Selection of parental strain on the sawdust cultivation and mycelial growth and cultural characteristics of Lentinula edodes hybrid strains (표고 톱밥재배용 모균주 선발과 교배균주의 균사배양 및 생육 특성)

  • In order to breed new strains of oak mushroom, Lentinula edodes, on the sawdust cultivation, we collected 10 from Korea, Taiwan and China respectively and examined somatic incompatibility, morphological features of fruiting body and productivity. Four strains(SANJO701HO, FMRI2534, FMRI2337 and FMRI2613) shown remarkable results, were confirmed with parental strains. 80 monokaryotic strains derived from the selected 4 parental strains, were selected and 117 hybrid strains were made by mono-mono mating. Aslo, Physiological characteristics were investigated. Average of the mycelial growth of hybrid strains of mating combination SANJO701HO-FMRI2337 was approximately 10% lower than other mating combinations. Overall, This study was founded to develop new varieties of L. edodes. The productivity of new 117 strains on sawdust cultivation needs continued research.

Occurrence of dsRNA Mycovirus (LeV-FMRI0339) in the Edible Mushroom Lentinula edodes and Meiotic Stability of LeV-FMRI0339 among Monokaryotic Progeny

  • dsRNA was found in malformed cultures of Lentinula edodes strain FMRI0339, one of the three most popular sawdust cultivated commercial strains of shiitake, and was also found in healthy-looking fruiting bodies and actively growing mycelia. Cloning of the partial genome of the dsRNA revealed the presence of the RdRp sequence of a novel L. edodes mycovirus (LeV), and sequence comparison of the cloned amplicon showed identical sequences sequence to known RNA-dependent RNA polymerase genes of LeV found in strain HKA. The meiotic stability of dsRNA was examined by measuring the ratio of the presence of dsRNA among sexual monokaryotic progeny. More than 40% of the monokaryotic progeny still contained the dsRNA, indicating the persistence of dsRNA during sexual reproduction. Comparing the mycelia growth of monokaryotic progeny suggested that there appeared to be a tendency toward a lower frequency of virus incidence in actively growing progeny.

Highly Accelerated SSFP Imaging with Controlled Aliasing in Parallel Imaging and integrated-SSFP (CAIPI-iSSFP)

  • Purpose: To develop a novel combination of controlled aliasing in parallel imaging results in higher acceleration (CAIPIRINHA) with integrated SSFP (CAIPI-iSSFP) for accelerated SSFP imaging without banding artifacts at 3T. Materials and Methods: CAIPI-iSSFP was developed by adding a dephasing gradient to the balanced SSFP (bSSFP) pulse sequence with a gradient area that results in $2{\pi}$ dephasing across a single pixel. Extended phase graph (EPG) simulations were performed to show the signal behaviors of iSSFP, bSSFP, and RF-spoiled gradient echo (SPGR) sequences. In vivo experiments were performed for brain and abdominal imaging at 3T with simultaneous multi-slice (SMS) acceleration factors of 2, 3 and 4 with CAIPI-iSSFP and CAIPI-bSSFP. The image quality was evaluated by measuring the relative contrast-to-noise ratio (CNR) and by qualitatively assessing banding artifact removal in the brain. Results: Banding artifacts were removed using CAIPI-iSSFP compared to CAIPI-bSSFP up to an SMS factor of 4 and 3 on brain and liver imaging, respectively. The relative CNRs between gray and white matter were on average 18% lower in CAIPI-iSSFP compared to that of CAIPI-bSSFP. Conclusion: This study demonstrated that CAIPI-iSSFP provides up to a factor of four acceleration, while minimizing the banding artifacts with up to a 20% decrease in the relative CNR.

Study of Viral Effects of the Mycovirus (LeV) and Virus-Free Commercial Line in the Edible Mushroom Lentinula edodes

Comparing the mycelia growth of monokaryotic progeny suggested that, although variations in the growth rate existed among progeny and virus infection was observed in highly actively growing progeny, there appeared to be a tendency toward a lower frequency of virus incidence in actively growing progeny. This study attempted to cure the edible mushroom L. edodes strain FMRI0339 of the L. edodes mycovirus (LeV) in order to obtain an isogenic virus-free fungal strain as well as a virus-infected strain for comparison. Mycelial fragmentation, followed by being spread on a plate with serial dilutions resulted in a virus-free colony. Viral absence was confirmed with gel electrophoresis after dsRNA-specific virus purification, Northern blot analysis, and PCR using reverse transcriptase (RT-PCR). Once cured, all of fungal cultures remained virus-free over the next two years. Interestingly, the viral titer of LeV varied depending on the culture condition. The titer from the plate culture showed at least a 20-fold higher concentration than that grown in the liquid culture. However, the reduced virus titer in the liquid culture was recovered by transferring the mycelia to a plate containing the same medium. In addition, oxygen-depleted culture conditions resulted in a significant decrease of viral concentration, but not to the extent seen in the submerged liquid culture. Although no $discernable phenotypic changes in colony morphology were observed, virus-cured strains showed significantly higher growth rates and mycelial mass than virus-infected strains. We were also explored effects of LeV on fruiting body formation and mushroom yield. The fruiting body formation yield of virus-free L. edodes was larger than virus-infected L. edodes. These results indicate that LeV infection has a deleterious effect on mycelial growth and fruiting body formation. In addition, we have been investigated host-parasite interaction between L. edodes and its mycovirus interaction to study viral mechanism by establishment of proteomics.

Comparison of methods for the proportion of true null hypotheses in microarray studies

  • We consider estimating the proportion of true null hypotheses in multiple testing problems. A traditional multiple testing rate, family-wise error rate is too conservative and old to control type I error in multiple testing setups; however, false discovery rate (FDR) has received significant attention in many research areas such as GWAS data, FMRI data, and signal processing. Identify differentially expressed genes in microarray studies involves estimating the proportion of true null hypotheses in FDR procedures. However, we need to account for unknown dependence structures among genes in microarray data in order to estimate the proportion of true null hypothesis since the genuine dependence structure of microarray data is unknown. We compare various procedures in simulation data and real microarray data. We consider a hidden Markov model for simulated data with dependency. Cai procedure (2007) and a sliding linear model procedure (2011) have a relatively smaller bias and standard errors, being more proper for estimating the proportion of true null hypotheses in simulated data under various setups. Real data analysis shows that 5 estimation procedures among 9 procedures have almost similar values of the estimated proportion of true null hypotheses in microarray data.

The Frequency Effect in the Somatosensory Cortex Response to Vibrotactile Stimulator in fMRI (기능적 자기공명영상에서 진동자의 자극 주파수가 감각피질의 반응에 미치는 영향)

  • The fMRI response of the somatosensory cortex was investigated with vibrotactile stimulation. Three different frequencies of 8, 15, or 25 Hz were applied in order to mainly focus on the hemodynamic response of Meissner corpuscles sensitive to frequencies of 5-40 Hz. A closed-system, pneumatically-driven, rubber diaphragm was fabricated that overcame many of the limitations of existing vibrotactile devices and produced robust sensory cortex activation in an fMRI experiment. Increasing frequency vs. activation area was analyzed in terms of signal percent change and number of pixels. Our preliminary results indicated that the distribution of the signal percentage change widened and more activated pixels were obtained with higher frequencies.

  • For better predictions and classifications in customer recommendation, this study proposes an integrative model that efficiently combines the currently-in-use statistical and artificial intelligence models. In particular, by integrating the models such as Association Rule, Frequency Matrix, and Rule Induction, this study suggests an integrative prediction model. Integrated models consist of four models: ASFM model which combines Association Rule(A) and Frequency Matrix(B), ASRI model which combines Association Rule(A) and Rule Induction(C), FMRI model which combines Frequency Matrix(B) and Rule Induction(C), and ASFMRI model which combines Association Rule(A), Frequency Matrix(B), and Rule Induction(C). The data set for the tests is collected from a convenience store G, which is the number one in its brand in S. Korea. This data set contains sales information on customer transactions from September 1, 2005 to December 7, 2005. About 1,000 transactions are selected for a specific item. Using this data set. it suggests an integrated model predicting whether a customer buys or not buys a specific product for target marketing strategy. The performance of integrated model is compared with that of other models. The results from the experiments show that the performance of integrated model is superior to that of all other models such as Association Rule, Frequency Matrix, and Rule Induction.

Reorganization of Motor Network and the Effect of Cross Education Derived From Unilateral Coordination Training (편측 협응훈련에 의한 운동신경망의 재조직 및 교차훈련의 효과 -사례연구-)

  • We report the reorganization of motor network resulted from intensive unilateral coordination training and the effect of cross education on the untrained side in patient with traumatic brain injury using functional magnetic resonance imaging (fMRI). A 22 year-old male patient who had suffered from diffuse axonal injury for 58 months showed coordination deficit in the left hand at initial examination. Intensive motor training including complex finger movements and coordination activities using a metronome was introduced to the patient 4 hours per day for a week. FMRI was performed on a 3T ISOL Forte scanner. All functional images were analyzed using SPM-99 software. Hand function was improved after training not only in the trained left hand, but also in the untrained right hand. There was no activation in the right primary motor area (M1) during left hand movement before training whereas robust activation of left M1 was demonstrated by the right hand movement. Profuse activation of bilateral prefrontal lobes was seen during both hand movements before training. After training of left hand, right M1 became prominently activated during the left hand motion. The activation of bilateral prefrontal lobes disappeared after training not only for the left hand movement but also for the right, which clearly demonstrated the effect of cross education. This case report demonstrated the learning-dependent reorganization of the M1 and the effect of cross education.
