The effects of the nonionic surfactant Tween 80 and a mixture of fibrolytic enzymes on total tract digestion, in situ disappearance (ISD) and ruminal fermentation characteristics of orchardgrass hay and barley grain were investigated in a 4${\times}$4 Latin square experiment with 4 non-lactating Holstein cows and 4 diets in 4 periods. Cows were offered a total mixed ration of 50% rolled barley grain and 50% orchardgrass hay treated with either 1) water (control), 2) 0.2% (vol/wt) Tween 80, 3) 0.2% (vol/wt) hydrolytic enzyme, or 4) 0.2% hydrolytic enzyme plus 0.2% Tween 80. Total tract digestibility coefficients of DM, nitrogen, NDF and ADF were not affected (p>0.05) by dietary treatment. Compared to the control, the rate of ISD of DM from orchardgrass hay was faster (p<0.05) in cows receiving diets treated with the enzyme alone or with enzyme plus Tween 80 (0.06/h vs. 0.076 and 0.069/h). The rate of digestion was lower (p<0.05) as compared to control when barley grain was treated with these additives. Ruminal fluid pH and concentrations of total VFA, acetate, isobutyrate and butyrate were not affected (p>0.05) by treatments. Cows that consumed diets treated with enzyme plus Tween 80 had higher (p<0.05) ruminal concentrations of propionate and isovalerate, and lower (p<0.05) acetate:propionate ratios. Compared to the control, microbial protein synthesis tended (p = 0.13) to increase with the addition of enzyme to the diet while nonammonia nitrogen flow to the duodenum increased (p<0.05) with both enzyme and Tween 80 treatments. The study indicated that fibrolytic enzymes alone or in combination with Tween 80 could enhance ISD of orchardgrass hay and ruminal concentrations of propionate, valerate and iso-valerate, but do not affect total tract digestibility.
Effects of particle size and physical effective fibre (peNDF) of rice straw in diets on chewing activities, feed intake, flow, site and extent of digestion and rumen fermentation in goats were investigated. A 4${\times}$4 Latin square design was employed using 4 mature Liuyang black goats fitted with permanent ruminal, duodenal, and terminal ileal fistulae. During each of the 4 periods, goats were offered 1 of 4 diets that were similar in nutritional content but varied in particle sizes and peNDF through alteration of the theoretical cut length of rice straw (10, 20, 40, and 80 mm, respectively). Dietary peNDF contents were determined using a sieve for particle separation above 8 mm, and were 17.4, 20.9, 22.5 and 25.4%, respectively. Results showed that increasing the particle size and peNDF significantly (p<0.05) increased the time spent on rumination and chewing activities, duodenal starch digestibility and ruminal pH, and decreased ruminal starch digestibility and $NH_{3}$-N concentration. Intake and total tract digestibility of nutrients (i.e. dry matter, organic matter, and starch) and ruminal fermentation were not affected by the dietary particle size and peNDF. Increased particle size and peNDF did not affect ruminal fibre digestibility, but had a great impact on the intestinal and total tract fibre digestibility. The study suggested that rice straw particle size or dietary peNDF was the important influential factor for chewing activity, intestinal fibre and starch digestibility, and ruminal pH, but had minimal impact on feed intake, duodenal and ileal flow, ruminal and total tract digestibility, and ruminal fermentation.
Four rumen fistulated swamp buffaloes were randomly assigned according to a $4{\times}4$ Latin square design to investigate the effects of Eucalyptus (E. Camaldulensis) leaf meal (ELM) supplementation as a rumen enhancer on feed intake and rumen fermentation characteristics. The dietary treatments were as follows: T1 = 0 g ELM/hd/d; T2 = 40 g ELM/hd/d; T3 = 80 g ELM/hd/d; T4 = 120 g ELM/hd/d, respectively. Experimental animals were kept in individual pens and concentrate was offered at 0.3% BW while rice straw was fed ad libitum. The results revealed that voluntary feed intake and digestion coefficients of nutrients were similar among treatments. Ruminal pH, temperature and blood urea nitrogen concentrations were not affected by ELM supplementation; however, ELM supplementation resulted in lower concentration of ruminal ammonia nitrogen. Total volatile fatty acids, propionate concentration increased with the increasing level of EML (p<0.05) while the proportion of acetate was decreased (p<0.05). Methane production was linearly decreased (p<0.05) with the increasing level of ELM supplementation. Protozoa count and proteolytic bacteria population were reduced (p<0.05) while fungal zoospores and total viable bacteria, amylolytic, cellulolytic bacteria were unchanged. In addition, nitrogen utilization and microbial protein synthesis tended to increase by the dietary treatments. Based on the present findings, it is suggested that ELM could modify the rumen fermentation and is potentially used as a rumen enhancer in methane mitigation and rumen fermentation efficiency.
Ten male crossbred (Thai $Native{\times}Anglo$ Nubian) goats with average live weight of $27{\pm}2$ kg were randomly assigned according to a $5{\times}5$ replicated Latin square design to receive five diets, $T_1$ = concentrate with 0% cassava chip (CC), $T_2$ = 25% CC, $T_3$ = 50% CC, $T_4$ = 75% CC and $T_5$ = 100% CC. Fresh elephant grass (FEG) was offered ad libitum as the roughage. A metabolism trial lasted for 21 days during which liveweight changes and feed intakes were measured. Based on this experiment, there were no significant differences (p>0.05) among treatment groups regarding DM intake and digestion coefficients of nutrients (DM, OM, NDF and ADF), except for $T_5$ (100% CC) which was lowest (p<0.05) in digestion coefficient of CP than $T_1$ and $T_3$. Rumen parameters (ruminal pH, $NH_3$-N and volatile fatty acids), blood urea nitrogen, blood glucose and packed cell volume were similar among treatments. Moreover, rumen microorganism populations were not affected (p>0.05) by cassava inclusion. The amount of N absorption and retention were similar among treatments, except for $T_5$ which tended to be slightly lower. Based on this experiment, it could be concluded that the optimal level of cassava inclusion to replace corn in goat diets was in the range of 25-75% of CC when fed with FEG and it was a good approach in exploiting the use of local feed resources for goat production.
This study evaluated microbial risk that could develop within soil microbial communities after amended with organic fertilizers from stabilized swine manure waste. For this purpose, we assessed the occurrences and competitiveness of antibiotic resistance and pathogenicity in soil microbial communities that were amended with swine manure wastes stabilized by a traditional lagoon fermentation process and an autothermal thermophilic aerobic digestion process, respectively. According to laboratory cultivation detection analysis, soil applications of the stabilized organic fertilizers resulted in increases in absolute abundances of antibiotic resistant bacteria and of two tested pathogenic bacteria indicators. The increase in occurrences might be due to the overall growth of microbial communities by the supplement of nutrients from the fertilizers. Meanwhile, the soil applications were found to reduce competitiveness for various types of antibiotic resistant bacteria in the soil microbial communities, as indicated by the decrease in relative abundances (of total viable heterotrophic bacteria). However, competitiveness of pathogens in response to the fertilization was pathogens-specific, since the relative abundance of Staphylococcus was decreased by the soil applications, while the relative abundance of Salmonella was increased. Further testes revealed that no MAR (multiple antibiotic resistance) occurrence was detected among cultivated pathogen colonies. These findings suggest that microbial risk in the soil amended with the fertilizers may not be critical to public health. However, because of the increased occurrences of antibiotic resistance and pathogenicity resulted from the overall microbial growth by the nutrient supply from the fertilizers, potential microbial risk could not be completely ruled out in the organic-fertilized soil samples.
This trial was to compare the digestive function of laboratory animals, which included omnivores (rats) and herbivores (rabbits, guinea pigs and hamsters). Ten weanling rats, rabbits, guinea pigs and hamster were randomly allocated in individual metabolic cages for a performance and a digestibility trial. Results showed that guinea pigs were significantly best in digestion of dietary crude protein and fiber. Rabbits, however, had the poorest nutrient digestion among the herbivores. Crude fiber digested significantly less by rats than by the herbivores (p<0.05). The digesta retention in the GI tract was longest in rabbits followed by guinea pigs, rats and hamsters. The relative length of the large intestine was significantly shorter in rats as compared to guinea pigs or hamsters. Both the weight and length ratio of the caecum or colon-rectum to the large intestine reflected the major site for fermentation that was the caecum in rabbits and the colon-rectum in guinea pigs. 10% of crude fiber diet did not result in damaged mucosa in any of the experimental animals in this trial.
Due to its carbon-neutral nature, biogas generated from anaerobic digestion or fermentation of biodegradable wastes is one of the important renewable energy sources to reduce global warming. It is mainly composed of methane and various inert gases such as $CO_2$ and $N_2$, and the actual composition of biogas significantly varies depending on the origin of anaerobic digestion process. Therefore, in order to effectively utilize this fuel as an energy source for electricity, it is important to develop power generation engines which can successfully apply biogas with significant composition variations. In this study, efforts have been made to develop a diesel-biogas duel fuel engine as a way to achieve such a stable power generation. The effects of diesel fuel injection quantity and pressure on stable combustion and engine performance were investigated, and an impact of diesel fuel atomization was discussed. The engine test results show that there exists a 2 stage combustion which consists of diesel pilot fuel burning and premixed biogas/air mixture burning in dual fuel engine operation and optimum diesel injection parameters were suggested for biogases with various compositions and heating values.
Four Murrah male buffalo calves with an average body weight of $188{\pm}1.6kg$ each fitted with rumen and abomasal cannula were subjected to defaunation followed by refaunation. The animals were offered wheat straw and a concentrate mixture. There was no difference in dry matter, starch and nitrogen intake in defaunated and refaunated buffalo calves. Production of ruminal total volatile fatty acid and acetate : propionate ratio decreased (p < 0.01) whereas, molar proportion of propionate increased (25.8 Vs 19.4% p < 0.01) in defaunated animals. Fermentation of starch in rumen increased (73.9 Vs 65.8%, p < 0.01) but in small intestine decreased (20.2 Vs. 28.2%, p < 0.05) in defaunated calves. The flow of non ammonia nitrogen (NAN) to abomasum (75.1 vs 68.6 g/d, p < 0.01) and its digestion in small intestine (37,6 vs 32.5 g/d, p < 0.01) was improved due to defaunation. However, No difference in the total tract digestibility of starch and nitrogen was found in defaunated and refaunated buffalo calves.
This study was performed to optimize the integrated anaerobic digestion (AD) and microbial electrolysis cells (MECs) for the enhanced hydrogen production. The optimum operational conditions of integrated AD and MECs were obtained using response surface methodology. The optimum substrate concentration and operational pH were 10 g/L and 6.8, respectively. In the confirm test, 1.43 mol $H_2/mol$ hexose was achieved, which was 2.5 times higher than only AD. After 40 to 60 hour at seeding, the volatile fatty acids (VFAs) in reactor of AD were not changed. However the VFAs of reactor of AD-MECs were reduced by 61.3% (acetate: 76.4%, butyrate: 50.0%, lactate: 55.0%).
This study was performed to evaluate the characteristics of start-up of anaerobic digestion from food waste with different inoculum ratios. The hydrogen yield was similar with different inoculum ratios. The hydrogen production rate increased with increasing inoculum ratio. But the specific hydrogen production rate decreased with increasing inoculum ratio. Total volatile fatty acids composition analysis showed that butyrate and acetate were the prevalent products in all reactors, followed by lactate and propionate. The acetate was most prevalent product in reactors at $X_0/S_0=0.080$ and 0.159. But in reactors at $X_0/S_0=0.239$ and 0.318, butyrate accounted for greater than 50% of the total volatile fatty acids.
