Identification and Expression of Retroviral Envelope Polyprotein in the Dogfish Squalus mitsukurii

  • Determining the infection history of living organisms is essential for understanding the evolution of infection agents with their host, particularly for key aspects such as immunity. Viruses, which can spread between individuals and often cause disease, have been widely examined. The increasing availability of fish genome sequences has provided specific insights into the diversity and host distribution of retroviruses in fish. The shortspine spurdog (Squalus mitsukurii) is an important elasmobranch species; this medium-sized dogfish typically lives at depths of 100~500 m. However, the retroviral envelope polyprotein in dogfish has not been examined. Thus, the aim of the present study was to identify and analyze the retroviral envelope polyprotein in various tissues of dogfish. The 1334-base pair full-length novel cDNA of dogfish envelope polyprotein (dEnv) was obtained by 3' and 5'-rapid amplification of cDNA end analysis from S. mitsukurii. The open reading frame showed a complete coding sequence of 815 base pairs with a deduced peptide sequence of 183 amino acids that exhibited 34~50% identity with other fish and bird species. It was also expressed according to reverse transcription and real-time polymerase chain reaction in the kidney, liver, intestine, and lung, but not in the gill. This distribution can be assessed by identifying and analyzing endogenous retroviruses in fish, which consists of three main genes: gag, pol and env. Dogfish envelope polyprotein sequence is likely important in evolution and induces rearrangements, altering the regulatory and coding sequences. This is the first report of the identification and molecular characterization of retroviral envelope polyprotein in various tissues of S. mitsukurii.

  • Polybrominated diphenyl ethers (PBDEs) as flame-retardant additives have been used in a wide array of products, including building materials, electronics, furnishings, motor vehicles, airplanes, plastics, polyurethane foams, and textiles. They are structurally similar to PCBs and other polyhalogenated compounds. PBDEs are found in a variety of foods, and 95% of all human exposure to POPs comes from food. The major food sources are fish/seafood and dairy products. A number of studies have reported high levels of PBDEs in animals, increasing the public's concern over PBDE levels in animals. This study evaluates the relationship between the level of PBDEs according to piglet diseases. Salmonella spp. and Streptococcus are bacterial diseases. Porcine respiratory reproductive syndrome (PRRS) is a viral disease. The concentration of PBDEs in the piglet liver from viral diseases was higher than that from bacterial diseases. BDE-47 and BDE-99 were detected in most samples. More PBDEs were detected in the piglet liver from PRRS of North American type than from that of European type.

Changes in Differentially Expressed Genes in the Liver of Oryzias latipes by Binary Exposure to Carcinogenic Polycyclic Aromatic Hydrocarbons

  • The biological effects of carcinogenic polycyclic aromatic hydrocarbons (cPAHs) including benzo[a]pyrene (BaP), dibenzo[a,h]anthracene (DBA), benzo[a]anthracene (BaA), benzo[b] fluoranthene (BbF), benzo[k]fluoranthene (BkF), and indeno[1,2,3-c, d]pyrene (InP) on transcriptomic changes were determined in the liver of Oryzias latipes. Differentially expressed genes (DEGs) by binary exposure to cPAHs (BaP+BaA, BaP+BbF, BaP+BkF, BaP+DbA, BaP+InP) were screened by annealing control primers-based polymerase chain reaction followed by sequence analysis and BLAST searching. The results showed that four DEGs were commonly expressed by cPAHs and they were identified as ribosomal protein S4, coagulation factor II, elongation factor 1 beta, and a predicted protein similar to human immunodeficiency virus type I enhancer binding protein 3. This finding suggests that binary exposure to cPAHs interferes protein synthesis required for fundamental liver functions in fish.

Effects of 4-Nonlyphenol Exposure on $P.$ $olivaceus$ and $S.$ $schlegeli$ Vitellogenesis

  • The effects of the estrogenic compound 4-nonylphenol (4-NP) on vitellogenesis in juvenile olive flounder ($Paralichthys$ $olivaceus$) and rockfish ($Sebastes$ $schlegeli$) exposed continuously at 10, 50 and $100{\mu}g{\ell}^{-1}$ levels for 7 days were compared. The expression of VTG mRNA level and protein using specific probes were examined. The levels of plasma estradiol-17 ${\beta}(E_2)$ and testosterone (T) were assessed by radioimmunoassay (RIA). Plasma $E_2$ concentrations increased significantly in two female fish species exposed to $100{\mu}g{\ell}^{-1}$ of 4-NP over concentrations in control fish. Plasma T concentrations increased in $P.$ $olivaceus$. Four days after exposure, the level of VTG mRNA expression increased in $P.$ $olivaceus$ and $S.$ $schlegeli$ exposed to $20{\mu}g{\ell}^{-1}$ of 4-NP. In addition, plasma VTG protein expression was seen in $P.$ $olivaceus$ and $S.$ $schlegeli$. In $S.$ $schlegeli$ and $P.$ $olivaceus$ exposed to 4-NP, the changes were noticed mainly in hepatocytic vaculation after 7 days of exposure. Thus, 4-NP may disrupt vitellogenesis in immature fish both directly and indirectly via disrupted steroidogenesis and liver pathology. Immature $S.$ $schlegeli$ were the most sensitive to 4-NP exposure in vitellogenesis.

Telomeres Distribution and Telomerase Activity During Chick Embryonic and Developmental Stages

  • Telomeres are the end of chromosomes and consist of a tandem repeat sequence of (TTAGGG)n and associated proteins. Telomerase is a ribonucleoprotein which act as a template for the synthesis of telomeric DNA. Telomeres are essential for chromosome stability and are related with cell senescence, apoptosis and cancer. Even though telomeres and telomerase have been studied extensively, very little is known about telomere dynamics in embryonic cells. This study was carried out to analyze the telomeres distribution and telomerase activity of chicken cells during embryonic and developmental stages. The target cells for analysing were sperms, ovulated ova, early embryonic cells and the cells from brain, heart, liver, kidney and germinal tissue in fetus. Telomeres distribution on target cells was analyzed by Q-FISH (Quantitation-Fluorescence in situ Hybridization) techniques using a chicken telomere repeat probe. Telomerase activity was performed by TRAP assay (Telomeric repeat Amplification Protocol) with target DNA. In results, the telomeres of chicken were found at the ends of all chromosomes. In addition, chicken had interstitial telomeres on chromosomes 1, 2 and 3. Telomerase activity was highly detectable in early embryonic cells, germinal tissues and kidney cells. Whereas telomerase activity was gradually down-regulated when the organs, including brain, heart, and liver, were developed from embryos. In the distribution of telomeric DNA on the embryonic and developmental stages, most of the cells was gradually decreased in telomere quantity during ontogenesis.

β-용혈성 Streptococcus iniae 불활화백신의 넙치에 대한 효능 평가 (Evaluation on efficacy of β-hemolytic Streptococcus iniae vaccine on olive flounder, Paralichthys olivaceus)

  • Olive flounder, Paralichthys olivaceus is one of the most important cultured fish in Korea, its farming has been negatively impacted by viral, bacterial and parasitic diseases. Streptococcal infection was considered as a serious problem because of significant economic losses in olive flounder farm industry. The development and evaluation of vaccine for protection against infection by this agent were required. We evaluated the safety and efficacy of ${\beta}$-hemolytic Streptococcus (S.) iniae vaccine on olive flounder Three hundreds of flounders (weight $119.8{\pm}20.7g$, body length $22.6{\pm}1.4cm$) were reared in 0.5 tons aquaria in land-marine tank system. Seawater was provided from the sea of Inchon in Korea, and water temperature was set to $22^{\circ}C$ and $25^{\circ}C$ in the vaccination and challenge test, respectively. We used the formalin-inactivated ${\beta}$-hemolytic S. iniae (F2K) vaccine (M VAC INIAE; Kyoritsu seiyaku, Japan) originated in Japan. The vaccine was intraperitoneally administered to fish. Both of vaccinated group and control group were challenged with intraperitoneally injection by virulent S. iniae SI-36 isolates with $1.0{\times}10^7CFU/fish$ at 3 weeks after vaccination. Difference on mortality of control and vaccinated group (90.0 and 15.0%, 76.5 and 8.0% respectively) at two trials were found significant (p<0.05), and relative percent survival were 83.4% and 89.5%, respectively. The dead fishes were showed dark pigmentation of skin, abdominal extension, hemorrhagic ascites, and liver necrosis, and isolated the S. iniae strain from ascites, liver and kidney. We confirmed the safety and efficacy of ${\beta}$-hemolytic S. iniae vaccine by determinations of the optimal management condition and artificial challenge test in olive flounder.

Suppression of Fatty Acid Synthase by Dietary Polyunsaturated Fatty Acids is Mediated by Fat itself, not by Peroxidative Mechanism

  • This study examined the effect of dietary polyunsaturated fatty acids (PUFA) that were supplemented with vitamin E on lipid peroxidation, glutathione-dependent detoxifying enzyme system activity, and lipogenic fatty acid synthase (FAS) expression in rat liver. Male Sprague-Dawley rats were fed semipurified diets containing either 1% (w/w) corn oil or 10% each of beef tallow, corn oil, perilla oil, and fish oil for 4 wk. Alpha-tocopherol was supplemented in perilla oil (0.015%) and fish oil (0.019%). Hepatic thiobarbituric acid reactive substances, an estimate of lipid peroxidation, were not significantly different among the dietary groups. The glutathione peroxidase, glutathione reductase, and glutathione S-transferase activities were all elevated by the polyunsaturated fats, especially fish oil. The activity of FAS was reduced in the polyunsaturated fat-fed groups in the order of fish oil, perilla oil, and corn oil. The mRNA contents decreased in rats that were fed the 10% fat diets, particularly polyunsaturated fats, compared with the rats that were fed the 1% corn oil diet. Similarly, the inhibitory effect was the greatest in fish oil. These results suggest that lipid peroxidation can be minimized by vitamin E; PUFA in itself has a suppressive effect on lipogenic enzyme.

식이지방에 첨가한 Conjugated Linoleic Acid가 지방산 조성, delta-5 Desaturase(D5D)와 Fatty Acid Desaturase2(FADS2) 유전자 발현에 미치는 영향 (Effects of Conjugated Linoleic Acid Supplement on the Composition of Fatty Acids, Expressions of delta-5 Desaturase (D5D) and Fatty Acid Desaturase2 (FADS2) Genes in Mice)

  • 본 연구에서 4주령 ICR 수컷 마우스에 쇠기름과 어유에 단독 또는 0.5% CLA를 첨가한 식이로 4주동안 사육하여 혈장과 간의 지방산 조성과 DSD와 FADS2의 유전자 발현을 관찰한 결과는 다음과 같다. CLA는 C18:2와 경쟁적으로 지방산의 elongation과 desaturation에 참가하며, CLA의 첨가는 BTC군에서 C20:4수준을 높여주는 효과가 있었다. 또, 지방산의 ${\beta}$-산화에 의해 생성되기도 하는 E22:6은 BTC군에서 간의 C22:6의 함량이 BT군보다 증가하였다. DSD와 FADS2는 쇠기름섭취군(BT, BTC)에서 발현이 유의적으로 증가하였는데, 특히 D5D의 발현은 BTC군이 다른 군보다 유의적으로 증가하였다. 이에 대한 지방산 전환 또한 BTC 군이 C18:2로부터 C20:4로로 불포화하고 elongation하는 비율이 혈장과 간 모두에서 가장 높았다. 결론적으로 BT에 CLA를 첨가한 경우에 지방산 조성의 변화와 FADS2와 DSD의 발현을 증가시켜 지방산의 불포화도를 높여서 eicosanoids 합성과 항상성에 영향을 미치는 것으로 나타났으나 FO에 CLA첨가시에는 영향을 미치지 않는 것으로 나타났다.

이리도 바이러스 감염에 의한 양식 돌돔, Oplegnathus fasciatus의 대량폐사 (Mass Mortalities of Cultured Striped Beakperch, Oplegnathus fasciatus by Iridoviral Infection)

  • 1998년 8~10월에 남해안 일원 해상 가두리 양식장에서 사육중인 돌돔, Oplegnathus fasciatus이 60%이상 폐사하였다. 병어의 체색은 검어지거나 퇴색되며 아가미는 빈혈증상이 심하고 비장은 특이적으로 비대되어 있었고, 비장, 신장, 심장 및 간장 조직에서 퓰겐양성과 호염기성의 비대세포가 관찰되었다. 병어의 비장조직 마쇄액을 GF세포에 접종배양한 결과, 세포가 구형화 되면서 커지는 세포변성효과가 나타나 감염된 GF 세포로부터 바이러스가 분리되었다. 분리된 바이러스 배양액($10^4TCID_{50}$/0.1 ml)을 건강한 돌돔 치어에 복강주사하면 자연감염어와 유사한 증상을 나타내면서 폐사가 일어났고, 또한 병어의 비장조직을 전자현미경으로 관찰한 결과, 조직내 비대세포의 세포질에서 직경이 120~130 nm이며 외막이 있는 정 20면체의 바이러스 입자가 관찰되었다. 그리고 genebank로부터 얻은 참돔 이리도바이러스(RSIV)의 ATPase gene의 DNA염기서열을 주형으로 제작된 primer를 사용하여 PCR을 수행한 결과, 500 bp의 PCR 생성물이 병어나 인위감염된 GF세포에서 얻어졌는데, 이들 PCR 생성물은 RSIV의 ATPase cDNA gene과 95%의 상동성을 나타내었다. 따라서 양식돌돔에 대량 폐사를 일으킨 원인 바이러스는 참돔 이리도바이러스(RSIV)와 유사한 이리도바이러스로 밝혀졌다.

심내막염과 척추 주위 농양을 동반한 황색포도알균에 의한 출어 중의 선상 패혈증 사망: 증례 보고 (Unexpected Death by Sepsis of Staphylococcus aureus with Infective Endocarditis and Paravertebral Abscess in a Fisherman during Sailing out for Fish: An Autopsy Case)

  • Staphylococcus aureus is an important cause of human infections, and it is also a commensal that colonizes the nose, axillae, vagina, throat, or skin surfaces. S. aureus has increasingly been recognized as a cause of severe invasive illness, and individuals colonized with this pathogen are subsequently at increased risk of its infections. S. aureus infection is a major cause of skin, soft tissue, respiratory, bone, joint, and endovascular disorders, and staphylococcal bacteremia may cause abscess, endocarditis, pneumonia, metastatic infection, foreign body infection, or sepsis. The authors describe a case of a fisherman who died of sepsis on a fishing boat during sailing out for fish. The autopsy shows paravertebral abscess, pus in the pericardial sac, infective endocarditis with vegetation on the aortic valve cusp, myocarditis, pneumonia and nephritis with bacterial colonization, and also liver cirrhosis and multiple gastric ulcerations.
