Evaluation of Physico-chemical Properties of Acrylic Resin Hydrogel and their Application to Transdermal Delivery System

  • Recently, many attempts have been made to use hydrogels of various polymers as delivery systems of various drugs and bioactive materials to prolong and control their phamacological activities. In this study, we have evaluated the physico-chemical properties of methacrylic acid-methyacrylic acid methyl ester copolymer 9Eudispert mv)m a acrylic resin hydorgel, and its application to transdermal delivery system. In the dissolution tests, the release rate of salicylic acid (SA) and sodium salicylate (SOd. SA) were faster than lidocain (LD) and lidocain-HCl(LD-HCl). As the concentration of Eudispert mv polymer increased, the extensibility of Eudispert mu hydrogel decreased, whereas the swelling ratio increased. The more NaOH and polymer concentration increased, the more osmotic pressure linearly increased. The skin permeation of Sod. SA, an acidic model drug, was remarkably enhanced by Eudispert mv hydrogel. All fatty acids, except for Sod. glycolate, dramatically increased the skin permeation flux in Eudispert mu hydrogel containing LD-Hcl, a basic model drug. Consequently, it is suggested that Eudispert mv hydrogel may be used as potential transdermal delivery vehicle.

한국산 호도의 단백질 및 지질의 조성에 관한 연구 (Studies on the Composition of Protein and Lipid from Korean Walnut (Juglans regia L.))

  • 한국산 호도의 단백질 및 지방성분을 gel filtration, polyacrylamide gel electrophoresis, 아미노산분석기, thin layer chromatography 및 gas liquid chromatography에 의하여 분석하였고 그 일반성분은 조단백질의 22.18%, 조지질이 64.23%였다. 호도 단백질의 sodium dodecyl sulfate polyacrylamide gel electrophoresis에 의한 결과 12개의 band를 나타내었고 분획된 주단백질의 수득율은 60.67%였으며 이것의 분자량은 43,000이었다. 아미노산 조성은 17종류로써 glutamic acid가 38.60%로써 가장 많았고 다음은 arginine이 9.45%, aspartic acid가 6.55% 순으로 나타났으며 필수아미노산이 고르게 함유되어 있었다. 호도기름에는 중성지질이 93.05%이나 복합지질은 약 7.0%에 불과하였으며 중성지질의 성분으로써 82.05%가 triglyceride였고 sterol ester 및 유리지방산이 각각 3.85% 및 4.80%였다. 호도의 총지질과 중성, 당 및 인지질의 지방산 조성은 oleic acid 및 linoleic acid가 각각 $13.89{\sim}15.36%,\;64.48{\sim}69.98%$였고 중성지질에서 분별된 triglyceride의 지방산 조성은 linoleic acid가 69.98%로 가장 높은 함량이었다.

한국산 마의 지질 성분의 분리 및 분석 (Fractionation and Quantitative Analysis of Lipid Components in Korean Yam (Dioscorea) Tubers)

  • 한국산 마의 주요 품종인 Dioscorea batatas, D. aimadoimo and D. japonica 3가지 품종의 시료들을 동결건조하여 분말로 만든 다음 지질의 추출, silicic acid column chromatography에 의한 지질의 분획, thin layer chromatography에 의한 지질의 분리 및 gas chromatography에 의한 지방산 조성 등을 행하였다. 총지질의 함량은 D. japonica가 다른 마에 비해 적은 편이었으며, 지질을 분획한 결과 3가지 품종 모두 중성지질이 가장 많았고 그 다음 당지질, 인지질 순으로 나타났다. 분획한 각 지방의 구성 지질로는 중성지질이 sterolester, triglyceride, 1,3-diglyceride, 1,2-diglyceride 및 monoglycerid였고, 당지질에서 acylsterylglycoside, sterylglycoside, digalactosyldiglyceride 및 sulfolipid였고, 인지질에서 phosphatidylethanolamine, phosphatidylcholine 및 phosphatidylinositol 이었다. GC에 의한 총지질의 지방산 조성은 팔미트산과 리놀레산이 가장 많은 부분을 차지하였고 불포화지방산 함량이 포화지방산 함량보다 많았다. 또한 중성지질, 당지질 및 인지질의 지방산 조성도 총 지질을 구성하는 지방산 조성과 같은 양상을 보여 주었다.

잣지질 성분의 분획정량 및 각 획분의 지방산 조성 (Lipid Contents and Fatty Acid Composition of Three Lipid Classes in Korean Pinenut)

  • 잣(Pinus koraiensis S & Z)에서 얻은 총지질 성분을 silicic acid column chromatography에 의해 중성지질, 당지질, 인지질로 분획 정량하고, 다시 이 분획된 성분을 thin layer chromatography 로써 재분리시켜 구성성분들을 분별하고 scanner에서 정량하였으며, 각 획분별 지방산 조성을 gas chromatography로써 분석한 결과는 다음과 같다. 1) 잣지질의 구성은 중성지질이 총지질의 $95.9{\sim}96.7%$, 당지질이 $2.5{\sim}3.2%$, 인지질이 $0.8{\sim}0.9%$였으며, 중성지질에서는 triglyceride(80.8%), diglyceride(6.3%), free fatty acid(5.2%), sterol(4.6%) ,sterol ester 및 hydrocarbon 등이 분별 정량되었으며, 이 중 triglyceride가 주된 물질이었다. 당지질에서는 monogalactosyl diglyceride 및 esterified sterylglycoside(23.5%), cerebroside 및 steryl glycoside(7.8%), digalactosyl diglyceride(2.3%) 등이 분별정량되였고, 인지질의 획분에서는 phosphatidyl cho-line(40.2%), phosphatidyl ethanolamine(19.4%), phosphatidyl inositol(7.3%), lysophosphatidyl choline(5.8%), phosphatidyl serine(2.6%) 등이 분별 정량되었다. 2) 각 분획된 지질의 지방산 조성은 중성지질의 경우 linoleic acid(48.7%), oleic acid(28.8%), arachidic acid(14.4%), palmitic acid(5.7%), stearicacid(1.84%), linolenic acid(0.7%), myristic acid(0.1% 이하), lauric acid(0.1% 이하) 등이 분리 정량되었으며, 당지질과 인지질에서도 linoleic acid, oleic acid등이 주성분으로 이루어져 있었다. 그러나 인지질의 경우 palmitic acid(17.7%)와 stearic acid(6.0%)의 함량이 다른 지질획분에 비해 다소 많았다.

Dietary supplementation of finishing pigs with the docosahexaenoic acid-rich microalgae, Aurantiochytrium limacinum: effects on performance, carcass characteristics and tissue fatty acid profile

  • Objective: The aim of this experiment was to evaluate the effect of dietary supplementation with the docosahexaenoic acid (DHA)-rich microalgae, Aurantiochytrium limacinum (AURA) on pig performance, carcass traits, and the fatty acid composition of pork Longissimus lumborum (LL) and backfat. Methods: A total of 144 Pig Improvement Company (PIC)${\times}$Goland finishing pigs (72 females and 72 castrated males) of mean weight 117.1 (${\pm}13.1$) kg were blocked by sex and body weight and provided with 0% or 1% AURA in isonutritive and isocaloric diets. A total of 24 pens provided 12 replicates per treatment. Animals were weighed on day 0 and 28 with feed and water intake recorded per pen. After 31 days supplementation (28 days of study and 3 days until the slaughtering date) three animals per pen (n = 72) were slaughtered and the LL and backfat thickness, lean meat content and dressing percentage were recorded for the carcasses. The fatty acid (FA) profile of the LL and backfat was established by direct FA methyl ester synthesis. Results: No differences were observed for any performance parameters or carcass traits. Supplementation with AURA resulted in significant changes to the FA profiles of both the LL and backfat with male and female pigs responding differently to supplementation in terms of particular FAs. Overall, pork LL samples had significantly higher eicosapentaenoic acid (p<0.001) and DHA concentrations (p<0.001), and higher omega-3 (n-3) FAs (p<0.001), as well as an increased omega3:omega6 (n-3:n-6) ratio (p = 0.001). For backfat, supplementation resulted in significantly higher amounts of DHA (p<0.001) and n-3 FAs (p<0.001). Conclusion: These results indicate that dietary supplementation with 1% AURA over a 31 day period can increase the FA composition of pork LL and backfat, specifically the DHA, with no major impact on growth performance and carcass traits.

닭에서의 지방산(脂肪酸) 생합성(生合成)에 미치는 급여지방산(給與脂肪酸)의 영향(影響) (Fatty Acid Biosynthesis of Chicken Fed Various Long Chain Fatty Acids)

  • 무지방사료(無脂肪飼料) 및 여기에 17:0, 18:0 혹은 18:2을 첨가(添加)한 사료(飼料)를 병아리에 급여(給與)해서 간장(肝臟) 및 혈장(血漿)의 각지질분획(各脂質分劃)의 지방산조성(脂肪酸組成)에 미치는 급여지방산(給與脂肪酸)의 영향(影響)을 조사(調査)하였다. 그 결과(結果) 간장중(肝臟中)의 총지질함량(總脂質含量) 및 각지질분획(各脂質分劃)의 함량(含量)은 급여지방산(給與脂肪酸)에 의해서 영향(影響)받지 않았다. 그러나 17:0을 급여(給與)하면 간장(肝臟) 및 혈장(血漿)의 각지질분획(各脂質分劃)에 17:0 및 17:1 출현(出現)하고, 혈장(血漿)트리그리세라이드에서는 17:0이 또한 혈장(血漿)콜레스테롤에스텔에서는 17:1이 다른 분획(分劃)보다도 유의(有意)하게 높았다. 더욱이 간장(肝臟)에서는 각분획(各分劃)사이에 양지방산(兩脂肪酸)의 분포치(分布値)에 큰 차이(差異)가 없었다. 18:2의 급여(給與)도 17:0을 급여(給與)한때와 같이 간장(肝臟) 및 혈장(血漿)의 각지질분획(各脂質分劃)에 18:2이 출현(出現)하고, 특(特)히 간장인지질분획(肝臟燐脂質分劃)에서 18:2의 분포치(分布値)는 다른분획(分劃)보다도 유의(有意)하게 높았다. 그러나 18:0을 급여(給與)해도 이것이 간장(肝臟) 및 혈장중(血漿中)에 특별적(特別的)으로 증가(增加)하지는 않았다. 또한 17:0 및 18:2과 같은 외인성지방산(外因性脂肪酸)의 급여(給與)에 의하여 내인적(內因的)으로 합성(合成)되는 16:1 및 18:1의 분포치(分布値)는 간장(肝臟)에서 감소(減少)하는 경향(傾向)을 나타내었다.

A comparison study of extraction methods for bio-liquid via hydrothermal carbonization of food waste

  • The hydrothermal carbonization method has received great attention because of the conversion process from biomass. The reaction produces various products in hydrochar, bio-liquid, and gas. Even though its yield cannot be ignored in amount, it is difficult to find research papers on bio-liquid generated from the hydrothermal carbonization reaction of biomass. In particular, the heterogeneity of feedstock composition may make the characterization of bio-liquid different and difficult. In this study, bio-liquid from the hydrothermal carbonization reaction of food wastes at $230^{\circ}C$ for 4 h was investigated. Among various products, fatty acid methyl esters were analyzed using two different extraction methods: liquid-liquid extraction and column chromatography. Different elutions with various solvents enabled us to categorize the various components. The eluents and fractions obtained from two different extraction methods were analyzed by gas chromatography with a mass spectrometer (GC/MS). The composition of the bio-liquid in each fraction was characterized, and seven fatty acid methyl esters were identified using the library installed in GC/MS device.

Comparative analysis of Bombyx batryticatus and Bombyx mori on α-glucosidase inhibition and their bioactive compositions

  • This study aimed to evaluate and compare the inhibitory effects of extracts from Bombyx batryticatus (BBE) and Bombyx mori (BME) on α-glucosidase, DPP-4, and LDL oxidation activities, focusing on their potential applications in managing postprandial hyperglycemia and metabolic syndrome. The results demonstrated that both BBE and BME effectively inhibited α-glucosidase and LDL oxidation, with BBE exhibiting higher inhibitory activity than BME. HPLC analysis identified linolenic acid, linoleic acid, linolenic acid ethyl ester, pheophorbide a, and pyropheophorbide a as key compounds contributing to these effects. Notably, the identified unsaturated fatty acids and pheophorbides showed strong α-glucosidase inhibitory activity, surpassing that of acarbose, a standard diabetic drug. These results suggest that, in addition to the previously reported 1-DNJ and fibroin proteins, unsaturated fatty acids and chlorophyll-derived pheophorbides may play significant roles in glycemic control. Compounds, particularly those from BBE, present promising opportunities for the development of natural therapeutic agents for diabetes management. The study concludes that BBE and BME have strong potential as functional ingredients in future diabetes treatment strategies, possibly offering enhanced efficacy over conventional inhibitors.

Production of Methyl Ester from Coconut Oil using Microwave: Kinetic of Transesterification Reaction using Heterogeneous CaO Catalyst

  • Methyl ester derived from coconut oil is very interesting to study since it contains free-fatty acid with chemical structure of medium carbon chain ($C_{12}-C_{14}$), so the methyl ester obtained from its part can be a biodiesel and another partially into biokerosene. The use of heterogeneous catalysts in the production of methyl ester requires severe conditions (high pressure and high temperature), while at low temperature and atmospheric conditions, yield of methyl ester is relatively very low. By using microwave irradiation trans-esterification reaction with heterogeneous catalysts, it is expected to be much faster and can give higher yields. Therefore, we studied the production of methyl ester from coconut oil using CaO catalyst assisted by microwave. Our aim was to find a kinetic model of methyl ester production through a transesterification process from coconut oil assisted by microwave using heterogeneous CaO catalyst. The experimental apparatus consisted of a batch reactor placed in a microwave oven equipped with a condenser, stirrer and temperature controllers. Batch process was conducted at atmospheric pressure with a variation of CaO catalyst concentration (0.5; 1.0; 1.5; 2.0, 2.5%) and microwave power (100, 264 and 400 W). In general, the production process of methyl esters by heterogeneous catalyst will obtain three layers, wherein the first layer is the product of methyl ester, the second layer is glycerol and the third layer is the catalyst. The experimental results show that the yield of methyl ester increases along with the increase of microwave power, catalyst concentration and reaction time. Kinetic model of methyl ester production can be represented by the following equation: $-r_{TG}=1.7{\cdot}10^6{_e}{\frac{-43.86}{RT}}C_{TG}$.

Synthesis and biodistribution of 18F-labeled α-, β- and ω-fluorohexadecanoic acid

  • ${\omega}-[^{18}F]$-Fluorohexadecanoic acid (FHA) has been used for imaging of fatty acid metabolism of myocardium. To increase retention of radiolabeled fatty acid by blocking ${\beta}$-oxidation, methyl branched analogues have been used. In this experiment, we tried to synthesize 18F-labeled ${\alpha}-$, ${\beta}-$ and ${\omega}-FHA$ for imaging of the myocardial fatty acid metabolism. We synthesized ${\alpha}-$, ${\beta}-$ and ${\omega}$-mesylated methyl hexadecanoates and labeled with $^{18}F$ by reacting with $[^{18}F]$TBAF in acetonitrile at $80^{\circ}C$ for 10 min. Methyl ester group was removed by 1 M NaOH at $80^{\circ}C$ for 5 min. The yields of ${\alpha}-[^{18}F]$ and ${\omega}-[^{18}F]FHA$ were 25.5 and 45.5%, respectively [EOS]. However, ${\beta}-[^{18}F]FHA$ was not labeled at all due to a fast elimination reaction. The biodistribution study in ICR-mice showed that ${\omega}-[^{18}F]FHA$ has higher myocardial uptake and lower liver uptake than ${\alpha}-[^{18}F]FHA$. The electron-withdrawing effect of fluorine at ${\alpha}-$ position is believed to be the major factor affecting the biodistribution.
