Analysis of Residual Fluoroquinolones Contents in Fish (어류 중 Fluoroquinolone계 항균제의 분석 및 잔류량 조사)

  • The residual contents of fluoroquinolones in fish and shrimp were analyzed by using HPLC-FLD and LC-MS. The limit of detection (LOD) and limit of quantification (LOQ) for fluoroquinolones by HPLC-FLD were 0.0030-0.0083 and 0.0090-0.0252 mg/kg, respectively. Those by LC-MS were 0.0019-0.0040 and 0.0059-0.0122 mg/kg, respectively. Recoveries regarding ofloxacin, norfloxacin, pefloxacin, ciprofloxacin and enrofloxacin ranged from 76.4-104.6%. The detection rate of fluoroquinolones was 7.5% from a total of 268 samples. In only one Korean bullhead sample, the sum of ciprofloxacin and enrofloxacin contents was detected at a higher level than its maximum residue limits (MRL) of 0.1 mg/kg. Ofloxacin, norfloxacin and pefloxacin were not detected in any samples and detection rates of ciprofloxacin and enrofloxacin were 3.4% and 6.7%, respectively. In detection rate by species eel (2.6%), loach (2.2%), Korean bullhead (1.5%), carp (0.7%) and snakehead (0.4%) were in order.

A Survey of Zearalenone in Beans Using High Performance Liquid Chromatography-Fluorescence Detector (HPLC-FLD) and Ultra Performance Liquid Chromatography Tandem Mass Spectrometry (LC-MS/MS) (HPLC-FLD 및 LC-MS/MS에 의한 두류 중 제랄레논 오염실태 조사)

  • A survey for zearalenone contamination was conducted on 27 soy bean samples, 27 red bean samples, 16 black bean samples, 19 seoritae samples, 14 seomoktae samples, for a total of 127 commercial Korean samples. Zearalenone was quantified by the immunoaffinity column clean-up method with high performance liquid chromatography-fluorescence detection (HPLC-FLD), and was confirmed by liquid chromatography tandem mass spectrometry(LC-MS/MS). The limits of detection and quantification were $2.0{\mu}g/kg$ and $6.0{\mu}g/kg$, respectively. The recovery in the beans ranged from 82.2 to 98.4%. According to HPLC-FLD, zearalenone was detected in 13 samples (10.2% incidence), including 1 soybean and 12 red bean samples. The zearalenone contamination levels were in the range of 8.01${\sim}38.98{\mu}g/kg$. Finally, LC-MS/MS analysis was conducted in the contaminated samples to verify the results of HPLC-FLD. The LC-MS/MS results confirmed the presence of zearalenone in all 13 samples. The contamination level was lower than that of EU, which is below $100{\mu}g/kg$ for raw grains.

  • A survey for total aflatoxins (aflatoxins $B_1$, $B_2$, $G_1$, and $G_2$) was conducted on 245 cereals and processed cereal products, and 148 nuts and processed nut products in Korea, for a total of 393 commercialized ed samples. The total aflatoxins were quantified by the immunoaffinity column clean-up method with high performance liquid chromatography (HPLC) - fluorescence detection (FLD), and were confirmed by liquid chromatography tandem mass spectrometry (LC-MS/MS). Total aflatoxins(AFs) were detected in 37 samples (9.4% incidence), including 2 millet samples, 1 mixed cereal (sunsik), 1 powdered malt sample, 2 processed cereal products, 6 peanut samples, 22 peanut butter samples, and 1 sample each of almonds, adlay tea, and a processed nut product. The contamination levels were $0.04-2.65{\mu}g/kg$ for aflatoxin $B_1$, and $0.04-5.51{\mu}g/kg$ for total aflatoxins. Finally, LC-MS/MS analysis of the contaminated samples was conducted to confirm the detected aflatoxins, and all 37 samples showing aflatoxins by HPLC-FLD were confirmed by LC-MS/MS.

Prediction of the Forming Limit Diagram for AZ31B Sheet at Elevated Temperatures Considering the Strain-rate Effect - II (변형률속도 효과를 고려한 AZ31B 판재의 온간 성형한계도 예측 - II)

  • The purpose of this study is to predict the forming limit diagram (FLD) of strain-rate sensitive materials on the basis of the Marciniak and Kuczynski (M-K) theory. The strain-rate effect is taken into consideration in such a way that the stress-strain curves for various strain-rates are inputted into the formulation as point data, not as curve-fitted models such as power function. Tensile tests and R-value tests were carried out at several levels of temperature and strain-rate from $25^{\circ}C$ to $300^{\circ}C$ and 0.16 to 0.00016/s, respectively to obtain the mechanical properties of AZ31B magnesium alloy sheet. The FLD of this material was experimentally obtained by limit dome height tests with the punch velocity of 0.1 and 1.0 mm/s at $250^{\circ}C$. The M-K theory-based FLD predicted using Yld2000-2d yield criterion was compared with the experimental results.

  • This study aimed at providing an experimental database for the mechanical properties of AZ31B magnesium alloy sheet such as stress-strain curve, yield stress, R-value and forming limit diagram(FLD) at various strain-rates and temperatures. Tensile tests were carried out on specimens having the orientations of $0^{\circ}$, $45^{\circ}$ and $90^{\circ}$ to the rolling direction with different crosshead speeds in the range between 0.008 and 8 mm/s at temperature from 25(room temperature) to $300^{\circ}C$. The influence of the specimen gage length on the tensile properties was investigated. FLD tests were performed at punch speed of 0.1 and 1.0 mm/s in the same temperature range as that of the tensile tests. Swift cup tests were conducted to verify the usefulness of the material database and the reliability of the finite element analysis(FEA). The effects of strain-rate as well as temperature were taken into account in these simulations. It was shown that the FLD-based failure was reasonably well predicted by the thermal-deformation coupled analysis for this rate-sensitive material.

Forming Limit Diagram of Laser Welded Blank and Its Application to Forming Analysis of Stamping Dies (레이저 용접 합체박판의 성형한계도와 스탬핑 금형 성형해석에 적용)

  • The new FLD of the laser welded blank, which includes FLCs of welded zone and base metals, is introduced. For the forming limits of welded zone, the hemispherical dome punch tests were performed with various widths of asymmetric specimen. The FLC0 as well as the dome height at fracture associated with various specimen widths in the same and different thickness combinations were found to see the formability depending on thickness combinations. In order to show the application of the new FLD, the measured strains of squared cup drawing and simulated strains of door inner panel stamping were compared with those of FLCs. The successful prediction of fracture in the applications reveals that the forming limits of welded zone and base metals should be separately found for more accurate evaluation of the formability and workability of the laser welded blank.

Analysis of Neurotoxins, Anatoxin-a, Saxitoxin in Algae Cultured and Algae in Dam Reservoir and its Water Treatment (배양조류 및 댐 저수지 조체중 신경독소 Anatoxin-a, Saxitoxin류의 분석 및 수처리방안)

  • In this study we developed the analytical methods for the determination of three neurotoxin; anatoxin-a, saxitoxin and neosaxitoxin using HPLC/FLD system and this analytical methods were applied to real sample; algae culture and algae extracts. For the HPLC/FLD analysis of anatoxin-a samples were concentrated on WCX(Weak Cation Exchanger) SPE and then anatoxin-a in concentrate was derivatized with NBD-F solution. Supernatant was injected on HPLC system. For the HPLC/FLD analysis of saxitoxin and neosaxitoxin samples were separated on the column and then derivatizied by post column reactor for fluorescen detection. For post column reaction of saxitoxin we feed two kinds of reaction solution; Oxidizing Reagent of which composition was periodic acid(7mM) in 50mM potassium phosphate buffer, pH 9 and acidifying reagent of which Composition was 0.5M acetic acid. The LOD value for anatoxin-a, saxitoxin and neosaxitoxin in HPLC/FLD method was 24.3 ng. $35{\mu}g/L$, $27{\mu}g/L$ respectively. We determined the anatoxin-a content of lyophilized anabaena flos-aquae and $20{\mu}g/g$ d.w. of anatoxin-a was detected. We analyzed saxitoxin and neosaxitoxin in algae culture media and extracts of lypopyllized algal cell cultured and that of Deachung reservior. Saxitoxin and neosaxitoxin in real sample were below the limit of detection. Although there are various water treatment processes for removing neurotoxins were suggested no process give simultaneous and complete removal of neurotoxins. It was cocluded that nanofiltration which reject material by size can be a process for removal of neurotoxins.

Block Shear Behavior of Cold-Formed Duplex Stainless Steel (STS329FLD) Welded Connection with Base Metal Fracture (냉간성형 듀플렉스계 스테인리스강(STS329FLD) 용접접합부 모재 블록전단파단 거동)

  • Recently, lean duplex stainless steel, STS329FLD with less nickel (reduced to 0.5~1.5%) has been developed as a substitute of austenitic stainless steel (8%~10.5% nickel in STS304) and included in Korean standards (KS). This paper investigates the block shear behavior of cold-formed duplex stainless steel (STS329FLD, nominal plate thickness of 1.5mm) fillet-welded connection with base metal fracture. Main variables are weld lengths in the longitudinal and the transverse directions of applied force ranged from 20mm to 50mm. As a result, specimens failed by typical block shear facture (the combination of gross section tensile fracture and shear fracture or shear yielding) and ultimate strength of the specimens got higher with the increase of weld length. Block shear fracture strengths predicted by current design specifications (KBC2016/AISC2016 and EC3) and existing proposed equations for welded connections by Topkaya, Oosterhof & Driver and Lee et al. were compared with test strengths. KBC2016/AISC2016 and EC3 design specifications underestimated block shear strength of STS329FLD welded connections by on average 24%, 29%, respectively and Oosterhof & Driver, Topkaya and Lee et al's equations overestimated the ultimate strength of the welded connection by the range of 3% to 44%.

Determination of phenol using solid-phase extraction and HPLC/MSD/FLD in water (고체상추출법과 HPLC/MSD/FLD를 이용한 수질중의 페놀 분석)

  • An analytical method for determining phenol considered priority pollutants of the US EPA and precursor of toxic phenolic compounds by solid-phase extraction (SPE) and high performance liquid chromatographic systems (HPLC) equipped with fluorescence and mass selective detectors have been developed. The SPE process for sample preconcentration was performed on a commercially available Oasis HLB cartridge packed with polymeric sorbents. The effect of pH, elution solvent, and elution volume on the recoveries of the analytes were investigated with HPLC/FLD. Average recovery of >87.0% was achieved with 60 mg sorbents using 5 mL of methanol as an elution solvent at pH=3.

Simultaneous Determination of Synephrine and N-Methyltyramine in Orange Fruit and Juice from Korean Market by UPLC-FLD (UPLC-FLD를 이용한 국내 시판 오렌지 및 오렌지 주스 중 시네프린 및 n-메틸티라민 동시분석)

  • An accurate and sensitive analytical method was established for the simultaneous determination of synephrine and n-methyltyramine contents by ultra-performance liquid chromatography (UPLC) with a fluorescence detector (FLD). A 70:30 (v/v) mixture of 10 mM sodium dodecyl sulfate (SDS) and acetonitrile was used as the mobile phase. The coefficient of correlation ($r^2$) was 0.9999 for both synephrine and n-methyltyramine, and their limits of detection (LOD) were 0.02 and 0.01 mg/kg, respectively. The percentage recoveries for synephrine and n-methyltyramine were 96.4% and 100.9%, respectively, from bitter orange (Citrus aurantium) samples. The synephrine and n-methyltyramine contents were 38.07-118.21 mg/kg and 0.27-0.56 mg/kg, respectively, in the orange fruit samples, while they were 14.61-120.39 mg/kg and up to 3.34 mg/kg, respectively, in the tested commercial orange juice samples. The differences in synephrine and n-methyltyramine content between orange fruit and commercial orange juice were not significant (p<0.05). These results suggest that UPLC-FLD can be applied to develop an analytical method of quality control for commercial orange juice.
