  • A comparative study of free amino acid content in healthy and virus diseased tobacco leaves was carried out by author throughout the gorwing season from June to November of 1963. The methods of qualitative analysis of free amino acids applied in this experiment is followed by Moore and Stein. 1,2 Free amino acids determined in this experiment are shown in Fig. Ⅰ, Ⅱ and Table Ⅰ. As the figure and the table are shown, four more amino acids such as a spartic acid, glutamic acid, tyrosine and phenylalanine are detected in the healthy leaves; these four additional amino acids in the healthy leaves are conspicuous. More quantities of asparagine and alanine are detected in the diseased leaves than the healthy leaves and more quantities of tryptophan is detected in the healthy leaves. It is presumed that such amino acids as tyrosine and phenyllanine are decreased by the incooperation of free amino acid to TMV protein in the process of the process of the leaf protein metabolism which is caused by TMV-RNA trapping action in the diseased leaf protoplasm. It is thought that the decrease of asparagine and the increase of asparic acid in the healthy leaves are the results of in incooperaton of NH2, produced by the protein dissimilation in the diseased leaves, to aspartic acid; it's reaction is caused by the respiration of the diseased leaves accelerated by TMV attack. It is presumed, consequently, that the check of the diseased tobacco leave growth is influenced by the reduction of such amino acids as tryptophane and glutamic acid, which reduction may be due to the abnormal protein metabolism and the action of certain enzyme caused by TMV attack on host protoplast.

Fundamental study on the evapo-transpiration requirements of paddy rice plant (수도용수량계획상의 엽면증발량 및 주간수면증발량에 관한 기초적인 연구)

  • The purpose of this study is to find out the reasonable amount of evapo-transpiration required for the paddy rice plant during the whole growing season. So, on the basis of the 3year experimental data concerning the evapo-transpiration from 1966 to 1968, the author obtained the following results. 1) The leaf area index in the densely planted plot is generally higher than that in the conventionally planted one during the first half of growing season. So, the coefficient of transpiration in the former plot is some what higher than in the latter, and the coefficient of water surface evaporation under the plant cover has the inverse relation between both plots. 2) It is unreasonable that coefficient of evapo-transpiration is applied to the calculation of the evapo-transpiration requirments of each growing stage, because a certain degree of variation in meteorological factors and in the thickness of the plant growth is involved in it. 3) It is most reasonable that the rate of transpiration and of the water surface evaporation is applied to the calculation of the transpirated amount and evaporated one in each growing stage because it shows almost constant value in spite of any meteorological conditions in so far as the variety of rice, planted density and control of applying fertilizer are same and the disease and blight are negligible. 4) The ratio of the amount of transpiration to the weight of the whole air dried yields has the tendency of decreasing as that of the yields increase, having almost constant value despite the amount of pan evaporation; and the value is about 210 when the weight of root parts is included to that of the yields. 5) Although the required amount of transpiration during the whole growing season can be calculated with the above ratio, fig. 7 showing the relation between the amount of transpiration and the weight of the yields is more reasonable and will be convinient to find it. And the requirements of water surface evaporation during the same season can also be directly found with the weight of air dried straw refering to fig.8.

  • On the growing of the interspecific hybrid ginseng plant, the phenomena of hybrid vigoures are observed in the root, stem, and leaf, but it can not produce seeds favorably since the ovary is abortive in most cases in interspecific hybrid plants. The present investigation was undertaken in an attempt to elucidate the embryological dses of the seed failure in the interspecific hybrid of ginseng (Panax Ginseng ${\times}$ P. Quinque folium). And the results obtained may be summarized as follows. 1). The vegetative growth of the interspecific hybrid ginseng plant is normal or rather vigorous, but the generative growth is extremely obstructed. 2). Even though the generative growth is interrupted the normal development of ovary tissue of flower can be shown until the stage prior to meiosis. 3). The division of the male gameto-genetic cell and the female gameto-genetic cell are exceedingly irregular and some of them are constricted prior to meiosis. 4). At meiosis in the microspore mother cell of the interspecific hybrid, abnormal division is observed in that the univalent chromosome and chromosome bridge occure. And in most cases, metaphasic configuration is principally presented as 23 II+2I, though rarely 22II+4I is also found. 5). Through the process of microspore and pollen formation of F1, the various developmental phases occur even in an anther loclus. 6). Macro, micro and empty pollen grains occur and the functional pollen is very rare. 7). After the megaspore mother cell stage, the rate of ovule development is, on the whole, delayed but the ovary wall enlargement is nearly normal. 8). Degenerating phenomena of ovules occur from the megaspore mother cell stage to 8-nucleate embryo sac stage, and their beginning time of constricting shape is variously different. 9). The megaspore arrangement in the parent is principally of the linear type, though rarely the intermediate type is also observed, whereas various types, viz, linear, intermediate, Tshape, and I shape can be observed in hybrid. 10). After meiosis, three or five megaspore are some times counted. 11). Charazal end megaspore is generally functional in the parents, whereas, in F1, very rarely one of the center megaspores (the second of the third megaspore) grows as an embryo sac mother cell. 12). In accordance with the extent of irregularity or abnormality in meiosis, division of embryo sac nuclei and embryo sac formation cause more nucellus tissue to remain within th, embryo sac. 13). Even if one reached the stage of embryo sac formation, the embryo sac nuclei are always precarious and they can not be disposed to theil proper, respective position. 14). Within the embryo sac, which is lacking the endospermcell, the 4-celled proembryo, linear arrangement, is observed. 15). Through the above respects, the cause of sterile or seed failure of interspecific hybrid would be presumably as follows, By interspecific crossing gene reassortments takes place and the gene system influences the metabolism by the interference of certain enzyme as media. In the F1 plant, the quantity and quality of chemicals produced by the enzyme system and reaction system are entirely different from the case of the parents. Generally, in order to grow, form, and develop naw parts it is necessary to change the materials and energy with reasonable balance, whereas in the F1 plant the metabolic process becomes abnormal or irregular because of the breakdown of the balancing. Thus the changing of the gene-reaction system causes the alteration of the environmental condition of the gameto-genetic cells in the anther and ovule; the produced chemicals cause changes of oxidatio-reduction potential, PH value, protein denaturation and the polarity, etc. Then, the abnormal tissue growing in the ovule and emdryo sac, inhibition of normal development and storage of some chemicals, especially inhibitor, finally lead to sterility or seed failure. Inconclusion, we may presume that the first cause of sterile or seed abortion in interspecific hybrids is the gene reassortment, and the second is the irregularity of the metabolic system, storage of chemicals, especially inhibitor, the growth of abnormal tissue and the change of the polarity etc, and they finally lead to sexual defect, sterility and seed failure.

Density Effects on the Size of 2-1 Korean Pine and 1-1 Jack Pine Nursery Stock (잣나무 2-1 묘(苗)와 방크스소나무 1-1 묘(苗)의 생장(生長)에 미치는 밀도효과(密度効果))

  • One of the most common needle leaf species used in planting in Korea is korean pine (Pinus koraiensis S. et Z.), and jack pine (Pinus banksiana Lamb.) is one of the test species for suitability. The relation of nursery bed density of 2-1 korean pine and 1-1 jack pine was studied at the Kwang Nung Nursery, Central Branch Station of Forest Research Institute, and about 40km north of Seoul. Nursery bed density of 2-1 korean pine, which ranged from 36 to 324 trees per square meters and of 1-1 jack pine, which ranged from 25 to 169 trees per square meters, had a marked effect on caliper, height, dry weight and percent and amount of plantable stock. The soil physical and chemical properties is silt plus clay, 50.55 percent; organic matter, 2.09 percent; total nitrogen, 0.13 percent; available phosphorus, 253.25 ppm; exchangeable potash, 0.46 m.e/100g; and pH, 5.58. As the density of the nursery seedling stand of 2-1 korean pine increases, the average tree height increases (Fig. 1A), but in 1-1 jack pine density do not affect to increase or decrease the average tree height. As the density of nursery bed increases, the average stem caliper (at 2cm above ground line) and dry weight decrease (Fig. 1B), but the decreasing rate is more seriously in 1-1 jack pine than 2-1 korean pine (Fig.5). As increasing the density of nursery bed, the T/R ratio of trees of the test species increase. Also the dry weight of leaf, stem and root parts are decreasing in proportion to the increase of stand density, but the drop rate of jack pine is more rapid than korean pine (table. 1) The patent facts of difference of growth characteristics between 2-1 korean pine and 1-1 jack pine were studied. These facts should be used to select the scale of stand density at the nursery bed or the plantable site. Korean pine is demanded high density, on the other hand in jack pine low density are more suitable to manage the stand density. Stands of comparatively low density had the greatest percentage of high-quality stock, and the stands of high density had less than the high quality trees of low density. An important criterion of the best density is percent and number of high-quality trees produced per square meter of bed area. Stem caliper and stem height of seedling is used in most public nurseries to sort seedling into plantable grades. The stock grade standard has set at 4.5mm caliper and 16cm height of 2-1 korean pine as the minimum desired stem caliper and height. By the result studies, the plantable stock grade standards of 2-1 korean pine used at stem height 16cm and stem caliper 4.5mm from public nurseries should be reformed to stem height 18cm and stem caliper 4.0mm by the growth characteristics and the tree distribution of stem height and caliper of relation to density. For the 2-1 korean pine, best density should be about 160 to 200 trees per square meter according to soil fertility. For the 1-1 jack pine, the suitable standard of plantable stock should be at stem height 25cm and caliper 6mm (at 2cm above ground line) and best density was about 100 to 120 trees.

Biochemical Changes of Dissimilation and Assimitation in Zoysia japonica Steud during the Regrowth Process after mowing (한국잔디(Zoysia japonica Steud.)의 예초후 재생과정에 따른 이화적, 동화적 생화학변화에 관한 연구)

  • During the regrowth the process after mowing, NRA in the leaf was the highest activity from the 5th day to the 7th day. Before mowing, the NRA in the root was not almost detected. But, the NRA in root appeared a rapid increasing activity from the 3rd day to the 4th day after mowing ( Figs.27 ~ 32). During the regrowth process after mowing, a general tendency of AA in the aboveground parts appeared an increasing tendency from the 1st day to the 4th day, a rapid increasing tendency from the 7th day to the 8th day reaching its peak, and a decreasing rate on the 8th and 9th day reaching its peak, and a decreasing rate on the 8th and 9th day. But the AA in the root appeared rapid increasing rate from the 2nd day to the 7th day, the heginning of reagrowth, this tendency showed a similar figure in the case of Total Soluble Carbohydrate ( TSC) in the internode. Both AA and NRA were appeared recovery stage frorn the 8th day after mowing(Figs.15~20). During the regrowth process after mowing, changes of the maximum plant lengths were 18.27cm in the 6cm mowing plot on the 24th day after mowing, 18.83cm in the 3cm mowing plot on the 18th day after mowing, and 18.16cm in the 6cm mowing plot on the 14th day after mowing ( Fig.2). During the regrowth process after mowing. changes in Dry Matter (DM) contents in leaf and stem were a slow decreasing tendency from the 1st day to the 4th day. From the 5th day to the 8th day it appeared a rapid increasing tendency. And afterward until the 15th day. All treatments were reached at a steady state ( Figs.3 ~ 8). During the regrowth process after mowing, changes in the TSC contents of stem and crown were a slow decreasing tendency from the 1st day to the 5-6th day. Prom the 7th to the 8th day three was a rapid increasing tendency. And afterward until the 15th day there was a decreasing rate at a steadyv state. In root there was a similar tendency to that of leaf and stem organs. A general tendency in internode, the TSC content appeared a similar figure to increment of AA (Figs. 9 - 14). During the regrowth process after mowing, changes in te Crude Proem (CP) content of ahoveground parts appeared a slow increasing tendency from the 1st day to the 5-6th day, where it is peak. And afterward to the 15th day there was a decreasing rate at a steady state. But, in toot there were a contrary tendency to that of aboveground

Optimized Condition of Genomic DNA Extraction and PCR Methods for GMO Detection in Potato (유전자재조합 감자의 검정을 위한 DNA분리 및 PCR검출의 최적조건 탐색)

  • To compare the quality of genomic DNA extracted from potato for PCR detection, four different methods, such as silica-based membrane method, silica-coated bead method, STE solution treatment, and CTAB-phenol/chloroform method, were evaluated. Also, to remove an excessive carbohydrate from the potato, ${\alpha}$- and ${\beta}$-amylase were used individually and in combination. When used both silica-based membrane method and silica-coated bead method combined with enzymes, the genomic DNAs were extracted from the raw potato with high purity for PCR. However, the silica-coated head method combined with enzyme treatment was the most efficient for extraction of the genomic DNA from the frozen fried potatoes. When applied with STE solution, the highly purified DNA was extracted from the raw potatoes without enzyme treatment in adequate yield for PCR. In cases of processed potatoes, such as frozen-fried potato and fabricated potato chips, CTAB-phenol/chloroform method is mostly feasible for DNA extraction and PCR efficacy at high sensitivity. As the results of PCR amplification, 216bp of PCR product was detected on 2% agarose gel electrophoresis, but any amplicons derived from New leaf and New leaf Y gene was not detected in any sample.

Characteristics Comparison of Mutants Induced through Gamma Irradiation in 'Kardinal' Rose (감마선 조사로 유기한 장미 '카디날' 돌연변이체의 특성 비교)

  • This study was carried out to compare the pattern of mutant variation and to evaluate the characteristics of mutants obtained by gamma irradiation in rose 'Kardinal'. Forty four rooted cuttings of 'Kardinal' were irradiated at 70 Gy gamma-ray dose from a $^{60}Co$ source to induce mutants in 2002. The irradiated plants were planted in field, and observed spotting of petal color mutants from 2002 to 2004. Four different kinds of mutant twigs with each different color flower were obtained from the irradiated 'Kardinal' with red petal. After being identified to be a stable mutant from 2004 to 2008, each mutant line propagated by cutting was hydroponic-cultured to evaluate the characteristics in the greenhouse from 2008 to 2009. Four mutant lines obtained from 'Kardinal' with red petal (Red group, 44A, 45B) include KA1 with light pink petal (Red group, 55B-55D), KA2 with pink petal (Red group, 63A-63B), KA3 with deep pink (Red purple, N57A-N57C), and KA4 with orange red (Red group, 43A-43B). Diameters of each flower in four mutant lines were different from 'Kardinal'. The line KA1 was 9.5 cm wide, and it showed the smallest diameter when compared to other mutants. While the line KA2 was the largest one with 12.5 cm 'Kardinal'. Petal number per flower was also variable among the mutants. The line KA2 had 39.8 petals being the largest number among the mutants, while the line KA1 was the lowest one compared to 35.5 petals of 'Kardinal'. Petal color was measured by using colorimeter. Brightness (L) measured at each petal of four mutants increased more than 'Kardinal'. CIE Lab values, a and b decreased more than 'Kardinal' at the petal color of three mutants except the line KA4. Characteristics of shoot, leaf, etc. from four mutants were also different from the ones of 'Kardinal'. The line KA1 was shortest in shoot, node and peduncle length, and lowest in prickle number. The reverse side of leaves was reddish green color in 'Kardinal' as well as the line KA4, but green color in the line KA1, KA2, and KA3.

Studies on the Chilling Injury of Rice Seedlings II. Maximum Tolerance of the Recent Varieties from Indica ${\times}$ Japonica Cross to Chilling Stress at the 3rd Leaf-stage (수도의 유묘기 냉해에 관한 연구 II. 주요 신품종들의 제 3 엽기 내냉한계)

  • Comparative tolerance of the new rice varieties from Indicax Japonica cross to elastic and plastic chilling injuries was examined at the 3rd leaf-stage, the most sensitive stage to chilling during nursery, using a chilling chamber set 11\circ C, 13hrs. 20K lux day/5$^{\circ}C$ night. Seedlings were reared until 35 days after seeding in a 26\circ C, 13hrs. day / 18\circ C night chamber except the period of chilling treatment for 0, 3, 4, 6, 8 or 10 days. The new varieties were approximately twice susceptible to elastic injuries, and thrice susceptible to plastic injuries than the Japonica varieties in the term of chilling duration. Among the variables indicating apparant chilling injuries the reduction in dry matter responded most sensitively and differentiated well the varietal difference in chilling resistance even with shorter chilling period. Sequential aspects of the cause and effect relations involved in the development of the apparant chilling injuries are discussed and a schematic diagram is presented as Fig. 5 in the text.

Effects on Growth, Photosynthesis and Pigment Contents of Liriodendron tulipifera under Elevated Temperature and Drought (온도 증가와 건조 스트레스가 백합나무의 생장, 광합성 및 광색소 함량에 미치는 영향)

  • This study was conducted to investigate the effects of high temperature and drought on growth performance, photosynthetic parameters and photosynthetic pigment contents of Liriodendron tulipifera L. seedlings. The seedlings were grown in controlled-environment growth chambers with combinations of four temperature ($-3^{\circ}C$, $0^{\circ}C$, $+3^{\circ}C$, $+6^{\circ}C$; based on the monthly average for 30 years in Korea) and two water status (control, drought). Temperature rise increased growth, total dry weight and leaf area in all water status. Also photosynthetic rate, dark respiration, stomatal conductance and transpiration rate increased with increasing temperature. In contrast, growth and photosynthetic parameters of L. tulipifera seedlings were lower in $-3^{\circ}C$ than $0^{\circ}C$. But temperature rise decreased water use efficiency in all water status. Temperature rise increased photosynthetic pigment contents of leaf. Also chlorophyll a/b ratio increased with increasing temperature. In conclusion, the elevated temperature lead to causes growth increase through the increase of energy production by higher photosynthetic rate during a growth period of L. tulipifera seedlings.

Study of the mechanisms underlying increased glucose absorption in Smilax china L. leaf extract-treated HepG2 cells (청미래덩굴 잎 물추출물이 처리된 HepG2 세포에서의 포도당흡수기전 연구)

  • Purpose: Previous studies have shown that treatment with Smilax china L. leaf extract (SCLE) produces antidiabetic effects due to ${\alpha}$-glucosidase inhibition. In this study, we examined the mechanism underlying these antidiabetic effects by examining glucose uptake in HepG2 cells cultured with SCLE. Methods: Glucose uptake and glucokinase activity were examined using an assay kit. Expression of glucose transporter (GLUT)-2, GLUT-4, and HNF-$1{\alpha}$ was measured by RT-PCR or western blot. Results: Treatment with SCLE resulted in enhanced glucose uptake in HepG2 cells, and this effect was especially pronounced when cells were cultured in an insulin-free medium. SCLE induced an increase in expression of GLUT-2 but not GLUT-4. The increase in the levels of HNF-$1{\alpha}$, a GLUT-2 transcription factor, in total protein extract and nuclear fraction suggest that the effects of SCLE may occur at the level of GLUT-2 transcription. In addition, by measuring the change in glucokinase activity following SCLE treatment, we confirmed that SCLE stimulates glucose utilization by direct activation of this enzyme. Conclusion: These results demonstrate that the potential antidiabetic activity of SCLE is due at least in part to stimulation of glucose uptake and an increase in glucokinase activity, and that SCLE-stimulated glucose uptake is mediated through enhancement of GLUT-2 expression by inducing expression of its transcription factor, HNF-$1{\alpha}$.
