Effects of Dietary Fats and Fibers on Modulation of Biomarkers and Tumor Incidence in Rats during 1, 2-Dimethylhydrazine-Induced Colon Carcinogenesis

  • This study investigated the effect of different dietary fats and fibers on colon tumor incidence and cell proliferation, the levels of eicosanoids and polyamines in colonic mucosa of DMH-treated rats. The experiment was conducted on male Sprague Dawley rats using a 2 $\times$3 factorial design with two fats (corn oil and DHA-rich fish oil) and two fibers (cellulose and pectin) and a fiber-free control. The rats were find an experimental diet containing 15% (w/w) dietary fat and 6% (w/w) fiber for 25 weeks. Tumor incidence was Bower in rats fed fish oil as opposed to corn oil. The levels of arachidonic acid (AA) and eicosanoids ($PGE_2, and TXB_2$) in normal colonic mucosa were significantly lower in rats fed fish oil and there was a concomitant increase of docosahexaenoic acid (DHA). The levels of eicosanoids and AA in tumor tissues were significantly higher than those of normal colonic mucosa. The level of polyamines in normal colonic mucosa was not affected by dietary fats but was significantly lower than that in rumor tissues. Dietary fiber did not have a significant effect on rumor incidence and the levels of AA, eicosanoids and polyamines. Overall, fish oil rich in DHA reduced cell prolifiration and thus inhibited colon carcinogenesis through its effect on the distribution of AA and production of eicosanoids in normal colonic mucosa. However, its effect on colon carcinogenesis revealed a lack of consistency depending on the type of dietary fiber in diet.

  • This study was evaluated influence of fluidity properties according to basalt fiber and high volume fly ash in the mortar level, as part of a basic study for development of fiber reinforced concrete using basalt fiber and high volume fly ash. In the first step, it was evaluated that fluidity properties of mortar according to replacement ratio 6 level of fly ash(10, 20, 30, 40, 50 and 60mass%) and fluidity properties of mortar according to content 5 levels of SP(1.3, 1.5, 1.7, 1.9 and 2.1%) and content 5 levels of VA(0.2, 0.4, 0.6, 0.8 and 1.0%) for dispersion of the basalt fiber, in the second step, it was evaluated that fluidity properties of mortar using High-volume fly ash (50mass%) on 3 levels of basalt fiber length (6, 20 and 30mm). Results of assessment, if after a fiber mixed, it showed that viscosity agent is more effective to improve the fluidity and fiber dispersion than superplasticizer, high volume fly ash (50%) applying the mixing, due to three properties of fly ash, showed that the improved fiber dispersibility and flow improvement.

Quality Characteristics and Sensory Properties of Reduced-fat Emulsion Sausages with Brown Rice Fiber

  • We evaluated the effects of dietary fiber extracted from brown rice on the quality of reduced-fat emulsion sausage. Reduced-fat emulsion sausage was supplemented with brown rice fiber at levels of 0, 1, 2, 3, and 6%. Adding different levels of brown rice fiber affected the proximate composition of the reduced-fat emulsion sausages (p<0.05), except for protein content. Adding different levels of brown rice fiber influenced the physicochemical properties of the reduced-fat emulsion sausages. The cooking yield, emulsion stability, textural properties, and viscosity of the reduced-fat emulsion sausages containing brown rice fiber improved with increases in added brown rice fiber (p<0.05). The reduced-fat emulsion sausages containing 1-3% brown rice fiber generated sensory evaluation scores similar to those of the high-fat sausage control. These results indicate that reduced-fat emulsion sausages with 1-3% added brown rice fiber had the most acceptable sensory properties and highest quality characteristics compared to the others.

Effects of dietary fiber levels on cecal microbiota composition in geese

  • Objective: This study shows the effects of dietary fiber levels on cecal microbiota composition in geese at day 70 according to pyrosequencing of the 16S ribosomal RNA gene. Methods: A total of 468 1-day-old healthy male Yangzhou goslings with similar body weight were randomly divided into 3 groups with 6 replicates per group and 26 geese per replicate. Geese were fed diets with fiber levels of 2.5% (low fiber level diet, Group I) and 6.1% (Group III) during days 1-70, respectively, or 4.3% for days 1-28 and 6.1% for days 29-70 (Group II). Results: Low fiber level diet decreased body weight, average daily gain during, increased lower feed conversation rate of geese during day 1 to 70 (p<0.05). Low fiber level diet decreased the total operational taxonomic units, Chao1 index and Shannon index, whereas increased the Simpson index of cecal microbiota in geese at day 70. Low fiber level diet decreased the relative abundance of Bacteroidetes, Firmicutes, Bacteroides, and Paraprevotella in cecum of geese at day 70. The similarity of cecal microbiota between low fiber level diet group and other groups was smaller. Conclusion: This study indicates that the low fiber level diet decreased diversity of microbiota, and relative abundance of some beneficial microbiota in cecum of geese at day 70, implying that the low fiber level diet has negative influence on performance by altering the diversity and population of cecal microbiota in geese.

  • This study was to investigate the effect of dietary protein and fiber on the antioxidant enzyme activities of brain in acute or chronic ethanol-treated rats. Male Sprague-Dawley rats were fed on diets containing two levels of protein(7%, 20%) with two levels of fiber (5%, 10%) Rats were administered 40%(v/v) ethanol(5g/kg body weight)orally 90min before decaptiation in acute ethanol-treated groups and 25%(v/v) ethanol(5g/kg body weight) once a day for 5 weeks in chronic ethanol treated-groups. The rats were sacrificed after 5 weeks of feeding periods. Superoxide dismutase and gluthathione S-transferase activities were lower in chronic ethanol-treated groups than acute ethanol-treated groups whereas catalase and glutathuone peroxidase activities were significantly increased by chronic ethanol treatment. Low protein supplement accelerated to change of their activities however dietary fiber levels did not affect antioxidant enzyme activities. Chronic ethanol treatment and/or low protein supplement results in increasing the brain lipid peroxide content but in lowering glutathione level. (Korean J Nutrition 33(6) ; 613~618, 2000)

효소 레팅에 의한 케냐프 섬유의 분리 -킬레이터의 영향- (Effect of Enzyme Retting on the Fiber Separation of Kenaf Bast - influence of chelator -)

  • This research was aimed to investigate the effect of enzyme and the addition of chelators on rotting of the Kenaf bast. Enzyme rotting was effective only when the chelators were added with the enzyme. EDTA was a more effective chelator than oxalic acid under 1% concentration. There was no difference in the rotting effect under different enzyme concentration levels, and under different treatment time and temperature. Therefore, it was found that enzyme rotting can be carried out with low enzyme concentration(0.125%) at room temperature. Retting time can be shortened when higher enzyme concentration and higher temperature are applied. Cellulose I structure of kenaf fiber did not change after enzyme rotting, and different enzyme concentration did not affect the crytallinity structure. Non-cellulosic matters such as hemicellulose, lignin, and pectin were present in the descending order in the enzyme rotted kenaf fiber, and there were no differences in their amounts due to enzyme concentration levels. There was no difference in the dyeabilities of kenaf fiber rotted with different enzyme concentration levels. Enzyme rotted kenaf fiber showed better cyeability when pectin, lignin, and hemicellulose were removed.

Effect of different levels of fiber and protein on growth performance and fecal characteristics in weaning pigs

  • This experiment was conducted to evaluate the growth performance, fecal score, and fecal microbial shedding in weaning pigs fed diets with different levels of fiber and protein. A total of 96 weaning piglets ($7.41{\pm}0.71kg$) were used in a 5-week trial. Pigs were allotted to dietary treatments based on initial body weight in a $2{\times}2$ factorial design with the following factors: dietary fiber (100 and 200 g/kg, respectively, during days 0 to 14; 175 and 300 g/kg, respectively, during days 14 to 35) and dietary protein (170 or 200 g/kg). There were 6 replicates with 4 pigs per pen. On day 14, pigs fed high protein or high fiber diets had heavier body weight (p < 0.05). During days 0 to 14, pigs fed high protein or high fiber diets grew faster (p < 0.05). Additionally, during days 14 to 35, an interactive effect of fiber and protein was found (p < 0.05) on average daily gain. The different levels of protein and fiber in diet did not affect the pigs' fecal scores (p > 0.05). However, feces from the high fiber group showed lower concentration of Escherichia coli (p < 0.05). In conclusion, the results of the present study indicate that a high protein diet improves the growth of weaning pigs especially during the first two weeks. Moreover, the increments in fiber level, even in the high protein diet, favorably decreased the number of E. coli.

지방의 섭취량과 첨가된 섬유소의 종류가 흰쥐의 체내 지질 수준에 미치는 영향 (The Effect of Dietary Fat Levels and Sources of Dietary Fiber on Serum and Liver Lipids of Rats)

  • 식이중 지방 수준과 fiber 종류가 흰 쥐의 혈청과 간 조직내의 지질 함량에 미치는 영향을 알아보기 위해 지방 수준을 0% , 5% , 20% 수준으로 달리 하였으며 fiber 종류는 고지방 식이군( 20% 지방 )에 pectin과 cellulose를 각각 10% 수준으로 첨가하였다. 또 식이내 cholesterol첨가의 효과를 관찰하기 위해 고지방 식이군에 0.5% cholesterol을 첨가한 후 pectin과 cellulose를 10%씩 넣어 전체적으로 7군으로 나누어 각 각 다른 식이로 4주간 사양한 후 혈청과 간에서 지질 함량을 조사하였으며 간 조직의 형태학적 조사를 실시하여 다음과 같은 결과를 얻었다. 1) 식이 섭취량은 지방 수준이 낮을수록 높았으나 뚜렷한 차이는 보이지 않았으며 pectin과 cellulose 첨가로 인해서 섭취량이 줄었다. 증체량은 지방 수준이나 fiber종류에 따라 큰 차이를 보이지 않았다. 변의 무게는 지방 수준에 따른 차이는 없었으나 fiber 첨가로 인해서는 pectin은 약간의 감소를 cellulose 는 뚜렷한 증가를 보였다. 또 cholesterol을 첨가한 군 간의 비교에서도 pectin첨가군은 감소를 cellulose첨가군은 증가를 나타냈다. 간과 epididymal fat pad의 무게는 지방 수준이 높아짐에 따라 증가하였으며 fiber 종류에 따라서는 pectin첨가군이 cellulose 첨가 군 보다 낮았다. 2) 혈청 분석 결과는 식이내 지방 수준이 높을수록 total lipids수준이 높아지는 경향을 보였으며 pectin이 cellulose보다 혈청내 지질 수준을 뚜렷하게 낮추었다. Total cholesterol 및 phospholipids 량에 있어서도 지방 수준이 증가함에 따라 높아졌으며 cellulose보다 pectin이 첨가 되었을때 현저한 감소를 나타냈다. HDL- cholesterol수준은 무지방군에서는 매우 높게 나타났으나 표준군과 고지방군간에 는 명확한 결과를 얻을 수 없었다. 3) 간 분석 결과는 지방수준이 높을수록 total lipids량이 높아지는 경향을 보였다. 또 pectin 첨가가 cellulose첨가군보다 현저히 낮은 lipids수준을 나타냈다. 간의 total cholesterol함량은 지방 수준이 높아짐에 따라 증가하였으나 뚜렷한 차이는 아니었으며 pectin첨가가 cellulose첨가보다 cholesterol 수준이 상당히 낮았다. 4) 간 조직의 현미경 관찰 결과는 지방 수준이 높아짐에 따라 조직내 지방 침착이 심해졌으며 fiber 의 첨가로 인해서는 pectin은 뚜렷한 지방 침착의 감소를 보였으나 cellulose는 거의 감소 효과를 보이지 않았다.

Influence of pre-compression on crack propagation in steel fiber reinforced concrete

  • In this study, a new understanding is presented on the microcracking behavior of high strength concrete (HSC) with steel fiber addition having prior compressive loading history. Microcracking behavior at critical stress (σcr) region, using seven fiber addition volume of 0.5, 0.75, 1.0, 1.25, 1.5, 1.75, and 2.0% was evaluated, at two aspect ratios (60 and 75). The specimens were loaded up to a specified compressive stress levels (0.70fc-0.96fc), and subsequently subjected to split tensile tests. This was followed by microscopic analyses afterwards. Four compressive stress levels as percentage of fc were selected according to the linearity end point based on stress-time (σ-t) diagram under uniaxial compression. It was seen that pre-compression has an effect on the linearity end point as well as fiber addition where it lies within 85-91% of fc. Tensile strength gain was observed in some cases with respect to the 'maiden' tensile strength as oppose to tensile strength loss due to the fiber addition with teething effect. Aggregate cracking was the dominant failure mode instead of bond cracks due to improved matrix quality. The presence of the steel fiber improved the extensive failure pattern of cracks where it changes from 'macrocracks' to a branched network of microcracks especially at higher fiber dosages. The applied pre-compression resulted in hardening effect, but the cracking process is similar to that in concrete without fiber addition.

A Fiber Spool's Vibration Sensitivity Optimization Based on Orthogonal Experimental Design

  • A fiber spool with ultra-low vibration sensitivity has been demonstrated for the ultra-narrow-linewidth fiber-stabilized laser by the multi-object orthogonal experimental design method, which can achieve the optimization object and analysis of influence levels without extensive computation. According to a test of 4 levels and 4 factors, an L16 (44) orthogonal table is established to design orthogonal experiments. The vibration sensitivities along the axial and radial directions and the normalized sums of the vibration sensitivities are determined as single objects and comprehensive objects, respectively. We adopt the range analysis of object values to obtain the influence levels of the four design parameters on the single objects and the comprehensive object. The optimal parameter combinations are determined by both methods of comprehensive balance and evaluation. Based on the corresponding fractional frequency stability of ultra-narrow-linewidth fiber-stabilized lasers, we obtain the final optimal parameter combination A3B1C2D1, which can achieve the fiber spool with vibration sensitivities of 10-12/g magnitude. This work is the first time to use an orthogonal experimental design method to optimize the vibration sensitivities of fiber spools, providing an approach to design the fiber spool with ultra-low vibration sensitivity.
