Isolation of Steroids and Flavonoids from the Herbs of Hypericum ascyron L. (물레나물로부터 Steroid 및 Flavonoid 성분의 분리)

  • A sterol mixture, 3-O-glucosides of these sterols, 6'-O-fattyacyl ester of these sterol glucosides, kaempferol, quercetin and isoquercitrin were isolated from the whole plants of Hypericum ascyron L. The sterols were found to be a mixture of ${\beta}-sitosterol$, campesterol and stigmasterol by GC-MS. The kinds of fatty acids linked at 6'-OH of sterol glucoside ester mixture were shown to be palmitic acid, stearic acid, oleic acid and linoleic acid by GC-MS. Three flavonoids were identified by spectroscopic methods and comparisons of mixed mp and co-TLC with authentic specimens.

The Chemotoxonoic Relationship of Vibrio cholerae non-O1 by Fatty Acid Compositions (지방산 조성에 의한 vibrio cholerae non-O1의 화학분류학적 관계)

  • The authors attempted utilization of fatty acid composition of vibrios as a tool for identification of the strains. Fatty acid of 49 strains of Vibrio cholerae non-O1, V. cholerae O1, V mimicus, V vulinificus and V parahaemolyticus was analyzed by gas-liquid chromatography column. According to the statistical analysis of the fatty acid data, the relationship between the Vibrio species and serotypes of the strains was discussed. Forty one kinds of fatty acid were detected from the tested strains and 35 kinds of fatty acids among the detected fatty acids were significant factors to identify the vibrios. The predominant fatty acids were 16:0, 16:1 cis 9, 18:1 trans 9/6/cis 11 and 15:0 iso 2OH/16:1 cis 9 as above about 20% in total. Fatty acid compositions of the Vibrio species were an important factor in identifying their subspecies either predominant fatty acids or minor ones. According to the analysed results by a conventional statistical processing method (UPGMA) and prepared dendrogram, V cholerae non-01 had more closer relationship with V. mimicus compared with V. cholerae 01. Moreover, the distribution of hydroxy acid was a significant factor for identifying V cholerae subspecies. Comprising all the 10 serotypes detected from V. cholerae non-01 examined such as O2, O5, O8, O10, O14, O27, O37, O39, O45 and O69, we could group them into seven subspecies by cluster analysis with the similarity value of fatty acid composition as above 92%. It means that there is a significant relationship between serotypes and fatty acid composition of V. cholerae. These results indicated that numerical analysis of fatty acid composition data of V cholerae non-01 could classifY them into subspecies, and also which may provide a useful epidemiologic information or a basis for further analysis such as PCR and DNA probe analysis.

Reduction of Saturated Fatty Acid Methyl Esters of Biodiesel Produced from Beef Tallow by Acetone Fractionation (우지로부터 합성된 바이오 디젤의 포화도 감소를 위한 용매 분별 연구)

  • It is known that the content of saturated fatty acids methyl ester (SFAME) affect the pour point of biodiesel at low temperature. In this study, biodiesel (BD) was produced from beef tallow (TAL) by alkali catalyst. To reduce the saturation in BD, acetone fractionation was applied. Besides, TAL was also solvent-fractionated to reduce the saturated fatty acid (SFA) content for further producing BD. With acetone, TAL or TAL methyl ester (5:1 v/w) were fractionated at 10, 0, -10, and $-15^{\circ}C$, respectively. At $-10^{\circ}C$, 17.35% of SFA was observed in fractionated TAL (liquid part, -10TAL) when 5:1 solvent ratio was used for 24 hr. Under the same condition, fractionated BD (liquid part, -10BD) showed SFA (33.14%) with 78wt % yield. Also, fractionation of BD with different concentration of crystallizer 209 (0.1, 0.5, and 1%) along with different time (2, 6, 12, and 24 hr.) was observed. The best condition for reducing the SFA was 0.5% of crystallizer 209 addition for 12 hr of fractionation time at $-10^{\circ}C$, in which 30.14% of SFA content was observed in BD (liquid part). Among different crystallizer, ps 66 showed the least content of SFA content (23.28%) in BD after fractionation ($-10^{\circ}C$ and 24 hr) with 0.5wt% addition.

Changes in Fatty Acid and Cholesterol Composition of Koran Styled Beef Broths (Gom-Guk) during Cooking (쇠고기 곰국의 조리중 지방산 및 Cholesterol 조성변화)

  • The content of total lipid, cholesterol and fatty acid in Korean styled various beef broths was examined before and during toiling to determine the influence of cooking time. The values obtained from the broths of brisket, ox-tail and small-intestine were determined by using G. C. The highest total lipid value among small-intestine, brisket and ok-tail was in the small-intestine. The content of total lipid in the boiled meat decreased during cooking with the exception of the brisket. Total lipid content found in the broth of brisket meat increased until 6hr and thell decreased at cooking time of 8hr and 10 hr. The highest total lipid contents in the broths of brisket, ox-tail and small-intestine were found when the cooking time were 6 hr, 10 hr and 4 hr, respectively. The main fatty acids found in the broths of brisket, ok-tail and small-intestine were $C_{14}$, $C_{16}$, $C_{18}$, $C_{18:1}$, and $C_{18:1}$. The content of $C_{18:1}$ and $C_{18:2}$ from the brisket meat increased until 4hr's cooking, especially $C_{18:2}$ increased significantly until 6 hr cooking. The highest ratios of unsaturated to saturated fatty acid (UNS/S) in the meat of brisket and ox-tail were found in the 2hr and 4hr cooking, respectively. The ratios of UNS/S in the broths of both of brisket and small-intestine, and ok-tail were highest in the 4 hr and 2 hr cooking, respectively. The content of total cholesterol (TC) was the highest in the small-intestine and the highest percentage of ester cholesterol was 5.4 in the brisket. The content of TC in small-intestine, brisket and ox-tail was decreased significantly after 2 hr cooking. Precentage of the ester in the brisket was the highest when the cooking time was 8 hr. Only trace amount of total cholesterol was found in the broth.

Recovery of Covalently Linked Fatty Acid Monolayer on the Hair Surface Using Biomimetic Lipid (생체모사 지질을 이용한 모발 표면에 공유 결합된 지방산 단분자층의 회복)

  • There is a unique type of fatty acid in the hair surface. 18-methyleicosanoic acid (18-MEA) is an unusual anteiso fatty acid covalently linked to the outermost surface of hair cuticle. A layer of 18-MEA is located in the upper ${\beta}$-layer of the CMC that is responsible for the low surface energy and low friction resistance of the hair's outer surface. The high mobility of 18-MEA molecule facilitates spreading of extraneous lipid by decreasing interfacial shear strength. In this study, we introduced N-hydroxyl succinimidyl ester functional group to the one end of C10 - 40 isoalkyl acid for regenerating hair surface with covalently bound fatty acid layer. The re-hydrophobicization of hair surface has been investigated by contact angle measurement. The inner moisture content of hair at different levels of humidity (40, 55, 70 %RH) was measured by electric moisture analyzer. Treatment with Hydroxysuccinimidyl C10 - 40 Isoalkyl Acidate (HCIA) was supposed to make hair surface smoother by filling the cracks between cuticles with covalently bound fatty acid monomolecular layer like cuticle glue. This glue effect was also confirmed with line profile of AFM images. Therefore, the moisture and structural components of inner hair were not easily flown out and the optimum moisture content could be kept constantly though the outside humidity level was changed. The lateral force microscopy (LFM) by using atomic force microscope showed that the friction force of hair surface treated with HCIA was decreased. It also showed the constantly sustained friction value even after shampooing repeated 15 times.

Improvement of Low Temperature Property of Biodiesel from Palm Oil and Beef Tallow Via Urea Complexation (요소 착물형성에 의한 포화지방산 고함유 팜유 및 우지 유래 바이오디젤의 저온유동성 개선효과 연구)

  • Biodiesel is non-petroleum based fuel produced from vegetable oils or animal fats through transesterification. The compositions of saturated and unsaturated fatty acids in the feedstocks are important factors for biodiesel quality in terms of low-temperature fluidity and oxidative stability. The goal of this study is to improve the cold flow property of biodiesel from vegetable and animal origin containing highly saturated methyl esters (approx. 50%). In this purpose poly-saturated methyl esters in palm and tallow biodiesel were removed via urea-based fractionation and then the recovered fractions (enriched unsaturated fatty acid methyl esters) were supplemented with cold flow improvers. The highest concentration of unsaturated fatty acid methyl esters (93.8%) was obtained using a urea/fatty acid ratio of 3:1 at the crystallization temperature of $0^{\circ}C$ for 17 hours in incubation, with recovery of 71% and the addition of cold flow improver (Flozol$^{(R)}$ 515, 3,000 ppm) to the enriched poly-unsaturated fatty acid methyl esters reduced the CFPP(cold filter plugging point) of palm biodiesel from $12^{\circ}C$ to $-42^{\circ}C$. In tallow biodiesel both the enrichment of unsaturated fatty acid methyl esters (93.71%) and the addition of cold flow improver (Infineum R408, 3,000ppm) reduced the CFPP from $10^{\circ}C$ to $-32^{\circ}C$.

Enhanced Uptake of Modified Low-Density Lipoprotein by Eicosapentaenoic Acid-Treated THP-1 Macrophages

  • Animal and clinical studies as well as epidemiological data have provided convincing evidence that n-3 polyunsaturated fatty acids can protect against atherosclerosis. However, the effects of the fatty acids on atherogenesis are contradictory. This discrepancy could derive from great susceptibility of the fatty acids to oxidation. We investigated the effect of eicosapentaenoic aced(EPA) on cellular atherogenesis via the scavenger receptor of THP-1 derived macrophages. THP-1 cells were fully differentiated into macrophages by incubating with phorbol 12-myristate 13-acetate for seven days. Atherogenic features of EPA were compared by subsitituting for linoleic acid (LA). Macrophages were also incubated without treatment of the fatty acids as controls. EPA (5-50 nmol/mL) was not cytotoxic and did not measurably induce cellular oxidation compared to bovine serum albumin (BSA) vehicle or identical doses of LA. EPA increased macrophage uptake and degradation of acetylated LDL(AcLDL) up to 14% and 88%, respectively. EPA increased markedly total cellular sterol synthesis and heparin-releasable lipoprotein lipase activity of macrophages, indicating that EPA may enhance accumulation of cellular cholesteryl ester and possibly facilitate formation of foam cells. These results demonstrate that EPA promotes the modified LDL-triggered atherosclerotic process by the modulation of the scavenger receptor and the activation of LPL in macrophages.

Synthesis of ${\alpha}-Sulfonated$ Fatty Acid Polyethylene Glycol Ester (알파술폰 고급지방산 폴리에틸렌 글리콜 에스테르류의 합성)

  • ${\alpha}-sulfonated$ fatty acid polyethylene glycol esters with polyethylene oxide(addition, 3, 5, 10mol) were synthesized through esterification of ${\alpha}-sulfonated$ fatty acid methyl esters with alkyl chain length $C_{12}{\sim}C_{18}$. Their compounds were separated with column chromatography, and confirmed by TLC. Quantitative analysis of all the sulfonates were performed according to JIS K-3362 method, and ethylene oxide unit number were determined by ISO 2270 method. Structural properties of ${\alpha}-sulfonated$ fatty acid methyl esters and their derivatives were also identified from IR, and $^1H$ NMR spectra.

  • Through an experiment with gas chromatography carried out using diethylene glycol succinate(DEGS) as the packing material of the column, we have obtained the correction factor between the weight ratio and the peak dimension of the saturated fatty acid methyl esters of C10, C12, C14, C16, and C18 and unsaturated fatty acid methyl esters of oleic acid, linoloic acid, and linolenic acid, employing the detector of thermal conductivity type. Quantitative analysis of the fatty acids contained in rice Bran oil was performed with the above correction factor and the results are as follows; 1. Main components were found to be palmitic acid, oleic acid sand linolenic acid. No trases of capric acid (C10) lauric acid (C10) were found. 2. It was confirmed that there were straight line relation between the logarism retention time of each fatty acid and the number of carbon of saturated fatty acid or the number of double bond of other fatty acids having the same number of carbon. 3. The correction factor became larger as to the number of carbon increased up to C18 in case of saturated fatty acids, end as for other fatty acids, and as for other fatty acids of the same carbon number, it became larger according as the number of double bond increased.

Structural Analyses of the Novel Phosphoglycolopids Containing the Unusual very Long Bifunctional Acyl Chain, α,ω-13,16-Dimethyloctacosanedioate in Thermoanaerobacter ethanolicus

  • Novel membrane lipids containing the unusual very long chain fatty $acid{\alpha}{\omega}-1316-dimethyloctacosanedioate$, dimethyl. Ester (DME C30) was isolated and purified from thermophilic anaerobic eubacterium, Thermoanaerobacter ethanolicus. Structures of the lipids containing the bifunctional fatty acyl components were proposed by various analyses such as $^1H,\;^{13}C,\;^{31}P$ nuclear magnetic resonance (NMR), Fourier transform infrared(FTIR) spectroscopy, gas chromatography/mass spectrometry (GC/MS) and fast atom bombardment mass spectrometry (FAB/MS). Combined with the GC/MS, $^1H,\;and\;^{13}C$NMR data, we confirmed that the head groups of the lipids contained the glycerol and/or glucosamine molecules. $^{31}P$ NMR spectrum also showed that the lipids contained phosphate in a phosphodiester linkage. The proposed structures of these novel lipid components were the ones in which two head groups were linked by the membrane spanning fatty acyl component(DME C30)and regular chain fatty acids on glycerol moiety of each head group.
