Histologic Characteristics and Mechanical Properties of Bovine Pericardium Treated with Decellularization and ${\alpha}$-Galactosidase: A Comparative Study

  • Background: Bioprostheses for cardiovascular surgery have limitations in their use following as calicification. ${\alpha}$-galactosidase epitope is known as a stimulant of immune response and then shows a progressing calcification. The objective of this study was to evaluate histologic characteristics and mechanical properties of decellularization and treated with ${\alpha}$-galactosidase. Materials and Methods: Bovine pericardial tissues were allocated into three groups: fixation only with glutaraldehyde, decellularization with sodium dodesyl sulfate and decellularization plus treatment with ${\alpha}$-galactosidase. We confirmed immunohistological characteristics and mechanical properties as fatigue test, permeability test, compliance test, tensile strength (strain) test and thermal stability test. Results: Decellularization and elimination of ${\alpha}$-gal were confirmed through immunohistologic findings. Decellularization had decreased mechanical properties compared to fixation only group in permeability (before fatigue test p=0.02, after fatigue test p=0.034), compliance (after fatigue test p=0.041), and tensile strength test (p=0.00). The group of decellularization plus treatment with ${\alpha}$-galactosidase had less desirable mechanical properties than the group of decellularization in concerns of permeability (before fatigue test p=0.043) and strain test (p=0.001). Conclusion: Favorable decellularization and elimination of ${\alpha}$-gal were obtained in this study through immunohistologic findings. However, those treatment including decellularization and elimination of ${\alpha}$-gal implied the decreased mechanical properties in specific ways. We need more study to complete appropriate bioprosthesis with decellularization and elimination of ${\alpha}$-gal including favorable mechanical properties too.

Effects of Alpha Phase on the Fatigue Properties of Fe-29%Ni-17%Co Low Thermal Expansion Alloy (Fe-29%Ni-17%Co 저열팽창 합금의 피로 특성에 미치는 알파상의 영향)

  • The effect of alpha phase on the fatigue properties of Fe-29%Ni-17%Co low thermal expansion alloy was investigated. Two kinds of alloys (Base alloy and Alpha alloy) were prepared by controlling the minimal alloy composition. Microstructure observation, tensile, high-cycle fatigue, and low-cycle fatigue results were measured in this study. The Base alloy microstructure showed typical austenite ${\gamma}$ phase. Alpha alloy represented the dispersed phase in the austenite ${\gamma}$ matrix. As a result of tensile testing, Alpha alloy was found to have higher strengths (Y.S. & T.S.) and lower elongation compared to those of the Base alloy. High cycle fatigue results showed that Alpha alloy had a higher fatigue limit (360MPa) than that (330MPa) of the Base alloy. The Alpha alloy exhibited the superior high cycle fatigue property in all of the fatigue stress conditions. SEM fractography results showed that the alpha phase could act to effectively retard both fatigue crack initiation and crack propagation. In the case of low-cycle fatigue, the Base alloy had longer fatigue life in the high plastic strain amplitude region and the Alpha alloy showed better fatigue property only in the low plastic strain amplitude region. The fatigue deformation behavior of the Fe-29%Ni-17%Co alloy was also discussed as related with its microstructure.

  • Recently the steel parts used at automobiles are required to be used under high stress more than ever before in need of the weight down. To achieve this requirement of a high strength steel, it must be necessary to decrease inclusion content and surface defect as like decarburization, surface roughness etc.. In this study, the surface conditions are measured to know the influence on fatigue properties by two cases of shot peening of two-stage shot peening and single-stage shot peening. And for this study, two kinds of spring steel (SUP7, 50CrV4) are used. This study shows the outstanding improvement of fatigue properties at the case of two-stage shot peening in the rotary bending fatigue test and this is assumed to be from on low stress condition, the 1st stage shot peening is not affected by nonmetallic inclusion under metal. it is possible that the 2nd stage shot peening increases the fatigue life and the high stress but that is affected by nonmetallic inclusion under metal. so far beeasily 50CrV4 have made high stress. But, results also show fatigue failures originated at inclusion near surface, and this inclusion type is turned out to be a alumina of high hardness.

To achieve this requirement of the high strength steel, it must be necessary to decrease inclusion contents and surface defects as like decarburization, surface roughness etc. In this study, the surface conditions are measured to know the influence on fatigue properties by two cases of two-stage shot peening and single-stage shot peening. And for this study, three kinds of spring steel (JISG408l-SUP7, SAE 9254 and DIN 50CrV4) are shaped. This study shows the outstanding improvement of fatigue properties at the case of two-stage shot peening in the rotating bending fatigue test and it results from (1) decreasing the surface roughness (2) unchanging the surface hardness (3) increasing the compressive residual stress. Moreover, results also show fatigue failures originated at the inclusion near the surface, and this inclusion type is turned out to be an alumina of high hardness.

  • In order to estimate the fatigue life of mufflers at the early stage of researches and designs, the new prediction process was developed by the artificial neural network, which has the algorism of weldment properties. Bending fatigue test was carried out for defining the characteristics of muffler weldment fatigue life and damage. For considering and predicting mechanical and fatigue properties of the muffler, the maximum stress of weldment was adapted as the variable of artificial neural network training. Also, it was compared with the fatigue life predicting results using fatigue notch factors, for proving the newly developed process of the artificial neural network.

  • In this study, low fatigue behavior of laser welded sheet metal were investigated. Before welding, the cross section of butt joint was prepared only by fine shearing without milling process. Specimens were same sheet metal and welding condition that using automobile manufacturing company at present. Butt joint of cold rolled sheet metal was welded by $CO_2$ laser. It is used that welding condition such as laser welding speed was 5.5m/sec and laser output power was 5kW for 0.8mm and 1.2mm sheet metal. The laser weldments were machined same or different thickness and same or different material. In order to mechanical properties of around welding zone, hardness test was performed. Hardness of welding bead is about 2 times greater than base material. We performed the low cycle fatigue tests for obtaining fatigue properties about thickness and the weld line direction of specimen. The results of strain controlled low cycle fatigue test indicate that all specimens occur cyclic softening, as indicated by the decrease in stress to reach a prescribed strain.

  • In this study, the fatigue characteristic of spot weld specimen was studied by using the various specimen. The specimen types were tensile shear specimen welded one spot and two spot, and cross tension. The tensile tests and fatigue tests were executed to know the mechanical properties under static and fatigue load condition. In addition, the relationship was illustrated by finite element method.

  • A computer software was developed for predictions of fatigue crack initiation life of notched members under variable loadings. The software was constructed based on a new fatigue life prediction method utilizing modified .epsilon.-N curves, which can account for the stress interaction effect. The effect of mean plastic strain on low-cycle fatigue life was also incorporated in the software. The software can be utilized for the first step approximation when fundamental data of material fatigue properties are not available.
