Spermatogenesis of Black Molly and Sailfin Molly (Poeciliidae, Teleostei) (경골어류 난태생 송사리과에 속하는 black molly와 sailfin molly의 정자형성과정)

  • Black molly (Poecilia sphenops) and sailfin molly (Poecilia latipinna) are a teleost belonging to Poeciliidae. The spermatogenesis between two species were investigated by light and electron microscope. The whitish testes of both black molly and sailfin molly were located between intestine and air bladder. The size of testis was major axis 7 mm, minor axis 2 mm. The testis contained numerous testicular cysts. In both black molly and sailfin, primary spermatocytes were comparatively large ellipsoidal, and mitochondria showed a marked development. The secondary spermatocyte was smaller than that of primary spermatocyte, highly condensed according to their development. The nucleus with electron-dense was round shape and flagella started to be formed. In spermiogenesis, chromatin was more condensed. The mitochondria were rearranged along the tail. The number of mitochondria was 2 to 4 in cross section and 8 to 10 in longitudinal section. The head of mature sperm was long cone shape and had not acrosome. The microtubules of flagella were arranged 9+2 structure. Also, the tail of sperm have lateral fins. In conclusion, spermatogenesis and sperm morphologies of these two species were same. These morphological similarity seems to be an indication of the Poeciliidae.

Soybeans Fermented with Bacillus po/yfermenticus KJS-2 Protects Oplegnathus fasciatus from Iridovirus and Pathogenic Bacterial Infection (Bacillus polyfermenticus KJS-2에서 발효된 콩의 돌돔에 대한 이리도바이러스 및 병원성균에 대한 예방효과)

  • Lyophilized powder (BP2FS) of soybeans fermented with Bacillus polyfermenticus KJS-2 (B. polyfermenticus KJS-2) exhibited in vitro antibacterial activities against eight pathogenic bacteria. BP2FS was used as a fodder additive for Oplegnathus fasciatus (0. fasciatus) culture. One group (UFD) of O. fasciatus was fed a commercial fodder, while another group (FD) was fed the same fodder, but including BP2FS ($6{\times}10^{4}$ cfu $g^{-1}$ fodder), two times daily for 120 days. The mean body weight of the FD group (67.29${\pm}$12.62 g) was higher than that of the UFD group (56.56${\pm}$8.21 g) after 120 days. The survival rate of FD was 80% compared to 40% for the UFD group. Cumulative mortalities in the FD and UFD groups were 18.95% and 60.98% respectively. B. polyfermenticus KJS-2 was isolated from the intestines of the FD group and the number of viable colonies was estimated to be $1.04{\times}10^{4}$ cfu $g^{-1}$. Iridovirus and Vibrio vulnificus was detected in the organs of the UFD group but not in the FD group. All of the infected fish showed typical clinical symptoms of hemorrhage in their tail fins. Dissection of the infected internal organs revealed liver congestion and spleen enlargement - typical symptoms caused by iridovirus infection. These results clearly show that BP2FS is highly beneficial in preventing O. fasciatus from iridovirus infection.

Gill Disease of Pseudobagrus fulvidraco Fingerlings by Deficiency of Pantothenic acid (Pantotheic acid 결핍에 의한 동자개(Pseudobagrus fulvidraco)치어의 사료성 아기미병)

  • A new nutritional disease has occurred among the hatchery-reared Korean bullhead fingerlings (Pseudobagrus fulvidraco) in the Chonbuk Province in September 1997. Diseased fish were all dead within 3-7 days, showing sluggish behavior, head up and tail down swimming. Most characteristic clinical signs were anaemia, clubbed and fused gill, skin desquamation. haemorrhage around the mouth and at the base of pectoral fins. Any causative bacteria and parasites were not isolated from the lesions and internal organs of the diseased fish. The hepatosomatic index, red blood cell count, hematocrit, hemoglobin and erythrocytes size of peripheral blood in the diseased fish were remarkably decreased compared with those of normal fish. In the histopathological observations, epithelial hyperplasia of the gill filaments initiated at the base of the gill was pronounced. This symptom was the characteristic appearence of all the diseased fish. A 0.6% saline bath and feeding a pantothenic acid-supplemented diet were conducted to decrease the mortality. Ten days after 0.6% saline bath or 25 days after feeding a pantothenic acid supplemented diet resulted in decreasing in the mortality. Microscopic appereance of the gill from the recovered fish was similar to that of the gill from healthy fish. These results indicate that the disease was caused by deficency of pantothenic acid in their diet and that 0.6% saline bath or supplementation of pantothenic acid in the diet was an effective way to decrease the mortality.

Wet Surface Air-Side Performance of Fin-and-Tube Heat Exchangers Having Sine Wave Fins and Oval Tubes (사인 웨이브 핀과 타원관으로 구성된 핀-관 열교환기의 공기측 습표면 성능)

  • Experiments were conducted on sine wave fin-and-tube heat exchangers having oval tubes under wet condition. Oval tubes having an aspect ratio of 0.6 were made, by deforming 12.7mm round tubes. Twelve samples, having different fin pitch and tube row, were tested. Results showed that, for oval tube samples, the effect of fin pitch on j and f factor was not significant. As for the effect of tube row, the lowest j factor was obtained for one row configuration(81% of two row configuration), which is clear contrast to round tube samples, where the highest j factor was obtained for one row configuration. Possible reasoning is provided considering the flow and heat transfer characteristics of sine wave channel combined with connecting oval tubes. Comparison of $j/f^{1/3}$ with plain fin-and-tube heat exchanger having 15.9mm O.D. round tube reveals that present oval fin-and-tube heat exchanger shows superior thermal performance except for one row configuration. In other words, $j/f^{1/3}$ of the two row oval tube heat exchanger was 1.6~2.5 times larger than those of round tube heat exchanger, 1.4~2.4 times larger for three row configuration and 1.2~2.8 times for four row configuration.

Air-side Heat Transfer and Friction Characteristics of Fined-tube Heat Exchangers under Heating Condition (핀-관 열교환기의 난방운전 시 공기측 열전달 및 마찰특성)

  • An experimental study has been performed to investigate the characteristics of air-side heat transfer and friction of a fined tube heat exchanger under heating conditions. Air enthalpy calorimeter was used to obtain the performance evaluation and analysis of the fined tube heat exchanger. Eight finned tube heat exchangers with slit fin, louver fin, and plain fin were used. The air-side heat transfer coefficient was calculated by the log-mean-temperature-difference. Air-side heat transfer and friction were presented in terms of j factor and friction factor on Reynolds number. From the experimental result, it was found that the variations of air-side heat transfer and friction of fined tube heat exchanger with the change of the fin configuration, row number, fin pitch, and tube circuit were obtained. j factor and friction factor decreased with Reynolds number increased. The tube circuit affected the air-side heat transfer and friction. In the case of slit and louver fin, j factor of 1st row was higher than that of 2nd row. But, with increasing Re, j factor was reversed. The characteristics of j factor and friction factor of 2nd row heat exchanger were different according to the kind of fins.

Chilodonella hexasticha(Protozoa, Ciliata) from Korean freshwater fish (한국산 담수 어류에 기생하는 Chilodonella hexaticha에 관하여)

  • During a study to document more fully the parasitic ciliates of Korean freshwater fishes, we found numerous small ciliates in the gills and skin of crucian carp, Carassius carassius. Cursory examination indicated that the ciliate were very similar with Chilodonella sp. In this study, we described the morphometrics of this ciliate in detail and compared with other species of Chilodonella in the world. Numerous small ciliates were observed in the body surface, fins and gills of infected fish and excessive mucus production were seen in those fish. Sometimes ulcer was observed in the body of moribund fish. From the scrutinized observation of the parasite, it was identified as Chilodonella hexasticha. The parasite was dorsoventrally flattened body, without a notch in the posterior margin, and it measured 30-$45{\mu}m$ long and 15-$30{\mu}m$ wide. In number of kineties, the right band ciliature was 3-5 and the left band ciliature was 3-5. The right ventral ciliary band shaped arch and was longer than the left, straight band. It had a single oval macronucleus, 8-$15{\mu}m$ in diameter, a single micronucleus, 2-$4{\mu}m$ in diameter. The cytopharynx was reinforced by 10-16 conspicuous nematodesmata, which shaped like a tube and curved at its inner end. Two contractile vacuoles, one anteriorly at right and the second posteriorly at left were observed in wet mounts. This parasite reproduced by a simple division at $22{\pm}1^{\circ}C$.

Biological characterization of Tenacibaculum maritimum isolated from cultured olive flounder in Korea and sensitivity against native plant extracts (한국의 양식넙치에서 분리한 Tenacibaculum maritimum의 특성과 자생식물 추출물에 대한 감수성)

  • Tenacibaculum maritimum (formerly Flexibacter maritimus) is the aetiological agent of an ulcerative and necrotic disease commonly called tenacibaculosis in marine fish. Tenacibaculosis is an economically important disease in a great variety species in Jeju Island cultured fish and leading to this pathogen initially affected by skin, mouth, fins, tail causing severe necrotic and ulcerative lesions on the body surface. In the present study, A-7 strain was isolated from Paralichthys olivaceus showing symptoms of tenacibaculosis and identified as T. maritimum by morphological, biochemical and molecular biological analysis. T. maritimum A-7 is experimentally infected through immersion route in Paralichthys olivaceus which the disease outbreaks in land-based fish tanks of Jeju Island. Up to data a number of treatments proposed for the tenacibaculosis outbreaks are based on the immersion administration of drugs in tank. Oxytetracycline is the most widely used disinfectants in fish farms. However, most of fish farms manager and consumers have expressed concern as bioaccumulation in tissue and its environmental. In addition, this antimicrobial compounds is expensive in fish farmers. The overcome of this problem is desired the application of natural plant derived products. To obtain as 70% EtOH extract antimicrobial compounds against tenacibaculosis from 35 species of Jeju Island native plants were screened for antimicrobial activity against T. maritimum. In the present study were identified most of the plant extracts were better antimicrobial activity against T. maritimum.

Lipid Class and Fatty Acid Composition of Muscle of Common Squid Todarodes pacificus (살오징어 (Todarodes pacificus)의 근육부위별 지질 Class 및 지방산 조성)

  • The chemical components, lipid class, and fatty acid composition of muscle from male and female common squid, Todarodes pacificus, were examined to evaluate the potential utilization of muscle from fin, head, arms, and tentacles, which consumers usually like less than the mantle. The mantle was found to constitute 47-49% of the total muscle and the proportion was slightly higher in females than in males. For the remaining 51-53% of the muscle, the only gender difference was that the arms of males contained approximately 3% more muscle than those of females (P<0.05). The protein content was higher in the mantle, arms, and tentacles than in the fin and head in both males and females (P<0.05), and was slightly higher in males (15.7-20.7%) than in females (15.1-19.2%). By contrast, the lipid content was slightly higher in females (1.82-2.54%) than in males (1.01-2.37%), and the fins in both males and females contained the most lipids (2.37-2.54%) of all muscle. The prominent lipid classes in the muscles were free sterol (males 81.5-91.9% vs. females 84.9-91.8% for the non-polar lipid content), phosphatidylcholine (PC, males 59.3-62.4% vs. females 49.2-57.8% for the phospholipid content) and phosphatidylethanolamine (PE, males 22.0-28.8% vs. females 25.6-33.8% for the phospholipid content). The percentage of PC was approximately 5-10% higher in males (P<0.05), especially in the fin, while that of PE was approximately 3-5% higher in female (P<0.05), especially in the head. All of the squid muscle contained 52.1-54.9% of n-3 polyunsaturated fatty acids (PUFA), such as docosahexaenoic acid (DHA) and eicosapentaenoic acid (EPA). Males contained slightly more DHA, whereas female contained more EPA. The total percentage of n-3 PUFA differed little among muscles within the same gender.

ON THE MORPHOLOGY OF LARVAL AND YOUNG STAGES OF CHAMICHTHYS DOLICHOGNATHUS HILGENDORF (점망둑 Chasmichthys dolichognathus HILGENDORF의 자치어기의 형태)

  • Morphological changes of early post-larval and young stages of Chasmichthys dolichognathus HILGEnDORF (Family Gobiidae) have been studied based on the samples of 953 individuals collected in June 1968, July 1969 and July 1971. Particular emphasis was paid on the development of fin rays, chromatophore patterns and ventral fins. The primordial fin rays of the first dorsal fin appear in the post-larvae of around 8.0 mm in total length, and dorsal fin fully develops in the larvae of around 9.2 mm. In the early young stages of 17.0 mm in total length fin rays have completely developed. According to chromatophore patterns the larvae are grouped into three successive groups. The larvae at the early stages of 6.3-14.2 mm have melanophores on the whole dorsal surface, the posterior ventro-lateral part of the tail and the basal part of the caudal fin. In the later larval stages of 17.0-24.4 mm a group of melanophores are added on medio-lateral part of the tail. These melanophores extend anteriorly and eventually cover the medio-lateral part of the whole body. In the early young stages of 97.2-34.8 mm the chromatophores cover the whole body surface in cloudy and H-shaped patterns. The chromatophore patterns of this stage are distinctive as generic characters of the fish. Fin membranes of the ventral fin appear in the post-larval stage (ca. 7.4 mm), and the primordial fin rays develop in the late post-larval stages (ca. 14.2 mm). The fin rays develop into a complete sucker in the young fish stage of around 30.0 mm in total length.

Ontogenetic Color Variation of Abudefduf notatus (Pomacentridae: Perciformes) Revealed by 16S rRNA Sequences Analysis (미토콘드리아 16S rRNA 염기서열 분석에 의해 밝혀진 동갈자돔 치어의 성장에 따른 체색변이)

  • Seven individuals (16.1~29.1 mm in length) which were estimated with fishes of genus Abudefduf were collected in Seogwipo, Jeju Island in the summer of 2011 and 2012. Among them, five individuals (20.8~29.1 mm SL) are similar to Abudefduf notatus, based on morphological characters such as yellow transverse band on the body, one small black spot on the base of the pectoral fin, and yellow caudal fin. On the other hand, two individuals (16.1 mm, 17.0 mm SL) differ from them in several light transverse bands and transparent rays of all fins except for pelvic fin, and anterior transparent head with only one (16.1 mm). According to the results of molecular analyses of 578 base pairs of mtDNA 16S rRNA sequences, these individuals correspond to A. notatus adult, with a high bootstrap value of 95% (genetic distance, d=0.002). Therefore, this study shows that the individuals more than 20.8 mm similar to adult body color but the 16.1 mm individual differs to that of adult. We confirm that this species changes to body color during their early life stages. This result regards as a survival strategy to protect themselves against their predator during their early life stages.
