We developed and evaluated a method of short-term acute toxicity testing based on the feeding behavior of Ceriodaphnia dubia. In prior toxicity tests, neonates of C. dubia were hatched and cultivated with the addition of yeast only for the preparation of the transparent daphnid's gut. Scenedesmus subspicatus was supplied as food after 1 to 6 h of exposure to toxicants. The effects of 1-h and 6-h exposure time on test sensitivity did not significantly differ. A comparison of the short-term l-h acute toxicity test developed in this study to the standard 48-h acute toxicity test using heavy metals, cyanide, and pentachlorophenol indicated that the 1-h test provided an acceptable sensitivity level in toxicity testing of C. dubia..
Two methods, a Ceriodaphnia algal uptake suppression test (CAUST) and a new toxicity test based on temperature control (TTBTC) which are based on feeding behaviour and temperature control, respectively, were developed and compared for the adoption as the better methodology for short-term toxicity screening. As previously published by Lee et aI., (1997), the CAUST method is based on the feeding behaviour of C. dubia and requires as little as 1 hour of contact time between C. dubia neonates and toxicant. However, even though CAUST requires only 1 hour of contact time, this method still take many hours for the preparation and measurement. Before the test starts, neonate digestive tracts were cleared by feeding yeast to the daphnids, Neonates were then exposed to toxicant, followed by addition of Scenedesmus subspiatus into the bioassay vessels. Daphnids were examined under the bright-field microscope with the presence of algae (indicated by a green colored digestive tract) or the absence of algae. Uptake indicated no toxic effect, whereas, absence of uptake indicated toxic inhibition. Unlike CAUST, the newly developed method (TTBTC) is based on just temperature control for the toxicity test of C. dubia. Initially, neonates are exposed to toxicants while the temperature of water bath containing media increased to $35.5^{\circ}C$. After 1.25 hour of contact time, the number of the daphnids, either live (no toxic effect) or dead (toxic effect), is counted without the aid of any instrument. In both methods, median effective concentrations ($EC_{50}$ values) were computed based on the results over a range of dosed toxicant concentrations. It showed that TTBTC was as sensitive as the standard 48-hour acute bioassay and CAUST. TTBTC and CAUST were much more sensitive than the I-hour I.Q. test and 30-minute Microtox. This study indicates that TTBTC is an easier and more rapid toxicity test than the standard 48-hour acute bioassay and even CAUST.
Honey bees (Apis mellifera) forage in agricultural areas, and are exposed to diverse pesticide poisoning. Toxic effects on Apis mellifera of different groups of pesticides were tested in the laboratory; fungicide (Metconazole), herbicide (Glyphosate), acaricide (Amitraz), organophosphate insecticide(Fenitrothion) and neonicotinoid insecticides(Thiacloprid, Thiamethoxam, Imidacloprid, Acetamiprid, Dinotefuran and Clothianidin). Commercial formulations were serially diluted from the recommended concentration (RC) to 10-6 times to carry out feeding and contact tests. Toxicity was transformed into lethal dose (LD50) and hazard question (HQ). The acute toxicity of pesticides showed similar patterns between feeding and contact tests. But feeding tests showed greater toxic to honey bee than contact test. The organophosphate and nitro-neonicotinoid insecticides were highly toxic with HQ values ranging greater than 1. However, cyano-neonicotinoids of Thiacloprid and Acetamiprid showed low toxicity. Even at the RC, 24 hr mortalities were 18 and 30%. The acaricide (Amitraz) showed intermediate level of toxicity at RC but negligible at the concentration lower than 10-1 times. A fungicide(Metconazole) and herbicide(Glyphosate) showed minimal impacts. The results imply that the selective use of pesticides could help conservation of pollinators in agricultural production systems.
Over the past few years, anthocyanin has been demonstrated to exert potentially anti-oxidative and bioavailability effects including anti-cardiovascular disease and cancer-preventive effects in humans. This study was conducted to assess the toxicity of anthocyanin extracts from black soybean seed coats via oral administration in a mice feeding test. The final anthocyanin-containing products induced signs of toxicity on mortality in the 4-week feeding test. The incidence of clinical signs and changes in body and organ weight were also not observed in all anthocyanin-treated groups as compared with the control groups. In hematology analysis, RBC indices concluding MCV, MCH, and MCHC and WBC differential counting such a NEU, LYM, MONO, EOS, and BASO evidenced no significant differences between the anthocyanin treatment and control groups. The biochemical parameters in serum analysis - ALT, AST, BUN, Crea etc.- were also not significantly altered in those groups. Absolute and relative organ weights were not increased after 4 weeks of treatment with anthocyanin extracts in mice. In conclusion, anthocyanin extracts from the black soybean has biological activity without any toxic effects, which also suggests that the consumption of soy containing anthocyanin products may be a good source for health and medical supplies.
시안의 만성적 조건에서 이스라엘 잉어의 생존, 섭이 및 성장에 미치는 독성영향을 파악하기 위하여 유수식방법에 의해 6주 동안 실험을 실시하였다. 시안의 만성적 조건하에 노출시킨 이스라엘잉어의 생존율은 시안농도 $151{\mu}$g/$\ell$이상에서 유의한 감소를 나타내었고, 실험종료시 시안농도 151, $289{\mu}$g/$\ell$에서 대조구에 비해 각각 15.2, $24.5\%$가 감소하였다. 섭이율은 시안농도 $151{\mu}$g/$\ell$에서는 3주, $289{\mu}$g/$\ell$에서는 1주 후부터 실험종료까지, 사료효율은 시안농도 $151{\mu}$g/$\ell$이상에서 2주 후부터 실험종료시까지 대조구에 비해 유의한 감소가 인정되었다. 성장률 및 일일성장율은 시안농도 $73{\mu}$g/$\ell$에서 6주 후에, 시안 농도 $151{\mu}$g/$\ell$이상에서는 2주 후부터 실험 종료시가지 유의한 감소를 나타냈다.
The water flea has been used as a test organism of toxicity test for surface water. Toxicity test with water flea is categorized into two parts. One is acute toxicity test with observing immobility and mortality and the other is chronic toxicity test determined by survival and reproduction of water flea. Heartbeat measurement of water flea was designed as a short-term toxicity test in this study. Direct measurement of heartbeat under microscope by aid of video camera gives and early diagnosis of mortality in short time. Therefore, the effects of measuring illumination, measuring time, and non-feeding during the test on heartbeat of water flea was evaluated to establish a new test approach. Test organisms used in this study are Daphnia magna, a well standardizes toxicity test organism, and Simocephalus mixtus, a newly refined organism $IC_{50}$ values of these test organism by heartbeat measurement were compared and discussed. It was found that toxicity test by heartbeat measurement was a reproducible, easy and simple method accomplished in a few hours.
This study designs to investigate effects of onion diet on carbon tetrachloride toxicity of rats. Experiments were performed with week's feeding, body weight, food intake, ratio of orgen weight/dody weight, serum lipid levels, superoxide dismutase and catalase activity and malondialdehyde(MDA) content in liver and kidney were determined. The content of serum total cholesterol in each group were lower than those of control group(p<0.05), especially at onion juice treated group. Serum HDL-cholesterol level of CBB and CBJ groups was significantly lower than that of control group (p<0.05). Superoxide dismutase activities of liver and kidney were significantly increased by carbon tetrachloride treatment and decreased by onion feeding. MDA contents in liver and kidney of CCl4 treated rats were significantly decreased by boiled and fresh onion fed group, compared with CCl4 treated control group. This result suggested that onion diet has a protective effect of CCl4 induced hepatotoxicity and nephrotoxicity of rat.
This preliminary feeding trial was conducted to study the effects of different dietary selenium (Se) levels on growth performance and toxicity in juvenile black seabream, Acanthopagrus schlegeli (Bleeker). Fish averaging $7.0{\pm}0.1g$ ($mean{\pm}SD$) were fed one of the five semi-purified diets containing 0.21, 0.30, 0.52, 1.29 and 12.3 mg sodium selenite ($Na_2SeO_3$)/kg diet (Se 0.21, Se 0.30, Se 0.52, Se 1.29 or Se 12.3) for 15 weeks. After the feeding trial, weight gain (WG), feed efficiency (FE), specific growth rate (SGR) and protein efficiency ratio (PER) of fish fed Se 0.21, Se 0.30, Se 0.52 and Se 1.29 diets were not significantly different, however fish fed Se 12.3 diet showed significantly lower WG, FE, SGR and PER than those of fish fed the other diets (p<0.05). Fish fed Se 0.21, Se 0.30, Se 0.52, Se 1.29 and Se 12.3 diets showed no significant differences in hematocrit (PCV), hemoglobin (Hb) and red blood cells (RBC), however fish fed Se 12.3 diet showed lower values of PCV, Hb and RBC than those of fish fed the other diets. Histopathological lesions such as tubular necrosis and polycystic dilation of tubules in the kidney tissues were observed in fish fed Se 12.3 diet. Se was accumulated in a dose-dependent manner in the liver, kidney, muscle and gill tissues. Based on the results of this preliminary feeding trial, a dietary Se level of 0.21 mg $Na_2SeO_3/kg$ diet could be optimal for proper growth performances, and a dietary Se level of 12.3 mg $Na_2SeO_3/kg$ diet may ultimately be toxic to juvenile black seabream, Acanthopagrus schlegeli.
Kyoaesamultang(KAT) has been used as an important prescription for various diseases including threatened abortion, associated with pregnancy in traditional medicine. In oder to identify the safety of KAT, this study was designed to determine mutagenicity and hepato-toxicity. In Rec-assay, Bacillus subtills H-17($Rec{^+}$) and M-45($Rec{^-}$) strains were used to clarify the DNA damage property. In Ames test, Salmonella typhimurium TA98 and TA100 were used for mutagenicity testing. In SOS umu test, Salmonella typhimurium TA1535 containing plasmid pSK1002 was used as a tester strain, and the levels of umu operon expression were monitored by measuring the $\beta$-galactosidase activity. From tested results, KAT did not show DNA damage and mutagenicity. On the other hand, hepato-toxicity of KAT to female ICR mice was monitored by the measurements of s-GOT, s-GPT and LDH activities after oral feeding for 15days. KAT showed 34% increase of s-GOT and s-GPT activities, also exhibited 35% increase of LDH activity in mice sera.
The purpose of the present study was to investigate the effects of non-watering, fasting and non-watering fasting for 16-17 hrs on haematological and biochemical parameters in SD rats. 1. Liver weight was decreased in the order of in the fasting group, non-watering fasting group and non-watering group in both sexes. 2. In fasting group and non-watering fasting group, haematologic parameters of male (HGB and MCH) and femal (MCHC) were trends to decrease compared with normal feeding & watering group. 3. In the differential leucocyte counts, there were no significant differences compared with noraml feeding % watering group.
