Characterization and Expression in Escherichi coli of Streptococcus pneumoniae FtsH

  • FtsH is a membrane-bound, ATP-dependent metalloprotease that is involved in a variety of cellular functions including the regulation of responses to heat and stress shock. Previously, we had cloned and sequenced pneumococcal ftsH gene whose deduced amino acid sequence was very similar to those of several gram-positive bacteria and Escherichia coli, except for the N-terminal domain that was responsible for membrane anchoring. In order to better understand the role of Streptococcus pneumoniae FtsH, we expressed pneumococcal ftsH gene in Escherichia coli. When it was expressed from a strong promoter, $P_{tac}$, a considerable amount of the recombinant FtsH was produced, although the prolonged induction resulted in not only accumulation of breakdown products but also ceasing of the further growth of E. coli host. This indicated that the expression of the exogenous ftsH gene was tightly regulated since the excessive FtsH appeared detrimental to bacterial cells. In Western blotting, the pneumococcal FtsH protein, whether native or recombinant, was reactive to anti-E. coli FtsH serum. The observation that FtsH proteins were well conserved throughout the bacterial kingdom and its expression level was fine-tuned suggests an important role for this protein in the stress adaptation which may be related to infecting process by pneumococci.

Cloning, Expression, and Polymerization Assay of FtsZ Protein from Staphylococcus aureus (Staphylococcus aureus FtsZ의 클로닝, 발현 및 폴리머 형성 활성 분석)

  • Cytokinesis is the final stage of cell division, dividing one mother cell into two daughter cells. For the cutting of a plasma membrane during bacterial cytokinesis, a tubulin homolog FtsZ protein is recruited from the cytoplasm to the division site. FtsZ protein polymerizes in a GTP-dependent manner and its N-terminal domain has a GTPase activity. In this study, we have begun to characterize FtsZ from Staphylococcus aureus (SA). Full-length SA FtsZ was cloned into pRSFDuet-1 vector and the clone was transformed into a BL21 (DE3) star cell. The recombinant SA FtsZ protein was purified using Ni-NTA affinity chromatography and dialysis. Using a spectrofluorometer, we showed that SA FtsZ undergoes a GTP-dependant polymerization in vitro. The polymer of the SA FtsZ protein disappeared after a few minutes, suggesting that the polymer is degraded as the GTP is consumed. This assay system may well be applied for inhibitor screening targeting S. aureus FtsZ.

  • This paper describes the design and implementation for spread spectrum FTS encoder and decoder. The FTS command format is defined by 64 bit encrypted packet that contains all required information relayed between the ground and the vehicle. Encryption is accomplished using the Tripple-DES encryption algorithm in block encryption form. The proposed FTS encoder and decoder is using the Convolution Encoding and Viterbi Decoding for forward error correction. The Spread Spectrum Modulation is done using a PN code, which is 256 bit gold code. The simulation result shows that the designed FTS decoder is compatible with the designed FTS encoder.

Effects of the Method of FTSs on the Performance of HPAs (FTS 방식이 전력증폭기의 성능에 미치는 영향)

  • It is necessary to develop the next generation FTS which is suitable for our environment and effectively operates many launch vehicles. Standard tone, Secure tone, MHA, EFTS and DSSS are studied for the next generation FTS. FTS requires a high quality of performance and reliability because of their specific mission. And few FTSs are needed but the price is very expensive. Therefore we must investigate a part of the FTS whether the part can be reused for a part of the next FTS. In this paper, we use CCDF of the transmitted signal from FTS as the method to study a possibility of reusing HPA used in the present system. The simulation results show that PEP of Standard tone is 0.21dB and Secure tone and MHA has the same PEP. CPFSK's PEP is 1.81dB and PEP of DSSS using BPSK modulation is 2.6dB.

Effects of Launching Vehicle's Velocity on the Performance of FTS Receiver (발사체의 속도가 FTS 수신기의 성능에 미치는 영향)

  • A doppler shift is generated by moving a transmitter or receiver operated in communication systems. The doppler frequency shift between a transmitter and a receiver or the frequency offset present in transceivers must be removed to get the wanted system performance. FTS is used for preventing an accident from operating abnormally and for guaranteeing public protection. A launching vehicle's initial velocity is very fast in order to escape the earth and the amount of doppler shift is large. Recently many studies to adopt the next generation FTS are ongoing. To introduce new FTS, the effects of doppler shift on the performance of the new FTS must be studied. In this paper the doppler effect caused by launching vehicle's velocity affecting the performance of FTS receiver is investigated into two cases, one is for EFTS as a digital FTS and the other is for FTS using a tone signal. Noncoherent DPSK and noncoherent CPFSK are considered as the modulation methods of EFTS. In the cases of the doppler frequency shift of 200Hz present in EFTS using noncoherent DPSK and noncoherent CPFSK are simulated. Simulation results show that $E_b/N_o$ of 0.5dB deteriorates in the region of near BER of about $10^{-5}$ in RS coding. And there is no performance variation in $E_b/N_o$ or $E_b/N_o$ is worsened about 0.1dB in the same BER region for the case of using convolutional and BCH coding. Quadrature detector used in FTS using tone signals is not influenced by the doppler frequency shift.

Membrane-Bound Protease FtsH Protects PhoP from the Proteolysis by Cytoplasmic ClpAP Protease in Salmonella Typhimurium

  • Among the AAA+ proteases in bacteria, FtsH is a membrane-bound ATP-dependent metalloprotease, which is known to degrade many membrane proteins as well as some cytoplasmic proteins. In the intracellular pathogen Salmonella enterica serovar Typhimurium, FtsH is responsible for the proteolysis of several proteins including MgtC virulence factor and MgtA/MgtB Mg2+ transporters, the transcription of which is controlled by the PhoP/PhoQ two-component regulatory system. Given that PhoP response regulator itself is a cytoplasmic protein and also degraded by the cytoplasmic ClpAP protease, it seems unlikely that FtsH affects PhoP protein levels. Here we report an unexpected role of the FtsH protease protecting PhoP proteolysis from cytoplasmic ClpAP protease. In FtsH-depleted condition, PhoP protein levels decrease by ClpAP proteolysis, lowering protein levels of PhoP-controlled genes. This suggests that FtsH is required for normal activation of PhoP transcription factor. FtsH does not degrade PhoP protein but directly binds to PhoP, thus sequestering PhoP from ClpAP-mediated proteolysis. FtsH's protective effect on PhoP can be overcome by providing excess ClpP. Because PhoP is required for Salmonella's survival inside macrophages and mouse virulence, these data implicate that FtsH's sequestration of PhoP from ClpAP-mediated proteolysis is a mechanism ensuring the amount of PhoP protein during Salmonella infection.

Characterization of ZTO Thin Films Transistor Deposited by On-axis Sputtering and Facing Target Sputtering(FTS) (On-axis 스퍼터링과 FTS 공정으로 증착한 ZTO 박막트랜지스터의 특성)

  • We have investigated the properties of thin film transistors(TFT) fabricated using zinc tin oxide(ZTO) thin films deposited via on-axis sputtering and FTS methods. ZTO thin films deposited by FTS showed lower root-mean-square(RMS) roughness and more uniformity than those deposited via on-axis sputtering. We observed enhanced electrical properties of ZTO TFT deposited via FTS. The ZTO films were deposited at room temperature via on-axis sputtering and FTS. The as-deposited ZTO films were annealed at $400^{\circ}C$. The TFT using the ZTO films deposited via FTS process exhibited a high mobility of $12.91cm^2/V.s$, a low swing of 0.80 V/decade, $V_{th}$ of 5.78 V, and a high $I_{on/off}$ ratio of $2.52{\times}10^6$.

  • Flight termination system(FTS) is used to terminate safely the launch vehicle's flight when it faces an emergency situation by transmitting termination command from ground FTS. RCC standard IRIG tone method has been used widely for FTS commands method in foreign ranges and Naro Space Center, but this method has a weakness for security of command signal. Therefore RCC had studied more secured EFTS standard and chose CPFSK digital modulation method. This paper describes basic concept and types of FTS and FTS types which foreign ranges had applied and describes standardization of EFTS in the U.S. based on RCC EFTS study reports.

  • The growth of the AR/VR market due to the advent of the 4th Industrial Revolution begins with the development of the display industry. The development of OLED and flexible displays is further accelerated by the development of R2R technology. Micro-processing technology using a fast tool servo (FTS), the core technology in R2R processes, is making technological progress in increasingly diverse ways. This paper proposes a method to develop an FTS for horizontal driving and presents this method through experiments and analyses. To develop a swing-type FTS based on a seesaw motion, a rotational moment hinge structure was designed for each type, and research was conducted to determine an effective design method. A cantilever-based swing-type FTS was developed in two variations: one with single-side hinges and another with dual-side hinges. The parameters in the design of the swing-type FTS are rotational moment, natural frequency, and material selection. In conclusion, an FTS with a single-side hinge demonstrates the high performance required for micro processing.

  • The preparation and characteristics of ITO anode films grown using a linear facing target sputtering (FTS) technique for use in organic light emitting diodes (OLED) and flexible OLED is described. The electrical, optical, and work function of the ITO anode, which was prepared by linear FTS at room temperature, were comparable to those of commercial ITO anode films. In particular, linear FTS-grown ITO films shows very smooth surface without defects such as pin hole and cracks due to low substrate temperature. Furthermore OLED with the linear FTS-grown ITO anode film shows comparable electrical and optical properties to those of OLED with the commercial crystalline-ITO anode film. This suggested that linear FTS is promising thin film technology for preparing high quality anode film in OLEDs and flexible OLEDs.
