체액이 초기배의 발생생리에 미치는 효과에 관한 연구. I. 생쥐 1- 및 2-세포배의 체외발생에서 배양액과 단백질원의 효과 (Studies on the Effects of Body Fluids on the Developmental Physiology of Early Preimplantation Embryos. I. Effect of Serum on In Vitro Development of 1- and 2-Cell Mouse Embryos)

  • In vitro developmental ability of early preimplantation monse embryos was shown to be depend on the embryonic stages, media and snpplements and their interaction(Experiment 1). The development of I-cell embryos were more promoted in the complex medinm(Ham's Fl0) than in the simple one(m-KRB), but that of 2-cell embryos showed the reverse effect. The bovine serum albumin(BSA) as a medium snpplement more promoted the development of I- and 2-cell embryos, compared with human fetal cord serum(HCS). On the other hand, the harmful effect of HCS was especially shown on the early cleavage in the embryonic development of the two stages. The effect of serum, in the respect of interaction between media and snpplements. was also more significantly appeared in m-KRB than Ham's Fl0. In the experiment 2, when the harmful effect of HCS was compared with that of fetal bovine serum(FBS), the former more promoted the development of l - and 2-cell embryos than the latter. The effect of HCS was more significantly shown in the development of I-cell than that of 2-cell embryos. Conclusively, as I- and 2-cell embryos were different in the requirements for the in vitro development. the optimal medium and supplement have to be selected for each embryonic stage. It is also respected to the better result if it take into consideration into the kinds of sera when serum is used for culture of early preimplantation embryos.

Effect of Mitogens, Supplement of β-mercaptoethanol and Fetal Bovine Serum Supplementation in Whole Blood Culture Medium for Bovine Chromosome Analysis

  • We investigated the effects of the mitogen supplements of 3 types, pokeweed mitogen (PWM), phytohemagglutinin (PHA) and concanavalin A (ConA), to a whole blood culture system on the number of metaphase spreads obtained in perinatal bovine chromosome analysis. In addition, the supplementation of ${\beta}$-mercaptoethanol (${\beta}$-ME) and FBS was examined in such system. Significant differences (p<0.05) were seen in the number of metaphase spreads with PHA stimulation compared to both PWM and ConA stimulation. When examined the effects of ${\beta}$-ME supplementation, the number of metaphase spreads was significantly (p<0.05) increased at $30{\mu}M$ ${\beta}$-ME compared to control. When evaluated FBS supplementation during PWM stimulation, no significant effect of the supplementation was found. Finally, the effects of the cortisol concentration (10-20, 20-30 and >30 ng/ml) of the blood samples were examined. There was no significant effect of cortisol concentration (p>0.05) among these 3 cortisol concentration groups. The mean percentages of normal metaphase plates (2n=60) from each calf 1) with ${\beta}$-ME, 2) without ${\beta}$-ME and 3) with FBS stimulated with PWM were not significantly different (p>0.05). In conclusion, these findings may be useful in cytogenetic screening programs for not only perinatal calves but also for mature cattle.

A Spatial-domain Fourier Transform Infrared Spectrometer: Application for Analyte Measurement in Cell Culture Media

  • A spatial-domain Fourier Transform (FT) infrared (IR) spectrometer coupled with a PtSi Schottky­barrier IR detector plane was developed in the spectral range of $2.0-2.5{\mu}m$ for noninvasive measurement of analyte concentrations in cell culture media during cell culture processing. A key optical component of the spectrometer is a Savart plate which is a birefringent polarizer generating coherent two rays for interfering. The spectral resolution of the spectrometer was determined as $71cm^{-1}$ (${\~}0.05{\mu}m$ at $2.5{\mu}m$). Clear IR fringe patterns were imaged on the IR detector plane. The feasibility of the spectrometer for our application was investigated by measuring absorbance spectra of glucose and fetal bovine serum (FBS) which are important compounds in cell culture media. Experiment results show that the spectral quality of glucose and FBS was comparable with the standard spectra acquired with a commercial FT-IR spectrometer, presenting the feasibility of the spectrometer to perform analyte measurement in cell culture media.

Cell Cycle Analysis of Bovine Cultured Somatic Cells by Flow Cytometry

  • The cell cycle phase in which donor nuclei exist prior to nuclear transfer is an important factor governing developmental rates of reconstituted embryos. It was suggested that quiescent G0 and cycling G1 cells could support normal development of reconstituted embryos. In a quest of optimized donor nuclei treatment prior to nuclear transfer, this study was undertaken to examine the cell cycle characteristics of bovine fetal and adult somatic cells when cultured under a variety of culture treatments and the cell cycle change with the lapse of time after trypsinization. This was archived by measuring the DNA content of cells using flow cytometry, Cultured fetal fibroblast cells, adult skin and muscle cells, and cumulus cells were divided by 3 culture treatments; 1) grown to 60-70% confluency (cycling), 2) serum starved culture, 3) culture to confluency. Trypsinized cells were fixed by 70% ethanol and stained with propidium iodide. For one experiment, trypsinized cells were resuspended in DMEM+10% FBS and incubated for 1.5, 3 and 6 h with occasional shaking before ethanol fixation. Cell cycle phases were determined by flow cytometry enabling calculation of percentages of G0+G1, S and G2+M. The majority of cells were in G0+Gl stage regardless of origin of cells. Cultures that were serum starved or cultured to confluency contained significantly (P<0.05) higher percentages of cells in G0+G1 (89.5-95.4%). For every cell lines and culture treatments, percentages of cells in existing in G0+G1 increased with decreasing of the cell size from large to small. In the serum starved and confluency groups, about 98% of small cells were in G0+G1 Serum starved culture contained higher percentages of small-sized cells (38.5-66.9%) than cycling and confluent cultures regardless of cell lines (P<0.05). After trypsinization of fetal fibroblast and adult skin cells that were serum starved and cultured to confluency, the percentages of cells in G0+G1 significantly increased by incubation for 1.5(95.7-99.5%) and 3.0 h (95.9-98.6%). The results suggest that the efficient synchronization of bovine somatic cells in G0+G1 for nuclear transfer can be established by incubation for a limited time period after trypsinization of serum starved or confluent cells.

다양한 혈청 물질의 첨가가 염소 수정란의 체외배양에 미치는 영향 (Influence of various serum supplement on in vitro culture for goat embryos)

  • 본 연구는 염소 수정란의 체외배양용 배지에 FBS, gBS 및 PVA 첨가시 배발달 및 apoptosis 발생률을 분석하여 체외배양시 각 첨가물의 효과를 조사하였다. 체외성숙 후 체외수정을 실시하여 체외배양 배지에 10% FBS, 10% gBS 및 10% PVA를 첨가하고 배발달 효율과 배반포의 품질을 확인하기 위해 TUNEL assay를 통해 apoptosis 발생 비율을 조사한 결과, 난할률 및 배반포 형성률에서 처리군 모두 대조군(무혈청) 보다 유의적으로 높은 결과를 보였다. 특히 gBS와 PVA 처리군에서 각각 31.95, 35.29%로 유의적으로 가장 높은 배반포 형성률을 보였다. 또한 TUNEL assay의 결과에서도 배발달 비교실험에서와 같이 대조군에서 가장 적은 총세포수와 가장 높은 apoptosis 비율을 보였으며, gBS와 PVA 처리군에서 가장 많은 총세포수 및 가장 적은 apoptosis 비율을 보였다. 염소의 체외배양 효율 향상을 위한 본 연구의 결과, 무혈청 배지나 FBS첨가 보다는 gBS나 PVA의 첨가가 배발달 효율뿐만 아니라 배반포 품질에도 긍정적임을 알 수 있었으나, gBS내 확인되지 않는 물질들의 위험성을 감안했을 때 PVA가 보다 안전하고 효율적일 것으로 생각된다.

Tomplasma gondii의 숙주세포 침투를 억제하는 우태아혈청 성분 (Inhibition of entry of Toxoplasma gonldii into MDCK cells by fetal bovine serum)

  • 우태아혈청의 성분이 Toxoplasma의 숙주세포 침투를 억제하는 현상을 보여 이를 보고하고자 한다. MDCK세포를 숙주로 하여 Toxoplasma의 RH주를 첨가하였을 때, 우태아혈청 농도 1%(v/v) 에서 억제 효과가 나타나기 시작하여 5%일때 침투능을 반감시켰다. 우태아절청의 비동화 여부는 억제 효과에 영향을 미치지 못하였으며, 우태아혈청을 $95^{\circ}C$에서 10분간 처리하여 억제물질의 기능을 파괴시킬 때 억제 효과는 약간 감소되었다. Toxoprasma를 먼저 우태아혈청으로 처리한 후 숙주 세포에 첨가하였을 때 침투능에는 효과를 미치지 못하였으나, Tomplasma를 숙주세포와 배양하면서 우태아혈청을 첨가하였을 때에는 시간의 경과에 따라 침투능을 억제시켰다. 이상의 결과로 우태아혈청에 Toxoplasma의 숙주세포 침투를 억제하는 성분이 존재하는 것을 확인하였으며, 억제 방법이 침투하는 순간에 일어나며, 억제물질의 기능을 통해서라기 보다는 구조를 통해서 이루어진다고 추정할 수 있었다.

  • The study evaluated the effect of donor cell treatments for G0/Gl synchronization and the donor ceil type on development and incidence of apoptosis in cloned cattle embryos. Primary cultures were established from a female fetus on day 50 of gestation and adult ear skin biopsies. Cells were randomly allocated into 3 experimental treatment groups after 6~8 passages. Group 1 (Confluent), cells were cultured in DMEM supplemented with 10% FBS until 90% confluent. Group 2 (Serum-starvation), cells were cultured in DMEM Supplemented With 0.5% FBS for 5 days. Group 3 (Roscovitine), Cells were cultured in DMEM supplemented with 10% FBS and 30 $\mu$M Roscovitine for 12 h. Cell cycle and apoptosis were analyzed using flow cytometry after labelling with DAPI and YO-PRO-1. At 19 h post-maturation (hpm), enucleated oocytes were reconstructed with donor cells and fused by a single DC pulse (1.6 kV/cm, 60 $\mu$sec). (중략)

TGF-$\beta$1와 IGF-I이 소 난포란의 체외성숙 및 체외수정란의 배양에 미치는 영향 (Effect of TGF-$\beta$1 and IGF-I on Bovine In Vitro Maturation and Embryo Culture)

  • 본 연구는 혈청첨가 또는 무첨가에 따른 소 난포란의 체외성숙에 있어서 참가된 TGF-$\beta$1과 IGF-I이 그후의 수정 및 발생에 미치는 영향과, 이들 growth factor의 농도에 따른 8세포기 소 체외수정란의 발달에 미치는 영향을 조사하고자 실시하였다. 도축장에서 얻어진 난소로부터 채취된 난포란을 20% FBS가 첨가 또는 첨가되지 않은 TCM-199에 TGF-$\beta$1, IGF-I 또는 TGF-$\beta$1+IGF-I을 각각 10ng/ml 첨가하여 38.5$^{\circ}C$에서 24시간 배양하여 체외성숙을 유기하였다. 성숙된 난자를 1$\times$106/ml 정자농도로 수정후 24시간에 glucoserk 첨가되지 않은 CZB 배양액으로 옮겨 48시간 배양한 다음, TCM-199+20%FBS에서 96시간 추가배양하였다. 본 연구에서 혈청이 첨가된 난포란의 체외성숙배양액에 첨가된 growth factor들은 수정후의 배분할 및 배발생에 영향을 미치지 않았다. 혈청이 첨가되지 않은 경우에서는 TGF-$\beta$1의 첨가는 배분할 및 배발생율을 향상시켰다(P<0.05). 한편 TCM-199+20%FBS에 5, 10ng/ml의 TGF-$\beta$1 및 5, 10, 50, 100ng/ml의 IGF-I을 각각 첨가후 8세포기 체외수정란을 배양한 결과, 10ng/ml TGF-$\beta$1의 첨가는 배반포기로의 발생율을 향상시켰다(P<0.05). 결론적으로, 혈청이 포함되지 않은 소 난포란의 체외성숙 배양액, 또는 수정란의 체외배양액에 10ng/ml TGF-1의 첨가는 배반포기로의 발생율을 향상시킨다.

Efficient Cryopreservation of Porcine Blastocysts produced by In Vitro Fertilization

  • Cryopreservation has been applied successfully in many mammalian species. Nevertheless, pig embryos, because of their greater susceptibility to cryoinjuries, have shown a reduced developmental competence. The aim of this study was to evaluate the survival status of vitrified-warmed porcine embryos. Forced blastocoele collapse (FBC) and non-FBC blastocysts are vitrified and concomitantly cultured in culture media which were supplemented with/without fetal bovine serum (FBS). Porcine vitrified-warmed embryos were examined in four different methods: group A, non-FBC without FBS; group B, non-FBC with FBS; group C, FBC without FBS; group D, FBC with FBS. After culture, differences in survival rates of blastocysts derived from vitrified-warmed porcine embryos were found in group A~D (39.5 (A) vs 52.5 (B) and 54.8 (C) vs 66.7% (D), respectively, p<0.05). Reactive oxygen species (ROS) level of survived blastocysts was lower in group D than that of another groups (p<0.05). Moreover, total cell number of survived blastocysts was higher in group D than that of other groups (p<0.05). Otherwise, group D showed significantly lower number of apoptotic cells than other groups ($2.0{\pm}1.5$ vs $3.2{\pm}2.1$, $2.8{\pm}1.9$, and $2.7{\pm}1.6$, respectively, p<0.05). Taken together, these results showed that FBS/FBC improves the developmental competence of vitrified porcine embryos by modulating intracellular levels of ROS and the apoptotic index during the vitrification/warming procedure. Therefore, we suggest that FBS and FBC are effective treatment techniques during the vitrification/warming procedures of porcine blastocysts.

다양한 배양 조건에서 제대혈 유래 CD34+ 조혈 세포의 체외 증식 (Ex vivo Expansion of CD34+ Hematopoietic Cells from Cord Blood in Various Culture Environments)

  • 본 연구에서는 제대혈 유래의 조혈줄기세포를 효과적으로 배양하기 위한 선행 연구로서 세포 배양 환경에 따른 조혈줄기세포 증식능의 변화를 관찰하였다. 제대혈의 단핵구 세포에서 분리한 $CD34^+$ 세포를 성장인자 조성-I(이하, coc-I) (EPO, GM-CSF, SCF, IL-3) 및 성장인자 조성-II(이하, coc-II) (TPO, G-CSF, SCF, IL-6, Flt3/Flk-2 ligand)가 포함되어 있는 IMDM(Iscove's modified Dulbecco's medium) 및 무혈청 배지(serum free media, SFM)에서 배양하였으며, 우태아혈청(FBS)의 첨가 영향, 2차원 및 3차원 배양 후 각 조건에서의 세포 증식 및 콜로니 형성능을 비교하였다. 일반적으로 coc-I에서의 세포 증식 및 콜로니 증식이 coc-II에서보다 높았다. 3차원 배양(methocult)에서는 가장 높은 세포 증식($2,258{\pm}456$배)을 나타냈으며, 같은 조성의 2차원 배양(IMDM + coc-I + FBS)에서는 가장 높은 콜로니 증식(BFU-E: $652{\pm}19$, CFU-GM: $520{\pm}58$, CFU-GEMM: $339{\pm}100$배)이 나타났다. 배지를 기준으로 보면, coc-II 조성에 우태아혈청이 포함되지 않은 경우를 제외한 모든 경우에서 세포 증식 및 콜로니 증식이 무혈청 배지에서보다 IMDM에서 높았다. 결론적으로, 모든 배양 조건 중에서 'IMDM + coc-I + FBS' 및 'IMDM + coc-I'에서 가장 좋은 콜로니 증식을 보였으며, 우태아혈청의 첨가 및 2차원 배양 조건이 콜로니 증식에 더 효과적인 것으로 확인되었다. 본 연구 결과는 앞으로 조혈줄기세포의 체외 증식에 필요한 공정개발이나 생물반응기 설계에 유용한 정보를 제공할 수 있을 것으로 사료된다.
