수 처리 방법이 근적외선분광법을 이용한 옥수수 사일리지의 화학적 조성분 및 발효품질의 예측 정확성에 미치는 영향 (Mathematical Transformation Influencing Accuracy of Near Infrared Spectroscopy (NIRS) Calibrations for the Prediction of Chemical Composition and Fermentation Parameters in Corn Silage)

  • 본 연구는 국내산 원물 옥수수 사일리지의 품질을 신속하게 분석 평가하기 위한 NIRS DB 구축과 원물시료의 분석 예측능력을 향상시키기 위한 근적외선 스펙트라의 적정 수 처리 방법을 구명하기 위하여 수행되었다. 옥수수 사일리지는 전국 사료작물 사일리지 품질경연대회에 출품된 시료와 2014년부터 2015년까지 전국 조사료 품질검사 시범사업에 참여한 조사료 생산경영체, 농축협 TMR회사 및 생산농가에서 407점을 수집하였다. 옥수수 사일리지의 품질평가를 위한 NIRS DB 구축을 위해 수집된 시료를 근적외선 분광기를 이용하여 스펙트라를 측정하고 측정된 시료는 실험실에서 화학적 분석을 실시하였다. 다양한 수 처리 방법에 따른 사료가치 및 발효품질의 예측정확성을 평가하기 위하여 원시 스펙트라를 미분처리하여 최적의 수처리 방법을 구명하였다. 옥수수 원물 사일리지의 수분함량 예측능력은 1차 미분처리(1, 16, 16)한 것으로 나타났으며 NDF와 ADF의 최적 수 처리는 두 성분 모두에서 2, 16, 16 처리가 예측 정확성이 가장 높게 나타났다. 조단백질 함량의 예측능력은 1차 미분처리(1, 4, 4)가 SECV 0.51과 $R^2{_{cv}}$ 0.72로 가장 우수한 예측능력을 나타내었다. 옥수수 사일리지의 발효산물인 산도(pH) 예측 정확성은 원물 스펙트라를 1차 미분처리(1, 8, 8)한 것으로 나타났으며 젖산과 초산의 예측능력은 2차 미분처리(2, 16, 16)에서 각각 SECV 0.81% 및 0.71%의 분석오차로 높게 나타났다.

  • This study investigated the effect of coated feed using mixed oil on rumen fermentation characteristics. Two experiments were conducted based on materials that were mixed. First, cashew nut and soybean oils were mixed with white mineral oil. And second, different plant extracts were mixed with white mineral oil. At first experiment, inclusion levels of mixed oil on diet (0.03%, 0.1%, 0.5%) were applied as variables. A coated diet was fermented with rumen inoculum according to in vitro rumen fermentation and its parameters were investigated. In the result of first experiment, no negative effects on rumen pH were found. Significantly decreased dry matter digestibility was detected at 0.5% treatment (P<0.05). Total gas productions in control and 0.03% were significantly greater than those of others (P<0.05). Significantly reduced methane productions were found in all treatments compared to the control (P<0.05). Inclusion of mixed oil did not affect on ammonia production. Total volatile fatty acid production was also not influenced by coating with mixed oils. Rumen fermentation parameters were greatly changed according to introduced plant extracts at second experiment. The significantly lowest and greatest ammonia productions were found at treatments with Ixeris dentata and Plantago asiatica, respectively (P<0.05). The significantly greatest acetate and propionate productions were detected at treatments with Crucuma longa and Zizyphus vulgaris 1, respectively (P<0.05). All treatments, except Chrysanthemum idicum, Euyale ferox seed, Moringa leaf and fruit and Zizyphus vulgaris 1, showed significantly increased total volatile fatty acid production compared to the control (P<0.05). Only Paeonia lactiflora showed significantly lesser gas production than the control (P<0.05). In methane production, Ceramium, Zizyphus, Paeonia, Agrimonia, Torilis, Mugwort, Foeniculum, Euphorbia, Taraxacum, Artemisia, Momordica, Curcuma and Moringa reduced methane significantly compared to the control (P<0.05).

Prediction of Chemical Composition and Fermentation Parameters in Forage Sorghum and Sudangrass Silage using Near Infrared Spectroscopy

  • This study was conducted to assess the potential of using NIRS to accurately determine the chemical composition and fermentation parameters in fresh coarse sorghum and sudangrass silage. Near Infrared Spectroscopy (NIRS) has been increasingly used as a rapid and accurate method to analyze the quality of cereals and dried animal forage. However, silage analysis by NIRS has a limitation in analyzing dried and ground samples in farm-scale applications because the fermentative products are lost during the drying process. Fresh coarse silage samples were scanned at 1 nm intervals over the wavelength range of 680~2500 nm, and the optical data were obtained as log 1/Reflectance (log 1/R). The spectral data were regressed, using partial least squares (PLS) multivariate analysis in conjunction with first and second order derivatization, with a scatter correction procedure (standard normal variate and detrend (SNV&D)) to reduce the effect of extraneous noise. The optimum calibrations were selected on the basis of minimizing the standard error of cross validation (SECV). The results of this study showed that NIRS predicted the chemical constituents with a high degree of accuracy (i.e. the correlation coefficient of cross validation ($R^2{_{cv}}$) ranged from 0.86~0.96), except for crude ash which had an $R^2{_{cv}}$ of 0.68. Comparison of the mathematical treatments for raw spectra showed that the second-order derivatization procedure produced the best result for all the treatments, except for neutral detergent fiber (NDF). The best mathematical treatment for moisture, acid detergent fiber (ADF), crude protein (CP) and pH was 2,16,16 respectively while the best mathematical treatment for crude ash, lactic acid and total acid was 2,8,8 respectively. The calibrations of fermentation products produced poorer calibrations (RPD < 2.5) with acetic and butyric acid. The pH, lactic acid and total acids were predicted with considerable accuracy at $R^2{_{cv}}$ 0.72~0.77. This study indicated that NIRS calibrations based on fresh coarse sorghum and sudangrass silage spectra have the capability of assessing the forage quality control

혐기성 발효에 의한 다시마 추출물로부터 휘발성 유기산 제조: 휘발성 유기산 생산성에 대한 환경적 영향인자 평가 (Volatile Fatty Acid Production from Saccharina japonica Extracts by Anaerobic Fermentation: Evaluation of Various Environmental Parameters for VFAs Productivity)

  • 본 연구에서는 거대 갈조류 대표종인 다시마(Saccharina japonica)로부터 물리화학적 전처리 방법, 미생물 접종비율, 다시마 추출물의 농도 및 pH 조건에 따른 휘발성 유기산(volatile fatty acids, VFAs) 생산 가능성 확인과 생산 효율을 평가하고자 하였다. 물리화학적 전처리 방법에 따른 휘발성 유기산의 최대 농도는 황산, 아임계수, 지질 추출 후 아임계수 전처리 순으로 나타났다. 황산 전처리 방법에서 미생물 접종비율(유효용적(WV)/미생물 부피(M) = 10~30), pH (6.0~7.0) 및 다시마 추출물의 농도(18.0~72.0 g/L)의 혐기성 발효 조건에 따른 휘발성 유기산 생성 농도에 미치는 영향을 확인한 결과, 발효 온도 $35^{\circ}C$, 미생물 접종비율 15, pH 7.0, 발효시간 372시간에서 다시마 추출물의 농도가 18.0, 36.0, 54.0, 72.0 g/L일 때, 휘발성 유기산의 최대 농도가 각각 9.8, 13.9, 18.6, 22.3 g/L로 확인되었다. 생산된 휘발성 유기산의 조성은 pH가 높을수록 아세트산과 프로피온산의 생산 비율이 높았으며, pH가 낮을수록 부티르산의 비율이 높게 확인되었다. 생산된 저농도의 휘발성 유기산은 농축 및 분리공정과 연계하여 향후 기초화학 원료와 바이오연료 등으로 사용될 수 있으므로, 기존 화석연료의 대체에너지 생산에 기여할 수 있을 것으로 기대된다.

  • The research described here was undertaken with the aim of monitoring, optimizing and ultimately controlling the production of heterofermentative microbes used as starters in the salami industry. The use of starter cultures in the fermented meats industry is a well-established technique used to shorten and standardize the ripening process, and to improve and control the organoleptic quality of the final product. Starter cultures are obtained by the submerged cultivation of suitable microorganisms in stirred, and sometimes aerated, fermenters where monitoring of key physiological parameters such as the concentration of biomass, substrates and metabolites suffers from the general lack of real-time measurement techniques applicable to aseptic processes. In this respect, the results of the present work are relevant to all submerged fermentation processes. Previous work on the application of on-line NIR spectroscopy to the lactic acid fermentation (Dosi et al. - Monreal NIR1995) had successfully used a system based on a measuring cell included in a circulation loop external to the fermenter. The fluid handling and sterility problems inherent in an external circulation system prompted us to explore the use of an in-line system where the NIR probe is immersed in the culture and is thus exposed to the hydrodynamic conditions of the stirred and aerated fluid. Aeration was expected to be a potential source of problems in view of the possible interference of air bubbles with the measurement device. The experimental set-up was based on an in-situ sterilizable NIR probe connected to the instrument by means of an optical fiber bundle. Preliminary work was carried out to identify and control potential interferences with the measurement, in particular the varying hydrodynamic conditions prevailing at the probe tip. We were successful in defining the operating conditions of the fermenter and the geometrical parameters of the probe (flow path, positioning, etc.) were the NIR readings were reliable and reproducible. The system thus defined was then used to construct and validate calibration curves for tile concentration of biomass, carbon source and major metabolites of two different microorganisms used as salami starters. Real-time measurement of such parameters coupled with the direct interfacing of the NIR instrument with the PC-based measurement and control system of the fermenter enabled the development of automated strategies for the interactive optimization of the starter production process.

Comparison of Rabbit Caecal Content and Rabbit Hard Faeces as Source of Inoculum for the In vitro Gas Production Technique

  • In order to find an alternative source of inoculum to caecal content for studying the fermentation activity of rabbit hindgut, caecal content and faeces of 25 hybrid Hyla rabbits were used as inocula for an in vitro gas production trial. About 1 g of three substrates (dehydrated alfalfa meal, dehydrated beet pulp, barley) was weighed, in quadruplicate per inoculum, in 120 ml bottles; 75 ml of anaerobic medium and 4 ml of reducing solution were added and bottles were placed at $39^{\circ}C$. Caecal content and faeces were diluted respectively 1:2 (CI) and 1:8 (FI) with anaerobic medium and were introduced in the respective bottles (10 ml). Gas production was recorded 20 times at 2-24 h intervals throughout fermentation (96 h). The fermentation characteristics (i.e. degraded organic matter, OMd; potential gas production, A; fermentation rate, Rmax; time at which it is reached, Tmax; pH, volatile fatty acid, VFA) were studied by inoculum and feedstuffs. The feedstuffs, according to their chemical composition, showed very different fermentation characteristics. In particular, OMd, A and Rmax allowed feedstuff classification as follows: barley>beet pulp>alfalfa. The inocula differ (p<0.05) in Tmax, were higher for CI (15.53 vs. 11.96 h) and in VFA production. In particular, CI produced higher levels of acetate (38.9 vs. 33.4 mM/g OM incubated, p<0.01) and isobutyrate (0.72 vs. 0.42, p<0.01) but less propionate (7.1 vs. 10.3, p<0.01) and butyrate (11.3 vs. 14.0, p<0.01). However, the trend of gas production, similar for the inocula according to the fermented substrate, and the good regression equation to estimate some caecal fermentation parameters from faeces suggest that, after standardisation, the faeces could be used as an alternative inoculum for gas tests in rabbit.

Effects of Polyurethane Coated Urea Supplement on In vitro Ruminal Fermentation, Ammonia Release Dynamics and Lactating Performance of Holstein Dairy Cows Fed a Steam-flaked Corn-based Diet

  • Three experiments were conducted to investigate the effects of polyurethane coated urea on in vitro ruminal fermentation, ammonia release dynamics and lactating performance of Holstein dairy cows fed a steam-flaked corn-based diet. In Exp. 1, a dual-flow continuous culture was run to investigate the effect of polyurethane coated urea on nutrient digestibility, rumen fermentation parameters and microbial efficiency. Three treatment diets with isonitrogenous contents (13.0% CP) were prepared: i) feedgrade urea (FGU) diet; ii) polyurethane coated urea (PCU) diet; and iii) isolated soy protein (ISP) diet. Each of the diets consisted of 40% steam-flaked corn meal, 58.5% forages and 1.5% different sources of nitrogen. PCU and FGU diets had significantly lower digestibility of NDF and ADF (p<0.01) than the ISP diet. Nitrogen source had no significant effect (p = 0.62) on CP digestibility. The microbial efficiency (expressed as grams of microbial N/kg organic matter truly digested (OMTD)) in vitro of the PCU diet (13.0 g N/kg OMTD) was significantly higher than the FGU diet (11.3 g N/kg OMTD), but comparable with the ISP diet (14.7 g N/kg OMTD). Exp. 2, an in vitro ruminal fermentation experiment, was conducted to determine the ammonia release dynamics during an 8 h ruminal fermentation. Three treatment diets were based on steam-flaked corn diets commonly fed to lactating cows in China, in which FGU, PCU or soybean meal (SBM) was added to provide 10% of total dietary N. In vitro $NH_3-N$ concentrations were lower (p<0.05) for the PCU diet than the FGU diet, but similar to that for the SBM diet at all time points. In Exp. 3, a lactation trial was performed using 24 lactating Holstein cows to compare the lactating performance and blood urea nitrogen (BUN) concentrations when cows were fed PCU, FGU and SBM diets. Cows consuming the PCU diet had approximately 12.8% more (p = 0.02) dietary dry matter intake than those consuming the FGU diet. Cows fed the PCU diet had higher milk protein content (3.16% vs. 2.94%) and lower milk urea nitrogen (MUN) concentration (13.0 mg/dl vs. 14.4 mg/dl) than those fed the FGU diet. Blood urea nitrogen (BUN) concentration was significantly lower for cows fed the PCU (16.7 mg/dl) and SBM (16.4 mg/dl) diets than the FGU (18.7 mg/dl) diet. Cows fed the PCU diet had less surplus ruminal N than those fed the FGU diet and produced a comparable lactation performance to the SBM diet, suggesting that polyurethane coated urea can partially substitute soybean meal in the dairy cow diet without impairing lactation performance.

호박을 첨가하여 제조한 고추장의 숙성 중 성분 변화 및 관능적 특성 (Sensory Evaluation and Changes in Physiochemical Properties, and Microflora and Enzyme Activities of Pumpkin-added Kochujang)

  • 순창식 식혜고추장의 담금 과정에서 식혜액 제조시 찹쌀의 일부를 건조완숙호박으로 대체하여(고추장 총 무게비로 1, 2, 5% 수준) 제조한 고추장을 $25^{\circ}C$에서 120일간 숙성시키면서 30일 간격으로 이화학적 특성 및 미생물 그리고 효소활성도의 변화를 측정하였으며 관능적특성은 숙성 120일째에 조사하였다. 숙성 중 고추장의 수분함량은 전반적으로 감소하였다. pH는 담금 초기부터 서서히 감소하다가 숙성 90일에 약간 상승하는 경향을 보였으나 그 후 다시 감소하였다. 적정산도는 숙성 중 서서히 증가하여 숙성 90일에 가장 높은 값을 나타내다가 숙성 120일에는 약간 감소하였다. 호박 첨가에 의해 고추장의 pH는 낮아지고 적정산도는 높아지는 것으로 나타났는데 이는 호박중의 유기산에 기인되는 것으로 판단된다. 숙성이 진행되는 동안 환원당 함량은 계속 증가하여 숙성 120일에는 대조구가 20.82%, 1% 호박첨가구가 21.34%, 2% 호박첨가구가 22.73% 그리고 5% 호박첨가구가 24.48%로 분석되었다. 아미노태 질소는 숙성 기간 중 서서히 증가하여 숙성 120일에 최고치를 나타내었다. 환원당과 아미노태 질소는 호박의 첨가량에 비례하여 높아졌으며, 에탄올 함량도 호박 첨가에 의해 증가하는 것으로 나타났다. 그러나 효모수나 세균수 그리고 효소 활성도는 호박 첨가에 의해 크게 영향을 받지 않았다. 관능적 특성에 있어 호박첨가는 고추장의 색에 좋지 않은 영향을 끼치는 것으로 나타났다. 냄새, 구수한 맛, 매운맛, 단맛, 짠맛 그리고 전체적인 기호도 등의 항목에서는 대조구, 1% 호박첨가구, 2% 호박첨가구 그리고 5% 호박첨가구 사이에서 통계적 유의성을 보이지는 않았으나 단맛의 경우에는 호박첨가량에 비례적하여 높아지는 경향을 보였고 구수한 맛의 경우에도 비슷한 경향을 보였으나 호박첨가량이 낮은 수준에서는 오히려 대조구에 비해 낮게 평가되는 등 다소 복잡한 양상을 보였다. 예상과는 달리 고추장의 전체적인 기호도는 호박첨가에 의해 향상되는 것으로 나타나지는 않았다.

흰쥐에서 가시오가피 발효물의 단회 및 반복투여 독성평가 (Single- and Repeated-dose Toxicities of Acanthopanax senticosus Fermentation Products in Rats)

  • 본 연구에서는 사용된 가시오가피 영지버섯 발효물과 가시오가피 상황버섯 발효물에 대한 열수추출물을 최고 2 g/kg의 농도로 단회 투여한 SD-rats에 체중 변화, 임상증상 등에서 어떠한 독성도 관찰할 수 없었으며, 1 g/kg 이하의 농도로 4주간 반복 투여한 SD-rats에서도 체중변화, 임상증상, 장기 무게, 혈액학적 성상, 혈액생화학적 성상 등에서 어떠한 독성과 비정상적인 소견을 관찰할 수 없었다. 따라서, 본 연구의 결과로 볼 때 가시오가피 영지버섯 발효물 열수추출물(FM-5111)과 가시오가피 상황버섯 발효물 열수추출물(FM-5131)은 본 연구에서 사용한 농도 내에서의 안전성을 확인할 수 있었다. 황 함유 아미노산의 결핍은 여러 가지 간질환 등의 증상을 야기한다(27). 또한, Choi와 Ahn(28)은 오가피 부위별 50% (v/v) 메탄올 추출액의 전자공여능은 줄기 추출물에서 90.21%, 뿌리 추출물에서 85.71%로 높은 활성을 보고하여 오가피의 항산화능을 규명하였다. 따라서, 본 연구에 사용된 시료 또한 항산화능과 같은 기능성을 가질것으로 사료되며, 추가 연구로 아미노산 분석을 통해 황 함유 아미노산의 함량 변화를 분석하고 간 기능 개선 효능을 평가하여 안전성이 확보된 기능성 식품소재 개발 연구를 진행할 계획이다.

Enhancing Butyrate Production, Ruminal Fermentation and Microbial Population through Supplementation with Clostridium saccharobutylicum

  • Butyrate is known to play a significant role in energy metabolism and regulating genomic activities that influence rumen nutrition utilization and function. Thus, this study investigated the effects of an isolated butyrate-producing bacteria, Clostridium saccharobutylicum, in rumen butyrate production, fermentation parameters and microbial population in Holstein-Friesian cow. An isolated butyrate-producing bacterium from the ruminal fluid of a Holstein-Friesian cow was identified and characterized as Clostridium saccharobutylicum RNAL841125 using 16S rRNA gene sequencing and phylogenetic analyses. The bacterium was evaluated on its effects as supplement on in vitro rumen fermentation and microbial population. Supplementation with $10^6CFU/ml$ Clostridium saccharobutylicum increased (p < 0.05) microbial crude protein, butyrate and total volatile fatty acids concentration but had no significant effect on $NH_3-N$ at 24 h incubation. Butyrate and total VFA concentrations were higher (p < 0.05) in supplementation with $10^6CFU/ml$ Clostridium saccharobutylicum compared with control, with no differences observed for total gas production, $NH_3-N$ and propionate concentration. However, as the inclusion rate (CFU/ml) of C. saccharobutylicum was increased, reduction of rumen fermentation values was observed. Furthermore, butyrate-producing bacteria and Fibrobacter succinogenes population in the rumen increased in response with supplementation of C. saccharobutylicum, while no differences in the population in total bacteria, protozoa and fungi were observed among treatments. Overall, our study suggests that supplementation with $10^6CFU/ml$ C. saccharobutylicum has the potential to improve ruminal fermentation through increased concentrations of butyrate and total volatile fatty acid, and enhanced population of butyrate-producing bacteria and cellulolytic bacteria F. succinogenes.
