과발현 형질전환벼에서 CCCH type zinc-finger protein 유전자 OsZF2 기능 분석 (Functional characterization of a CCCH type zinc-finger protein gene OsZF2 by ectopic overexpression of the gene in rice)

  • 벼의 저온처리 cDNA 은행에서 분리된 CCCH 형태 zinc finger 단백질인 OsZF2의 기능을 분석하기 위하여 벼에서 CaMV 35S 프로모터 조절하에 OsZF2가 발현(35S:OsZF2)되는 형질전환벼 식물체를 개발하였다. 35S:OsZF2 형질전환벼에 대한 하이그로 마이신저항성 검정을 통해 동형접합체 계통을 선발하고 Northern 발현분석에 의해 OsZF2 유전자가 형질전환체에서 과발현되는 것을 확인하였다. 형질전환체와 대조구인 낙동벼를 100 mM NaCl 첨가 MS 배지에서 키운 후 잎과 뿌리의 길이를 측정하여 내염성 검정을 수행한 결과 대조구에 비해 형질전환체 생육이 다소 양호 한 것으로 나타났다. GMO 포장에서 생육상태를 관찰 한 결과 형질전환체는 생육지연으로 인한 왜화 현상을 나타내며 출수기 또한 열흘 정도 지연되나 등숙기에는 대조구와 같은 초장을 보였다. zinc finger 유전자는 식물체의 발달과 분화 단계 및 환경 스트레스 반응 등 중요한 역할을 하는 것으로 알려져 있으므로 유전체발현 분석으로 하위단계에서 조절되는 유전자 발현 양상을 분석하였다. 35S:OsZF2 전환체에서 낙동벼보다 4배 이상 발현이 증가된 유전자 중에서 게놈 주석에 기초한 기능을 유추하면 신호전달과 관련된 protein kinase, DNA 결합단백질과 대사에 관련된 효소 유전자, 스트레스 반응에 관여하는 일부 유전자 및 병 저항성과 관련된 유전자들의 발현이 증가되었다. 따라서 벼에서 분리된 OsZF2 CCCH type zinc finger 유전자는 벼 성장 발달과 스트레스에 반응하는 상위 조절자로서 기능을 할 것으로 추측된다.

손기능과 손의 특성과의 관계 (The Relationships between Hand Function and Characteristics of Hand)

  • Purpose. This study was to investigate the relationship between hand function and characteristics of hand. Methods. Participants were 56 occupational therapy students who studied in M colleges in Gyengsangnam-do, april 20, 2015 from 1 May, and hand circumference and length (vertical, w), the width was measured. A hand grip on the measurement subject was measured by the Grip dynamometer and Pinch gauge. Results. The results was hand length, width, circumference long, broad, thick, and hand strength was a greater correlation emerged. Grip strength was showed a significant correlation to the size and grip strength was also correlated. Conclusion. This study was correlation of length of the hand, a length wide and strength of the finger circumference.

비행장갑설계를 위한 사용실태분석 및 개선안 개발 (Usage Analysis and Design Development for Pilot Gloves)

  • This study was conducted to analyze the wearing condition on gloves for pilots and to suggest the developments for pilot gloves design. This survey was performed with 99 pilots and the questionnaires included items about discomfort, materials and problems when wearing pilot gloves. The respondents felt uncomfortable on the finger tip, the hollow area of the hand and the end of thumb when they move their hands with the gloves. They replied that the finger length, palm length, and circumference of the fingers did not match well. Also they complained there was severe abrasion in the tip of the thumb, finger tips and the middle of the palm of the gloves. The result of these analyses supports the following suggestions. It is necessary to improve durability by applying the additional materials to the severely worn parts. Punching on the glove materials could improve sweat rate and ventilation, and careful washing should be accompanied to prevent the occurrence of linters and transformation after laundering. And it is needed to prepare an accurate and more diversified size system and to design the gloves fitted to the individual hands. With these in mind, it is required to propose solutions for pilot gloves with reference to the survey results in order to design appropriate pilot gloves in terms of movement, size and material. It was concluded that an analysis of the wearing condition and developments are useful for ergonomics pilot gloves design.

Functional Analysis of Pepper Cys2/His-Type Zinc-Finger Protein Promoter Region in Response to Bacterial Infection and Abiotic Stresses in Tobacco Using Agrobacterium-Mediated Transient Assay

  • The promoter region flanking the 5’ CAZFP1 coding region was isolated from the genomic DNA of Capsicum annuum. To identify the upstream region of the CAZFP1 gene required for promoter activity, a series of CAZFP1 promoter deletion derivatives was created. Each deletion construct was analyzed by Agrobacterium-mediated transient transformation in tobacco leaves after infection by Pseudomonas syringae pv. tabaci, or treatment with methyl jasmonate (MeJA), ethylene, abscisic acid (ABA), salicylic acid (SA), cold and wounding. Promoter fragments of 685 bp or longer showed 7-fold or greater induction after P. s. pv. tabaci infection and MeJA treatment. The CAZFP1 full-length promoter (-999 bp) also showed 6-fold induction in response to ethylene. The transiently transformed tobacco leaves with the CAZFP1 full length promoter fused-GUS gene showed more than 5-fold induction in response to SA, ABA and cold. These results suggest that the CAZFP1 promoter contains responsive elements for pathogen, MeJA, ethylene, SA, ABA and cold.

손톱위 피부피판을 이용한 손톱형태 복원의 치험례 (Restoration of the Injured Fingertip with Eponychial Cutaneous Flap)

  • The restoration of the fingernail is not simple in case of amputated distal fingertip which involved the pulp and nail. The nail should maintain a length of at least 2 mm from the eponychium for an adequate grip and decent appearance. Various methods to reconstruct the fingernail bed are available. The nail bed graft from amputated finger or great toe, and free onychocutaneous flap are commonly used. The nail bed of the injured tip tends to be atrophied, deformed and failed as a graft. And the great toe is often turned down as a donor. We have restored satisfactorily the nail beds of three injured finger tips with eponychial cutaneous flaps. The pulps were reconstructed with either a reverse dorsal digital island flap or free pulp graft. Repeated again. A mean follow- up was six months. The nail grew up to the average of 3.7 mm. All patients were satisfied with the length of the nail and met with good cosmetic results. An eponychial cutaneous flap is useful to restore the nail of the distal fingertip amputation. The procedure is relatively simple and morbidity is minimum.

한국인 손 치수의 추세 변동 및 외국 자료와의 비교 (Trend of hand dimensions for Koreans and Comparison with foreigners' data)

  • The purposes of this are (1) to present representative five hand dimensions by measurement year, (2) to investigate hand characteristics based on relative ratios of varying hand dimensions and (3) to compare Korean hand dimensions to other countries' data. Comparing to those of 1979, statures of 2004 increased by 3.0~3.5%, while males' hand dimensions of hand length, medius finger length, hand circumference, hand breadth and hand thickness decreased by 2.0~5.0%. Ratios of hand dimensions to those of medius finger were not significantly different depending upon age group and gender, but were significantly different depending upon races of Korean and American. Hand lengths of Koreans were shorter by about 4.0% than those of Japanese irrespective of gender, while the other compared dimensions for Koreans were larger than those of Japanese. Hand dimensions used in this study of Koreans were generally smaller that those of Americans. It is recommended from this study that when designing hand relevant things including glove, computer keyboard, smart phone key pad, etc., the different hand characteristics found in this study be fully reflected to develop competitive products.

인간-기계 인터페이스를 위한 근전도 기반의 실시간 손가락부 힘 추정 (EMG-based Real-time Finger Force Estimation for Human-Machine Interaction)

  • In this paper, we describe finger force estimation from surface electromyogram (sEMG) data for intuitive and delicate force control of robotic devices such as exoskeletons and robotic prostheses. Four myoelectric sites on the skin were found to offer favorable sEMG recording conditions. An artificial neural network (ANN) was implemented to map the sEMG to the force, and its structure was optimized to avoid both under- and over-fitting problems. The resulting network was tested using recorded sEMG signals from the selected myoelectric sites of three subjects in real-time. In addition, we discussed performance of force estimation results related to the length of the muscles. This work may prove useful in relaying natural and delicate commands to artificial devices that may be attached to the human body or deployed remotely.

Lycopersicon Eculentum C2H2-type Zinc Finger Protein Induced by Oxidative Stress Especially

  • A tomato zinc-finger protein gene, LeZFP1, encoding the Cys2/His2-type zinc-finger transcription factor was searched from cDNA microarray analysis of gene expression following induction of the overexpressed tomato transgenic plants showing resistance for pathogen and abiotic stresses. The full-length cDNA of LeZFP1 encoded a protein of 261 amino acid residues. Analysis of the deduced amino acid sequence of LeZFP1 revealed that it shares high sequence identity with pepper CAZFP1 (81% identity). We found that single copy of LeZFP1 gene is present in the tomato genome through southern blot analysis. The LeZFP1 transcripts were constitutively expressed in the tomato mature and young leaves, but were detectable weakly in the flower, stem and root. The LeZFP1 transcripts were significantly reduced in treated leaf tissues with NaCl and mannitol. The LeZFP1 gene was induced by oxidative stress especially. Our results indicated that LeZFP1 may play a role function involved in oxidative stress signaling pathways.

소아맥진(小兒脈診)의 특성(特性)에 대한 소고(小考) (A study on The Characteristic of Traditional Pediatric Pulse Diagnosis)

  • Objective : The region, observation period, figuration of pulse, pulse frequency, process of diagnosis and related diagnostic methods in traditional pediatric pulse diagnosis were studied in this article. Method : The articles related to traditional pediatric pulse diagnosis in Traditional medical classics, and analysis to the characteristics of it was done, then lastly, the principle of pulse manifestation and the meaning of application to modern times were considered. Result & Conclusion : It is difficult to catch the figuration of pediatric pulse because of dynamic change of body conditions, fast heartbeat and narrow region of pulse. Therefore, the pulse diagnosis appropriated for children have been developed and combined with the special diagnosis methods, for example, palpation of forehead and observation of index finger. The traditional pediatric pulse diagnosis is done with just one finger palpation in the period of 3 to 5 years old to identify the speed, length, size and height of pulse. The standard average of pulse frequency is 7~8 par breath in the period of 3 to 5 years old according to traditional medical classic.

유리 반전 전완피판술을 이용한 수지부 결손의 치료 (Reconstruction of the Finger Defect with Free Vascularized Reversed Radial Forearm Flap)

  • Radial forearm flap is one of the most useful skin flap in hand reconstructuion with distally based reverse pedicled or free vascularized fashion. Athors modified that flap into reverse pedicled and free vascularized flap which has advantages of both methods. The modification composed with harvesting flap on recipient side distal forearm just as free flap, than apply it as reverse distal pedicled flap fashion with microvascular anastomosis with distal vascular stump of donor radial vessels. We underwent this method in 5 cases in finger reconstruction from 1996, all of the cases had sucessful results. The advantages of this method are: 1. Thin flap which is compatible to finger skin can harvest from distal forearm with very long vascular pedicle that can be passed under the subcutaneous tunnel which avoid additional skin incisions on the hand. 2. The vessels of donor site and recipient site are same vessel in effected side of forearm, which can preserve contralateral side forearm and hand keep intact. 3. The flap can cover the defects on distal portion of the fingers which is difficult in conventional reversed radial forearm pedicled flap because of limited mobilization of flap due to limitation of pedicle length reach to tip of the fingers.
