Antioxidant and Anti-Diabetic Activities of Soybean Fermented with Monascus (홍국발효 대두의 항산화 및 항당뇨 활성)

  • This study was performed to investigate the antioxidant and antidiabetic activities of soybean fermented with Monascus. Also, the changes in the content of isoflavones and Monacolin K were analyzed. It was observed that the glycoside forms of daidzin and genistin were converted to aglycones of daidzein and genistein within 6 days of fermentation. The product can be used as a health functional material that can increase bioavailability. Monacolin K production was found to increase significantly with the progression of fermentation with an increase to 0.04 mg/g and 0.44 mg/g on 6 and 12 days of fermentation, respectively. The DPPH radical scavenging activity of soybean fermented with Monascus was significantly increased compared to that of soybean. The protein expression of inflammation-related genes (TNF-α, IL-6, and COX-2) in the MIN cell was significantly increased in the presence of alloxan compared to the normal group, but a decrease was observed in the presence of soybean fermented with Monascus. In conclusion, soybean fermented with Monascus showed the highest antidiabetic and antioxidant effects. These results suggest that soybean fermented with Monascus has the potential to be used as a beneficial ingredient with antidiabetic and antioxidant effects.

Antimutagenic Effects on Aflatoxin $B_1$ of Soybean Pastes Fermented by Bacillus Strains

  • Antimutagenic effects of methanol extracts of various soybean pastes against aflatoxin B1 were examined using Salmonella typhimurium TA98 and TA100. Antimutagenic activities of boiled soybeans, Japanese Miso, traditional Korean soybean pastes, soybean pastes fermented by wild type strains, and soybean pastes fermented by mutants, transformants, and cell fusants were 53.6 to 54.6%, 73 to 79.7%, 78.3 to 95.7%, 85 to 97.1%, 71.9 to 78.3%, 65.5 to 77.7%, and 73.4 to 79.0%, respectively. Soybean pastes fermented by wild type strains showed higher activities than those fermented by mutant, transformant, and cell fusant strains.

  • Yeast strains are capable of hydrolyzing non-digestible saccharides, such as melibiose, raffinose, and stachyose, found in soy meal components. This study revealed the biochemical properties of fermented soybean meal during 72 hours with kefir. Starchyose and raffinose, non-digestible components, were almost digested in kefir 150 mL + soybean meal 500 g + water 70 mL and galactose was produced. Proteolysis of the soybean meal produced most of the small molecule peptides in kefir 150 mL + soybean meal 500 g + water 70 mL. The production of the vitamin B group and C were the highest in kefir 250 mL + soybean meal 500 g. The yeast number of the fermented soybean meal was 7.0 × 106 CFU·mL-1 which was the highest in kefir 250 mL + soybean meal 500 g. The lactic acid bacteria of the fermented soybean meal was the highest at 3.5 × 109 CFU·mL-1 in kefir 70 mL + soybean meal 500 g. The antioxidant effect was the highest at 57% in kefir 250 mL + soybean meal 500 g. Expression of inflammation-related cytokine (interleukin [IL]-1β, tumor necrosis factor [TNF]-α, and interleukin [IL]-6) was significantly inhibited in fermented soybean meals with different treatments. These results suggest that fermented soybean meal by kefir has an antiinflammatory and anti-oxidation activity and could be utilized in feed manufacturing, and inhydrolyzing non-digestible soy meal components.

Bacterial Diversity in a Korean Traditional Soybean Fermented Foods (Doenjang and Ganjang) by 16S rRNA Gene Sequence Analysis

  • The bacterial diversity in Korean soybean-fermented foods was investigated using a PCR-based approach. 16S rRNA sequences were amplified and cloned from two different soybean-fermented foods such as doenjang (soybean paste), and ganjang (soybean sauce). Staphylococcus equorum (60.6%), Tetragenococcus halophila (21.2%), Leuconostoc mesenteroides (9.1%), Lactobacillus sakei (6.1%), and Bacillus subtilis (3.0%) were detected among clones isolated from soybean paste samples and Halanaerobium sp. (37.5%), Halanaerobium fermentans (37.5%), T. halophila (12.5%), Staphylococcus sp. (6.3%), S. equorum (3.1%), and B. subtilis (3.1%) were detected among clones isolated from soybean sauce. Our approach revealed different bacterial distributions and diversity from those previously obtained using culture-dependent methods.

Manufacturing and Quality Characteristics of the Doenjang made with Aspergillus oryzae Strains Isolated in Korea (국내에서 분리된 황국균을 활용한 된장 제조 및 특성 분석)

  • This study was conducted to evaluate the possible utility of 3 Aspergillus oryzae strains (designated as SCF-6, SCF-37, and JJSH-1), isolated from Korean traditional fermented materials, as starter cultures in the soybean paste industry. Doenjang (fermented soybean paste) was made with the A. oryzae strains described above, and its quality attributes were analyzed during a 60-day aging period. No significant differences in pH, moisture, or salt content were detected among the doenjang varieties made with the 3 Aspergillus strains. The concentration of amino-nitrogen, an indicator of doenjang aging, increased in each sample during the aging period. After the 60-day aging period, the contents of amino-nitrogen and free amino acid in doenjang made with SCF-6 showed the highest concentrations among the tested doenjang products: 971.6 and 8,064.9 mg%, respectively. Measurements of the color of doenjang showed that lightness and yellowness decreased during the aging period, but redness increased. After the 60-day aging period, the ${\gamma}$-aminobutyric-n-acid (GABA) concentrations in doenjang made with SCF-6 and SCF-37 were 61.3 and 53.7 mg%, respectively. In doenjang samples, aflatoxin was not detected and the concentrations of biogenic amines (histamine and tyramine) were 2.55-5.60 mg/kg and 3.70-5.87 mg/kg, respectively. These results indicated that A. oryzae SCF-6 isolated from traditional fermented foods could be useful as a starter culture in the soybean paste industry.

The Quality of Doenjang (Soybean Paste) Manufactured with Bacillus brevis (Bacillus brevis 로 제조한 된장의 품질)

  • We investigated the quality of soybean paste(Doenjang) fermented by BAcillus brevis. The results obtained were as follows : soybean paste fermented by Bacillus brevis had alkaline pH and yellow ochre color. Dextrinizing activity was about 98 D.P. unit from 5th to 25th day of fermentation at 3$0^{\circ}C$ and after that day somewhat decreased . Saccharifying activity was respectively 6.1, 7.2, 6.8, 6.4 S.P. unit on 5, 15, 25 and 35th day of fermentation. Protease activity suddenly increased after 15th day of fermentation and was 250, 275, 299 unit on15, 25, 35 th day of fermentation , respectively. The most abundant free amino acid was found to be glutamic acid (561.8mg%) in soybean paste fermented by Bacillus brevis. In case of free sugar and non-volatile organic acid, fructose and oxalic acid showed highest content of 10.25mg% on 25th day and 12.20mg% on 15th day. The contents of free amino acids, free sugars, organic acids in soybean paste fermented by B.brevis were most abundant after 25 days of fermentation and this results were similar to that of traditional soybean paste. The odor of soybean paste fermented by Bacillus brevis was improved to be a nice soybean paste odor on 25th day of fermentation. However, sensory evaluation value of the taste of it decreased after 10 th day of fermentation.

Activity of Superoxide dismutase(SOD) by fermented soybean (발효 대두 식품의 Superoxide dismutase(SOD) 활성)

  • This study was performed to evaluate the inhibition effects of fermented soybean on lipid perosidation and antioxidative relative enzyme activity. in vivo. Fermented soybean was induced the high SOD activity, while significantly inhibited on the peroxide value of linoleic acid and lipid perxidation from rat microsome induced by Fe$^{2+}$ ascorbate system, Sprague-Dawley(SD) male rats were fed basic diet, and experimental diets group added 200 or 500 mg/kg fermented soybean for 2 weeks. The effect of fermented soybean is also significantly increased catalase and glutathione peroxidase activities, while significantly inhibited the lipid peroxidation of rat liver microsome in a dose dependent manner. Therefore, these results suggest that fermented soybean has antioxidative activity which is related enzyme to prevention of oxidative stress.s.

Production of Bioactive Components and Anti-Oxidative Activity of Soybean Grit Fermented with Bacillus subtilis HA according to Fermentation Time (Bacillus subtilis HA를 이용한 soybean grit의 고체발효 기간에 따른 생리활성물질 생산 및 항산화 효과)

  • Soybean grits, fortified with various bioactive components, were produced by solid-state fermentation using Bacillus subtilis HA. ${\alpha}$-Amylase activity gradually increased during fermentation over 5 days. Fibrinolytic and protease activities were highest in the soybean grits fermented for 7 days. The grits fermented for 5 days also showed the highest tyrosine content, indicating a higher peptide content. Peptides of low molecular weight (below 3,000 daltons) and browning pigments increased with increasing fermentation time. The fermented soybean grits showed higher contents of total phenolic compounds, to approximately 18 mg/g. DPPH free radical scavenging effects were higher in the soybean grits fermented for 3 days. Also, ABTS radical scavenging effects were greater in the fermented grits compared to the unfermented grits. Overall, the soybean grits fermented by solid-state fermentation for 5 days showed enhanced production of bioactive compounds and greater antioxidant properties.

Traditional Fermented Food Products in Korea (한국의 발효식품에 관하여)

  • Fermented foods available in Korea may be classified into four groups, namely, fermented soybean products, fermented cereal products, fermented vegetable products, and fermented fishery products based on raw materials used. The representative fermented foods based on soybean are Kanjang(soysauce), Doenjang(fermented soybean pastes), and Kochujang(red pepper added-fermented soybean paste). Such fermented products are made using Meju(functioning as a starter and prepared by fermentation of steamed soybean mash) as an essential ingredient, and used widely as a soup base and/or in seasoning side-dishes for everyday meals year around. Excepting Sikhae, all fermented products based on rice and other cereal grains are of alcoholic nature. Takju (Makgeolli) used to be made using rice as the major raw material, however, mainly due to the shortage of rice in recent years, other cereals, such as, barely, corn, and wheat flours are also used to replace rice today. Owing to such changes in the raw materials. the popularity of Takju has been somewhat reduced, yet it is still widely consumed in rural areas. Although Chungju is a popular rice wine with superior qualify over Takju, the amount consumed is considerably limited. The highest qualify rice wine, Bupju, in particular, is made by a low temperature fermentation using glutinous rice. Kimchi is an unique fermented vegetable product of long tradition in Korea. Although it was for consumption mainly in winter season serving as a source of vitamins, today it is widely used throughout the year. Except Kkakdugi and Dongchimi, all of the fermented vegetable products contain salted Korean cabbage as an essential item, while they abound in varieties depending on material composition and methods of processing, and also on seasons and localities Next to Kimchi in this category is Kkakdugi made of raddish in popularity and quantity consumed. The four groups of fermented food products described above are reviewed in some detail and evaluated in terms of their nutritional significances, processes and microorganisms involved. and their commercial potentials. Jeotkal (or Jeot) is a name given to all fermented products of fishery origin. A number of Jeot can be prepared by adding salt and allowing fermentation to the raw materials such as shrimp, anchovy octopus, clam, oyster, etc.

Effects of fermented soybean meal on growth performance, nutrients digestibility, blood profile and fecal microflora in weaning pigs

  • A nutrition study on weaned pigs using fermented soybean meal was done to determine the effect on growth performance, nutrients digestibility, blood profile and fecal microflora. A total of 100 weaning pigs with an initial average body weight (BW) of 8.27 ± 1.10 kg were randomly allotted into 1 of 2 dietary treatments in a 6-week feeding trial. There were 10 replicate pens in each treatment with 5 pigs per pen. The dietary treatments included: 1) control: Basal diet (CON); 2) fermented soybean meal (FSBM): Basal diet supplemented with 5% fermented soybean meal. The average daily feed intake (ADFI) was significantly improved (p < 0.05) with the dietary supplementation of the FSBM compared with the control meal during phase 2. The dietary supplementation with 5% FSBM had a significant effect (p < 0.05) on gain to feed ratio (G : F) during the overall experiment period. Collectively, the results of this study indicate that dietary supplementation of 5% fermented soybean meal improved the body weight and average daily gain (ADG), ADFI, and feed efficiency of the weaning pigs; however, there were no supplementation effects on total tract digestibility of dry matter (DM), nitrogen, energy, blood profile and fecal microflora.
