Comparison of Free Sugar Content in Grains Fermented with Mycelia of the Basidiomycetes (담자균이 배양된 곡물의 유리당 조성 변화)

  • The grains were used as solid-substrate for the cultivation of basidiomycetes. The grains were fermented with Ganodema lucidum 7094, Fomitella fraxinea 81003, Phellinus igniarius 26005, and its free sugar composition was investigated. For the mass cultivation of mycelia, the hydrated grains with cold water were put into the plastic bottle. The mycelial growth rate in the bottled grains was high in the early stage with inoculation of homogenized mycelium. The activity of mycelium was maintained by adding sterilized water in the middle of cultivation. There was marked difference in the content of total free sugar and composition ratio of free sugar according to kinds of basidiomycetes. The content of free sugar increased far more in grains fermented with mycelium than in grains which was not fermented.

Studies on the Artificial Substrates with Rice Straw and the Spawning for Pleurotus florida in Korea (볏짚을 이용(利用)한 사철느타리버섯의 배지(培地) 제조(製造) 및 종균재식(種菌栽植)에 관한 연구(硏究))

  • This study was performed to examine the cultivation of Pleurotus florida that forms pinhead between 5 and $25^{\circ}C$, on rice straw substrates in Korea. The highest yield of P. florida sporophores was obtained from the fermented rice straw substrates added 5% rice bran to the rice straw by dry weight basis at the starting of fermentation. The pile turning of materials for uniform fermenting was made three times when the temperature of the pile reached $60^{\circ}C$ during out door composting. The $60^{\circ}C$ row steam was treated 6 hrs for pasterization of the substrates in the cultivating house. The quantity of spawn affected the mycelial growth and the yield of P. florida. The optimum amount of spawn was 10% to the rice straw by dry weight basis mixed with the substrates. P. florida produced normal sphorophores from September to middle of July of the following year.

Change of Chemical and Microbial Properties during Fermentation of Cotton Waste for Oyster Mushroom Cultivation (느타리 재배용 폐면 발효 중의 화학성 및 미생물 상의 변화)

  • The changes of microflora and chemical characteristics during fermentation process of cotton waste for oyster mushroom cultivation were investigated with 5 l bench-scale reactors placed in an incubator at different temperatures ($40,\;50\;and\;60^{\circ}C$). Cotton waste was wetted to 70% moisture, and air flow rates to the substrate were 50, 100 and 300 cc/min. In processing of composting, the mesophilic bacterial population decreased sharply but thermophilic bacterial population increased. In case of fungi, both mesophilic and thermophilic population decreased. The daily $CO_2$ evolution showed little difference in all treatments, while $NH_3$ dropped sharply after 3 days. The desirable composting temperature and air flow based on the mycelial growth of oyster mushroom were $50^{\circ}C$ and 100 cc/min, respectively.

Antifungal Activity of Streptomyces pad anus isolate TH04 against Monilinia fructicola, Brown rot Fungus on Stone-fruits (잿빛무늬병균에 대한 Streptomyces padanus isolate TH04의 항균활성)

  • The Streptomyces padanus isolate TH04, isolated from mummified peaches, showed strong antifungal activity to Monilinia fructicola. The inhibition activity of the isolate TH04 to mycelial growth and spore germination at 1% concentration of sub-antifungal powder made from culture suspension (CS) was ranged from 79.8% to 81.0% and from 73.9% to 75.8% to M. fructicola four strains, respectively. In the test of antifungal activity in mixed culture of the isolate and M. fructicola, inhibition rate was 7.5%, 86.8% and 94.0% in 0.01, 0.1, and 1% concentration of CS containing bacterial cell of the isolate, respectively. On apples (cultivar; Fuji), the control values of the isolate TH04 crude filtrates (0.1 and 1%) were 85.9% and 100%, respectively. The results suggest that the isolate TH04 indicate development possibility as biocontrol agent of brown rot caused by M. fructicola with the study on delivery method and fermentation condition to produce an antifungal compound.

Quality Characteristics of White Bread made with Flour of Wheat Cultured with Pleurotus ostreatus (Pleurotus ostreatus을 배양한 밀 분말 첨가가 식빵의 품질 특성에 미치는 영향)

  • This study was carried out to investigate the quality characteristics of white bread made with flour of wheat cultured with Pleurotus ostreatus(POW). The property of the wheat used for the solid-state cultivation of the Pleurotus ostreatus was examined. The hydration time of wheat with cold water was 10 hours, with a final moisture content of 39%, which was much less than the optimum moisture content for mycelial growth. However, hot water soaking reduced the required hydration time of the wheat, and elevated the moisture content to 65% within 90~120 min. The POW was composed of 7.2% moisture, 15.4% protein, 1.1% fat, 1.7% ash and 2.9% fiber. The POW was substituted for wheat flour in white bread recipes at weight percentages of 0%, 2.5%, 5%, 7.5% and 10%. The weight, volume and fermentation of the dough tended to decrease with increasing POW content. The color of the bread crust and crumb darkened with increasing POW content. A sensory evaluation showed a high preference for the white bread made with 5% POW, indicating that the addition of 5% POW to flour for bread dough could improve the quality characteristics of white bread. These study results also provide useful information for using the flour of wheat cultured with Pleurotus ostreatus as an ingredient source for food processing.

Antifungal Activity of Benzoic Acid from Bacillus subtilis GDYA-1 against Fungal Phytopathogens (Bacillus subtilis GDYA-1로부터 분리한 benzoic acid의 식물병원성 곰팡이에 대한 항균활성)

  • A bacterial strain antagonistic to some fungal phytopathogens was isolated from the stem of a Persimmon tree in Yeongam, Korea. This bacterium was identified as Bacillus subtilis by 16S rRNA gene sequencing and designated as B. subtilis GDYA-1. In in vivo experiment, the fermentation broth exhibited antifungal activities against Magnaporthe oryzae on rice plants, Phytophthora infestans on tomato plants, and Puccinia recondita on wheat plants. We isolated one antifungal compound and its chemical structure was determined by mass and $^1H$-NMR spectral data. The antifungal substance was identified as benzoic acid. It inhibited mycelial growth of M. oryzae, Rhizoctonia solani, Sclerotinia sclerotiorum, and P. capsici with minimum inhibition concentration (MIC) values, ranging from 62.5 to 125 ${\mu}g/ml$. Moreover, the substance effectively suppressed Phytophthora blight of red pepper caused by P. capsici in a pot experiment. To the author's knowledge, this is the first report on the antifungal activity of benzoic acid against phytopathogenic fungi. Benzoic acid and B. subtilis GDYA-1 may contribute to environmental-friendly protect crops from phytopathogenic fungi.
