Isolation and Identification of a Lactic Acid Bacterial Strain KJ-108 and Its Capability for Deodorizing Malodorous Gases Under Anaerobic Culture Conditions

  • A number of different sources, such as composts, leachates, and pig feces samples, were collected from different pig farms in Korea, and several microorganisms were screened for their ability to deodorize the malodorous gases. Consequently, a novel malodorous gases-deodorizing bacterial strain, KJ-108. was isolated, because it was highly abundant in nitrate-supplemented minimal medium ($MM-NO_3^-$) under anaerobic culture conditions. Airtight crimp-sealed serum bottles containing $MM-NO_3^-$ , medium were inoculated with KJ-108. Nitrate concentration was decreased rapidly after 20 h of incubation, and incubation was carried out until nitrite production reached almost zero. Taxonomic identification, including 16S rDNA base sequencing and phylogenetic analysis, indicated that the isolate had $100\%$ homology in its 165 rDNA base sequence with Lactobacillus pentosus. Among the volatile fatty acids, acetic acid contained in large amounts in fresh piggery slurry was decreased by about $40\%$ after 50 h incubation with strain KJ-108. n-Butyric acid, n-valeric acid, and isovaleric acid were gradually decreased, and isobutyric acid and capronic acid were dramatically eliminated at theinitial period with the treatment. Moreover, NH, removal efficiency reached a maximum of $98.5\%$ after 50 h of incubation, but the concentration of $H_2S$ was not changed.

Effects of Fructans and Isomaltooligosaccharide on Large Bowel Mass and Plasma and Fecal Immunoglobulin A in Rat

  • There are increasing evidences that prebiotics can modulate various properties of the immune system. This study was conducted to investigate effects of three kinds of fructans (chicory inulin, chicory inulin oligosaccharide and fructooligosaccharide) and a glucose oligomer(isomaltooligosaccharide) in large bowel mass and innnunoglobulin A (IgA) in rats. Forty five Sprague-Dawley male rats weighing about 1909 were randomly sorted to receive one of the five treatments, which were control diet, control diet+6% isomaltooligosaccharide (IMOS), control diet+6% fructooligosaccharide (FOS), control diet+6% chicory inulin oligosaccharide (CIOS), or control diet + 6% chicory inulin (CI). Rats were pair-fed and received the experimental diets for 5 weeks. Cecal and colonic wall weights were significantly higher in fructan (FOS, CIOS, CI)-fed groups compared with control and IMOS groups, and the length of colon was elevated in FOS and CIOS groups compared with control group. Fecal concentrations of acetic acid and total short-chain fatty acids (SCFAs) were significantly elevated in fructan-fed groups. Plasma and cecal levels and fecal excretion of immunogiobulin A (IgA) in rats were not significantly different among groups. However, fructooligosaccharide tended to increase IgA level in cecum. Cecal IgA level was significantly negatively correlated with pH of cecal content (r=-0.337), positively correlated with acetic acid level (r=0.310). Fecal IgA excretion was positively correlated with total SCFA (r=0.311) and propionic acid (r=0.400) level in feces. These results indicate that fructooligosaccharide and chicory inulin oligosaccharide exerted trophic effects in large bowel wall, increased production of SCFAs and decreased pH, which were conditions positively associated with cecal and colonic IgA secretion.

Effects of Feeding Dried Food Waste on Growth and Nutrient Digestibility in Growing-Finishing Pigs

  • A total of thirty-six gilts $(24.78{\pm}1.39kg)$ were employed for 38 days (Exp. 1), and twenty-four gilts $(46.50{\pm}1.90kg)$ for 43 days (Exp. 2) to determine growth performance and carcass characteristics in pigs fed dried food waste (DFW). Pigs were allocated by dietary treatments: 0%, 20% and 40% DFW. For nutrient digestibility, twelve female pigs $(50.20{\pm}0.52kg)$ were used in individual pens to collect feces. Food wastes were collected from restaurants and apartment complex areas and dried in a drum-type dryer at $115{\pm}2{^{\circ}C}$. Experimental diets for feeding trials were formulated to contain 3,400 kcal DE/kg and 17% crude protein. The tested DFW contained 2,858 kcal DE/kg, 25.0% crude protein, 17.3% crude fat, 1.37% Ca, 1.28% P, and 3.28% NaCl. As compared to crude protein content, the limiting amino acids (i.e., 0.90% lysine and 0.52% methionine) were low. The digestibilities of energy, ash, calcium and phosphorus in the DFW were generally lower than those in the grower diets, but the digestibilities of crude protein and crude fat were higher in DFW than the grower diet. Feeding DFW in pigs had a linear (p<0.01) effect on ADG and feed/gain as the inclusion levels of DFW were increased (Exp. 1). The ADG of pigs fed 40% DFW was also poorer (p<0.05) than that fed the control diet (Exp. 2). Carcass characteristics in terms of backfat and dressing percentage were not affected by dietary treatments. In conclusion, it seems that the optimal dietary inclusion level of the DFW is about 20% in the diet for growing-finishing pigs.

EFFECT OF SUPPLEMENTING RUMEN-PROTECTED LYSINE AND METHIONINE ON RUMINAL CHARACTERISTICS AND NUTRIENT DIGESTIBILITY IN SHEEP

  • This experiment was conducted to investigate the protein sparing effect of rumen protected lysine(RPLys) and methionine hydroxyl analogue(MHA) in sheep. The treatments were $T_1$ (CP 15% + RPLys 0%), $T_2$ (CP 12% + RPLys 0%), $T_3$ (CP 12% + RPLys 0.4%) and $T_4$ (CP 12% + RPLys 0.4% + MHA 0.3%). Ruminal characteristics, in situ and in vitro digestibility and nitrogen retention rate were measured in sheep receiving different combinations of dietary supplement. The results are summarized as follows: 1. Ruminal pH and VFA concentrations were not affected by the treatments. Ruminal ammonia-N concentration was high in sheep fed diets $T_2$, $T_3$ and $T_4$ with the highest value in the $T_4$ treatment(p<0.05). 2. The digestibilities of dry matter and organic matter were not affected by the treatments. 3. Nitrogen losses through feces and urine were the highest with $T_1$ (p<0.05) and nitrogen retention rates of groups $T_1$, $T_2$, $T_3$ and $T_4$ were 18.6, 32.4, 35.5 and 27.5% of nitrogen intake, respectively, indicating that RPLys supplementation improved nitrogen retention in sheep.

Response of Feeding Amino Acids Supplemented Cottonseed Meal on Growth Performance and Digestibility of Early Weaned Cow Calves

  • This study was designed to examine the response of feeding early weaning diets without and with amino acids supplemented cottonseed meal on growth performance and digestibility of early weaned cow calves. Fifteen 14-21 days old cross bred cow calves were randomly allotted to three experimental diets. Diet A comprised of milk replacer and concentrate feed whereas two isocaloric and iso-nitrogenous early weaning diets viz., B and C were prepared without and with lysine and methionine supplemented cottonseed meal, respectively. These early weaning diets were used as substitute of milk replacer for calves. Calves were placed in individual cages and fed twice daily for a period of 60 days. Daily feed intake, feed refused and weekly weight gain was recorded. Two digestibility trials I and II were performed at 5th and 9th week of the experiment, respectively. During the digestibility trial I, calves were fed on ad libitum basis whereas in trial II, calves were fed at 90% of their voluntary DMI. Feed, orts and feces samples were collected, weighed daily, composited, subsampled and analysed for DM and CP. Results indicated that weight gain of calves was (p<0.05) higher on diets A (0.63 kg/d/calf) and C (0.64 kg/d/calf) compared to calves on diet B (0.57 kg/d/calf). Significantly (p<0.05) less daily DMI was observed on diet A (1.48 kg) compared to early weaning diets B (1.70 kg) and C (1.72 kg). The feed efficiency was (p<0.05) better on diet A (2.33) compared to diets B (2.95) and C (2.65). The economic efficiency was noticed to be better on diets B and C compared to diet A. In trial I, digestibility of DM and CP of diet A was (p<0.05) higher than diets B and C. Whereas in trial II, digestibility of DM and CP of diet A was (p<0.05) less than diets B and C. It was concluded that early weaning diet based on lysine and methionine supplemented cottonseed meal produced better weight gain and feed efficiency compared to non-supplemented cottonseed meal based diet.

AMINO ACIDS DIGESTIBILITY TO PIGS IN VARIOUS FIBER SOURCES : 1. APPARENT DIGESTIBILITY OF AMINO ACIDS IN ILEAL DIGESTA AND FECES

  • Four fibrous feedstuffs from alfalfa meal (AFM), cassava leaf meal (CLM), rubber seed meal (RSM) and leucaena meal (LM) were included in semi-purified diets for growing pig (45 kg body wt.) at 20%, to investigate the effects of these fiber sources and fractions on amino acid digestibility. Cellulose (C), a purified fiber source was included in another diet at 5% level for comparison. The barrows fitted with ileal T-cannula were used in the digestion trials with latin square design. The digestibilities of amino acids were measured at both terminal ileum and fecal level. NDF and hemicellulose content were the highest in AFM-diet whereas LM-diet had the highest ADF and lignin content. RSM-diet contained the highest crude fiber and cellulose content. The digestibilities of amino acids at ileal level were found the highest with CLM-diet, while LM-diet was the least. At fecal level, control diet and CLM-diet were the highest in amino acid digestibility while AFM-diet was the least. The digestibility of amino acids was higher at ileal than fecal level. The digestibility of arginine was not affected with fiber fractions but was found to be the most disestible across all diets. The most depressed amino acid was methionine at both levels; praline and glycine, in the dispensable amino acid group, were depressed at ileal and fecal level, respectively. Lignin did not depress amino acid digestibility in general but specifically depressed methionine, histidine, isoleucine and threonine digestibility. Cellulose content did not affect amino acid digestibility but undesirable factors might be responsible.

단백질분해효소(bromelain) 처리 우모분이 Broiler의 생산성과 영양소 이용율에 미치는 영향 (Effect of Dietary Protease(bromelain) Treated Feather Meal on the Performance and Nutrient Utilization in Broilers)

  • This study was carried out to investigate the effect of replacing fish meal or soybean meal with feather meal or bromelain treated feather meal in broiler diets on the performances and nutrient utilization. Two hundred and twenty-five broilers were randomly allotted to five dietary treatrnents((1) control, basal diet; (2) PFM 50, 50 % of the fish meal replaced with bromelain treated feather meal; (3) PFM 100, 100% of the fish meal replaced with bromelain treated feather meal; (4) PSM 20, 20 % of the soybean meal replaced with bromelain treated feather meal; and (5) PSM 40, 40% of the soybean meal replaced with bromelain treated feather meal) in a 5-week feeding trial. In the overall period, body weight gain of the PFM 50(1,807 g), PSM 20(1,816 g) and PSM 40(1,823 g) were the highest and that of the PFM 100 was 1,744 g. The body weight gain of the control(1,698 g) was the lowest(p < 0.05) among treatments. Feed conversion was significantly(p< 0.05) improved when bromelain treated feather meal replaced 20% of the fish meal in the basal diet. Digestibilities of dry matter, ether extract, organic matter and phosphorus were not different among the treatments. Digestibility of crude protein of PFM 50(65.87 %), PSM 20(67.18 %) and PSM 40(67.56%) were the highest, and that of the control(54.49%) was the lowest(p < 0.05) among treatments. Arrunonia and hydrogen sulfide gases from the feces were significantly(p < 0.05) decreased in chicks fed the PFM 50, PSM 20 and GFM 40 diets, when observed after 3 weeks of feeding trials. Feed costs of the control and PFM 50 were 604 and 629 won, respectively but that of PSM 50 was 820 won. Therefore, replacement of fish meal with bromelain treated feather meal in the diets for chicks could be useful for economic production.

Effect of dual-type oligosaccharides on constipation in loperamide-treated rats

  • BACKDROUND/OBJECTIVE: Constipation is a condition that can result from intestinal deformation. Because humans have an upright posture, the effects of gravity can cause this shape deformation. Oligosaccharides are common prebiotics and their effects on bowel health are well known. However, studies of the physiological functionality of a product that contains both lactulose and galactooligosaccharides are insufficient. We investigated the constipation reduction effect of a dual-type oligosaccharide, Dual-Oligo, in loperamide-treated rats. MATERIALS/METHODS: Dual-Oligo consists of galactooligosaccharides (15.80%) and lactulose (51.67%). Animals were randomly divided into four groups, the normal group (normal), control group (control), low concentration of Dual-Oligo (LDO) group, and high concentration of Dual-Oligo (HDO) group. After 7 days of oral administration, fecal pellet amount, fecal weight, watercontent of fecal were measured. Blood chemistry, short-chain fatty acid (SCFA), gastrointestinal transit ratio and length and intestinal mucosa were analyzed. RESULTS: Dual-Oligo increased the fecal weight, and water content of feces in rats with loperamide-induced constipation. Gastrointestinal transit ratio and length and area of intestinal mucosa significantly increased after treatment with Dual-Oligoin loperamide-induced rats. A high concentration of Dual-Oligo tended to produce more acetic acid than that observed for the control group, and Dual-Oligo affected the production of total SCFA. Bifidobacteria concentration of cecal contents in the high-concentration oligosaccharide (HDO) and low-concentration oligosaccharide (LDO) groups was similar to the result of the normal group. CONCLUSIONS: These results showed that Dual-Oligo is a functional material that is derived from a natural food product and is effective in ameliorating constipation.

한국인 유아 분변에서 분리한 Lactobacillus acidophilus KY1909의 프로바이오틱 특성 (Probiotic Characteristics of Lactobacillus acidophilus KY1909 Isolated from Korean Breast-Fed Infant)

  • 발효 및 유산균 제품에 모유만을 먹은 생후 3개월된 유아의 분변으로부터 bromocresol pule을 함유한 MRS 배지에서 노란색의 집락을 형성하는 균주들을 내산성 및 담즙산 내성이 우수한 13종의 Lactobacillus 균주들을 분리하였으며 당발효성 및 생화학적 실험을 토대로 분리된 유산균들을 동정하였다. 최종적으로 선발한 균주를 L. acidophilus로 KY1909로 명명하였다. Lactic acid 생성 능력을 평가한 후 이성질체를 분석한 결과 상업적으로 이용되는 L. acidophilus NCFM, L. acidophilus CH5, L. acidophilus NOCKS 균주들은 전체 lactic acid 중에서 L(+) form이 차지하는 비율이 각각 52.49, 40.63 및 63.13%로 나타났으나, L. acidophilus KY1909는 L(+) lactic acid의 비율이 93.96%로 나타났다.

양돈농가에서의 돈사배출수 발생량 및 이화학적 특성 (Characteristics and Quantity of Wastewater Produced by Swine Farms)

  • 양돈농가에서의 계절별 돈사배출수 발생량은 돈사내에서 스크레퍼 및 인력으로 돈분을 분리수거하여 분은 퇴비화하고 배출수는 활성오니법을 이용하여 정화처리하고 있는 농가를 6농가를 선정하여 돈사배풀수랸 및 배출수의 특성을 조사한 결과는 다음과 같다. 1. 돈사배출수량의 평균배출량은 4.15 l/일/두였으며, 계절별로는 여름철 5.02, 겨울철 4.11, 겨울철에 3.44, 봄철에 4.04 l/일.두로 여름철에 가장 많은 세정수가 발생되었다. 2. 돈사배출수 조사농가의 돈분은 수분 함량은 평균 82.9%였고 돈분의 BOD 및 SS농도는 각각 56,847mg/l, 119,750mg/l였으며, 돈사배풀수의 BOD, SS농도는 각각 2,951, 1,983mg/$\ell$였다.
