Effect of High Water Temperature on the Growth and Lipid Compositions of the Sea Cucumber Apostichopus japonicus (고수온에서의 해삼(Apostichopus japonicus) 성장과 체지방 조성)

  • The optimum growth temperature of sea cucumber is $18^{\circ}C$, above $20^{\circ}C$, the appetite declines and the digestive tract is atrophied. At $25^{\circ}C$ or more, it is known that the digestive tract is degenerated as growth is stopped by minimizing metabolic activity. Because of these physiological and ecological characteristics, the growth period of sea cucumber Apostichopus japonicus is relatively short in the natural environment of Korea where four seasons are clear. Therefore, maintenance of water temperature in sea cucumber culture is very important. This experiment was conducted to investigate the growth and lipid compositions of sea cucumbers at high temperature. Results showed that the growth and survival rates of sea cucumber were not significantly different until $26^{\circ}C$ (P>0.01). The fatty acid compositions of total lipid of sea cucumber tended to increase with saturated fatty acid content and decrease with n-3 fatty acid content at higher water temperature.

Analysis of Biochemical Compositions and Nutritive Values of Six Species of Seaweeds (해조류 6종의 생화학적 조성 및 영양적 가치 분석)

  • The biochemical compositions and nutritive values of six species of seaweeds were analyzed to determine their applicability in functional foods or ingredients. The biochemical compositions (moisture, ash, protein, lipid, and dietary fiber) and fatty acid contents were determined for the following seaweed extracts: Phaeophyceae (Laminaria japonica, Hizikia fusiformis, and Undaria pinnatifida), Rhodophyceae (Porphyra tenera and Gracilaria verrucosa), and Chlorophyceae (Ulva lactuca). The moisture content (% dry weight) ranged from 11.47% to 13.94%, ash from 19.15% to 26.50%, protein from 5.08% to 15.44%, lipid from 2.75% to 4.43%, and dietary fiber from 36.84% to 52.98%. C14:0, C16:0, C18:0, C16:1, C18: 1n-3, C18:2n-6, C18:3n-6, C20:4n-6, and C20:5n-3 represented the predominant proportions of fatty acids. Interestingly, docosahexaenoic acid (C22:6n-3, DHA) was either not found or only detected in trace amounts in the analyzed seaweeds. The levels of n-3 fatty acid were higher than other polyunsaturated fatty acids, and the n-6/n-3 ratio was very low. These results indicate that seaweed inhabiting Korean coastal areas will be beneficial to human health.

Contents of Total Lipids and Their Composition in Colored and Aromatic Rices Cultivars (유색미와 향미 품종의 지방질함량 및 지방질의 조성)

  • The contents of total lipids and their components of brown rice grain were studied for 5 colored rice cultivars, 1 aromatic rice cultivar and 1 normal rice cultivar grown in Korea. The total lipid contents ranged 2.04 to 4.69% based on total dry weight. The Tohoku #149 showed the highest content which was followed by Hansanheukmi and Sanghaehyang-hyeolna. The lowest content were observed in three cultivars, Heuknambyeo, Hyangnambyo and Dongjinbyeo which showed no difference among cultivars. The main classes of the total lipid were triglyceride, diglyceride, free sterol, free fatty acid and sterol esters in all cultivars examined. The ratio of triglyceride in total lipid was high in colored rice and aromatic rice compared to Dongjinbyeo which is normal rice cultivars, but the ratio of diglyceride was high in Dongjinbyeo. The major fatty acid compositions in the cultivars examined were linoleic acid, oleic acid and palmitic acid. There was a varietal difference in fatty acid composition, for example, erucic acid was detected in aromatic and colored rices cultivars, while none of this compound was found in Dongjinbyeo.

The Role of Fatty Acid Binding Protein in the Fatty Liver Induced by Alcohol or High Cholesterol Diet in Rats (알코올 및 고콜레스테롤 식이로 유도된 흰쥐의 지방간에서 지방산 결합단백질의 역할 및 특성)

  • There is a marked increase in geriatric disease, especially liver disease, due to the continuous increase in alcohol and fat consumption. Since the fatty liver, induced by alcohol or fat, is basically from abnormalities in the lipid metabolism, it is possible that fatty acid binding protein(FABP) which is related to the fatty acid metabolism may also be abnormal in these livers. FABP is a small molecular weight protein family present in cytosol in high concentration. It has been proposed as a fatty acid transfer protein and as a binding protein responsible for controlling intracellular free fatty acid concentration. In this research, we have examined the relationship between liver FABP and fatty liver induced by alcohol or high cholesterol diet. Rats were fed one of either semipurified liquid diets; control diet containing 65% carbohydrate, 20% protein, and 15% fat or high cholesterol diet containing 1%(w/w) cholesterol or alcohol diet containing 37% of alcohol instead of carbohydrate. After 5 weeks of feeding period, all rats received commercial chow diet for 5 weeks to examine recovery effect. Liver and blood samples were collected at 0, 1, 3, 5 and 10 weeks to analyze lipid compositions. FABP was purified from liver cytosol and injected to rabbit to obtain antiserum. Liver FABP amount was determined by SDS-PAGE and western blotting methods. Fatty acid binding capacity was determined by binding of 14Cpalmitate with the delipidated liver cytosol. Consumption of alcohol increased serum cholesterol, triglyceride concentration and decreased HDL-cholesterol concentration after 5 weeks. Serum apolipoprotein B concentration increased after 3 weeks and LDL-cholesterol and apolipoprotein A concentration changed after 1 week. Liver cholesterol and triglyceride concentration increased after 3 weeks. Consumption of high cholesterol diet changed liver and serum lipid composition after 3 weeks. Swiching to normal diet for 5 weeks did not normalize most of lipid composition in serum and liver except serum and liver except serum cholesterol, triglyceride and liver cholesterol. Liver cytosol FABP content and the fatty acid binding capacity decreased dramatically after 1 week with alcohol consumption. This results indicate that FABP content changes before the changes before the changes of blood or liver lipid composition, suggesting changes of FABP may cause development of the fatty liver induced by alcohol and can be used as an index of detecting a early development of fatty liver.

EPA, DHA and Tocopherol Contents in Fish Oil Products and Fishes (어유제품과 생선의 EPA, DHA 및 토코페롤 함량)

  • Six brands of encapsulated fishoil products and five fishes were analyzed for their contents of eicosapentaenoic acid (EPA 20 :5 n-3) , docosahexaenoic acid(DHA 22: 6 n-3) and tocopherols. In both of the fish oil products and the fishes, major fatty acids were palmitic acid, oleic acid, eicosapentaeoic acid and docosahexaenoic acid and fatty acid compositions were also similar pattern each other. EPA showed variable amounts from 19.2 to 50.3% in the oil products whereas DHA were 13.2% to 28.3% inthe fishes. Tocopherols were studied in relation to the oxidative stability of fish products no relation was observed. However the amount of tocopherols in fish oil proudcts were higher than that of fishes. Contents of EPA, DHA and tocopherols in encapsulated fishoil products oil products were variable comparing with manufactures' claimed contents.

Changes during Storage of Rice Germ Oil and Its Fatty Acid Composition (쌀눈 유지의 저장중 변화와 지방산 구성)

  • Rice germ oils were extracted from common and dried rice germ $(80^{\circ}C,\;6\;hr)$, and their oxidative stabilities were evaluated by measuring acid value, peroxide value and fatty acid composition during storage at $40^{\circ}C\;and\;60^{\circ}C$. The acid values of all lipids were slightly changed during storage, but peroxide values (POV) were greatly dependent on storage temperature. The POVs of the dried germ oil and the refined rice bran oil were 146.2 meq/kg and 15.1 meq/kg, respectively after 31 days storage at $40^{\circ}C$. However after 24 days of storage at $60^{\circ}C$, the POVs of the dried germ oil and the refined rice bran oil were 151.7 meq/kg oil and 219.6 meq/kg oil, respectively. Major fatty acids were linoleic (39.8%) and oleic acid (34.7%) in rice germ oil, and oleic (40.1%) and linoleic acid (38.1%) in rice bran oil. The major fatty acid compositions were not greatly influenced by drying and storage temperature but linoleic acid decreased about half during storage.

Lipid and Fatty Acid Composition in Nannochloropsis oculata Cultured in Varying Salinities (염분농도에 따른 해양미세조류(Nannochloropsis oculata)의 지질 및 지방산의 변화)

  • The quality and quantity of food organisms in fish seed production are important. The marine microalgae Nannochloropsis oculata are used as initial food organisms in the field. We investigated the effects of salinity (0, 10, 20, 30, 40 and 50 psu) on the lipid and fatty acid composition of N. oculata. Cultivation of N. oculata at varying salinities showed the highest growth rate at 20 psu. Total lipid content ranged from 17.26 to 18.63% at salinities from 0 to 50 psu). The nonpolar lipid content increased markedly at 30 psu and was highest at 15.55%. The polar lipid content was lowest at 30 psu, by 84.45%. It was also found that the omega-3 and EPA contents were inversely proportional to salt concentration. For the polar and nonpolar lipid compositions, there was no significant effect of salinity. Omega-3 polyunsaturated fatty acid content especially the content of EPA in the seawater larvae is the essential fatty acid in this food organism. It is thus advantageous to culture N. oculata at 20 psu.

Effect of High Temperature and Growth Light Intensity on Fatty Acid Composition of Panax ginseng leaf (고온(高溫)과 재배광도(栽培光度)가 인삼(人蔘) 잎의 지방산(脂肪酸) 조성(組成)에 미치는 영향(影響))

  • Fatty acid compositions of Panax ginseng leaves (6 year) grown under different light intensity in field and of the detached leaves exposed to high temperature (20 hours) were investigated by gas chromatography. Linoleic, linolenic, palmitic and palmitoleic acid were the major components(80%) of leaf lipid. The higher the growth light intensity, the lower the percentage of unsaturated acids or bonds, indicating metabolic adaptation to high temperature. Pattern similarity of fatty acid composition was little changed until 20% light but significantly different at 30%, suggesting 20% as limitation light intensity. The close similarity of fatty acid composition between the loaves grown under 30% light and the one at harvest rises uncertainty between adaptation to high temperature and senescence. Total fatty acid content decreased with the increase of light intensity. Short term high temperature $(25^{\circ}C\;or\;35^{\circ}C)$ increased total fatty acid content, unsaturated acid percentage and insignificant difference in pattern similarity of composition.

  • These experiments were carried out to compared with components of AGM and raw beef. The results obtained were summarized as follows: 1) In proximate compositions of raw beef and AGM, water, protein, and fat were 71.21% and 48.47%, 29.35% and 21.85%, 5.36% and 14.95%, respectively. 2) Total amounts of amino acids of AGM were higher than raw beef, and glutamic acid was the highest in both, but methionine was the lowest in AGM, and cystine, in raw beef. 3) In fatty acids composition, unsaturated fatty acids were 90.4%, and linoleic acid (57.63%), among them, was the highest in AGM. But in raw beef, saturated fatty acids were 35.2%, and oleic acid, among them, was the highest content.

  • Agaricus blazei Murill, a mushroom which is well known as a potent antitumor agent, contains abundant pharmaceutical substances. To identifY the major components of Agaricus blazei Murill fruiting bodies cultivated in a Korean local farm was analyzed. There were not large differences in chemical compositions of inorganic substances, carbohydrates, amino acids and fatty acids between Agaricus blazei Murill fruiting bodies cultivated in Korea and those in Japan. However, carbohydrate composition was significantly lower in Agaricus blazei Murill of Korea compared to those of Japan. Total amino acid contents were 280.75 mg% in Agaricus blazei Murill fruiting bodies. Sixteen species of amino acid were identified by high performance liquid chromatography except cysteine. Linoleic acid, the most abundant fatty acid contained in Agaricus blazei Murill fruiting bodies, was estimated to be 78.3% of total fatty acids.
