Relationship between Cholesterol and Oxidative Potential from Meat Cooking (고기구이 초미세먼지 내 콜레스테롤 및 산화 잠재력과의 관계)

  • Identification of the major sources contributing to PM is of importance in order to understand their quantitative contributions to atmosphere. In the viewpoint of the meat cooking in Korea, only a few analyses of organic molecular markers have been conducted due to analytical difficulties. In this study, ten different parts of meat (i.e., blade shoulder, belly, and arm shoulder of pork; ribeye roll, top blade muscle, and short plate of beef; leg quarter, breast, and wing of chicken; duck; mackerel) were pyrolyzed to generate the cooked PM using an electronic heating plate. Generated PM were collected by the pyrolysis sampling system to identify total carbon (TC) using a carbon analyzer and cholesterol using a Liquid chromatography tandem-mass spectrometry (LC-MSMS) based on fragmentor voltage (FV), precursor ion, collision energy, product ion. In addition, oxydative potential (OP) analysis using dithiothreitol (DTT) method were discussed to investigate the toxicity relates. Highly correlated pairwise scatterplots between the cholesterol and TC indicate that oxydative potential was highly associated with different parts of meat. This study provides insight into the meat cooking components of PM, which could be drivers of the oxidative potential relates.

Central Venous Access in Children (소아의 중심정맥로 설치술)

  • To evaluate the effectiveness of central venous catheters(CVCs) in children, 320 CVCs placed in 255 neonate and children over a 10-year period were analyzed retrospectively. CVC was placed by one pediatric surgeon for a total of 6, 116 patients days. Catheters were placed preoperatively for TPN or chemo in 223 cases. CVC was solely for TPN in 57 cases and for chemotherapy in 40. Local anesthesia was utilized in 71 cases, and the general anesthesia was administered in the remainder of the patients. The subclavian vein was catheterized(SCV) in 202 cases(82 infants and neonates), tunneled external jugular venotomy(EJV) was utilized in 38, tunneled internal jugular venotomy(UV) in 2, the facial venotomy(FV) was used in 3, and the umbilical vein was catheterized UVC) with vein transposition in 74 infants. In neonates, 72 UVCs were placed during laparotomy. SCV was increased with ages, from 3 kg of minimal body weight. The average catheter-periods over-all were 19.1 days, SCV 17 days, EJV 40, IJV 60 and UVC 14. Technical complications were; arterial puncture(6), puncture failure(5) and abnormal location(12) in SCV; insertion failure(3) in EJV; abnormal location in the portal vein(4) and the liver parenchyma(2) cystic fluid accumulations in UVC. Twelve migrations(3.8 %) out of position occurred; SCV(2), EJV(1) and UVC(9). There were 4 cases(1.2 %) of catheter obstruction and 11(3.4 %) of catheter infection(3 SCV, 2 EJV and 6 UVC). Rescue procedures were utilized with some success. There was one mortality(0.3 %) due to deep sedation in a 1.06 kg baby during placement of an EJV. The surgeon's experience, proper catheter selection and following safety rules are the most important factors for successful CVCs.

  • In this study, a fully automatic pose and expression invariant 3D face alignment algorithm is proposed to handle frontal and profile face images which is based on a two pass course to fine alignment strategy. The first pass of the algorithm coarsely aligns the face images to an intrinsic coordinate system (ICS) through a single 3D rotation and the second pass aligns them at fine level using a minimum nose tip-scanner distance (MNSD) approach. For facial recognition, multi-view faces are synthesized to exploit real 3D information and test the efficacy of the proposed system. Due to optimal separating hyper plane (OSH), Support Vector Machine (SVM) is employed in multi-view face verification (FV) task. In addition, a multi stage unified classifier based face identification (FI) algorithm is employed which combines results from seven base classifiers, two parallel face recognition algorithms and an exponential rank combiner, all in a hierarchical manner. The performance figures of the proposed methodology are corroborated by extensive experiments performed on four benchmark datasets: GavabDB, Bosphorus, UMB-DB and FRGC v2.0. Results show mark improvement in alignment accuracy and recognition rates. Moreover, a computational complexity analysis has been carried out for the proposed algorithm which reveals its superiority in terms of computational efficiency as well.

Characterization of a QTL associated with chlorophyll content using progeny from an interspecific cross in rice (Oryza Sativa L.)

  • Rice (Oryza sativa L.) is the world's most important cereal crop. In crop plant, chlorophyll content and leaf senescence could affect grain filling and yield. We analyzed a QTL associated with chlorophyll content and delayed leaf senescence using high chlorophyll near isogenic line (HC-NIL). HC-NIL derived from a cross between Oryza sativa cv. Hwaseong as a recurrent parent and wild species O. grandiglumis as a donor parent showed higher chlorophyll content than Hwaseong. To identify QTL associated with chlorophyll content, 58 $F_3$ and 38 $F_4$ lines were developed from a cross between HC-NIL and Hwaseong. For QTL analysis, simple sequence repeat (SSR) markers were used for genotyping and one-way ANOVA was conducted. A QTL for chlorophyll content (qCC2) was detected in chromosome 2 and explained 24.63% of phenotypic variation. The senescence effect of the qCC2 was examined in dark-induced incubation (DII). Detached leaves from Hwaseong and HC-NIL were incubated on 3mM MES buffer (pH 5.8) at $27^{\circ}C$ under complete dark condition. After 3 days of incubation, the Hwaseong leaves turned yellow, but the HC-NIL leaves were green. HC-NIL has higher chlorophyll content with delayed senescence than Hwaseong. These results indicated that qCC2 is associated with stay-green phenotype. To know whether the qCC2 is responsible for leaf functionality, ion leakage test and Fv/Fm measurement were performed. Both experiment results showed that differences were observed between Hwaseong and HC-NIL but it was not statistically significant. These results might suggest that the qCC2 is possibly related to chlorophyll content and non-functional stay-green phenotype.

The effects of zeatin on Cd2+ -induced physiological toxicities in Commelina communis L (Zeatin이 Cd2+에 의한 닭의장풀의 생리적 독성에 미치는 영향)

  • Three-week old Commelina communis was transferred and grown in Hoagland solution containing $100{\mu}M\;Cd^{2+},\;100{\mu}M\;Cd^{2+}+100{\mu}M\;kinetin,\;100{\mu}M\;Cd^{2+}+100{\mu}M\;zeatin\;and\;100{\mu}M\;Cd^{2+},\;200{\mu}M$ zeatin for 7 days, and then a number of physiological activities were investigated. In control, the length of the stem of plants was increased to 4.7cm, but in $Cd^{2+},\;Cd^{2+}+kinetin,\; Cd^{2+}+100{\mu}M\;zeatin\;and\;Cd^{2+}+200{\mu}M$ zeatin treatments, the growth of plants were increased to 1.5cm, 2.1cm, 3.9cm and 4.3 em, respectively. In the treatments of $Cd^{2+},\;Cd^{2+}+kinetin,\;Cd^{2+}+100{\mu}M\;zeatin\;and\; Cd^{2+}+200{\mu}M$ zeatin, total chlorophyll contents were reduced to $26\%,\;24\%,\;15\%\;and\;3\%$, respectively, on the contrast to the control. In chlorophyll fluorescence experiments, Fv/Fm ratios were also reduced to $44\%,\;21\%,\;17\%\;and\;5\%$ in the light intensity of $2100{\mu}Mmole\;E\;m^{-2}s^{-1}\;by\;Cd^{2+},\;Cd^{2+}+kinetin,\;Cd^{2+}+$100{\mu}M\;zeatin\;and\;Cd^{2+}+200{\mu}M$ zeatin treatments on the contrast to the control. Water stresses were increased to 2.6, 1.7 and 1.2 times by $Cd^{2+},\; Cd^{2+}+kinetin\;and\;Cd^{2+}+{\mu}M$ zeatin. On the other hand, combination of $Cd^{2+}+200{\mu}M$ zeatin reduced water stress to $0.12\%$. In $Cd^{2+}$ accumulation experiments $Cd^{2+}$transports were inhibited to $33\%\; 48\%\;and\;70\%\;by\;Cd^{2+}+kinetin,\;Cd^{2+}+100{\mu}M\;zeatin\;and\;Cd^{2+}+200{\mu}M$ zeatin. Therefore, it could be concluded that zeatin clearly reduced the toxicities of $Cd^{2+}$ by reducing the absorption of $Cd^{2+}$.

Effects of pH and Salinity on the Cultivated Brown Alga Sargassumfulvellum and Associated Animals (갈조류 양식 모자반(Sargassum fulvellum)과 해적생물에 대한 pH와 염분의 효과)

  • The effects pH and salinity on the brown alga Sargassum fulvellum were investigated in the context of the removal of two major associated animals, Caprella scaura and Gammaropsis utinomi. Optimum quantum yield (Fv/Fm) of S. fulvellum was also examined in the same experimental conditions as an index of stress. Experiments on pH and salinity tolerances of the two animal species indicated that mortality was more than 80% at extremes of pH (2, 3, 4, 10, 11, 12) and salinity (0, 3.5, 7, 10, 44 psu) after a 5 min treatment. Lethal time (LT50) from pH 2 to pH 4 was less than 90 sec in C. scaura, and less than 70 sec in G. utinomi. From 0 to 10 psu, LT50 was less than 20 sec in C. scaura and less than 60 sec in G. utinomi. The quantum yield of S. fulvellum was not significantly different from controls within the pH range 4~10, and within the salinity range of 7-40 psu. When exposed to pH and salinity conditions outside these ranges, the effect of these factors on the removal of two animal species was higher, but quantum yield was highly reduced. These results indicate that the optimal conditions for removing the animal species without affecting optimum quantum yield were pH 4-10, and salinities 7-10 psu and 44 psu.

Localized Surface Plasmon Resonance (LSPR) Biosensors on Metal Nanoparticles with the Design of Bioreceptors

  • Label-free biomolecular assay based localized surface plasmon resonance (LSPR) of noble metal nanoparticles enables simple and rapid detection with the use of simple equipment. Nanosized metal nanoparticles exhibit a strong absorption band when the incident light frequency is resonant with the collective oscillation of the electrons, which is known as the LSPR. Here we demonstrate localized surface plasmon resonance (LSPR) substrates such as plasmonic Au nanodisks fabricated by a nanoimprinting process and gold nanorod-immobilized surfaces and their applications to highly sensitive and/or label-free biosensing. To increase detection sensitivity various bioreceptors weree designed. A single chain variable fragment (scFv) was used as a receptor to bind C-reactive protein (CRP). The results of this effort showed that CRP in human serum could be quantitatively detected lower than 1 ng/ml. Aptamers, which were immobilized on gold nanorods, were used to detect mycotoxins. The specific binding of ochratoxin A (OTA) to the aptamer was monitored by the longitudinal wavelength shift of LSPR peak in the UV-Vis spectra resulting from the changes of local refractive index near the GNR surface induced by accumulation of OTA and G-quadruplex structure formation of the aptamer. According to our results, OTA could be quantitatively detected lower than 1 nM level. Additionally, aptamer-functionalized GNR substrate was quite robust and can be regenerated many times by rinsing at 70 OC to remove bound target. During seven times of washing steps, the developed OTA sensing system could be reusable. Moreover, the proposed biosensor exhibited selectivity over other mycotoxins with an excellent recovery for detection in grinded corn samples, suggesting that the proposed LSPR based aptasensor plays an important role in label-free detection of mycotoxins.

Centralized Control Algorithm for Power System Performance using FACTS Devices in the Korean Power System

  • This paper presents a centralized control algorithm for power system performance in the Korean power system using Flexible AC Transmission Systems (FACTS) devices. The algorithm is applied to the Korean power system throughout the metropolitan area in order to alleviate inherent stability problems, especially concerns with voltage stability. Generally, control strategies are divided into local and centralized control. This paper is concerned with a centralized control strategy in terms of the global system. In this research, input data of the proposed algorithm and network data are obtained from the SCADA/EMS system. Using the full system model, the centralized controller monitors the system condition and decides the operating point according to the control objectives that are, in turn, dependent on system conditions. To overcome voltage collapse problems, load-shedding is currently applied in the Korean power system. In this study, the application of the coordination between FACTS and switch capacitor (SC) can restore the solvability without load shedding or guarantee the FV margin when the margin is insufficient. Optimal Power Flow (OPF) algorithm, for which the objective function is loss minimization, is used in a stable case. The results illustrate examples of the proposed algorithm using SCADA/EMS data of the Korean power system in 2007.

Determination of Analytical Approach for Ambient PM2.5 Free Amino Acids using LC-MSMS (LC-MSMS를 이용한 대기 중 PM2.5 유리아미노산 분석 방법 연구)

  • Atmospheric nitrogen containing organic compounds(e.g. amino acids) has attracted considerable attention from the viewpoint of the oceanic biogeochemical cycle of nitrogen as well as the long range transfer. However, only a few measurements of organic nitrogen compounds have been conducted due to analytical difficulties. In this study, total of nine amino acids such as Glutamic acid, Histidine, Arginine, Tyrosine, Cystine, Valine, Methionine, Phenylalanine, Lysine have been analytically determined by Liquid Chromatography - Mass Spectrometry Mass Spectrometry (LC-MSMS). As results, Fragmentor Voltage (FV), Precursor Ion, Collision Energy, Product Ion related to individual amino acid compounds are shown. Based on the operational conditions, Lysine, Glutamine Acid, Tyrosine were analyzed during the China Oriented Smog Period. High concentrations of Lysine, Glutamine Acid, and Tyrosine are discussed with organic carbon (OC), elemental carbon (EC), and water soluble ions. The results can provide to understand the sources with aging process related to amino acids influenced by the long-range transport from the Yellow Sea area.

Environmentally Friendly Phytal Animal Removal for Re-use of Holdfasts of Sargassum fusiforme (Harvey) Setchell: pH and Salinity (갈조류 톳의 포복지 재활용을 위한 친환경적 해적생물 구제: pH와 염분)

  • The brown seaweed Sargassum fusiforme is an edible and highly valued in Korea. During the summer season, phytal organisms graze heavily on young algal blades and holdfastsof the species and substantially reduce harvestable biomass. Here, in this study, we investigated the effects of pH (range: 2~13) and salinity (range: 0~44 psu) on the removal of two major phytal animals, Caprella scaura and Gammaropsis utinomi, associated with S. fusiforme. We also examined the optimum quantum yield (Fv/Fm) of algae in the same experimental conditions to quantify the tolerance of algae to acid and salinity treatments. It was observed that the phytal animals showed more than 80% mortality at pH lower that pH 4 and the extreams of salinity (0~10 psu and 44 psu) after a 5 min of immersion. However, the quantum yield of S. fusiforme was not significantly different from controls within the pH 3~11 range, and the 0~44 psu salinity range. Precisely, if the pH and salinity conditions outside these ranges were used in comercial Sargassum culture, the removal of the animal species would be higher, but with reduced quantum yield of algae. Taken together, our study results indicated that the pH and salinity treatments could allow multiple harvests from the same holdfast of S. fusiforme.
