Plant cell culture strategies for the production of natural products

  • Plants have evolved a vast chemical cornucopia to support their sessile lifestyles. Man has exploited this natural resource since Neolithic times and currently plant-derived chemicals are exploited for a myriad of applications. However, plant sources of most high-value natural products (NPs) are not domesticated and therefore their production cannot be undertaken on an agricultural scale. Further, these plant species are often slow growing, their populations limiting, the concentration of the target molecule highly variable and routinely present at extremely low concentrations. Plant cell and organ culture constitutes a sustainable, controllable and environmentally friendly tool for the industrial production of plant NPs. Further, advances in cell line selection, biotransformation, product secretion, cell permeabilisation, extraction and scale-up, among others, are driving increases in plant NP yields. However, there remain significant obstacles to the commercial synthesis of high-value chemicals from these sources. The relatively recent isolation, culturing and characterisation of cambial meristematic cells (CMCs), provides an emerging platform to circumvent many of these potential difficulties.

Forward osmosis membrane filtration for microalgae harvesting cultivated in sewage effluent

  • The purpose of this study is to evaluate the performance of forward osmosis (FO) system for harvesting microalgae cultivated in secondary sewage effluent. Microalgae species used in this study were chlorella sp. ADE4. The drawing agents used for forward osmosis system were seawater and concentrate of sea water reverse osmosis (SWRO) system. Chlorella sp. ADE4 cultured in secondary sewage effluent illustrated moderate efficiency in removal of total nitrogen (TN) (68%) and superior performance in total phosphorus (TP) removal (99%). Comparison of seawater and SWRO concentrate as drawing agent were made in FO membrane separation of the microalgae. The result from this study depicts that SWRO concentrate is strong drawing agent in FO membrane system providing an average dewatering rate of $4.8L/(m^2{\cdot}hr)$ compared to seawater with average dewatering of $2.9L/(m^2{\cdot}hr)$. Results obtained from this study indicated that FO system could be viable option for harvesting the microalgae for further biodiesel production. SWRO concentrate as a drawing agent could be very important finding in field of membrane technology for disposal of SWRO concentrate.

노린재류의 알에 기생하는 Trissolcus 속(벌목: 납작먹좀벌과)의 숙주 (Host Records of Trissolcus (Hymenoptera: Platygasteridae: Telenominae) Parasitizing Eggs of Stink Bugs in Korea)

  • 납작먹좀벌과(Platygasteridae)의 큰검정알벌속(Trissolcus)에 속하는 T. japonicus (Ashmead) (썩덩큰검정알벌, 신칭)이 과수원에서 과일에 심한 피해를 일으키는 해충인 썩덩나무노린재의 알에 기생하는 것으로 확인되었고, 생물적 방제인자로써 가능성이 기대된다. 또한 천마큰검정 알벌(T. itoi Ryu)이 북쪽비단노린재 알을, 검은다리큰검정알벌(T. nigripedius Nakagawa)이 알락수염노린재 알을 숙주로 각각 기생하는 것이 확인되었다. 동정을 위한 진단형질 및 영상자료와 한국에 분포하는 큰검정알벌속(Trissolcus)의 종들에 관한 숙주목록을 제공하였다.

Evaluate of Electrochemical Characteristics in Electrolyzed Reduced Water

  • Active oxygen species or free radicals are considered to cause extensive oxidative damage to biological macromolecules, which brings about a variety of diseases as well as aging. Electrolyzed reduced water(ERW) has been regarded as a ideal antioxidative agent in recent years. ERW is produced by passing a diluted salt solution through an electrolytic cell, within which the anode and cathode are separated by membrane. It can be produced reactive materials in ERW near the cathode during the electrolysis of water. ERW have the following advantages over other traditional cleaning agents: effective antioxidative agent, easy preparation, inexpensive, and environmentally friendly. The main advantage of ERW is its safety and antioxidative effect. ERW with strong reducing potential can be used to remove dirt and grease from items such as cutting boards and other kitchen utensils. The primary aim of this study is the activation mechanism of oxidation reduction potentials, ion conductivity, pH, and electrochemical properties with reactive materials in ERW.

제주 감귤원에서 귤굴나방에 대한 포식 기생자 천적의 자연 기생율 (Natural parasitism of parasitoids on Phyllocnistis citrella (Lepidoptera: Gracillaridae) in Jeju citrus orchards in Korea)

  • 본 연구는 감귤의 해충인 귤굴나방(Phyllocnistis citrella Stainton) 의 포식 기생자 천적의 종류와 기생률을 조사하기 위하여 수행하였다. 2013~2014 제주 감귤원에서 발견된 귤굴나방 포식 기생자 천적은 좀벌상과에 속한 총 7종을 발견되었다. 그 중 좀벌과에 속한 Sympiesis striatipes가 73%, Quadrastichus sp.가 22%로 우점하였다. 나머지 종은 Neochrysocharis sp., Ageniaspis sp., Pnigalio sp., Holcopelte sp., Trichomalopsis sp. (Pteromalidae) 등이었다. 천적의 평균 기생률은 관행방제 과원에서 10.7%, 무농약 과원에서는 32.2% 이었다.

Suppression of Bipolaris Stem Rot on Cactus by Heat-inactivated Conidial Suspension of Bipolaris cactivora

  • The heat-inactivated (at $121^{\circ}C$ for 20 min) conidial suspension of Bipolaris cactivora (HICS) was evaluated for the control of Bipolaris stem rot of cactus caused by B. cactivora. Severe rot symptoms were developed on the cactus stem discs inoculated with B. cactivora from 5 days after inoculation. However, only small brownish spots developed on the stem discs treated with HICS 2 days prior to the pathogen inoculation. HICS also reduced symptom development on cactus stem discs inoculated with other fungal pathogens such as Alternaria alternata, Colletotrichum gloeosporioides, and Fusarium oxysporum, suggesting its disease-inhibitory efficacy may not be pathogen-specific. HICS significantly reduced severities of the stem rot disease on several cactus species including Hylocereus trigonus, Cereus peruvianus, Chamaecereus silvestrii and Gymnocalycium mianovichii, but not on Cereus tetragonus. Extensive wound periderms were formed in the stem tissues of inoculation and/or wounding sites on C. peruvianus treated with HICS alone or prior to the pathogen inoculation, but not on C. tetragonus, indicating the structural modifications may be related to the mechanism of disease suppression by HICS. HICS also reduced the disease development on the grafted cactus (H. trigonus stock and G. mianovichii scion) with the control efficacy nearly equivalent to the application of a commercial fungicide. All of these results suggest HICS can be used as an environmental-friendly agent for the control of the cactus stem disease.

쑥 추출액을 이용한 Microcystis aeruginosa 제거 및 성장억제 연구 (Removal and Regrowth Inhibition of Microcystis aeruginosa using Artemisia asiatica Extracts)

  • Microcystis aeruginosa (M. aeruginosa) is a cyanobacterium species that can form harmful algal blooms in freshwater bodies worldwide. The use of Artemisia asiatica extracts to control M. aeruginosa inhibition will be environmentally friendly and promising. Artemisia asiatica extracts removed successfully upto 88% of M. aeruginosa pH 8 at $25^{\circ}C$ of temperature. These results was indicated that the amount of 2.24 g/L Artemisia asiatica extracts was removed 1g dryweight/L of M. aeruginosa. The kinetic data showed substrate inhibition kinetics and maximum growth rate was obtained when the M. aeruginosa was grown in medium containing 2.5 g/L of initial concentration of Artemisia asiatica extracts. In the various growth control models, Luong model showed the highest correlation coefficient of 0.9916. Therefore, the Luong model was the most suitable control model for the growth control of M. aruginosa using Artemisia asiatica extracts. In conclusion, the growth control of M. aruginosa using Artemisia asiatica extracts can be applied in the field without controlling the temperature and pH of rivers and streams, and it is possible to control the growth of M. aruginosa efficiently in a short time. The natural extract, Artemisia asiatica extracts, can be a promising inhibition due to its high efficiency and low dose requirements.

Effect of Carbonization Temperature on Hygric Performance of Carbonized Fiberboards

  • Increases of public attention on healthy environment lead to the regulation of indoor air quality such as Clean Healthy House Construction Standard. This standard covers emission of total volatile organic compounds (TVOCs) (e.g., formaldehyde, benzene, and toluene), ventilation, and use of environmentally-friendly products or functional products. Moisture absorption and desorption abilities are a recommended functionality for improving indoor air quality. In this study, moisture absorption and desorption capacities of carbonized board from wood-based panels and other materials were determined by using UNT-HEAT-01 according to ISO 24358:2008. Pine had higher moisture absorption and desorption capacities ($49.0g/m^2$ and $35.3g/m^2$, respectively) than hinoki cypress, cement board, gypsum board, oriented strand board, and medium density fiberboard (MDF). The moisture absorption and desorption capacities differed considerably according to the wood species. After carbonization process at $400^{\circ}C$, the absorption and desorption ability of MDF increased to 38% and 60%, respectively. However, moisture absorption and desorption capacities decreased with increasing carbonization temperature, but they were still higher than original MDF. Therefore, it is suggested that carbonization below $600^{\circ}C$ can improve moisture absorption/desorption capacities.

Phytotoxic Effect of 5-Aminolevulinic Acid, a Biodegradable Photodynamic Biomaterial, on Rice and Barnyardgrass

  • ALA (5-aminolevulinic acid) has been proposed as a tetrapyrrole-dependent photodynamic herbicide by the action of the protoporphyrinogen IX oxidase (Protox IX). A study was conducted to determine photodynamic herbicidal effect of ALA on seedling growth of rice (Oryza sativa L.) and barnyard grass (Echinochloa crus-galli Beauv. var. oryzicola Ohwi) under dry and wet conditions. ALA effect on early plant growth of rice and barnyardgrass was greatly concentration dependant, suggesting that it promotes plant growth at very low concentration and inhibits at high concentration. No significant difference in herbicidal activity of biologically and synthetically produced ALAs on plant lengths of test plants was observed ALA exhibited significant photodynamic activity regardless of PSDIP and its duration. Significant shoot growth inhibition by ALA soaking treatment exhibited apparently, indicating that ALA absorbed through root system was translocated into shoot part of plants. ALA reduced plant heights of rice and barnyardgrass seedlings by 6% and 27%, respectively, showing more tolerant to ALA in rice under wet condition. Leaf thickness was reduced markedly by ALA with increasing of ALA concentration, due to mainly membrane destruction and severe loss of turgidity in mesophyll cells, although the epidermal was little affected. It was observed that photodynamic herbicidal activity of ALA applied by pre-and post-emergence application exhibited differently on plant species, and that the activity of ALA against susceptible plants was highly correlated with growing condition.

Biological Control of the Pentatomid Stink Bug, Eocanthecona furcellata(Wolff.), by using their Parasitoid, Psix striaticeps Dodd, in Sericulture

  • Stink bug, Canthecona furcellatta (Hemiptera: Pentatomidae), is an important predator of silkworm larvae. Nymphs and adult attack the early stage silkworm larvae and causes about 10-15 per cent loss to silk industry. Synthetic organic pesticides has tremendous impact on minimizing the pest population but repeated and frequent use has created problems of residual toxicity, development of resistance to insecticides, pest resurgence and out break, phyto-toxicity and hazards to non target species and beneficial organism. Silkworms are very sensitive to pesticides; therefore, attempt has made to control the bug population through introduction of its native natural enemies in the silkworm-rearing field. Biological control has tremendous scope in sericulture because it is eco-friendly in nature and non-harmful farmers. Native natural enemies have been screened. Psix striaticeps, Trissolcus spp. and Telenomus spp. have been recorded as the most potential parasitoid against pentatomid bug. Life cycle, sex ratio and other various attributes of the par-asitoids have been recorded. The parasitization potential of the parasitoid is very high and they have the ability to discriminate between parasitized and unpar-asitized host. Mass propopagation technique under laboratory condition has been standaydized.
