구형 SiO2@Y2O3: Eu 코어-쉘 복합체 형광체 합성 및 특성 (Synthesis and Characterization of Spherical SiO2@Y2O3 : Eu Core-Shell Composite Phosphors)

  • The monodisperse spherical $SiO_2$ particles were overcoated with $Y_2O_3:Eu^{3+}$ phosphor layers via a Pechini sol-gel process and the resulting $SiO_2@Y_2O_3:Eu^{3+}$ core-shell phosphors were subsequently annealed at $800^{\circ}C$ at an ambient atmosphere. The crystallographic structure, morphology, and luminescent property of core-shell structured $SiO_2@Y_2O_3:Eu^{3+}$ phosphors were characterized by X-ray diffraction (XRD), field emission scanning electron microscopy (FE-SEM), and photoluminescence (PL). The spherical, nonagglomerated $SiO_2$ particles prepared by a Stober method exhibited a relatively narrow size distribution in the range of 260-300 nm. The thickness of phosphor shell layer in the core-shell particles can be facilely controlled by varying the coating number of $Y_2O_3:Eu^{3+}$ phosphors. The core-shell structured $SiO_2@Y_2O_3:Eu^{3+}$ phosphors showed a strong red emission, which was dominated by the $^5D_0-^7F_2$ transition (610 nm) of $Eu^{3+}$ ion under the ultraviolet excitation (263 nm). The PL emission properties of $SiO_2@Y_2O_3:Eu^{3+}$ phosphors were also compared with pure $Y_2O_3:Eu^{3+}$ nanophosphors.

  • The objective of the present study was to establish the method of measurement of hydrogen peroxide and to estimate the anti-oxidative effect of genistein in the skin. UVB induced skin oxidation and anti-oxidative effect of genistein formulations were evaluated by determining levels of hydrogen peroxide. The mechanism involved in the determination of hydrogen peroxide is based on a color reaction between ferric ion ($Fe^{3+}$) and xylenol orange, often called FOX assay and subsequent monitoring of absorbance values of the reactant at 540 nm. The reaction was to some extent pH-dependent and detection sensitivity was greatest at pH 1.75. Genistein liposomal gel demonstrated better anti-oxidative effect with regard to lowering hydrogen peroxide levels elevated by UVB irradiation compared to genistein-suspended gel. A linear relationship has been observed between anti-oxidative effect of genistein and drug deposition in the skin tissue. Genistein liposomal gel resulting in the localization of the drug in the deeper skin led to improved anti-oxidative effect compared to genistein gel. The suggested method for evaluation of oxidation of the skin can be used as a tool to screen effective anti-oxidative agents and their delivery systems acting on the skin.

Layer-by-Layer 코팅법을 적용한 복합막 제조와 투과성능 평가 (Preparation and Performance of Composite Membrane Prepared by Layer-by-Layer Coating Method)

  • 폴리설폰막 위에 친수성 고분자를 Layer-by-Layer법으로 코팅하여 복합막을 제조하였다. FE-SEM 분석을 통하여 복합막 표면과 기공 내 코팅층을 확인하였다. 또한 100 ppm NaCl 용액에 대한 복합막의 투과성능 평가를 실시하였다. 복합막 제조를 위한 코팅 고분자는 PVSA, PEI, PAA, PSSA, PSSA_MA를 사용하였다. 폴리설폰막 표면에 8,000 ppm PAA (이온세기 0.35) 수용액을 3분 동안 코팅한 뒤 10,000 ppm PEI 수용액을 4분 동안 코팅하였다. 그 결과 PAA-PEI 복합막의 투과도는 101 LMH, 제거율은 66.7%로 가장 좋은 투과성능을 나타내었다. PAA-PEI 복합막의 투과성능은 도레이케미칼의 NE 4040-70 (투과도 = 30 LMH, 염 배제율 = 40~70%) 제품과 유사한 성능을 보여주는 우수한 투과 특성을 나타내었다.

  • 투명 전극(transparent conducting oxide, TCO)은 높은 전기전도도 및 낮은 비저항 ($10^{-4}{\sim}10^{-3}\;{\Omega}cm$)과 가시광영역에서의 우수한 광투과도(> 80%) 특성을 가지며, 주로 디스플레이, 태양전지, 가스 센서 소자 등에 쓰인다. 투명전극으로 쓰이는 대표적인 물질로서는 ITO, ZnO, $SnO_2$ 등이 있으며, ITO는 전기적 특성이 우수하여 널리 사용되고 있으나 가격이 비싸고 화학적으로 불안정하고, ZnO는 ITO에 비해 가격이 저렴하지만 고온에서 불안정한 특성을 가지고 있다. 반면, $SnO_2$는 ITO와 ZnO에 비해 전기적 특성은 떨어지지만, 우수한 열적, 화학적 안정성 및 높은 내마모성을 가지고 제조단가가 저렴하여 TCO 재료로 많은 연구가 진행되고 있다. TCO 박막을 증착시키는 방법으로 CVD, ion plating, sputtering, spray pyrolysis 등이 있으며, 이 중 sputtering 방법은 균일한 입자로 균질의 박막을 입힐 수 있고 우수한 재현성과 낮은 온도에서도 증착이 가능하여 박막 제조 방법으로 널리 이용되고 있다. 본 연구에서는 $SnO_2$ 박막을 실리콘 (100) 및 글라스 (Eagle 2000) 기판 위에 RF magnetron sputtering 방법을 사용하여 제작하였다. 박막 증착을 위해 99.99%의 2 인치 un-doped $SnO_2$ 타겟을 사용하였고, 기판은 20 rpm 으로 회전시켜 균일한 박막이 형성될 수 있도록 하였으며, 초기 진공도는 $1{\times}10^{-6}\;Torr$가 되도록 하였다. 증착 변수로 기판-타겟간 거리, RF 파워, $O_2/(Ar+O_2)$ 비, 공정압력, 기판 온도 등을 각각 변화 시키며 $SnO_2$ 박막을 증착하였다. 증착된 박막의 구조적 및 광학적 특성을 분석하기위해 FE-SEM, AFM, XRD, UV/VIS spectrophotometer, Photoluminescence 등을 사용하였다.

Purification and characterization of an extracellular protease from culture filtrate of salmonella schttmulleri

  • An extracellular protease of Salmonella schottmulleri was purified from culture filtrate by using 0-75% ammonium sulfate precipitation, DEAE Sepharose Fast Flow ion exchange chromatography, Ultrogel HA chromatography and Sephacryl S-200 HR molecular sieve chromatography. To measure enzyme activity, synthetic dipeptide substrate (CBZ-arg-arg-AFC) with low molecular weight was employed as substrate. The molecular weight of the purified enzyme was approximately 80 kDa when determined by gel filtration on Sephacryl S-200 HR and 73 kDa when estimated by SDS-PAGE. The isoelectric point was 5.45. The activity of the purified enzyme was inhibited by metal chelating agesnts such as EDTA and 1.10-phenanthroline. The divalent cations, such as Ca$\^$2+/, Zn$\^$2+/, Fe$\^$2+/, Mg$\^$2+/ enhanced its activity. These results suggested that it was a metalloprotease. It had a narrow pH optimum of 6.5-7.5 with a maximum at pH 7.0 and a temperature optimum of 40.deg.C. It was stable at least for 1 week at 40.deg.C and maintained its activity for 24 hours at 50.deg.C, but it was rapidly inactivated at 65.deg.C. Purification and Characterization of a Novel Salt-tolerant Protease Produced by Saccharomyces sp. B101 Isolated from Baker's Dough Yeast

  • The proteolytic enzyme from Saccharomyces sp. B101 was purified to homogeneity by ammonium sulfate fractionation, ultrafiltration, diethyl aminoethyl (DEAE)-Sephadex A-50 ion-exchange chromatography, and Sephadex G-100 gel filtration chromatography from the culture supernatant of Saccharomyces sp. B101. The specific activity and the purification fold of the purified enzyme were 4,688.9 unit/mg and 18, respectively. The molecular weight of the purified enzyme was estimated to be 33 kDa by sodium dodecylsulfate-polyacrylamide gel electrophoresis (SDS-PAGE). The optimum pH and temperature for the enzyme activity were pH 8.5 and $30^{\circ}C$, respectively. The enzyme activity was relatively stable in the pH range of 6.5-8.5 at below $35^{\circ}C$. The salt-tolerance and stability for the enzyme activity were relatively stable even at NaCl concentrations of 10 and 15%. The activity of enzyme was inhibited by $Ag^{2+}$ and $Fe^{2+}$, and activated by $Mn^{2+}$. In addition, the enzyme activity was potently inhibited by ethylenediaminetetraacetic acid (EDTA) and phenylmethyl sulfonylfluoride (PMSF). Based on these findings we concluded that the purified enzyme was a serine protease. Km and Vmax values for hammastein milk casein were 1.02 mg/mL and 278.38 unit/mL, respectively.

  • Artemia franciscana의 cyst에서 protease를 정제하가 위해 세포를 파쇄하여 원심 분리한 후 상등액을 회수하였다. 그상등액에서 40-60% 황산암모늄을 첨가하여 침전시켜서 염을 제가한 후, Sphadex G-200 및 DEAE-Sephadex A-50 column chromatography를 하여 정제한 후 본 효소의 특성을 조사한바 그 특성은 아래와 같았다. 정제 과정을 거치는 동안, 비활성은 13.64배 증가하였고, 수율은 7%였다. 그리고 이효소는 pH 6에서는 안정하였으나, pH 8이상에서는 대부분의 활성이 실활 되었고, 최적 pH는 3.0이었다. 따라서 이 효소는 전형적인 산성 pro-tease로 판정되었다. 본 효소는 60$^{\circ}$C이상의 온도에서 대부분 실활 되었고, 최적 활성 반응 온도는 35$^{\circ}$C였다. 금속 이온의 영향을 알아본 결과, $Cu^{++}$, Z$Zn^{++}$, $Fe^{++}$가 효소의 활성을 저해하였고, 중금속 chelator인 EDTA는 반대로 효소 활성을 증가시켰다. 단백질분해효소에 특이적인저해제를 첨가하여 실험한 결과 thiol protease ingibitor인 antipain, chymostatin, leupeptin, E-64, iodoacetate등에 효과적으로 효소활성이 저해받으므로 본 효소는 acid thiol protease라 판정했다.

Magnetite for phosphorus removal in low concentration phosphorus-contained water body

  • Magnetite was chosen as a typical adsorbent to study its phosphate adsorption capacity in water body with low concentration of phosphorus (below $2mg\;PL^{-1}$). Magnetite was collected from Luoyang City, Henan Province, China. In this research, three factors have been studied to describe the adsorption of phosphate on magnetite, which was solution concentration (concentration ranging from 0.1 to $2.5mg\;PL^{-1}$), suspension pH (1 to 13) and temperature (ranging from $10^{\circ}C$ to $40^{\circ}C$). In addition, the modified samples had been characterized with XRD and FE-SEM image. The results show that iron ions contains in magnetite were the main factors of phosphorus removal. The behavior of phosphorus adsorption to substrates could be fitted to both Langmuir and Freundlich isothermal adsorption equations in the low concentration phosphorus water. The theoretical saturated adsorption quantity of magnetite is 0.158 mg/g. pH has great influence on the phosphorus removal of magnetite ore by adsorption. And pH of 3 can receive the best results. While temperature has little effect on it. Magnetite was greatly effective for phosphorus removal in the column experiments, which is a more practical reflection of phosphorous removal combing the adsorption isotherm model and the breakthrough curves. According to the analysis of heavy metals release, the release of heavy metals was very low, they didn't produce the secondary pollution. The mechanism of uptake phosphate is in virtue of chemisorption between phosphate and ferric ion released by magnetite oxidation. The combined investigation of the magnetite showed that it was better substrate for water body with low concentration of phosphorus.

Citric Acid Reduces Alkaline Stress-induced Chlorosis, Oxidative Stress, and Photosynthetic Disturbance by Regulating Growth Performance, Antioxidant Activity and ROS Scavenging in Alfalfa

  • Pollution of agricultural soil by alkaline salts, such as Na2CO3, is a critical and long-lasting problem in cultivable land. The aim of the study was to examine the putative role of citric acid (CA) in alleviating Na2CO3-stress in alfalfa. In this study, Na2CO3 significantly induced leaf chlorosis, inhibited plant growth and photosynthesis related parameters, increased hydrogen peroxide (H2O2) and reduced major antioxidant enzymes (SOD, CAD, APX) in alfalfa. However, the presence of CA these negative effects of Na2CO3-stress largely recovered. Interestingly, expression of antioxidant and ion transporter genes (Fe-SOD, CAT, APX, DHAR and NHX1) involved in Reactive oxygen species (ROS) homeostasis and oxidative stress tolerance in alfalfa. These findings suggest that CA-mediated Na2CO3 stress alleviation is an ecofriendly approach that would be useful to local farmer for alfalfa and other forage crop cultivation in alkaline soils.

Electrochemical Characteristics of Water-Soluble Phosphate-Functionalized Naphthalene- and Perylene-Bisimides and Their Zirconium Bisphosphate Multilayers on ITO Electrode

  • N,N'-bis(ethyldihydrogen phosphate)-1,4,5,8-naphthalene bis(dicarboximide) (EPNI) and N,N'-bis(ethyldihydrogen phosphate)-3,4,9,10-perylene bis(dicarboximide) (EPPI) and their zirconium bisphosphate multilayers (Zr-EPNI and Zr-EPPI), that had been briefly reported by us, were further investigated in terms of their electrochemical properties. EPNI in aqueous solution showed typical two reversible reductions at ITO electrode but the reductions were strongly dependent on solution pH while EPPI showed only an irreversible reduction. The single and mixed multilayers of Zr-EPNI and Zr-EPPI were well constructed on ITO electrode by the alternate adsorptions of zirconium ion and the bisimides. While Zr-EPNI multilayer on ITO electrode showed single broad reversible reduction with $E_{1/2}=-0.68V$, Zr-EPPI gave two separated reductions at $E_{1/2}=-0.54$ and -0.81 V vs SCE, quite differently from the solution properties. The average layer densities of the multilayers were estimated as $1.5{\times}10^{-10}$ and $2.3{\times}10^{-10}mol/cm^2$ for Zr-EPNI and Zr-EPPI, respectively. Both the monolayers of Zr-EPNI and Zr-EPPI could not completely block the electron transfer between $Fe(CN){_6}^{3-}$ in solution and ITO electrode but 3-5 layers of Zr-EPNI and Zr-EPPI could block it completely and mediated the one-way electron transfer at the potential shifted to their reduction potentials. When the monolayer of zirconium 1,10-decanediylbisphosphonate (Zr-DBP) was used as a sublayer of Zr-EPNI and Zr-EPPI layers, the mediated electron transfer became prominent without any direct electron transfer.
