Effect of Exposure Chlorpyrifos (CPF) on Survival, Hatching rate, Reproductivity and Histological Changes of Japanese Medaka, Oryzias latipes (Chlorpyrifos(CPF)가 송사리, Oryzias latipe의 생존, 부화율, 번식력에 미치는 영향 및 조직학적 변화)

  • Chlorpyrifos (CPF) is a widely used organophosphate insecticide. Japanese medaka, Oryzias latipes was selected to investigate the effects of chlorpyrifos on survival rate, hatching rate, reproductivity and histopathological changes. Adult fish were exposed to CPF at concentrations of 20, 40, 80 and $160{\mu}g/L$, and the test duration was 4 weeks. An experiment with eggs was also performed under CPF exposure continued for 7 days covering from the fecundation to end-hatching stages. Survival rate of the adult medaka decreased in CPF $80{\mu}g/L$ and $160{\mu}g/L$ treatment groups compared with control, solvent control, $20{\mu}g/L$ or $40{\mu}g/L$L treatment group. Spontaneous start of feeding significantly decreased compared with the control (p < 0.01) when exposed to CPF at above 40 $\mu g/L$. Total spawning decreased by the 20 or $40{\mu}g/L$ CPF treatment compared with the control or solvent control in the test with eggs obtained from untreated adult medaka, and abnormal eggs increased in those CPF-treated groups. There were certain adverse effects at above $40{\mu}g/L$ determined from liver and gonad histological examinations. The results indicate that CPF has insignificant toxic effects in Japanese medaka at less than $20{\mu}g/L$ in long term exposure.

A Biological Activity of Serratia marcescens Strains Isolated from Dead Larva of the Diamondback Moth, Plutella xylostella (Plutellidae, Lepidoptera) (배추좀나방(Plutella xylostella)의 죽은 유충에서 분리한 Serratia marcescens 균주의 생물활성)

  • The cause of death was investigated with several dead cabbage moth larvae in breeding box. Bacterial strains were isolated and selected from the dead larvae by bioassay. One of them was identified as Serratia marcescens used by morphological characteristics and gene sequencing. S. marcescens were cultured by Luria Bertani (LB) media broth for bioassay. When 100-fold dilution of culture broth to third larvae of diamondback moth, Plutella xylostella, it was showed a 100% mortality at 2 days after treatment, and only 10-fold dilution of supernatant liquid was showed 86.6% mortality. When the culture broth of S. marcescens was applied to the larvae of beet armyworm, Spodoptera exigua, contact and feeding toxicity were 20 and 8% of mortality, respectively. Otherwise, when the culture broth of S. marcescens was applied to 5 major plant pathogens, antibacterial activities against Fusarium oxysporum, Rhizoctonia solani, Colletotrichum gloeosporioides, Phytophthora capsici and Sclerotinias clerotiorum were 4.7, 11.3, 20, 15.7 and 42.6%, respectively. Also, degradation ability of S. marcescens against protein and chitin were examined.

An Overview of Problems Cyanotoxins Produced by Cyanobacteria and the Solutions Thereby (남조류에서 발생하는 독소의 문제점과 대책)

  • Cyanobacteria frequently dominate the freshwater phytoplankton community in eutrophic waters. Cyanotoxins can be classified according to toxicity as neurotoxin (Anatoxin-a, Anatoxin-a(s), Saxitoxins) or hepatotoxin (microcystins, nodularin, cylindrospermopsin). Microcystins are present within cyanobacterial cells generally, and they are extracted by the damage of cell membrane. It has been reported that cyanotoxins caused adverse effects and they are acculmulated in aquatic oganisms of lake, river and ocean. In natural, microcystins are removed by biodegradation of microorganisms and/or feeding of predators. However, in process of water treatment, the use of copper sulfate to remove algal cells caused extraction of a mess of microcystins. Microcysitns are removed by physical, chemical and biological methods according to reports. The reduction of nutrients (N and P) inflow is basic method of prevention of cyanobacteria bloom formation. However, it is less effective than investigation because nutrients already present in the eutrophic lake. In natural lake, cyanobacteria bloom are not formed because macrophytes invade from coastal lake by eutrophication. Therefore, a coastal lake has to recover to prevent of cyanobacteria bloom formation.

Persistence of Chlorfenapyr in Paprika Leaf and Its Residual Biological Activity to Two Spotted Spider Mite, Tetranychus urticae (파프리카 잎 중 Chlorfenapyr의 잔류량 변화와 점박이응애에 대한 잔효 생물 활성)

  • A comparative experiment was conducted to investigate the persistence of chlorfenapyr residue and its biological activity to two spotted spider mite, Tetranychus urticae, after treatment of chlorfenapyr suspension concentrate (10% SC) on paprika plants at recommended (2000 times dilution) and double dosage. Biological half-lives of chlorpfenapyr were estimated as 8 and 5 days in leaves and fruits of paprika plants, respectively. While initial deposits of chlorfenapyr residues in leaves at recommended and double dosages were 22.22 and 37.75 ${\mu}g\;g^{-1}$ at the time of application, its residue decreased to 1.56 ${\mu}g\;g^{-1}$ and 3.62 ${\mu}g\;g^{-1}$ at 29 days after treatment, respectively. Residual biological activity of chlorfenapyr SC to T. urticae at recommended dosage lasted for 7 or 15 days on the basis of 24 or 48-hrs mortality test assayed with feeding on excised leaf chlorfenapyr-treated. The control efficacy to the mite showed a good agreement with the persistence of chlorfenapyr residues in leaves.

Effects of TBTCl on the Behavior and Reproduction of the Guppy (Poecilia reticulata) (TBTCl이 구피(Poecilia reticulata)의 행동과 번식에 미치는 영향)

  • Sexually matured guppies (Poeiria reticulata) were exposed to TBTCI (0.1, 0.32, 1, 3.2, 10, 25, 32, 50, 75 and 100 $\mug/l$) for 144 hours to determine the bioaccumulation rate and effects on the reproduction and behavior. The ratio of TBT residues to $\SigmaBTs\; (TBT:\SigmaBTs)$ was 67% or higher in all the guppies exposed to TBTCl, and the higher the level of TBTCl exposed, the higher the ratio of TBT:∑BTs, suggesting that the higher the level of TBTCl exposed, the lower the metabolism rate of the fish. TBTCl exposure led to a poor reproductivity and an abnormal sexual behavior in the fish, i.e. a reduced number of the male sexual sigmoid display and of spermatophore in the efferent duct was observed in the fish exposed to 0.1 $\mug/l$ and higher levels of TBTCl, and a decreasing ratio of the testicular spermatophore cyst to the whole germ cell cysts was observed in the fish exposed to 0.32∼10 $\mug/l$)of TBTCl. The reduced ratio of the spermatophore cyst seems to be an effect of the endocrine disrupter inhibiting spermiogenesis. In the fish exposed to 25 $\mug/l$ and higher levels of TBTCl, more serious effects, such as a rapid increase of mortality, the necrosis of most of the germ cells, great damages in Sertoli cells and epithelial cells of the efferent duct, a significant increase of abnormal swimming behavior, and a cessation of feeding were observed, which suggest the acute toxicity of TBTCl inhibiting not only the reproduction and behavior but also the survival of the fish itself.

  • An extracellular polysaccharide, methylan, was produced under the specific conditions by Methylobacterium organophilum from methanol. The specific growth rate of cells was approximately constant regardless of C/N ratio and the specific product yield was maximum at a C/N ratio of 30. Methylan production was suppressed by the deficiency of mineral ions such as Mn$^{++}$ or Fe$^{++}$ ion. The optimal pH for cell growth and methylan production was 7. Whereas the optimal temperature for cell growth was found to be 37$^{\circ}C$, that for methylan production was 3$0^{\circ}C$. The methanol concentration above 4% completely inhibited the cell growth. The initial methanol concentration for the maximal production of methylan was 0.5% (v/v) and above this concentration, methylan production was markedly inhibited. To overcome the substrate toxicity and inhibition for both cell growth and methylan production, a fed-bach culture of intermittent feeding within 5 g/l methanol was conducted under the optimal culture condition. Methylan production of was stimulated by nitrogen limitation and methylan was accumulated up to 8.7 g/1 and cell mass also increased up to 12.4 g/l.

  • We developed insect-resistant GM rice(Bt transgenic rice) by inserting the mCry1Ac1 a modified gene from the soil bacterium, Bacillus thuringiensis. The Bt transgenic rice expressing the Bttoxin mCry1Ac1 was tested for the effects on survival of Misgurnus anguillicaudatus and Cyprinus carpio, commonly used as a model organism in ecotoxicological studies. M. anguillicaudatus and C. carpio fed 100% ground rice in suspension, using either Bt rice or non-GM counterpart rice(Nakdong). The Bt rice used for the test were confirmed to have the mCry1Ac1 gene expression by the immuno-strip and ELISA analysis. Feeding test showed that no significant differences in cumulative immobility and abnormal response of M. anguillicaudatus and C. carpio fed on between Bt rice and non-GM counterpart rice. The 96hr-LC50 values showed no difference between Bt rice(>1,000mg/L) and non-GM rice(>1,000mg/L). We concluded that there was no significant difference in toxicity for non-target organisms(M. anguillicaudatus and C. carpio) between Bt rice and non-GM counterparts.

Establishment of Food Processing Methods for Larvae of Allomyrina dichotoma, Korean Horn Beetle (식약용으로의 장수풍뎅이 유충 전처리 방법에 대한 연구)

  • With the objective of developing a functional food source, we established optimal processing conditions for the larvae of Allomyrina dichotoma, which have been used in traditional medicine to treat hepatic disorders in Korea. Without suitable processing, the larvae are difficult to consume as a food because of their disgusting taste and smell; moreover, in this form they might be a potential microbial hazard. In this study, we investigated the effect of feeding material, sterilization, and powdering after freeze-drying on the food quality of the larvae of A. dichotoma and on cytotoxicity against Raw 264.7 cells. Three to five days feeding with the sawdust from discarded oak-trees is sufficient for the breeding process. The sawdust was sterilized by vapor for five minutes. Sterilization of the larvae at a high temperature ($115^{\circ}C$ for 5 min, 0.9 $kgf/cm^2$) is necessary to eliminate pathogenic bacteria and fungi. The results of the cytotoxicity assay showed no toxicity in the prepared extract from larvae of A. dichotoma. In addition, to prepare the larvae for human consumption, various feeds were used and the smell, color, and taste were evaluated. Our results suggested that larvae of A. dichotoma could be developed as food source when a suitable processing method is established.

A Study on the Acute Toxicity of AF-2 (AF-2의 독성(毒性)에 관(關)한 연구(硏究)(급성중독(急性中毒)))

  • This study was undertaken to investigate the toxic effect of AF-2, 2-(2-furyl)-3-(5-nitro-2-furyl) acrylamide, in the diet of rats (Splague Dowley) for 4 weeks. Comparisons were made of weight gains, organ weights and the cholesterol, phospholipid and triglyceride content of the blood serum and liver tissue. Rats were divided into three groups, a control group which was fed on a standard diet and two groups were fed with 0.04% (group I) and 0.2% (group II) of AF-2. Male rats weighing 370-410 g, female rats weighing 240-250 g were used in this experiment and each group was composed of 24 Albino rats. The results were as follows. 1. Comparisons of weight gains, control and experimental groups did not show significant differences. 2. An examination of organ weights, liver weights of experimental groups showed higher than that of the control group. 3. In the lipid content of blood serum, phospholipid contents of experimental groups were lower than that of the control, while cholesterol and triglyceride content of experimental groups were higher than that of the control. 4. In the lipid content of liver tissue, phospholipid and triglyceride content of experimental groups did not show considerable changes, but cholesterol content was increased in proportion to AF-2 content. From these results, the authors could observe the significant changes in weight gains of liver and cholesterol content after feeding. It is concluded that the toxic effect of group II showed higher than that of group I and AF-2 had a toxic effect on rat liver to a certain extent.

Management of the Development of Insecticide Resistance by Sensible Use of Insecticide, Operational Methods (실행방식 측면에서 살충제의 신중한 사용에 의한 저항성 발달의 관리)

  • An attempt was made to stimulate future research by providing exemplary information, which would integrate published knowledge to solve specific pest problem caused by resistance. This review was directed to find a way for delaying resistance development with consideration of chemical(s) nature, of mixture, rotation, or mosaics, and of insecticide(s) compatible with the biological agents in integrated pest management (IPM). The application frequency, related to the resistance development, was influenced by insecticide activity from potentiation, residual period, and the vulnerability to resistance development of chemical, with secondary pest. Chemical affected feeding, locomotion, flight, mating, and predator avoidance. Insecticides with negative cross-resistance by the difference of target sites and mode of action would be adapted to mixture, rotation and mosaic. Mixtures for delaying resistance depend on each component killing very high percentage of the insects, considering allele dominance, cross-resistance, and immigration and fitness disadvantage. Potential disadvantages associated with mixtures include disruption of biological control, resistance in secondary pests, selecting very resistant population, and extending cross-resistance range. The rotation would use insecticides in high and low doses, or with different metabolic mechanisms. Mosaic apply insecticides to the different sectors of a grid for highly mobile insects, spray unrelated insecticides to sedentary aphids in different areas, or mix plots of insecticide-treated and untreated rows. On the evolution of pest resistance, selectivity and resistance of parasitoids and predator decreased the number of generations in which pesticide treatment is required and they could be complementary to refuges from pesticides To enhance the viability of parasitoids, the terms on the insecticides selectivity and factors affecting to the selectivity in field were examined. For establishment of resistant parasitoid, migration, survivorship, refuge, alternative pesticides were considered. To use parasitoids under the pressure of pesticides, resistant or tolerant parasitoids were tested, collected, and/or selected. A parasitoid parasitized more successfully in the susceptible host than the resistant. Factors affecting to selective toxicity of predator are mixing mineral oil, application method, insecticide contaminated prey, trait of individual insecticide, sub-lethal doses, and the developmental stage of predators. To improve the predator/prey ratio in field, application time, method, and formulation of pesticide, reducing dose rate, using mulches and weeds, multicropping and managing of surroundings are suggested. Plant resistance, predator activity, selective insect growth regulator, and alternative prey positively contributed to the increase of the ratio. Using selective insecticides or insecticide resistant predator controlled its phytophagous prey mites, kept them below an economic level, increased yield, and reduced the spray number and fruits damaged.
