게 나타났다. 체중 증가량도 대체로 처리간에 통계적인 처리가 없었고, 단지 실험 1에서 동물성 지방급여구가 식물성 지방급여구보다 유의하게 적게 나타났다. 그리고 체중 100g당의 무게로 나타냈을때 간의 무게는 실험 1의 어유구와 실험2의 경화어유구에서 각각 가장 무거웠고, 심장의 무게는 실험2에서 두 어유구가 모두 가장 무겁게 나타났다. 한천 간장의 총지방 함량은 실험2에서 우지구가 팜유구보다 유의하게 더 많았다. 고리고 간장의 cholesterol 함량은 실험1에서만 어유구가 팜유나 우지구보다 더 높게 나타났고, 혈액 cholesterol 함량은 실험2에서 어유구가 가장 낮았다(P〈0.05). 또 간장의 TG함량은 실험2에서 채종유, 우지 및 경화어유구등이 팜유구보다 더 높았고, 실험3에서는 C/P 비율이 164인 구가 대조구보다 간에 더 많은 TG를 함유한 것으로 나타났다 (P〈0.05). 그러나 혈액 TG는 전반적으로 개체 변이가 심해서 유의한 차이를 볼 수 없었다. 이상의 결과를 다시 첨가된 각 지방별로 요약하면, 옥수수유, 대두유, 채종유등은 여러 측정치에서 유사한 결과를 보여주었고, 채종유의 경우에는 심잠의 무게를 증가시킬 것으로 예상했었으나 그렇지 않았다. 그리고 팜유는 우지나 경화어유보다 간의 총지방 함량이나 간장의 TG를 더 낮추는 효과를 보였고 cholesterol에 대한 효과도 다른 식물성 지방과 유사하게 나타났다. 어유는 실험 1에서 간의 중량을 무겁게 하고, 간내의 cholesterol 함량을 높였는데 실험2에서는 심장의 무게를 각각 무겁게 했고 한편 혈액 cholesterol을 가장 낮은 수준으로 떨어뜨렸다. 그런데 실험 1과 2에서 사용된 어유는 각각 다른 source에서 구한 것으로서 여러 어종의 것이 아니기 때문에 비교하기가 곤란하다.

고DHA(Docosahexaenoic Acid)어유가 첨가된 식이가 흰쥐의 항혈전 및 지질과산화물대사에 미치는 영향 (The Effect of Docosahexaenoic Acid Rich-Fish Oil Addition on Antithrombotic effect and Lipid Peroxidation in Rat)

  • This study was undertaken to elucidate the effect of DHA rich fish oil(DHA rich oil) added to different dietary fats on thrombosis and lipid peroxidation. Rats were fed perilla oil, sesame oil and beef tallow with or without DHA rich oil for 12 weeks. Bleeding time was the longest in Perilla oil groups with or without DHA rich oil. The productions of thromboxane B2(TX B2) and 6-keto Prostaglandin F1$\alpha$(6-keto PG F1$\alpha$) were the highest in Esame oil group without DHA rich oil. Bleeding time tended to be extened and group showed the most antithrombotic effect among three oil groups when DHA rich oil added. The antithrombotic effect by DHA rich oil addition seemed to be resulted from the increase of dietary n-3 fatty acid rather than DHA. And there was not the difference in antithrombotic effect between DHA and $\alpha$-linolinic acid. The level of TBARS(thiobarbituric acid reactive substances) in plasma and liver, and the activities of lipid peroxide metabolizing enzymes(catalase, superoxide dismutase and gluthathion peroxidase) in erythrocyte and liver were not affected by the dietry fat type and DHA rich oil addition, except that activity of hepatic catalase was increased by DHA rich oil addition. Therefore it revealed the DHA level added in this study seldom affected lipid peroxidation. However, it dose not conclude that DHA level of this study make low production of lipid peroxide because the peroid of our study was short.

Effect of the Mixed Oil and Monensin Supplementation, and Feeding Duration of Supplements on c9,t11-CLA Contents in Plasma and Fat Tissues of Korean Native (Hanwoo) Steers

  • The present study was conducted with twenty-four Korean native (Hanwoo) steers to observe the effect of mixed oil and monensin supplementation and duration of feeding on c9,t11-CLA content in plasma and fat tissues. The steers were randomly assigned to three groups of eight animals each according to body weight. Hanwoo steers in the control group were fed the commercial concentrate for the late fattening stage. The other groups of steers were fed the same diet as control steers, but the concentrate was supplemented with high-$C_{18:2}$ oil mixture (soybean oil, sunflower oil, safflower oil) and fish oil at 6% level of concentrate (DM basis), and monensin (20 ppm). The second and third group of steers was fed the oil mixture supplemented diet with monensin for the last 10 weeks and 20 weeks, respectively, prior to being slaughtered. The oil mixture consisted of 45% soybean oil, 20% sunflower oil, 20% safflower oil and 15% fish oil. Average daily gain (p<0.049) and feed efficiency (p<0.018) of the steers fed the diet supplemented with oil mixture and monensin (OM-M) for 20 weeks were higher than those of the other groups of steers. Dressing percent, fat thickness and longissimus muscle area were not affected by the OM-M supplementation and duration of its feeding. The OM-M supplementation increased the content of total-cholesterol (p<0.0001-0.0007) and HDL-cholesterol (p<0.0001) in the plasma of steers compared to the control diet. The steers fed the OM-M diet had a higher proportion of c9,t11-CLA in plasma (p<0.048-0.044) than the control steers. Feeding the OM-M diet for 20 weeks increased the proportion of CLA in intramuscular (p<0.015), intermuscular (p<0.039) and subcutaneous (p<0.001) fat tissues compared with both steers fed the control diet and the OM-M diet for 10 weeks. Increased (p<0.007) proportion of total unsaturated fatty acids in steers fed the OM-M diet for 20 weeks compared to those in control steers was related to the increased (p<0.001) $C_{18:2}$ and decreased (p<0.001) $C_{18:0}$ proportions in subcutaneous tissue.
