  • Objectives The purpose of this study is to confirm the efficacy and safety of the treatment of with fermented oyster extract on height growth in children with short stature. Methods A total of 100 people, between 6 and 11 years old, will be participated in a randomized, double-blind, and placebo-controlled human study. The fermented oyster group will take 500 mg of fermented oyster extract once a day for 24 weeks. The placebo group will take 3400 mg of fructooligosaccharide as placebo once a day for 24 weeks. The outcomes of the intervention will be measured at the baseline, 6 week, 12 week, 18 week, and 24 week. The primary outcome is the changes in height from the baseline. The secondary outcomes are growth rate, height SDS, bone age, GH, IGF-1, IGFBP-3, osteocalcin, BALP, DPD, and LH. Results This trial was approved by the institutional review board of Pusan National University Korean Medicine Hospital (registry number: PNUKHIRB-2019002). Recruitment of the research participants will be opened from May 2019 till December 2019. Conclusions This study will provide clinical information to determine the efficacy and safety of the treatment with fermented oyster extract on height growth in children with short stature.

Effect of fermented sarco oyster extract on age induced sarcopenia muscle repair by modulating regulatory T cells

  • Sarcopenia is an age-related, progressive skeletal muscle disorder involving the loss of muscle mass and strength. Previous studies have shown that γ-aminobutyric acid (GABA) from fermented oysters aids in regulatory T cells (Tregs) cell expansion and function by enhancing autophagy, and concomitantly mediate muscle regeneration by modulating muscle inflammation and satellite cell function. The fermentation process of oysters not only increases the GABA content but also enhances the content of branched amino acids and free amino acids that aid the level of protein absorption and muscle strength, mass, and repair. In this study, the effect of GABA-enriched fermented sarco oyster extract (FSO) on reduced muscle mass and functions via Treg modulation and enhanced autophagy in aged mice was investigated. Results showed that FSO enhanced the expression of autophagy markers (autophagy-related gene 5 [ATG5] and GABA receptor-associated protein [GABARAP]), forkhead box protein 3 (FoxP3) expression, and levels of anti-inflammatory cytokines (interleukin [IL]-10 and transforming growth factor [TGF]-β) secreted by Tregs while reducing pro-inflammatory cytokine levels (IL-17A and interferon [IFN]-γ). Furthermore, FSO increased the expression of IL-33 and its receptor IL-1 receptor-like 1 (ST2); well-known signaling pathways that increase amphiregulin (Areg) secretion and expression of myogenesis markers (myogenic factor 5, myoblast determination protein 1, and myogenin). Muscle mass and function were also enhanced via FSO. Overall, the current study suggests that FSO increased autophagy, which enhanced Treg accumulation and function, decreased muscle inflammation, and increased satellite cell function for muscle regeneration and therefore could decrease the loss of muscle mass and function with aging.

발효굴 섭취가 중년 여성의 신체조성, 근력 및 혈중 근성장 인자에 미치는 영향 (Effects of Fermented Oyster Extract Supplementation on Body Composition, Muscular Strengths and Blood Muscle Growth Fact ors in Elderly Women)

  • Crassostrea gigas were fermented using L. brevis BJ20 to prepare fermented oyster extract (FO). The participants of this study were randomly assigned to FO and placebo (CON) groups. The FO group was given 1.0 g of FO supplementation and the CON group was given sucrose each day for eight weeks. The effects of FO supplementation on body composition, muscula r strength, and blood factors associated with muscle growth were assessed. The FO supplement was enriched with arginine (6,183.3 mg), phenylalanine (217.9 mg), leucine (122.6 mg), isoleucine (59.8 mg), valine (16.4 mg), and γ-amino butyric acid (GABA, 1,053.7 mg). The total fat was significantly decreased in the FO group compared with the CON group (p < 0.05). 60D/S Ext.T/Wo rk and 60D/S Flex.T/Work concomitantly with 60D/S Flex.PeakTQ/BW were significantly increase d by FO treatment compared to CON group (p < 0.05). However, posture stability was not significa ntly different between the groups. The levels of angiotensin-converting enzyme were significantly decreased within the FO group (p < 0.05). The FO group showed significantly decreased levels of tumor necrosis factor-α and increased levels of human growth hormone compared with the CON group (p < 0.01). The levels of insulin-like growth factor-1 increased (p < 0.01) in the FO group while that of creatine kinase and triglyceride decreased significantly compared with the CON group (p < 0.05). These results demonstrated that FO supplementation is effective in preventing sarcopenic obesity and maintaining and strengthening muscular function in elderly wom en. Hence, FO supplements can be used as functional ingredients for these benefits.

Safety effect of fermented oyster extract on the endocrine disruptor assay in vitro and in vivo

  • Oyster (Crassostrea gigas) is a marine bivalve mollusk widely distributed in coastal areas, and have been long widely used in industrial resources. Several studies demonstrated that fermented oyster (FO) extract attribute to bone health, but whether administration of FO play as an endocrine disruptor has not been studied. Therefore, in the present study, we investigated the effect of FO on the endocrine system in vitro and in vivo. As the results of the competitive estrogen receptor (ER) and androgen receptor (AR) binding affinities, FO was not combined with ER-α, ER-β, and AR. However, 17β-estradiol and testosterone, used as positive control, were interacted with ER and AR, respectively. Meanwhile, oral administration of 100 mg/kg and 200 mg/kg of FO doesn't have any harmful effect on the body weight, androgen-dependent sex accessory organs, estrogen-dependent-sex accessory organs, kidney, and liver in immature rats. In addition, FO supplementation has no effect on the serum levels of luteinizing hormone (LH), follicle stimulating hormone (FSH), testosterone, and 17β-estradiol. However, the relative weight of androgen- and estrogen-dependent organs were significantly increased by subcutaneously injection of 4.0 mg/kg of testosterone propionate (TP) and by orally administration of 1.0 ㎍ of 17α-ethynyl estradiol (EE) in immature male and female rats, respectively. Furthermore, TP and EE administration markedly decreased the serum LH and FSH levels, which are similar those of mature Sprague-Dawley (SD) rat. Furthermore, the testosterone and 17β-estradiol levels were significantly enhanced in TP and EE-treated immature rats. Taken together, our findings showed that FO does not interact with ER and AR, suggesting consequentially FO does not play as a ligand for ER and AR. Furthermore, oral administration of FO did not act as an endocrine disruptor including androgenic activity, estrogenic activity, and abnormal levels of sex hormone, indicating FO may ensure the safety on endocrine system to develop dietary supplement for bone health.

친환경자재를 이용한 인삼 점무늬병과 탄저병의 발병억제효과 (Suppressive Effects of Homemade Environment-friendly Materials on Alternaria Blight and Anthracnose of Ginseng)

  • 본 연구는 인삼점무늬병과 탄저병에 대한 친환경자재로 만들어진 친환경 살균제의 억제효과를 알아보기 위해서 수행을 했다. 인삼 점무늬병과 인삼 탄저병들은 인삼뿌리가 비대하기 시작하기 전에 낙엽을 시켜서 수확량을 크게 감소하는 주요한 병들이다. 천연칼슘, 녹차추출물, 홍삼박 술등은 A. panax에 의해서 유발된 인삼 점무늬병에 대해서 상당한 억제효과를 보였고 게 껍질 발효액과 익모초자연즙액은 C. gloeosporioides에 의해서 유발된 인삼탄저병에 대해서 큰 억제효과를 나타내었다. 이들 친환경 살균제는 치료효능보다 대체적으로 예방효능이 더 우수 하였다. 그러므로 이들 친환경 자재들은 인삼 유기재배포장에서 사용할 수 없는 화학농약을 대체하는 살균제로써 이용할 수 있고 그들의 억제효과를 극대화하기 위해 발병 전에 처리를 해야 한다.

새우젓의 정미성분에 관한 연구 (THE TASTE COMPOUNDS FERMENTED ACETES CHINENSIS)

  • In Korea fermented fish and shellfish have traditionally been favored and consumed as seasonings or further processed for fish sauce. Three major items in production quantity among more than thirty kinds which are presently available in the market are fermented anchovy, oyster and small shrimp. They are usually used as a seasoning mixture of Kimchi in order to provide a distinctive flavor. Fermented small shrimp, Acetes chinensis is most widely and largely used ana occupies an important position in food industry of this country. But no study on its taste compounds has been reported. This study was attempted to establish the basic data for evaluating taste compounds of fermented small shrimp. The changes of such compounds during fermentation as free amino acids, nucleotides and their related compounds, TMAO, TMA, and betaine were analysed. In addition, change in microflora during the fermentation under the halophilic circumstance was also investigated. The samples were prepared with three different salt contents of 20, 30 and $40\%$ to obtain the proper degree of fermentation at a controlled tempeature of $20{\pm}2^{\circ}C$. The results are summarized as follows: Volatile basic nitrogen increased rapidly until 108 days of fermentation and afterwards it tended to increase slowly. Amino nitrogen also increased rapidly until 43 days of fermentation and then increased slowly. Extract nitrogen increased and marked the maximum value at 72 day fermentation and then decreased slowly. ADP, AMP and IMP tended to degrade rapidly while hypoxanthine increased remarkably at 27 day fermentation but slightly decreased at 72 day fermentation. It is presumed that the characteristic flavor of fermented small shrimp might be attributed to the relatively higher content of hypoxanthine. In the free amino acid composition of fresh small shrimp abundant amino acids were proline, arginine, alanine, glycine, lysine, glutamic acid, leucine, valine and threonine in order. Such amino acids like serine, methionine, isoleucine, phenylalanine, aspartic acid, tyrosine and histidine were poor. In small shrimp extract, proline, arginine, alanine, glycine, lysine and glutamic acid were dominant holding $18.5\%,\;14.6\%,\;10.8\%,\;8.7\%,\;8.1\%\;and\;7.7\%$ of total free amino acids respectively. The total free amino acid nitrogen in fresh small shrimp was $63.9\%$ of its extract nitrogen. The change of free amino acid composition in the extract of small shrimp during fermentation was not observed. Lysine, alanine glutamic acid, proline, glycine and leucine were abundant in both fresh sample and fermented products. The increase of total free amino acids during 72 day fermentation reached approximately more than 2 times as compared with that of fresh sample and then decreased slowly. Fermented small shrimp with $40\%$ of salt was too salty to be commercial quality as the results of organoleptic test showed. It is found that 72 day fermentation with $20\%\;and\;30\%$ of salt gave the most favorable flavor. It is convinced that the characteristic flavor of fermented small shrimp was also attributed to such amino acids as lysine, proline, alanine, glycine and serine known as sweet compounds, as glutamic acid with meaty taste, and as leucine known as bitter taste. The amount of betaine increased during fermentation and reached the maximum at 72 day fermentation and then decreased slowly TMA increased while TMAO decreased during fermentation. The amount of TMAO nitrogen in fermented small shrimp was $200mg\%$ on moisture and salt free base. Betaine and TMAO known as sweet compounds were abundant in fermented small shrimp. It is supposed that these compounds could also play a role as important taste compounds of fermented small shrimp. At the initial stage of fermentation, Achromobacter, Pseudomonas, Micrococcus denitrificans which belong to marine bacteria were isolated. After 40 day fermentation, they disappeared rapidly while Halabacterium, Pediococcus, Sarcian, Micrococcus morrhuae and the yeasts such as Saccharomyces sp. and Torulopsis sp. dominated. It is concluded that the most important taste compounds of fermented small shrimp were amino acids such as lysine, proline, alanine, glycine, serine, glutamic acid, and leucine, betaine, TMAO and hypoxanthine.

유산균 발효가 흰점박이꽃무지(Protaetia brevitarsis) 유충의 항산화활성에 미치는 영향 (Antioxidant Activities of Protaetia brevitarsis Larvae Fermented by Lactobacillus acidophilus)

  • 본 연구에서는 유산균 발효가 큰느타리버섯 수확후배지를 식이한 흰점박이꽃무지 유충의 항산화 활성에 미치는 영향을 평가하기 위하여 흰점박이꽃무지 유충을 유산균으로 발효시키고, 흰점박이꽃무지 유충(PLE)과 발효 흰점박이꽃무지 유충(FPLE) 추출물의 항산화 활성을 비교하였다. 추출물 PLE와 FPLE의 총 폴리페놀 함량은 각각 65.02±1.32 mg GAEs/extract g과 93.33±0.98 mg GAEs/extract g이었고, 플라보노이드 함량은 각각 18.3±1.57 mg QEs/extract g과 17.69±0.95 mg QEs/extract g이었다. 추출물 PLE와 FPLE의 DPPH에 의한 라디칼소거 활성은 2-4 mg/ml의 농도에서는 두 시료 간의 유의적 차이를 나타나지 않았지만, 8mg/ml 이상의 농도에서는 추출물 PLE에 비해 FPLE의 DPPH 라디칼 소거능이 우수하였다. 추출물 PLE와 FPLE의 환원력도 추출물 PLE에 비해 FPLE이 우수하였으며 1.6 mg/ml 이상의 농도에서는 PLE에 비해 FPLE의 환원력이 2배 이상 높게 나타났다. 추출물 PLE와 FPLE의 ORAC 지수는 각각 74.34±0.37 uM TEs/extract g과 79.22±0.72 uM TEs/extract g으로 추추물 PLE에 비해 FPLE의 ORAC 지수가 높게 나타났고, RAW 264.7 세포에 대한 추출물의 세포독성은 나타나지 않았다. 이상의 결과를 종합하면, 큰느타리버섯 수확후 배지를 식이한 흰점박이꽃무지 유충은 유산균 발효에 의해 항산화물질인 총 폴리페놀 함량 및 항산화 활성이 증가된 것을 확인할 수 있었으며, 발효법은 흰점박이꽃무지 유충을 식의약품 소재로 개발하는 과정에 필요한 전처리 과정이라고 판단된다.

  • 김치제조에 널리 쓰이는 젓갈류 즉, 멸치젓갈(액젓, 저염 액젓, 육젓), 새우젓갈 및 저염 까나리액젓과 젓갈 대용물로서 굴 가수분해물, 명태육 가수분해물 및 청각추출물을 첨가하여 제조한 김치를 $20^{\circ}C,\;10^{\circ}C$$4^{\circ}C$에서 각각 숙성시키면서 숙성에 따른 아미노질소의 함량과 ACE 저해효과를 살펴보았다. 그 결과, 젓갈류 첨가구의 경우, 아미노질소의 함량과 ACE 저해효과는 모든 숙성온도에서 숙성됨에 따라 어느 정도 증가한 후 대체로 일정한 수준을 유지하는, 유사한 경향을 보였다. 젓갈 대용물 첨가구의 경우, 아미노질소는 모든 숙성온도에서 각 시험구별 숙성에 따른 큰 변화 없이 $400{\sim}600\;mg/100\;g$의 함량을 보였으며 ACE 저해효과는 숙성 초기인 $1{\sim}2$일 사이에 급격한 상승 후 완만히 증가하는 경향을 보였다. 첨가된 젓갈 및 젓갈 대용물 중에서는 저염 멸치액젓 첨가구가 모든 숙성온도에서 아미노 질소의 함량(>600 mg/100 g)과 ACE 저해효과 (>80%)가 비교적 높았다. 그러나, 젓갈 대용물도 ACE 저해효과 면에서 젓갈류에 못지 않은 좋은 효과를 보임으로써 ACE 저해 기능성 김치용 조미소재로 젓갈류와 함께 이용 가능할 것으로 사료된다.

  • 김치제조에 널리 쓰이는 젓갈류 즉, 멸치젓갈(액젓, 저염 액젓, 육젓), 새우젓갈 및 저염 까나리액젓과 젓갈 대용물로서 굴 가수분해물, 명태육 가수분해물 및 청각 추출물을 첨가하여 제조한 김치를 $20^{\circ}C,\;10^{\circ}C$$4^{\circ}C$에서 각각 숙성시키면서 숙성에 따른 아질산염의 함량과 그 분해효과를 살펴보았다. 그 결과, 젓갈류 첨가 김치의 경우, 아질산염 함량은 $20^{\circ}C$ 숙성에서 새우젓갈 및 멸치액젓 첨가구가 한 때 증가하였으나 그 후 다시 감소하였으며 $10^{\circ}C$$4^{\circ}C$의 경우도 대체로 감소하였다(<5 ppm). 젓갈 대용물 첨가 김치의 경우, 아질산염 함량은 숙성초기에 크게 감소한 후 숙성됨에 따라 일정하게 낮은(<2 ppm) 함량을 유지하였다. 각 시험구의 아질산염 분해효과는 젓갈류 첨가구가 다소의 변동을 보였으나 숙성온도에는 크게 영향을 받지 않으면서 대체로 일정한 수준을 유지하였다. 젓갈류 중에서는 저염 멸치액젓 첨가구가, 젓갈대용물 중에서는 굴가수분해물 첨가구가 90% 내외의 효과를 보여 타 시험구 $(70{\sim}80%)$보다 높은 아질산염 분해효과를 보였다.

김치의 숙성과 칼슘함량에 미치는 조개류 껍질 물추출물 첨가효과 (Effect of Water Extracts of Shellfish Shell on Fermentation and Calcium Content of Kimchi)

  • 천연의 칼슘자원인 조개류 껍질 물추출물을 사용하여 보존성이 높고 체내 흡수력이 양호한 젖산칼슘의 함량이 높은 김치의 제조를 시도하였다. 시료는 꼬막, 맛조개, 모시조개, 바지락, 굴 및 소라껍질의 물 추출물을 사용하였으며 절임배추에 대하여 5%를 첨가하여 1$0^{\circ}C$에서 숙성시켰다. 조개류 껍질의 칼슘함량은25.57~38.78%이었다. 조개류 껍질 물추출물을 첨가한 김치의 pH는 전 숙성 기간을 통하여 대조구보다 높았으며 산도는 특히 숙성 후기 에 낮았다. 총 균수는 조개 껍질 물추출물 첨가구가 대조구보다 낮았으나 젖산균수는 첨가구에서 높았다. 숙성 7일 이후의 조회분 함량은 첨가구 (3.31~4.99%)가 대조구(2.75~3.01%)넓다 높았다. 칼슘의 함량은 첨가구(300.2~376.0 mg%)가 대조구(69.9~95.0mg%) 보다 높았으며, 그 중에서도 굴 껍질과 모시조개 껍질 물추출물을 첨가한 김치에서 높았다. 조개류 껍질 물추출물을 첨가한 김치는 대조구에 비하여 아삭아삭한 조직감(crispiness)이 높은 반면 산미는 약했으며, 종합적 인기호도는 담금일 에는 차이를 보이지 않았으나 숙성 14일 이후에는 전반적으로 첨가구에서 높았다.
