The effect of fermentation parameters and medium composition on the simultaneous mycelial growth and exo-polymer production from submerged cultures of Ganoderma applanatum was investigated in shake-flask cultures. The optimum initial pH for mycelial growth and exo-polymer production was 5.0 and 6.0, respectively. The optimum temperature was $25^{\circ}C$ and the optimum inoculum content was 3.0% (v/v). The optimal carbon and nitrogen sources were glucose and corn steep powder, respectively. After 12 days fermentation under these conditions, the highest mycelial growth was 18.0 g/l and the highest exo-polymer production was 3.9 g/l.

The objective of this study was to determine effects of corn germ meal(CGM) and condensed molasses fermentation solubles(CMS) as the replacement of rice bran on mycelial growth and density in various mushrooms. The results of this study showed that CGM and CMS might be utilized more efficiently than rice bran in culturing mushrooms. The results are as follows; 1. Mycelial growth of Pleurotus ostreatus was remarkably higher than that of control when the concentration of CGM and CMS was 10%, respectively. However, the mycelial density was lower than that of control. Thus, Considering the mycelial growth as well as density, it was found that 20% supplements of CGM and CMS resulted in the best with 78mm/10d and 71mm/10d, respectively. 2. Mycelial growth of Pleurotus eryngii in the treatment group of 10% CMS and 20% CGM showed 74mm/10d and 67mm/10d, respectively. This result tended to be equal or somewhat lower compared with that of control(74mm/10d). 3. Mycelial growth of Flamulina velutipes was 87mm/10d in the treatment of 10% CMS, 79mm/10d in the treatment of 20% CGM, which showed significantly higher than that of control(56mm/10d). When the contents of CGM and CMS were mixed with the amount of 10-30%, Thus, it was assumed that the quantity of mushrooms may increase with the supplements of CEM and CMS. 4. Mycelial growth of Lentinus edodes in the treatment group of 10-40% CMS and 10-30% CGM showed 58-52mm/10d and 56-49mm/10d, respectively. This result was significantly higher that control(47mm/10d). 5. When CMS and CGM were supplememted with the level of 10-30%, mycelial growth rate of Ganoderma lucidum showed 76-72mm/10d and 74-69mm/10d, respectively, which are higher than those of control.
The Study was carried out to investigate in the optimal mycelial growth and Exo-Polysaccharides of Fomitopsis pinicola. Jar fermentations were carried out to optimize the culture conditions for mycelial growth and exo- polysaccharide production. The optimal agitation speed and aeration rate were 200 rpm and 1.5 v.v.m., respectively. Under optimal culture conditions, the maximum mycelial growth and exo-polysaccharide production after 11 days with a 5 L jar fermenter containing the optimized medium were 10.21 g/L and 3.56 g/L, respectively. However, the fundamental information obtained this study is insufficient in the development of a efficient process for mycelial growth and exe-polysaccharide production from Fomitopsis pinicola.
To improve cultivation technique for the stable and high yield, the outdoor fermentation of rice straw bundles, and pasteurization and fermentation of rice straw compost in tunnel were carried out. 1) Mycelial growth of P. ostreatus was becoming rapid and dense according to prolongation of fermentation periods outdoor rice straw bundles, but days untill fully mycelial growth after spawning was short. 2) As the fermentation period of rice straw bundles was 6 days (three times turning), yield of P. ostreatus was the highest as $50.5kg/3.3m^2 and increased 24% of yield than those of conventional method. 3) When it was fermented rice straw compost in tunnel for 4 days, mycelial growth of P. ostreatus was rapid and dense, and also days untill budding after spawning, was shorter, yield $67.6kg/3.3m^2$, resulted in increasing 17% of yield than those of conventional method. 4) On being sterilization and fermentation of the rice straw compost in tunnel, the production cost was decreased 22.3% in compared with conventional method.
A carrier-supported mycelial growth of Penicillium chrysogenum was applied to penicillin fermentation system. Among various materials tested, celite was found to be most effective for both spore adsorption and bioparticle development. Hyphal growth through pore matrices of the material showed strong anchorages and provided highly stable biofilm growths. When 5-10% celite was employed, both cell growth and penicillin production were observed to increase significantly comparing to the dispersed filamentous growth. Specific productivity of penicillin, however. was found to be kept almost constant at a value of 1,900 unit/g cell/hr. A semicontinuous fermentation in a fluidized-bed reactor. using the tarrier-supported biofilm growth, was conducted successfully although free mycelia appeared in the late phase of the fermentation made the reactor operation difficult. Control of the size of bioparticles was considered as a major operating factor to maintain the reactor productivity at a desired level.
It was discovered that ammonium phosphate in the medium played an important role in both growing mycelium and producing exopolysaccharides (EPS) from G. lucidum. In lower concentration levels of ammonium phosphate (0-3 g/l), an improved mycelial growth was observed by maintaining more filamentous morphology than in high concentrations (5-11 g/l). In addition, it was confirmed by comparing the factual dimension and frequency of the area regarding the mycelial pellets. This must be attributed to limitations of nutrient transfer by maintaining filamentous mycelium during the cultivation in a low ammonium phosphate containing medium. On the other hand, the best EPS production was observed in medium with the absence or low concentration of ammonium phosphate. The shear stress of the culture broth was greatly affected by the shear rate, as compared with that of the culture broth with high ammonium phosphate concentration. The rheological characteristics of the fermentation broth and filtrate worked well according to the Herschel-Bulkley model. It was also found that the morphological changes of the mycelium resulting from the ammonium phosphate concentration directly affected the rheological characteristics of the system and resulted in reversely affecting the EPS production levels. Based on these results, it can be concluded that delicate regulation of the ammonium phosphate concentration in the culture media should be provided in order to obtain optimal mycelial growth and/or EPS production.
The chemical media composition and culture conditions were optimized for mycelial growth of Phellinus igniarius 26005. The method of solid-state fermentation, cultivation of basidiomycetal strains in various grains, was developed. Media composition for optimal growth of Phellinus igniarius 26005 was made of 7.0% malt extract, 0.3% bacto soytone, and 0.2% yeast extract. The optimum condition for mycelial growth was $28^{\circ}C$ and pH 7.0, respectively. For the mass cultivation of mycelia, the hydrated grains with cold water, were put into the plastic bottle. The mycelial growth rate in the bottled grains was high in the early stage with inoculation of homogenized mycelium. The activity of mycelium was maintained by adding sterilized water in the middle of cultivation. The glucosamine content which determins the mycelial growth rate in solid material was in the order of job's tears>barley>black soybean>wheat>malt soybean>brown rice>sorghum>glutinous rice.
Various conditions for mushroom were investigated to study their effects on the mycelial growth of oyster mushroom and pathogens. Our results show that the optimal pasteurization temperatures for mycelial growth and pathogens are $60^{\circ}C\;and\;121^{\circ}C$ respectively. Both were then decreased by heating over $60^{\circ}C$ and getting down temperature under $121^{\circ}C$, respectively. In the experiment for pasteurization time, it was showed that both 8 and 12 hours pasteurization were greater than 24 hours treatment for the mycelial growth. In contrast, we didn't find any particular effect by different soaking time of substrates. The pH In substrates was leveled well by pasteurization and after-fermentation even though the acidity was lowered by extending the soaking time. Our results also show that the mycelial growth is greatest at $25{\sim}30^{\circ}C$ regardless of pasteurization time for substrates, Through the experiment with paddy straw as a substrate, it was showed that two time pasteurization was greater than one time for mycelium growth and pro-fermentation of paddy straw before pasteurization have resulted in worth mycelial growth compared with that of normal condition.
The optimum conditions for the submerged mycelial culture of Lentinus edodes SR-1 were elucidated to be incubation temperature of 25C, initial pH 4.0, agitation of 300 rpm, inoculation of 10.0%(v/v), and aeration of 1.0 v/v/m in TGY medium. The optimum c/n ratio and economic yield coeffcient for the submerged mycelial culture were 13.1:1 and 0.45 respectively. As the plant growth hormones test, SCM medium containing 0.5ppm of 2,4-dicholorophenoxyacetic acid increased mycelial yield in 1.1%, but 6-benzylaminopurine was not effective.
A carriersupported mycelial growth of Sreptomyces erythreus was applied to erythromycin fermentation sistem using celite as a support material. Hyphal growth through the pore matrices of the materials showed anchorages and provided a stable biofilm growth. When the phospate concentration was limited to 0.8g corn steep liquor/L(corresponding to 40mg KH2PO4/L), the specific production rate of erythromycin was increased from 557$\mu$g/g-cell.hr under unlimited condition to 2, 898 $\mu$g/g-cell.hr. A fluidized-bed bioreactor was operated for erythromycin production by a repeated fed-batch mode. The control of free mycelial concentration and the extension of production phase were considered important to maintain the reactor productivity at a desired level. The erythromycin production under phosphate-limited condition could be maintained for at least 600hrs.
