Bioluminescence 반응에 의한 ATP 측정을 이용한 젖소 유방염 진단에 관한 연구 (Diagnostic test for bovine mastitis by the determination of ATP based on firefly bioluminescence)

  • This study was carried out to diagnostic test for bovine mastitis by the determination of adenosine triphosphate (ATP) based on firefly bioluminescence. The results obtained are follow; 1. The infection rate of bovine mastitis investigated with 521 cows in 47 dairy farms were found to be 3.6% of clinical form and 44.1% of subclinical form according to the degree of infection. 2. The light yield produced in firefly bioluminescence system was proportional to the concentration of ATP giving stright line within the range of 100PM~1uM. 3. When the number of somatic cell in milk was determined by the ATP assay and compared with three conventional methods such Fossomatic, California mastatic test (CMT), and rolling ball viscometer (RBV), it was shown that r=0.92 for Fossomatic, 0.63 for CMT and 0.7 for RBV. 4. The microorganisms causing mastitis were isolated Staphylococcus sp. (53.3%), Streptococcus sp. (17.9%), Micrococcus sp. (13.5%), Gram negative bacilli (6.3%), Gram positive bacilli (5.5%) and Yeast-like fungi (5.4%). 5. The endogeneous ATP levels of bacteria in a raw milk determined by the firefly bioluminescence system and compared with the results of the conventional methods. The correlation was 0.88 for raw milk.

Genomic Structure of the Luciferase Gene and Phylogenetic Analysis of the Firefly, Pyrocoelia rufa

  • We describe here the complete nucleotide sequence and the exon-intron structure of the luciferase gene of the firefly, Pyrocoelia rufa. The luciferase gene of the P. rufa firefly consisted of six introns and seven exons coding for 548 amino acid residues. From the translational start site to the end of last exon, however, the genomic DNA length of the P. rufa luciferase gene from the Korean and Chinese samples spans 1,968 bp and 1983 bp, respectively, and 3 amino acid residues were different to each other. Additionally, we also analyzed mitochondrial cytochrome oxidase I(COI) gene of the Chinese P. rufa fireflies. Analysis of DNA sequences from the mitochondrial COI protein-coding gene revealed 4 mitochondrial DNA sequence-based haplotypes with a maximum divergence of 0.7%. With the 20 P. rufa haplotypes found in Korea, phylogenetic analyses using PAUP and PHYLIP subdivided the P. rufa into three clades, termed clades A and B for the Korean sample, and clade C for the Chinese sample.

  • The factors affecting the shear strength of the angle shear connectors in the steel-concrete composite beams can play an important role to estimate the efficacy of a composite beam. Therefore, the current study has aimed to verify the output of shear capacity of angle shear connector according to the input provided by Support Vector Machine (SVM) coupled with Firefly Algorithm (FFA). SVM parameters have been optimized through the use of FFA, while genetic programming (GP) and artificial neural networks (ANN) have been applied to estimate and predict the SVM-FFA models' results. Following these results, GP and ANN have been applied to develop the prediction accuracy and generalization capability of SVM-FFA. Therefore, SVM-FFA could be performed as a novel model with predictive strategy in the shear capacity estimation of angle shear connectors. According to the results, the Firefly algorithm has produced a generalized performance and be learnt faster than the conventional learning algorithms.

  • 본 논문에서는 최근 Xin-She Yang에 의해 소개된 반딧불이 알고리즘(FA)에 휴리스틱을 적용하여 개선하는 방안을 제안한다. 또한 이를 위하여 기존의 FA를 이와 유사한 문제영역의 알고리즘인 Particle Swarm Optimization(PSO)와 정확도 측면, 수렴 시간 측면, 각 입자의 움직임 측면에서 비교 분석한다. 비교 실험 결과, FA의 정확도는 PSO보다 나쁘지 않았지만, 수렴 속도는 느린 것으로 나타났다. 본 논문은 이에 대한 직관적인 원인을 고찰하고, 이를 극복하기 위해, 기존의 FA에 부분 돌연변이 휴리스틱을 적용하여 개선된 FA(Improved FA)를 제안한다. 벤치마크 함수들을 최적화 하는 비교 실험 결과, 개선된 FA가 PSO와 기존의 FA보다 정확도와 수렴속도 측면에서 우수함을 보이고자 한다.

  • The Wireless Sensor Network (WSN), is constructed out of teeny-tiny sensor nodes that are very low-cost, have a low impact on the environment in terms of the amount of power they consume, and are able to successfully transmit data to the base station. The primary challenges that are presented by WSN are those that are posed by the distance between nodes, the amount of energy that is consumed, and the delay in time. The sensor node's source of power supply is a battery, and this particular battery is not capable of being recharged. In this scenario, the amount of energy that is consumed rises in direct proportion to the distance that separates the nodes. Here, we present a Hybrid Firefly Glow-Worm Swarm Optimization (HF-GSO) guided routing strategy for preserving WSNs' low power footprint. An efficient fitness function based on firefly optimization is used to select the Cluster Head (CH) in this procedure. It aids in minimising power consumption and the occurrence of dead sensor nodes. After a cluster head (CH) has been chosen, the Glow-Worm Swarm Optimization (GSO) algorithm is used to figure out the best path for sending data to the sink node. Power consumption, throughput, packet delivery ratio, and network lifetime are just some of the metrics measured and compared between the proposed method and methods that are conceptually similar to those already in use. Simulation results showed that the proposed method significantly reduced energy consumption compared to the state-of-the-art methods, while simultaneously increasing the number of functioning sensor nodes by 2.4%. Proposed method produces superior outcomes compared to alternative optimization-based methods.

Molecular Cloning of a Defensin Homologue Gene of a Novel Family Member from the Firefly, Pyrocoelia rufa

  • A cDNA encoding the defensin homologue of a novel family member was isolated from the cDNA library of the firefly,Pyrocoelia rufa. Sequence analysis of the cDNA encoding the defensin homologue of P. rufa resulted that the 165 bp cDHA has an open reading frame of 55 amino acid residues. The deduced amino acid sequences of the defensin homologue gene from P. rufa showed identity to known mammalian defensins. Also 6 cystein residues in the P. rufa defensin homologue gene were conserved in the same position as those of known mammalian defensins. The result suggested that P. rufa defensin homologue is a novel member of the insect defensin family. Southern blot analysis suggests that there may be a single copy number of the P.rufa defensin homologue gene and their fat body-specific expression pattern at the transcriptional level was confirmed by Northern blot analysis.

Quick Detection of Firefly Luciferase Gene Expression in Live Developing Bovine Embryos by Photoncounting

  • The present study was designed, fIrst to develop the new methodology to measure the bioluminescence activity easily in live developing bovine embryos by photoncounting, and secondly to compare the expression efficiency of four luciferase reporter genes in bovine embryos at four- to 16-cell stages. In experiment 1, equimolar pSVlacZ and pSVEluc were microinjected into the pronucleus of fertilized bovine oocytes. At 2 days after micro injection, bioluminescence activity of these embryos was measured by photoncounting with a luminometer for 1 min, and lacZ gene expression in the same embryos was assayed by X-gal staining. All the luciferase-positive oocytes showed some bacterial ${\beta}$-galactosidase activity irrespective of the intensity. In experiment 2, four firefly luciferase genes (pTKEluc, pTK6WEluc, pSVEluc and pMiwluc) were introduced by micro injection, and the injected embryos were cultured for the following 2 days. Detection of the luciferase gene expression was done by photoncounting at 5 to 55 min. Over the measurement period, the luciferase activity was almost constant irrespective of the transgenes microinjected. The luciferase activity and expression efficiency at 2 days after microinjection were not significantly affected by the difference in the microinjected transgenes. The present results demonstrated that the bioluminescence activity in live developing bovine embryos could be measured quickly by photoncounting.

Developmental Characteristics and Life History of the Korean Native Firefly, Pyrocoelia rufa

  • Development and life history of the Korean native firefly, Pyrocoelia rufa, were investigated throughout the indoor rearing. Average size of egg with an oval shape was 1.7 m and the hatchability of egg of P. rufa was approximately 88.5%. The larvae were pupated at the ${5^th}$ instar and the body length of the matured larvae was 27.1 mm. The total periods of larval stage to the end of the 5$^{th}$ instar were approximately 104.7 days. Average pupal period was 10.3 days and average number of eggs oviposited by a female was 87.3 eggs. The body size of female in the pupa and adult was larger than that of male. The wings of female adult were deteriorated.

  • Modern power systems driven by high-power converters have become inevitable in view of the ever increasing demand for electric power. The total power loss can be reduced by limiting the switching losses in such power converters; increased power efficiency can thus be achieved. A reduced switching frequency that is less than a few hundreds of hertz is applied to power converters that produce output waveforms with high distortion. Selective harmonic elimination pulse width modulation (SHEPWM) is an optimized low switching frequency pulse width modulation method that is based on offline estimation. This method can pre-program the harmonic profile of the output waveform over a range of modulation indices to eliminate low-order harmonics. In this paper, a SHEPWM scheme for three-phase three-leg neutral point clamped inverter is proposed. Aside from eliminating the selected harmonics, the DC capacitor voltages at the DC bus are also balanced because of the symmetrical pulse pattern over a quarter cycle of the period. The technique utilized in the estimation of switching angles involves the firefly algorithm (FA). Compared with other techniques, FA is more robust and entails less computation time. Simulation in the MATLAB/SIMULINK environment and experimental verification in the very large scale integration platform with Spartan 6A DSP are performed to prove the validity of the proposed technique.

Artificial Habitat Creation of Luciola lateralis (Coleoptera: Lamphyridae) and Research of Breeding Technique for Festival at Hwadamsup, Korea

  • This study created the running water (Bandibooliwon) and lentic water (Sooryunwon) artificial habitats of Luciola lateralis in Hwadamsup, Gwangju-si, Gyunggi-do from April 2014 to February 2016. Through various habitat types, metapopulation was formed, and more sound population was induced to be maintained via interactions between habitats. We investigated coenosite at the habitat creation-target site, and 20 species in the lentic water site and 38 species in running water site were found. After releasing L. lateralis, we checked the status of settlement of L. lateralis larvae through traps and macroscopy, controlled the flow velocity of the habitats in line with the life cycle of L. lateralis (hatching, larva period, landing, pupation, copulation, and oviposition), and shaped gentle slope at water side, and planted moss. We artificially supplied melanian snails (Semisulcospira sp.), the food source, in the running water site and induced stable feeding. Also, We created artificial insectary, bred larvae stably in winter, and secured firefly population for a festival. Through night time observation, the time most suitable for a festival was confirmed to be from 20:30 to 22:00.
