홍삼, 천마, 적하수오 병용투여에 의한 고지혈증 랫드에서의 콜레스테롤 및 발기부전 개선효과 (Beneficial effect of Combination with Korean Red Ginseng, Gastrodia Rhizoma and Polygoni Multiflori on Cholesterol and Erectile Dysfunction in Hyperlipidemia rats)

  • Objectives : This study was designed to investigate effects of the combination with Korean Red Ginseng (Panax ginseng C.A. Meyer), Gastrodia Rhizoma (Gastrodia elata Blume) and Polygoni Multiflori Radix (Polygonum multiflorum Thunberg) on metabolic disorders including cholesterol and erectile dysfunction in hyperlipidemia rats.Methods : Animals were divided into six groups; Control with normal diet, high fat/cholesterol-diet (HFCD), fluvastatin, Korean Red Ginseng treated (KRG), and the combination treated (Korean Red Ginseng, Gastrodia Rhizoma and Polygoni Multiflori Radix; 1:1:1 for KGP1 and 2:1:1 for KGP2). The experimental groups initially received HFCD for 10 weeks and then treated orally with fluvastatin, KRG, KGP1 and KGP2 during the final 6 weeks. Erectile function was determined by the measurements of intracavernosal pressure (ICP) and maximal arterial pressure (MAP) after electrical stimulation of the cavernosal nerve.Results : KGP2 decreased the level of total cholesterol and LDL cholesterol in the sera of HFCD rats without no changes of body weights. KRG, KGP1 and KGP2 decreased the level of C-reactive protein (CRP) levels except of fluvastatin, synthetic HMG-CoA reductase inhibitor. KRG, KGP1 and KGP2 significantly increased the ICP, ICP/MAP ratio, area under the curve (AUC) compared with those of normal rat. Morphometric analyses showed that KRG, KGP1 and KGP2 increased the volume of smooth muscle and the regular arrangement of collagen fibers in corpus cavernosum of HFCD rats. The penile expression of eNOS was increased by KRG, KGP1 and KGP2.Conclusions : Based on these results, we suggest that the combination with Korean Red Ginseng, Gastrodia Rhizoma and Polygoni Multiflori may improve hyperlipidemia through regulating the lipid profiles and erectile dysfunction in rats.

  • Mesenchymal stem cells(MSCs) have been though to be multipotent cells that can replicate that have the potential to differentiate into lineages of mesenchymal tissue including the bone, cartilage, fat, tendon, muscle, and marrow stroma. Especially, scaffolds to support cell-based tissue engineering are critical determinants of clinical efforts to regenerate and repair the body. Selection of a matrix carrier imvolves consideration of the matrix's role as a scaffold for physical support and host tissue integration as well as its ability to support of synergize the osteoinductive program of the implanted mesenchymal stem cell. The aim of this study is to evaluate the effect of autobone and Bio-$Oss^{(R)}$ to adherent mesenchymal stem cells as scaffolds on sinus augmentation with fibrin glue mixture in a rabbit model. 16 New Zealand White rabbits were divided randomly into 4 groups based on their time of sacrifice(1, 2, 4 and 8 weeks). First, mesenchymal stem cells were isolated from iliac crest marrow of rabbits and expanded in vitro. Cell culture was performed in accordance with the technique described by Tsutsumi et al. In the present study, the animals were sacrificed at 1, 2, 4 and 8 weeks after transplantation, and the bone formation ability of each sides was evaluated clinically, radiologically, histologically and histomorphologically. According to the histological observations, autobone scaffolds group showed integrated graft bone with host bone from sinus wall. At 2 and 4 weeks, it showed active newly formed bone and neovascularization. At 8 weeks, lamellae bone was observed in sinus graft material area. Radiologically, autobone with stem cell showed more radiopaque than Bio-$Oss^{(R)}$ scaffolds group. there were significant differences in bone volume between 4 and 8 weeks(p<0.05).

창원지역 일부 비만아의 식습관과 식행동 및 영양지식에 대한 조사연구 ( 1 ) (A Study Food Habits , Food Behaviors and Nutrition Knowledge among Obese Children in Changwon ( 1 ))

  • The purpose of this study was to obtain basic data of nutrition education for obese children and to examine food habits, food behaviors, and nutrition knowledge. A convenience sample of 84(male:62, female:22) obese children was selected from "98 Children Nutrition Camp". The survey design employed a structured questionnaire. The results of this study were summarized as follows. 1. The percentage of the subjects who overestimated their body weight was higher in female compared to male. Only 35.9% of highly-obese subjects considered them as 'very fat'. The experience of weight control was higher in female than in male, and it was also higher moderately- or highly-obese subjects compared to mildly obese. 2. 64.3% of subjects reported that they regularly eat breakfast. The percentage of the subjects who indicated that they eat at regular meal time or they eat very irregularly was highest in the highly-obese subjects. 3. 34.5% of subjects reported that they eat very fast. The eating speed was slower in female compared to male, and in obese subjects. 55.4% of subjects reported that didn't eating out. 4. 80.9% of subjects had snacks, and most of them had snack between lunch and dinner. The reason the subjects eat snack was hunger. The most favorite snack was cookies. 5. The most preferred and distasteful food was meats(32.1%) and vegetables(67.1%), respectively. 6. The score of food behaviors was relatively low in most subjects. 7. The level of nutrition knowledge was in order of fair(83.1%), poor(9.6%), excellent(7.3%), and was higher in male compared to female. Consequently the results of this study showed to disagreement among food habits, food behaviors and nutrition knowledge, and feel keenly the necessity of education connecting of these. This education had to be practical education to change food behaviors.

보리 파쇄립을 이용한 압출성형에 의한 후레이크 제조 (Preparation of Flakes by Extrusion Cooking Using Barley Broken Kernels)

  • 쌀보리와 거의 유사한 식품학적 특성을 지니지만 활용도가 극히 낮은 보리 파쇄립을 이용하여 압출성형에 의한 보리 후레이크를 개발하였다. 압출성형시 $100^{\circ}C$ 이상의 온도, 35% 이상의 수분함량에서 파쇄립이 충분히 호화되었으며, 메성보리 파쇄립의 경우 찰성보리에 비해 절단성, roll 분리성 등이 우수하였다. 메성 및 찰성보리 파쇄립의 혼합비율을 달리하면서 압출성형하여 제조한 pellet의 특성을 조사한 결과 찰성보리 파쇄립의 혼합비율이 증가할수록 RVA 상의 최고점도가 감소하였고 또한 겉보기 점도와 항복력이 감소한 반면 유동지수는 증가하였다. 그리고 이를 oil frying하여 제조한 후레이크의 특성을 조사한 결과 찰성보리 파쇄립의 혼합비율이 증가함에 따라 압착강도와 bulk density가 크게 감소하였고 단면상에서의 air cell 크기가 커지고 cell을 이루는 body가 얇아지는 것으로 나타났다. 그리고 관능검사 결과 찰성보리 파쇄립의 혼합비율이 증가할수록 조직감과 맛에 대한 기호도가 증가하였고 메성과 찰성보리 파쇄립의 혼합비율 (60-70):(30-40) 처리구의 경우 관능적으로 가장 바람직한 것으로 나타났다.

  • 누에(Bombyx mori)에서 short neuropeptide F (sNPF) receptor를 encoding하고 있는 cDNA를 cloning하여서 BsNPF-R라고 이름을 붙였다. BsNPF-R는 이미 보고된 sNPF-R들과 아미노산 수준에서 사람(36%), 쥐(34%), 제브라피쉬(35%), 초파리(51%)와 상동성을 보였다. BsNPF-R는 계산적으로 분자량이 42,731 Da이고 원형질막을 관통하는 단백질이다. BsNPF-R 유전자발현은 중장, 후견사선, 말피기관, 정소에서 강한 반면 지방체, 혈세포, 난소에서 약하게 발현하였다. 그리고 합성된 sNPF에 의해서도 BsNPF-R의 유전자발현이 조절되었다.

TM-25659-Induced Activation of FGF21 Level Decreases Insulin Resistance and Inflammation in Skeletal Muscle via GCN2 Pathways

  • The TAZ activator 2-butyl-5-methyl-6-(pyridine-3-yl)-3-[2'-(1H-tetrazole-5-yl)-biphenyl-4-ylmethyl]-3H-imidazo[4,5-b]pyridine] (TM-25659) inhibits adipocyte differentiation by interacting with peroxisome proliferator-activated receptor gamma. 1 TM-25659 was previously shown to decrease weight gain in a high fat (HF) diet-induced obesity (DIO) mouse model. However, the fundamental mechanisms underlying the effects of TM-25659 remain unknown. Therefore, we investigated the effects of TM-25659 on skeletal muscle functions in C2 myotubes and C57BL/6J mice. We studied the molecular mechanisms underlying the contribution of TM-25659 to palmitate (PA)-induced insulin resistance in C2 myotubes. TM-25659 improved PA-induced insulin resistance and inflammation in C2 myotubes. In addition, TM-25659 increased FGF21 mRNA expression, protein levels, and FGF21 secretion in C2 myotubes via activation of GCN2 pathways (GCN2-$phosphoelF2{\alpha}$-ATF4 and FGF21). This beneficial effect of TM-25659 was diminished by FGF21 siRNA. C57BL/6J mice were fed a HF diet for 30 weeks. The HF-diet group was randomly divided into two groups for the next 14 days: the HF-diet and HF-diet + TM-25659 groups. The HF diet + TM-25659-treated mice showed improvements in their fasting blood glucose levels, insulin sensitivity, insulin-stimulated Akt phosphorylation, and inflammation, but neither body weight nor food intake was affected. The HF diet + TM-25659-treated mice also exhibited increased expression of both FGF21 mRNA and protein. These data indicate that TM-25659 may be beneficial for treating insulin resistance by inducing FGF21 in models of PA-induced insulin resistance and HF diet-induced insulin resistance.

한 대학병원 건강검진센터에 내원한 성인 여성의 지방간과 관상동맥질환위험인자와의 관련성 (Relationship Between Fatty Liver and Coronary Risk Factors among Health Examined Adult Women in an University Hospital)

  • 본 연구는 복부초음파검사를 통하여 얻은 건강진단 결과로부터 지방간의 유소견자 비율을 파악하고, 지방간이 어느 정도 관상동맥질환위험인자와 관련하고 있는가를 검토하는 것을 목적으로 2009년 7월부터 2010년 6월까지의 기간에 한 대학병원 건강검진센터에서 종합건강검진을 받았던 30세 이상 여성 665명을 대상으로 하였다. 연구결과 전체 조사대상 여성의 지방간 유소견율은 11.6%이었으며, 저연령군보다 고연령군에서, 비만도가 증가할수록, 유소견율은 유의하게 증가하였다. 연령과 BMI를 보정한 상태에서의 체지방율, 중성지질, 총콜레스테롤, LDL-콜레스테롤, 공복시 혈당 및 ALT는 지방간군이 비지방간군보다 유의하게 높았으며, HDL-콜레스테롤은 비지방간군이 지방간군보다 유의하게 높았다. 연령과 BMI를 조정한 관상동맥질환위험인자와 혈액 검사치에 따른 지방간의 위험비는 TG, TC, LDL-C 및 ALT치가 정상군보다 비정상군에서 유의하게 높게 나타났다.

중년기 남성 근로자의 흡연상태에 따른 혈압, 혈청지질 및 비만지표의 검토 (Investigation of Blood Pressure, Serum Lipids, and Obesity Indices according to Smoking Status in Middle-Aged Males)

  • 본 연구는 중년기 남성 근로자의 흡연과 비흡연에 따른 혈압, 혈청지질치 및 비만지표와의 관계를 알아보고, 금연이 혈압, 혈청지질 및 비만도의 변화에 어느 정도 영향을 미치는 가를 검토하고자 시도하였다. 연구대상은 30~59세의 남성 근로자 2,172명이었으며, 조사항목으로는 피조사자들의 연령, 흡연상태, SBP와 DBP, TC, TG, HDL-C, LDL-C, 동맥경화지수(AI), BMI, 체지방률 및 허리둘레였다. 조사는 2011년 4월부터 7월까지의 기간 동안에 실시하였다. 연구결과, 각 조사항목의 평균치를 보면 수축기혈압과 확장기혈압은 비흡연군(흡연중단군 포함)에 비해 흡연군에서 유의하게 높았다. TC, TG, 동맥경화지수(AI)는 비흡연군에 비해 흡연군에서 유의하게 높은 반면, HDL-C는 비흡연군에 비해 흡연군에서 유의하게 낮았다. BMI는 흡연군에 비해 비흡연군에서 유의하게 높았다. 흡연상태에 따른 각 조사항목의 분포를 보면, 고혈압은 비흡연군에 비해 흡연군에서 유의하게 높았다. TC, TG 및 HDL-C의 비정상치는 비흡연군에 비해 흡연군에서 유의하게 높았으며, BMI에 의한 비만은 흡연군보다 비흡연군에서 유의하게 높았다. 연령을 조정한 상태에서의 비흡연군에 비해 흡연군에서 발생할 각 조사항목의 위험비를 보면, 고혈압 발생의 위험비는 3.59배, 고콜레스테롤혈증 발생의 위험비는 1.49배, 고중성지질혈증 발생의 위험비는 1.81배, 저HDL-콜레스테롤혈증 발생의 위험비는 1.58배, 고동맥경화지수 발생의 위험비는 1.27배 증가하였다. 위와 같은 결과를 볼 때 흡연은 고혈압과 고지혈증에 관계하고 있음을 시사한다.

황토 첨가가 육계의 생산성과 육질에 미치는 영향 (Effects of Dietary Supplementation of Loess on the Performance and Meat Quality of Broiler Chicks)

  • 본 연구는 황토의 첨가 급여가 육계의 생산성과 육질 및 혈액 성상에 미치는 영향을 구명하기 위하여 실시하였다. 시험설계는 황토의 첨가수준을 0, 1.5, 3.0및 $5.0\%$로 4개의 처리구로 6주간 실시하였다. 증체량과 사료 요구율은 모든 황토 첨가구 보다 대조구에서 높았으나 유의적 차이는 없었다. 계육의 수분 함량은 모든 황토 첨가구에서 유의적으로 (P<0.05) 높았고, 조지방 함량은 대조구에 비해 모든 황토 첨가구에서 유의적으로 (P<0.05) 낮았으나, 조단백질 함량과 조회분 함량은 처리구간의 차이가 없었다. 근육내 pH는 모든 처리구간에 차이가 없었으나, 보수력은 대조구에 비해 모든 황토 첨가구에서 높았다. 계육의 육색 중에서 명도는 모든 황토 첨가구에서 증가한 반면, 적색도와 황색도는 차이가 없었다. 불포화지방산 함량은 대조구에 비해 황토-$3\%$와 황토-$5\%$ 첨가구에서 높았다. 혈액성분 중에서 HDL-cholesterol 과 중성지방 함량은 대조구에 비해 모든 황토 첨가구에서 유의적으로 (P<0.05) 낮았다. 계분의 수분 함량은 대조구에 비해 모든 황토 첨가구에서 유의적으로 (P<0.05)낮았다. 위의 결과를 종합하여 볼 때 육계사료에 황토를 첨가하였을 때 생산성에는 큰 차이가 없었으나 육질개선에는 효과가 있다고 생각된다.

점토광물 급여가 한우교잡우의 산육능력 및 육질에 미치는 영향 (Effect of Scoria, Quartz Porphyry and Oak Charcoal Powder Feeding on the Meat Quantity and Quality of Fattening Cross Bred Hanwoo Bulls)

  • This study was conducted to investigate the effects of Scoria, Quartz porphyry and Oak charcoal powder feeding by 1% level of concentrate on the fattening cross bred bulls(5/8 Korean Native Cattle, 2/8 Charolais, 1/8 Brahman germ plasma) Feeding trial was conducted with 4 treatment groups which were T1(Full feeding of concentrate and roughage for 12 months), T2(T1 and Scoria addition), T3(T1 and Quartz porphyry addition) and T4(Oak charcoal addition) for 360 days, consisting of 32 heads whose initially weights were about 299.7kg. The results obtained are summarized as follows; 1. During the over-all period, average daily gains by T3, T4, T2 and T1 were 1.024, 0.987, 0.977kg and 0.964, respectively, without a significant difference by treatments. 2. Concentrate intakes per body weight and TDN intakes required per unit of kilogram gain were lower in addition agent groups than in control, and in all the treatments feed efficiency was higher at early stage for 360 fattening day than at end stage. 3. Carcass weight, dressing percent, back-fat thickness and loin eye muscle area by treatments were 330.9 to 348.4kg(average 340.3kg), 55.2~56.2(average 55.8%), 0.41~0.55(average 0.46cm)and 76.0~80.9$\textrm{cm}^2$(average 77.6$\textrm{cm}^2$), respectively, and were improved in addition agent groups than in control. 4. Meat quality grade is higher in addition agent groups than in control, and heads of 1st grade by treatments was 3, 2, 1 and none in T4, T2, T3 and T1, respectively. 5. Incidence heads of diseases by treatment were not apparently different, but diseases of digestive system was decreased in addition agent groups than in control. 6. The nitrate nitrogen content of fattening bull dung which collected in rectum were 0.082~0.089% (average 0.084%), and the content in addition agent groups was smaller than in controll. 7. Income was higher in order of T4, T3, T2, and T1 and incomes of treatment groups were grater than that of control group by 21.4 to 33.5 percent. According to the above results it may be concluded that fattening bulls may be required to feed no more than 1% of Scoria, Quartz porphyry and Oak charcoal powder based on the concentrate feeding level during the fattening period(12 to 24 month of age) to produce high quality meat and increase income.
