An Assay of L-Ascorbic Acid with Crude Enzyme from Cucumber(Cucumis sativus L.) (오이(Cucumis sativus L.)에서 추출한 조효소 용액을 이용한 L-Ascorbic Acid의 정량분석)

  • The test possibility of L-ascorbic acid(AsA) assay by ascorbate oxidase(A.O) solution obtained from cucumber was estimated. The results obtained were summarized as follow. The difference of absorbance before and after oxidation of AsA by A.O solution was proportional to the AsA concentration. Iso- AsA compared to same concentration of AsA was 97% response. Therefore, the 0 specificity on Iso- AsA a was deficient. On the sucrose, glucose, fructose were coexisted from 5 times, 500 times of AsA concentration, none of them were affected by any level of AsA concentration. In the case of EDTA, it was not nearly affected that EDTA was coexisted 50 times of AsA concentration, but EDTA Inhibited considerably from 500 times of AsA concentration. The effect of citric acid on the AsA concentration was not in the same level of AsA, however, it was slightly inhibited from 5 times, 50 times of AsA concentration and 500 times of AsA inhibited remarkably As known L-AsA was catalycally autooxidated by Cu2+ and Fe2+. They were not nearly affected In the above 0.01M level on the Ash assay. However, Fe3+ ion was neither nearly affected in the 0.001M level nor In the above 0.01M level on the AsA assay.

A Study of Greenish Pigments from Rhodopseudomonas viridis by Acetone Extraction: Characteristics of Potential Food Colorant (Acetone 추출한 Rhodopseudomonas viridis 녹색색소에 대한 연구: 식용색소로서의 일반적 성질)

  • A study of physical and chemical characteristics of pigments from Rhodopseudomonas viridis DSM 133 was carried out for development of natural greenish colorant. Through visible absorption scanning, it showed three main absorption peaks at 378, 414 and 677 nm with three minor peaks at 510, 540 and 618nm, and it was shown to be greenish color. These pigments were more stabilized in alkaline solutions than in acid of between pH 6 and 9, and it was shown to be stabilized at the temperature below $40^{\circ}C$. In the presence of light and oxygen, the stability of pigments rapidly degraded, and it became unstable in the presence of metal ion such as $Fe^{3+}$ and $Al^{3+}$. But in the presence of $Cu^{2+}$ were very stable. On the result of TLC analysis, pigments were shown to be composed of four color fractions and main color fractions were F-4 and F-2.

  • The sorption of UO$_{2}$$^{2+}$ onto goethite and kaolinite under various experimental conditions was successfully interpreted using surface complexation modeling (SCM). The SCM approach used in this work is the triple-layer model (TLM) in which weakly bonded ions are modeled as outer-sphere (ion-pair) complexes and strongly bonded ions as inner-sphere (surface coordination) complexes. The change of ionic strength did not affect the U(VI) sorption onto goethite, thus the formation of inner-sphere surface complexes, (FeO)$_2$UO$_2$ and (FeO)$_2$(UO$_2$)$_3$OH$_{5}$ was assumed to simulate the effects of ionic strength and goethite concentration. On the other hand, the U(VI) sorption onto kaolinite showed ionic strength dependence, thus the formation of AlO-UO$_{2}$$^{2+}$(outer-sphere complex) and SiO(UO$_2$)$_3$OH$_{5}$ (inner-sphere complex) was assumed to simulate the experimental data. In the presence of carbonates, the sorption of U(VI) onto kaolinite decreased in the weakly alkaline pH range. This was well simulated assuming the formation of a outer-sphere surface complex, A1OH$^{2+}$- (UO$_2$)$_2$CO$_3$OH$_3$. Since SCM approach uses thermodynamic data such as surface complexation constants, it is more predictive than empirical modeling approach in which conditional values such as partition coefficient are used. used.

  • The spectrophotometric determination of Al(III) with thorin have been investigated. The optimum condition of pH, concentration of ligand and surfactant, and stability were evaluated. The thorin ligand offers selective separation of Al(III) from sample solution containing Fe(III), Ni(II), Cu(II), Pb(II) and Cu(II). Various surfactants were tested and Triton X-100 showed the best stability and the maximum absorbance in an aqueous solution of Al(III)-Thorin-Triton X-100 complex appears about 526 nm. The method was applied for the determination of Al(III) in mixed sample solution. Separation and preconcentration was performed with a short column filled with resorcinol-formaldehyde resin. Control of the pH during the column operation is essential because the adsorption capacities are very sensitive to change in pH. Their separation was carried out in 0.2 M acetic acid-sodium acetate buffer solution (pH 4.5) and 1.0 M $HNO_3$media.

  • This study investigated the separation and the property of palladium precipitate formed by ascorbic acid in a simulated radioactive liquid waste, which was composed of 10 elements((Pd, Ru, Rh, Nd, Cs, Sr, Fe, Ni, Zr, Mo). Pd was separated selectively by using reduction characteristics of metal ions contained in the simulated waste with ascorbic acid. When the nitric acid concentration was 0.5 M, the Pd over 99.5% was precipitated by adding 0.04 M ascorbic acid. Nitric acid concentration is important at the reduction reaction of Pd ion. The precipitation yield of Pd was decreased as the concentration of nitric acid was increased. The Pd precipitate was re-dissolved in reaching at an equilibrium when the concentration of nitric acid was high and ascorbic acid was added with a small amount. The Pd precipitate formed by ascorbic acid was Pd metal and was aggregated by particles less than $1.0{\mu}m$.

  • Cytochrome cL (CytcL) is an essential protein in the process of methanol oxidation in methylotrophs. It receives an electron from the pyrroloquinoline quinone (PQQ) cofactor of methanol dehydrogenase (MDH) to produce formaldehyde. The direct electron transfer mechanism between CytcL and MDH remains unknown due to the lack of structural information. To help gain a better understanding of the mechanism, we determined the first crystal structure of heme c containing CytcL from the aquatic methylotrophic bacterium Methylophaga aminisulfidivorans MPT at 2.13 Å resolution. The crystal structure of Ma-CytcL revealed its unique features compared to those of the terrestrial homologues. Apart from Fe in heme, three additional metal ion binding sites for Na+, Ca+, and Fe2+ were found, wherein the ions mostly formed coordination bonds with the amino acid residues on the loop (G93-Y111) that interacts with heme. Therefore, these ions seemed to enhance the stability of heme insertion by increasing the loop's steadiness. The basic N-terminal end, together with helix α4 and loop (G126 to Y136), contributed positive charge to the region. In contrast, the acidic C-terminal end provided a negatively charged surface, yielding several electrostatic contact points with partner proteins for electron transfer. These exceptional features of Ma-CytcL, along with the structural information of MDH, led us to hypothesize the need for an adapter protein bridging MDH to CytcL within appropriate proximity for electron transfer. With this knowledge in mind, the methanol oxidation complex reconstitution in vitro could be utilized to produce metabolic intermediates at the industry level.

Purification and Some Properties of Polyphenol Oxidase from Arrowroot (칡 뿌리의 Polyphenol Oxidase의 정제 및 성질에 관한 연구)

  • Acetone powder was prepared from raw arrowroots and the polyphenol oxidases of crude enzyme prepared from acetone powder were identified 5 isoenzymes by staining with catechol containing 0.05% phenylene diamine. The crude enzyme was passed through the columns of ion exchangers and gel permeation to fractionate the polyphenol oxidases. The main fraction of polyphenol oxidase appeared to be purified by 94-fold, with the activity yield of 45.4%, and its molecular weight was determined as 38,500 by poly acrylamide gel electrophoresis. The optimal pH and temperature for the enzyme activity were pH 7.5 and $50^{\circ}C$, respectively. The purified enzyme showed a high affinity for catechol and pyrogallol. The Michaelis constant for catechol was calculated to be 16.67mM according to the Lineweaver-Burk method. The enzyme activity was strongly inhibited by L-ascorbic acid, sodium bisulfite, EDTA and KCN, and totally inhibited, by $Fe^{3+}$ at a concentration of 1mM. However the enzyme was activated by $Zn^{2+}$ approximately 1.7 times at the same concentration.

A Study on the Alkaline Protease Produced from Bacillus subtilis (Bacillus subtilis가 생산하는 Alkaline Protease에 관한 연구)

  • The alkaline protease producing bacteria isolated from soil and identified as Bacillus subtilis. The optimum medium for alkaline protease production from the microorganism was as follows; soluble starch, 1.5% ; proteose peptone, 0.5% ; $K_2HPO_4$, 0.1% ; $MgSO_4{\cdot}7H_2O$, 0.02% and sodium carbonate, 1.0%. The optimum temperature for alkaline protease production was $35^{\circ}C$, and the initial pH of medium was pH 10.5. The alkaline protease activity was about 2,300 U per ml of culture broth by Casein-Folin Method. A 9.2 fold purification of alkaline protease was obtained from culture broth. The recovery was 14% and purified enzyme was identified as single band, and its molecular weight was about 19,000. The optimum temperature for enzyme reaction was $70^{\circ}C$, and optimum pH was 12. The activity of purified enzyme was inhibited by metal ion ($Fe^{++}$), and Phenylmethylsulfonyl Fluoride, a serine protease inhibitor.

  • Water chemistry in the Munkyeong hot-spring expresses high values of EC(1,857 $mutextrm{s}$/cm), $HCO_3$(1,250 mg/l), $SO_4$(147.60 mg/l), Mg(43.05 mg/l), and Ca(279.43 mg/l). The precipitates of small quantity is formed in lower temperature, but much of in case apply heat by boiler. Although mineral that is settled from original ground water is most calcite, aragonite and calcite at the same time crystallized in boiler. The $CO_3$ is present predominantly as $HCO_3^{-}$ and $H_2$$CO_3$, $SO_4$, Mg and Ca are present as free ion. Ca is saturated with respect to carbonate such as aragonite and calcite but slightly undersaturated with respect to anhydrite and gypsum Al is saturated with diaspore and gibbsite. The precitptates are composed of carbonate such as calcite and aragonite and amorphous Fe-hydroxide.

Synthesis and Phosphorus Adsorption Characteristics of Zirconium Magnetic Adsorbent Having Magnetic Separation Capability (자기분리가 가능한 지르코늄 자성 흡착제의 합성과 인 흡착 특성)

  • The purpose of this study, is to separate magnetic separation devices using permanent magnets by using magnetization characteristics remaining in treated water after adsorption and synthesizing phosphorus adsorbent capable of magnetic separation for efficient removal of phosphorus. The synthesis of the adsorbent which set Zirconium(Zr) having high friendly features for phosphorus as an element, and by synthesizing Iron Oxide($Fe_3O_4$, another name of $Fe_3O_4$ is magnetite) being able to grant magnetism to Zirconium Sulfate($Zr(SO_4)_2$), zirconium magnetic adsorbent(ZM) were manufactured. In order to consider the phosphorus adsorption characteristics of adsorbent ZM, batch adsorption experiment was performed, and based on the results, pH effect, adsorption isotherm, adsorption kinetics, and magnetic separation have been explore. As the experiment result, adsorbent ZM showed a tendency that the adsorption number was decreased rapidly at pH 13; however, it was showed a high amount of phosphorus removal in other range and it showed the highest amount of phosphorus removal in pH 6 of neutral range. In addtion, the Langmuir adsorption isotherm model is matched well, and D-R adsorption isotherm model is ranged 14.43kJ/mol indicating ion exchange mechanism. The result shown adsorption kinetics match well to the Pseudo-second-order kinetic model. The adsorbent ZM's capablility of regenerating NaOH and $H_2SO_4$, was high selectivity on the phosphorus without impacts on the other anions. The results of applying the treated water after adsorption of phosphorus to the magnetic separation device by using permanent magnets, shows that capture of the adsorbent by the magnetization filter was perfect. And they show the possibility of utilization on the phosphorus removal in water.
