Fructooligosaccharides Alter Profiles of Fecal Short-Chain Fatty Acids and Bile Acids in Rats

  • We investigated the effects of fructooligosaccharides and chicory inulin on the profiles of cecal and fecal short-chain fatty acids (SCFAs) and fecal bile acids in rats. Thirty-six Sprague Dawley male rats weighing about 190 g were randomly divided among four treatments; control diet, control diet +6%(w/w) fructooligosaccharide (POS), control diet +6% chicory inulin oligosaccharide(CIOS), and control diet +6% chicory inulin(CI). The rats were pair-fed and experimental diets were maintained for 5 weeks. Cecal and fecal pH was significantly decreased in rats that were fed fructooligosaccharides and chicory inulin. Cecal propionate was significantly elevated in rats fed CIOS diets, and butyrate was lower in rats fed FOS and CI than control values. Cecal lactate was significantly higher in the FOS group than in the control group. The fecal excretions of acetate and total SCFA were 200-300% higher in rats that were fed fructooligosaccharides and chicory inulin than in the control group. Lactate excretion was highest in rats that were fed FOS, followed by those fed CIOS and CI. The cholic acid and total bile acid concentrations in feces were significantly lower in the rats that were fed fructooligosaccharides and chicory inulin. The deoxycholic acid concentrations in wet feces were significantly lower in the groups of rats that ate CIOS (0.186 mM), FOS (0.274 mM), and CI (0.362 mM) than in the control group (0.595 mM). Among the fructans, short-chain fructooligosaccharide was more effective at decreasing colonic pH and lactate production, but medium-chain chicory inulin oligosaccharide was more effective at increasing fecal butyrate and lowering the fecal secondary bile acid concentration.

BIOLOGICAL CONTROL OF GINSENG ROOT ROTS WITH SOIL AMENDMENTS

  • The phenomenon of 'soil sickness' is one of the most important limiting factors for ginseng(Panax ginseng) production in Korea. The principal cause is known to be due to the root rots caused by Cylindrocarpon destructans and Fusarium solani. Attempts were made to control the root rots with non-polluting cultural methods or soil amendments. Among the nine soil amendments tested, crab shell, cow bone and pig feces were selected for further testing. Each of the three amendments increased the populations or various actinomycetes in the range of 10-25 times over that of non-amended soil, whereas the population of C. destructans was reduced to about $50-70\%$ as compared with the control. Five isolates of Streptomyces with clear zones on chitin-agar medium were selected and then tested for their antagonistic effects on C. destructans. When anyone of the five isolates of Streptomyces and C. destructans was grown together in a modified peptone broth, growth of the latter was highly inhibited. When three levels of crab shell, cow bone, or pig feces were used to amend potted soil infested with C. destruetans, the root rot ratings of ginseng seedlings were reduced to less than one half in all the treatments as compared to the control. In another similar experiment, crab shell and cow bone amendments resulted in almost complete control of the seedling root rots in soil infested with C. destructans or F. solani. In conclusion, biological control with soil amendments of ginseng root rots caused by C. destructans and F. solani was successful. Further basic studies should be pursued using soil amendments for better control. In addition, field experiments are needed to complement the soil amendment control measures in an integrated pest control program.

Analysis of antibiotic resistance genes in pig feces during the weaning transition using whole metagenome shotgun sequencing

  • Antibiotics have been used in livestock production for not only treatment but also for increasing the effectiveness of animal feed, aiding animal growth, and preventing infectious diseases at the time when immunity is lowered due to stress. South Korea and the EU are among the countries that have prohibited the use of antibiotics for growth promotion in order to prevent indiscriminate use of antibiotics, as previous studies have shown that it may lead to increase in cases of antibiotic-resistant bacteria. Therefore, this study evaluated the number of antibiotic resistance genes in piglets staging from pre-weaning to weaning. Fecal samples were collected from 8 piglets just prior to weaning (21 d of age) and again one week after weaning (28 d of age). Total DNA was extracted from the 200 mg of feces collected from the 8 piglets. Whole metagenome shotgun sequencing was carried out using the Illumina Hi-Seq 2000 platform and raw sequence data were imported to Metagenomics Rapid Annotation using Subsystem Technology (MG-RAST) pipeline for microbial functional analysis. The results of this study did not show an increase in antibiotic-resistant bacteria although confirmed an increase in antibiotic-resistant genes as the consequence of changes in diet and environment during the experiment.

Strategies to Reduce Environmental Pollution from Animal Manure: Nutritional Management Option - Review -

  • The first option in manure management is developing an environmentally sound nutritional management. This includes proper feeding programs and feeds which will result in less excreted nutrients that need to be managed. Critical components that should be controlled are N, P and minerals that are used at supranutritional levels. Amino acid supplementation and protein restriction reduce N excretion in the monogastric animals. Supplementation with enzymes, such as carbohydrases, phytase and proteases, can be used to reduce excretion of nutrients and feces by improving digestibility of specific nutrients. Growth promoting agents, such as antibiotics, beta-agonists and somatotropin, increase the ability of animals to utilize nutrients, especially dietary protein, which results in reduced excretion of N. Some microminerals, such as Cu and Zn, are supplemented at supranutritional level. Metal-amino acid chelates, metal-proteinates and metal-polysaccharide complexes can be used at a much lower level than inorganic forms of metals without compromising performance of animals. Deodorases can be used to avoid air pollution from animal manure. Nutritional management increases costs to implement. It is necessary to assess the economics in order to find an acceptable compromise between the increased costs and the benefits to the environment and production as well.

Screening of Conjugated Linoleic Acid Producing Lactic Acid Bacteria from Fecal Samples of Healthy Babies

  • This study was carried out to obtain conjugated linoleic acid (CLA) producing lactic acid bacteria for further study on the enzymes related to the production of CLA which has gained considerable attention and on the development as a probiotic culture. Total 34 lactic acid bacteria were isolated from 19 feces samples of healthy babies. CLA forming ability was measured spectrophotometrically by the modification of linoleate 12-cis, 11-trans-isomerase activity measuring method, and CLA of the cultures were extracted, methylated, and examined by HPLC analysis. CLA methyl ester of only one culture showing the highest value of CLA forming ability could be detected by HPLC analysis. The culture was found to be Gram positive, rods and catalase negative. It grows at $45^{\circ}C$ but not at $15^{\circ}C$, and was identified to be Lactobacillus fermentum on the basis of the biochemical characteristics and the utilization of substrates. These results provide an efficient experimental method to screen CLA producing lactic acid bacteria.

Effects of feed intake on the diversity and population density of homoacetogens in the large intestine of pigs

  • Objective: Homoacetogens play important roles in the production of acetate in the large intestine of monogastric mammals. However, their diversity in the porcine large intestine is still unknown. Marker gene analysis was performed to assess the effects of energy level on the diversity and population densities of homoacetogens in porcine feces. Methods: Crossbred pigs were fed high or low energy-level diets. The high-intake (HI) diet was sufficient to allow a daily gain of 1.2 kg. The low-intake (LI) diet provided 0.6 times the amount of energy as the HI diet. Genetic diversity was analyzed using formyltetrahydrofolate synthetase gene (FHS) clone libraries derived from fecal DNA samples. FHS DNA copy numbers were quantified using real-time polymerase chain reaction. Results: A wide variety of FHS sequences was recovered from animals in both treatments. No differences in FHS clone libraries between the HI and LI groups were found. During the experimental period, no significant differences in the proportion of FHS copy numbers were observed between the two treatment groups. Conclusion: This is the first reported molecular diversity analysis using specific homoacetogen marker genes from the large intestines of pigs. There was no observable effect of feed intake on acetogen diversity.

Heat-Killed Enterococcus faecium KU22001 Having Effective Anti-Cancer Effects on HeLa Cell Lines at a Lower Temperature

  • The anti-cancer effects of heat-killed Enterococcus faecium KU22001 (KU22001), KU22002, and KU22005 isolated from human infant feces were investigated. The anti-proliferative activity of these strains against various cancer cell lines was evaluated using the MTT assay. To determine the production of exopolysaccharides (EPS) with potential anti-cancer effect, ethanol precipitation and phenol-sulfuric acid method was used with the cell free supernatant of strains grown at 25℃ or 37℃. The EPS yield of E. faecium strains was higher at 25℃ than at 37℃. Among these E. faecium strains, KU22001 grown at 25℃ was associated with the highest bax/bcl-2 ratio, effective apoptosis rate, cell cycle arrest in the G0/G1 phase, and condensation of the nucleus in the cervical cancer HeLa cell line. In conclusion, these results suggest that KU22001 can be beneficial owing to the anti-cancer effects and production of functional materials, such as EPS.

CROM를 이용한 부영양 저수지의 유기물 제어: 이매패의 종 특이성에 대하여 (Continuous Removal of Organic Matters of Eutrophic Lake Using Freshwater Bivalves: Inter-specific and Intra-specific Differences)

  • 부영양 저수지의 수질개선을 목적으로 연속형 유기물제어(CROM)기술을 이용하여 이미 제어능이 밝혀진 국내산 이매패 말조개와 펄조개를 대상으로 이종간 또는 동종간의 유기물 섭식 특성을 비교 조사하였다. CROM은 매일 저수지 표층수를 직접 실내로 유입하였으며 저장소 및 조절조, 처리조, 분석조 등으로 구성하였다. 섭식실험은 온도와 광도가 일정하게 조절된 실험실에서 실시하였으며, 패류 처리 후 여과율, 제어능, 수질(영양염) 및 배설물 생산량 등을 측정하였다. 실험결과 패각길이가 비슷할 경우 이종간에 유기물 제어능은 서로 비슷하였으나 여과율의 경우에는 말조개가 더 높게 나타났다. 동일종(동밀밀도)내에서는 패각길이가 긴 개체들이 더 높은 제어능을 나타냈으며, 반대로 여과율은 패각길이가 짧은 개체가 높았다. 한편 동일종에서 처리밀도가 다를 경우 저밀도 처리군이 고밀도 처리군 보다 더 높은 여과율을 나타냈다. 배설물 생산은 여과율과 유사한 패턴을 보였으며 처리밀도와 먹이농도 변화가 배설물 생산에 영향을 미치는 것으로 나타났다. 두 종류의 패류는 평균 70% 이상의 뚜렷한 유기물(Chl-$\alpha$, SS) 제어율을 나타냈으며 섭식활동에 따른 $NH_4$-N나 $PO_4$-P과 같은 무기영양염의 배출량은 패각길이나 처리밀도에 따라 크게 영향을 받는 것으로 밝혀졌다. 결론적으로 CROM기술을 이용한 부영양 저수지의 유기물 제어는 적절한 패류의 종류나 밀도의 결정을 통하여 충분히 현장에 적용 가능한 생물학적 기법으로 판단되며, 부산물 처리 및 자원화를 비롯한 추후 연구가 필요하다고 사료되었다.

  • In summary, progesterone is probably the only naturallly occurring progestational agent of any significance. Small amounts may be synthesized by the cells of the follicle in the provulatory swelling phase; however, the major production is by the corpus luteum cells of the ovary during the luteal phase of the cycle. It is constantly produced in small amounts by the adrenal gland and by the testis in the male. In the adrenal and the testis, it probably serves as the precursor for corticoids and androgens. It is transported in the blood by a specific binding protein and metabolized and conjugated in the liver into sodium pregnanediol glucuronide which also circulates in the blood. Approximately 20% is excreted in the urine as sodium pregnanediol glucronide; pregnanolone represents a minor metabolic product. The pregnanediol which is excreted in the bile is enzymically hydrolyzed by the gut so that the pregnanediol recovered in the feces is in the free form.

  • The effects of dietary fiber on true digestibility of amino acids by growing pigs were studied, using semi-purified diets formulated from alfalfa meal, cassava leaf meal, rubber seed meal and leucacna meal at 20% level. A protein-free diet including 5% cellulose was formulated for correcting the endogenous amino acid loss. Across all the diets, arginine was the most digestible while the least at ileal level was threonine; methionine and/or histidine at fecal level respectively. The true digestibility value of amino acids at ileal level were higher than at fecal level except control diet (cellulose). The true digestibility values at ileal level were similar for all diets but differed at fecal level in different magnitude. These results indicate that undigestible compound in individual feedstuff might confound. True digestibility should be studied together for accurate diet formulation as apparent digestibility decreased when their amino acid concentration in the diet was reduced.
