In order to develop a new improved selective medium for the Bifidobacterium sp. from the human fecal samples, one hundred eight Bifidobacterium strains were isolated and identified. Sensitivity test for the antibiotics and antimetabolites and test for the specific substrate were performed to obtain basic data for the development of the Bifidobacterium selective medium. TOS(transgalactosylated oligosaccharide) was shown to be preferentially used by Bifidobacterium sp.. Sodium propionate promoted the growth of Bifidobacterium while inhibiting other intestinal bacteria. Upon these results, TP medium was designed and shown to be very effective for the selection of Bifidobacterium and better than Mitsuoka BS medium.
In order to guarantee the safety of Korean Naeng Myon Broth in summer, pH and micrological quality of iced noodle gravy which is cooked and sold in public restaurant were investigated. And the variations of pH, taste and microorganism were studied with organic acid treatment during storage period. The micrological status of iced noodle gravy showed that average number of total aerobic bacteria was 2.7${\times}$105CFU/$m\ell$ and fecal coliform was 8.3${\times}$103CFU/$m\ell$ respectly. Food poison microorganisms not detected and average of pH was 4.8. pH changed slightly during storage period and deceased with increasement of organic acid added. Sour taste of organic acid increased acetic acid, citric acid, lactic acid order and there was no organic acid having with more sour taste than iced noodle gravy which is cooked and sold in public restaurant. Salmonella was destroyed when treated with 1% of acetic acid and lactic acid and Escherichia coli became extinct at treatment with 0.8% acetic acid and 1% lactic acid. in case of same concentration, inhibition effects to Escherichia coli multiplication show citric acid, lactic acid, acetic acid order. In conclusion, treatment with more than 0.8% acetic acid and above 1.0% lactic acid or more than 1.0% citric acid in case of production and consumption a day can prevent from Escherichia coli pollution in Korean Naeng Myon Broth.
The aim of this study was to assess the Clinical Usefulness of Helicobacter pylori Stool Antigen (HpSA) immunochromatographic assay for the diagnosis of H. pylori infection. In this study, we had compared HpSA-immunochromatographic assay with CLO test and UBT test. From a total of 140 patients (M:F=88:52) with upper endoscopy, biopsy specimens were obtained for CLO test. Stool specimens was collected from all patients and tested using a HpSA-immunochromatic assay. H. pylori infection status was defined as infected if the results of both CLO test and UBT test were positive. CLO test and UBT test findings showed that 92 patients were H. pylori positive and 48 patients were H. pylori negative. According to this definition, the sensitivity, specificity, and positive or negative predictive value (PPV, NPV) of HpSA-immunochromatographic assay were 97.8%, 100%, 100%, and 96%, respectively. Cross reactivity test of HpSA-immunochromatographic assay were performed with 10 enteric bacteria strains in fecal habitat, and there were no false positive reaction. We evaluated the usefulness of HpSA assay for eradication therapy with 10 of 92 H. pylori positive patients, positive results of them at pre-eradication therapy were converted to negative at post-eradication. The HpSA-immunochromatographic assay is a highly sensitive and specific non-invasive diagnostic method for detection of H. pylori infection, a useful diagnostic method for H. pylori in post eradication stage.
In this study, discharge of the pollutants from the rice paddies during cultivation and its pattern were investigated. The pH in the returned and rainfall water from ten different paddies was between 6.9 and 7.4, which is not associated with the cultivation time and farming style of individual paddy. TN and TP concentrations were highest in the beginning of first top dressing (fertilization), which indicates that proper top dressing and returned water management is crucial to reduce their discharge loads. In particular, TN concentration was more or less constant until tillering stage. This is thought to be due to the runoff of soil-adsorbed nitrogen and compost. Average SS concentration in the returned and rainfall water was 28 mg/L, and organics was predominantly soluble form, and highest COD 25 mg/L during transplanting, lowest 11.3 mg/L and average 11.3 mg/L. Density of TC (total coliform bacteria) was from 1000/100 mL to 114,000/100 mL and FC (fecal coliform) was 5 to 16% of TC density.
Lactobacillus spp. are the bacteria most commonly used as probiotics and it has been proven that they inhibit pathogenic bacterial growth and improve skin repair in humans. This study was conducted to investigate the growth inhibitory effect of Lactobacillus on Staphylococcus pseudintermedius, the most commonly isolated pathogen in canine pyoderma, and whether Lactobacillus could inhibit the adhesion capability of S. pseudintermedius to canine corneocytes. For this study, L. salivarius and S. pseudintermedius were isolated from healthy beagle fecal samples and the skin surface of dogs with skin infection, respectively. S. pseudintermedius was co-cultured with L. salivarius to assess the inhibitory effect. For the adhesion assay, corneocytes were collected from healthy beagle ventral abdominal skin. Both bacterial species attached to corneocytes and were assessed in number. As a result, L. salivarius significantly inhibited the growth of S. pseudintermedius in the culture medium. Moreover, L. salivarius reduced attachment of S. pseudintermedius in the adhesion assay. These results suggest that L. salivarius has an inhibitory effect on S. pseudintermedius and may be effectively used in the topical therapy of canine skin infections.
This review delved into the intricate relationship between the gastrointestinal microbiome and gastric cancer, particularly focusing on post-treatment alterations, notably following gastrectomy, and the effects of anticancer therapies. Following gastrectomy, analysis of fecal samples revealed an increased presence of oral cavity aerotolerant and bile acid-transforming bacteria in the intestine. Similar changes were observed in the gastric microbiome, highlighting significant alterations in taxon abundance and emphasizing the reciprocal interaction between the oral and gastric microbiomes. In contrast, the impact of chemotherapy and immunotherapy on the gut microbiome was subtle, although discernible differences were noted between treatment responders and non-responders. Certain bacterial taxa showed promise as potential prognostic markers. Notably, probiotics emerged as a promising approach for postgastrectomy recovery, displaying the capacity to alleviate inflammation, bolster immune responses, and maintain a healthy gut microbiome. Several strains, including Bifidobacterium, Lactobacillus, and Clostridium butyricum, exhibited favorable outcomes in postoperative patients, suggesting their potential roles in comprehensive patient care. In conclusion, understanding the intricate interplay between the gastrointestinal microbiome and gastric cancer treatment offers prospects for predicting responses and enhancing postoperative recovery. Probiotics, with their positive impact on inflammation and immunity, have emerged as potential adjuncts in patient care. Continued research is imperative to fully harness the potential of microbiome-based interventions in the management of gastric cancer.
We investigated the quality of 10 commercial eorigul-jeot, salt-fermented oysters with seasoning, by measuring their chemical composition and bacterial concentrations. The Eorigul-jeot had 5.07-6.06 pH (mean, 5.63), of 1.92-4.74% salinity (mean, 3.36%), 7.01-14.70 mg/100 g volatile basic nitrogen (mean, 11.91 mg/100 g), 139.22-267.11 mg/100 g amino acid nitrogen (mean, 212.69 mg/100 g), and 1.02-1.65 g/100 g total acidity (mean, 1.24 g/100 g). The total viable and lactic acid bacterial counts were 5.7×104-8.7×105 and 2.7×103-2.0×105 CFU/g, respectively, and fecal coliform was detected in only one Eorigul-jeot sample. Bacillus cereus and Clostridium perfringens were detected in two samples, and all Eorigul-jeot samples were negative for Staphylococcus aureus and Vibrio parahaemolyticus. These results strongly suggest the need to monitor food-poisoning bacteria in commercial Eorigul-jeot to ensure consumer health.
The chickens fed with the feeds supplemented with green tea(GT) and lactic acid bacteria(LB) were infected orally with 10,000 oocysts per chicken of E. maxima. The groups administered with the feeds supplemented with GT by 0.5% and 2.0% of feed showed the significant levels of decreasing in the number of oocysts shed for 5 days after E. maxima infection. The feeds supplemented with LB by 0.1% and 0.5% of feed were less effective in reducing the number of the fecal oocyst, compared with the groups administered with GT. To evaluate the immunoregulatory effects of the feed additives, the expression patterns of IL-2 and IFN-r in spleen cells were studied by RT-PCR and ELISA. The higher levels of IL-2 transcripts after E. maxima infection were observed in the groups with n, compared with the groups with LB and the mixture of GT and LB. The $IFN-\gamma$ mRNA bands were observed in the all of experimental groups except the uninfected control. The culture supernatants of Con A-stimulated spleen cells($5{\times}10^6cells/ml$) were measured for the concentration of IL-2 and $IFN-\gamma$ by ELISA. The levels of IL-2 and $IFN-\gamma$ on days 3 and 7 after E. maxima infection were significantly augmented in the groups with n. These results indicated that GT-supplemented feeds resulted in higher reduction of oocyst-shedding and more enhanced immune responses in the chicken infected with E. maxima, as compared with LB-supplemented feeds. According to the results, it was implicative that the supplements could be utilized for development of feed additives for anti-coccidiosis.
BACKGROUND: Irrigation water is known to be one of the major sources of bacterial contamination in agricultural products. In addition, anti-microbial resistance (AMR) bacteria in food products possess serious threat to humans. This study was aimed at investigating the prevalence of foodborne bacteria in irrigation water and evaluating their anti-microbial susceptibility. METHODS AND RESULTS: Surface water (n = 66 sites) and groundwater (n = 40 sites) samples were collected from the Gyeongi and Gangwon provinces of South Korea during April, July, and October 2019. To evaluate the safety of water, fecal indicators (Escherichia coli) and foodborne pathogens (E. coli O157:H7, Salmonella spp., and Listeria monocytogenes) were examined. E. coli isolates from water were further tested for antimicrobial susceptibility using VITEK2 system. Overall, detection rate of foodborne pathogens in July was highest among three months. The prevalence of pathogenic E. coli (24%), Salmonella (3%), and L. monocytogenes (3%) was higher in surface water, while only one ground water site was contained with pathogenic E. coli (2.5%). Of the 343 E. coli isolates, 22.7% isolates were resistant to one or more antimicrobials (ampicillin (18.7%), trimethoprim-sulfamethoxazole (7.0%), and ciprofloxacin (6.7%)). CONCLUSION: To enhance the safety of agricultural products, it is necessary to frequently monitor the microbial quality of water.

This study evaluated the effect of lactic acid bacteria (LAB, a mixture of Enterococcus faecium and Lactobacillus plantarum) supplementation, the storage temperature, and storage period on the fermentation characteristics and in vitro ruminal digestibility of a total mixed ration (TMR). The TMR was prepared into two groups, namely, CON (control TMR without the LAB) and ML (supplementing a mixture of E. faecium and L. plantarum in the ratio of 1% and 2% (v/w), respectively). Both groups were divided and stored at 4℃ or 25℃ for 3, 7, and 14 d fermentation periods. Supplementing LAB to the TMR did not affect the chemical composition of TMR except for the lactate and acetate concentration. Storage temperatures affected (p<0.05) the chemical composition of the TMR, including pH, lactate, and acetate contents. The chemical composition of TMR was also affected (p<0.05) by the storage period. During in vitro rumen fermentation study, the ML treatment showed lower (p<0.05) dry matter digestibility at 24 h incubation with a higher pH compared to the CON. There was no difference in the in vitro dry matter digestibility (IVDMD) of TMR between the CON and ML treatment however, at 24 h, ML treatment showed lower (p<0.05) IVDMD with a higher pH compared to the CON. The effects of storage temperature and period on IVDMD were not apparent at 24 h incubation. In an in vivo study using Holstein steers, supplementing LAB to the basal TMR for 60 d did not differ in the final body weight and average daily gain. Likewise, the fecal microbiota did not differ between CON and ML. However, the TMR used for the present study did include a commercial yeast in CON, whereas ML did not; therefore, results were, to some extent, compromised in examining the effect of LAB. In conclusion, storage temperature and period significantly affected the TMR quality, increasing acetate and lactate concentration. However, the actual effects of LAB supplementation were equivocal.
