  • For the chemical assessment of soybean sprouts as a protein food, the changes of amino acid compositions were investigated in soybean as well as both in cotyledon and axis at various growing stages. The total amino acid content per soybean sprout was not changed until 2 days, and thereafter decreased. In cotyledon it is decreased but increased in axis. The ratio of essential amino acid to total amino acids decreased rapidly after 4 days, especially in axis. Aspartic acid increased considerably while glutamic acid decreased. The assessment of soybean sprout with chemical score, A/E and A/T, methods showed that the values were low and that there was no consistency among two methods for sprout products. The limiting amino acid was methionine. Both chemical scores by FAO reference protein and requirement pattern showed similar decreasing order of 2 day-sprout>soybean>4 day-sprout>8 day-sprout>6 day-sprout, while both essential amino acid index and requirement index showed 2 day-sprout>soybeans>4 day-sprout> 6 day-sprout>8 day-sprout.

Trace Metal Contents in Fishes and Shellfishes and Their Safety Evaluatins (우리나라 어패류중 미량금속 함량 및 안전성 평가)

  • This study was carried out to estimate the contents of trace metals in the fishes and shellfishes available on Korean markets. The maples were digested with acids, then analyzed by ICP (INductively Coupled Plasma Spectrometer) and AAS (Atomic Absorption Spectrometer) for the contents of lead (Pb), cadmium (Cd), arsenic (As), copper (Cu), manganese (Mn) and zinc (Zn). The contents of mercury (Hg) were determined using a mercury analyzer. The contents of trace metals [minimum~ maximum (mean), mg/kg] in fishes were as follows; Hg;0.004~0.500(0.082), Pb;N.D.~ 1.87 (0.29), Cd;N.D. ~0.094 (0.020), As;0.01~5045 (0.84), Cu;0.04~3.74 (0.66), Mn; N.D. ~7.56 (0.57), Zn;0.92~18.33(6.03). Those contents in shellfishes were as follows (mg/kg); Hg;N.D.~0.221 (0.029), Pb;0.01~1.51 (0.38), Cd;0.02~1.93 (0.51), As;0.18~3.07 study showed that trace metal contents in both fishes and shellfishes on our markets of lead, cadmium and mercury from fishes and shellfishes takes about 1~13% of PTWI(Provisional Tolerable Weekly Intakes) that FAO/WHO Joint Food Additive and Contaminants Committee has set to evaluate their safeties.

Trace Metal Contents in Tea Products and Their Safety Evaluations (우리나라 다류의 중금속 함량 및 안전성 평가)

  • This study was conducted to estimate the contents of heavy metals in tea products (n=200) and to provide a scientific basis for standardization of heavy metals in the Korea Food Code. The contents of mercury (Hg), lead (Pb), cadmium (Cd), arsenic (As), copper (Cu), manganese (Mn), zinc (Zn), and aluminium (Al) were determined using a mercury analyzer, an AAS (atomic absorption spectrophotometer), and ICP (inductively coupled plasma spectrometer). The values of heavy metals in tea products were [minimum-maximum (mean), mg/kg] Hg: $N.D.{\sim}0.02$ (0.003), Pb: $N.D.{\sim}0.35$ (0.03), Cd: $N.D.{\sim}0.07$ (0.007), As: $N.D.{\sim}0.19$ (0.006), Cu: $N.D.{\sim}24.41$ (2.73), Mn: $N.D.{\sim}767.88$ (66.91), Zn: $N.D.{\sim}18.60$ (4.54), Al: $N.D.{\sim}1304.30$ (85.53). These results showed that metal contents in tea products in our markets were similar to those reported in other countries. The average weekly intakes of lead, cadmium, and mercury from tea products were $0.1{\sim}0.2%$ of PTWI (Provisional Tolerable Weekly Intake) that the JECFA (Joint FAO/WHO Expert Committee on Food Additives) has set to evaluate their safeties.

Hazardous Heavy Metal Contents of Mushrooms from Retail Markets in Seoul (서울 지역에서 유통되는 버섯류의 유해중금속 함량 조사에 관한 연구)

  • This study determined the hazardous heavy metal content of mushrooms from markets in Seoul, Korea. One hundred and fifty mushroom samples were digested by the microwave method then the hazardous heavy metal (Pb, Cd, As, Cr, Ni and Hg) content was determined by the inductively coupled plasma-mass spectrometry (ICP-MS) and mercury analyzer. The average values of heavy metals in the samples were as follows: (mean [minimum-maximum], ${\mu}g/kg$); Pb 7.8 (1.9-30.0), Cd 45.3 (0.7-292.4), As 54.6 (24.2-229.3), Cr 42.6 (8.4-334.3), Ni 22.4 (up to 180.4), and Hg 8.9 (2.0-25.0). The contents of hazardous heavy metal in mushrooms were lower than those reported previously and also lower than the Provisional Tolerable Weekly Intake (PTWI) limit prescribed by the FAO/WHO. Thus, it could be suggested that the hazardous heavy metals levels in the mushrooms of retail market were adequately safe for consumption.

Assessment of Energy Intake and Energy Expenditure in Obese Elementary Students (비만 초등학생의 에너지 섭취량 및 에너지 소비량 평가)

  • The purpose of this study was to evaluate the energy intake and energy expenditure of obese children. The body weight and height were measured, and the daily energy intake and activity pattern assessed for one day, using the 24 hour recall method and interviews in 83 elementary school children (59 boys and 24 girls). The average age, height, weight, ideal body weight and obesity index in boys and girls were 11.8 years, 144.8 cm, 54.2 kg, 39.9 kg and 35.9% and 11.5 years, 140.8 cm, 48.9 kg, 36.9 kg and 33.7%, respectively. The mean daily energy intakes of boys and girls were 1,670.5 and 1,656.5 kcal; these being 76.4 and 88.5% of estimated energy requirement for Koreans, respectively. The energy intake ratio from carbohydrates, proteins and fats in boys and girls were 61.5:14.7:23.6 and 62.0:14.7:23.7, respectively. The resting energy expenditure, as estimated by the Harris-Benedict, WHO/NAO/FAO and Korean DRI formulae in boys and girls were 1,452.7, 1,239.2 and 1,629.2 kcal and 1,323.9, 1,156.0 and 1,252.4 kcal, respectively. The activity coefficient for both the boys and girls was 1.39. The daily energy expenditure of boys (1,990.7 kcal) was significantly higher than that of girls (1712.1 kcal). No significant correlations were found between the obesity index, energy intake and energy expenditure. These results suggested that a nutrition and exercise program is required to increase the energy expenditure of elementary school children. Further research will be needed to assess multiple dietary factors and dove]lop a standard method for assessing the energy expenditure of obese children.

A Study of Germination Characteristics of Native Plants to be Utilized in DMZ Barren Land (불모지 내 활용 가능한 자생식물의 발아특성 연구)

  • This study suggested suitable soil textures that is proper to propagate native plants to manage and restore barren land in DMZ. Germination tests were conducted for 16 native herbaceous plants growing in the DMZ border area in accordance with FAO-BI (Biodiversity International) standards, and the germination rate and T50 in vitro were investigated. In order to examine the germination characteristics according to the soil textures, we used gravel, bed and mixed soil and investigated the germination characteristics under ordinary room temperature conditions in the greenhouse. As a result, it was observed that the germination rate in the greenhouse was significantly decreased compared to the germination rate in vitro of the species advertised due to soil textures. T50 between the in vitro and each soil texture showed significant differences whereas T50 between soil textures alone did not in all species advertised. The germination rate in vitro of Aster koraiensis, Dendranthema zawadskii var. latilobum, Hosta clausa, and Hosta minor there was no significant difference compared to ordinary room temperature conditions. In addition, as the germination rate is demonstrated more than 70%, which is relatively higher than other species advertised, it is considered to have strong environmentally adaptable. On the other hand, considering that the 6 species of Leontopodium coreanum, Plantago major, Potentilla chinensis, Sedum kamtschaticum, Sedum latiovalifolium, and Veronica kiusiana demonstrated less than 50% of germination rate in vitro, it is expected to be difficult to propagate without pre-treatment. In order to use these 6 species as restoration material plants, it needs to be considered to pre-treat to improve germination rate, or to enhance the vitality of seeds by improving the seed gathering period and storage method.

The Composition of Candida guilliermondii Grown in Soybean Whey (Soybean Whey에 배양(培養)한 Candida guilliermondii의 균체조성(菌體組成))

  • A strain of yeast (Candida guilliermondii KFCC 35120) was cultivated by using the soybean whey with supplement of blackstrap molasses (4%) and ammonium acetate (0.5%). By the cultivation continued for 48 hours, 8.1g of dry cell was produced. The cell was consisted of carbohydrate 39.3%, protein 40.5%, lipid 3.9%, nucleic acid 9% and ashes 6%. The protein was analyzed to contain 18% of glutamic acid and 10% of lysine. Other amino acids showed a content near that of FAO reference protein except Methionine. Total lipid content was varied by nutritional condition especially by the kind of carbon source, however it showed a content 1.7 to 2.5 times higher than that of Saccaromyccs cerevisiae. The lipid contain 48% of oleic acid, 17% linoleic acid and 4% linolenic acid. The content of linoleic and lonolenic acid was higher than that of S.cerevisiae by 9 and 24 times, respectively.

  • Gentamicin은 임상에서 많이 사용되는 aminoglycoside계 항생물질로서 세균의 세포막 단백질 합성을 억제하여 살균작용을 나타낸다. 최근 Gentamicin이 동물사료에 포함되거나 동물약품으로 많이 사용되어, 이를 복용한 식용가축에서의 잔류 량에 대한 인체유해성이 WHO/FAO 식품첨가물 전문가 협의회에서 논의되고 있다. Gentamicin의 육가공류의 잔류허용량 기준설정을 위한 독성 재평가의 일환으로 in vivo. in vitro 염색체이상시험을 실시하여 다음과 같은 결론을 얻었다. 1. 체외 염색체이상시험에서는 포유동물 배양세포인 chinese hamster lung cell을 배양하여 gentamicin sulfate 및 gentamicin을 최고 처리농도 5mg/$m\ell$부터 세포독성시험을 실시한 결과, 세포독성을 나타내지 않았다. 본 시험에서는 5mg/$m\ell$를 최고농도로 2.5, 1.25mg/$m\ell$의 3농도를 직접법 및 대사활성화법으로 각 농도당 2매의 플레이트씩 슬라이드를 제작, 결과를 판독한 결과, 직접법 및 대사활성화법 모두에서 전 농도 군에서 음성대조군과 같은 정도의 염색체이상을 유발하여 유전독성이 없음을 나타내었다. 2. 체내 염색체 이상시험에서는 ddY마우스를 이용하여 gentamicin sulfate의 LD$_{50}$의 1/2에 해당하는 200mg/kg을 최고농도로 gentamicin 과 gentamicin sulfate를 암수 각각 3마리씩 공비 2의 3농도로 투여한 후, 24시간째 골수세포의 염색체 표본을 제작하여 관찰한 결과, 세포독성 및 염색체 이상을 유발하지 않았다. 또한 동물약품으로 사용되는 치료용량 및 투약방법에 근거하여 10mg/kg 및 5, 2.5mg/kg을 1일 1회씩 4회 투여한 군에서도 암수에 상관없이 전 농도 군에서 염색체이상을 나타내지 않아 유전독성을 나타내지 않음을 관찰하였다.

Trace Metal Contents in Cereals, Pulses and Potatoes and Their Safety Evaluations (우리나라 곡류, 두류 및 서류중 중금속 함량 및 안전성 평가)

  • 국내에서 생산된 곡류 9종 416건, 두류 5종 296건 및 서류 2종 156건에 대해 수은 함량은 Mercury Analyzer로, 납, 카드뮴, 비소, 구리, 망간, 아연 등은 습식분해 후 ICP로 분석하였다. 본 연구 결과, 곡류중 중금속 함량[최소~최대(평균),mg/kg]은 다음과 같았다. 수은 0.0001~0.051(0.007), 납 0.01~0.39(0.13), 카드뮴 0.001~0.098(0.023), 비소 0.01~0.38(0.09), 구리 0.06~11.85(1.73), 망간 0.91~39.15(7.47), 아연 1.35~24.15(9.32) mg/kg으로 나타났다. 또한 두류중 중금속 함량(mg/kg)은 수은 0.0002~0.031(0.005), 납 0.01~0.38(0.12), 카드뮴 0.005~0.098(0.030), 비소 0.01~0.37(0.10), 구리 0.03~6.56(2.44), 망간 0.85~22.97(8.16), 아연 2.40~40.18(11.25) mg/kg이었다 서류중 중금속 함량(mg/kg)은 수은 0.002~0.036(0.017), 비소0.01~0.20(0.08), 구리 0.02~2.91(0.84), 망간 0.26~9.48(2.54), 아연 0.35~6.11(2.23)mg/kg이었다. 본 연구에서 얻어진 분석치들은 국내외 다른 연구자들의 분석치와 비슷한 것으로 나타났으며, 이는 우리나라에서 생산된 곡류, 두류, 서류중의 중금속 함유량은 오염된 것이 아닌 자연함유량 수준인 것으로 파악되어 우리나라 사람이 이들 농산물에서 섭취하는 중금속량으로 인한 위해성은 없는 것으로 판단된다. 또한 우리나라 국민이 곡류, 두류, 서류 등을 통해 섭치하는 납, 수은, 카드뮴 등의 중금속 주간섭취량은 FAO/WHO에서 중금속 안전성 평가를 이해 정한 잠정주간섭취혀용량인 PTW1의 0.2~19%를 차지하고 있다.

Distribution of Nitrate Content in Vegetables for Kimchi Raw Material and Changes of Nitrate Content by Salting of Chinese Cabbage and by Cooking Process of Kimchi (김치 모재료 채소의 질산염 함량 분포와 배추 염장처리 및 김치 조리방법에 따른 질산염 함량 변화)

  • This study aims to find out the devices to minimize the amount of nitrate ingested from kimchi, the main way of vegetable intake for Koreans, and the basic data to deliver to calculate the total daily intake for Korean, investigating the year-round changes of nitrate content in vegetables for Kimche raw material, and changes of nitrate content by salting of chinese cabbage, and stewed kimchi through the use of different cooking methods. The results obtained were summarized as follow: over between 205-6655mg/kg f.w. in chinese cabbage, 480-3970mg/kg f.w. in chinese radish, 157-5820mg/kg f.w. in lettuce and 29-520mg/kg f.w. in cucumber respectively. Therefore it was strongly adviced to introduce the nitrate limit value of vegetables in Korea if the nitrate content in Kimchi should be reduced to meet the nitrate ADI(Acceptable Daily Intake, 219mg60kg b.w) of FAO.WHO, because the nitrate content in Kimchi reflects the nitrate content in vegetables. In order to keep the low nitrate content in Kimchi. it was adviced to remove the outer leaf which contains 2-3 times higher nitrate content compare to the inner leaf at the time of preparation, i.e. chinese cabbage, before the soaking treatment in salt solution for Kimchi making process. The dehydration by soaking treatment in salt solution occurred at 0.9%~4.7% in leaf midrib and more than 13%~24% in leaf blade. The nitrate content after soaking in salt solution was increased 107%~123% compared with before soaking, increasing rates of nitrate content in the outer and inner leaf midrib were higher than those in leaf blade. The increase of nitrate content in salt solution after soaking due to the dehydration of chinese cabbage by soaking treatment. The Kimchi stew(Kimchi JJige) was processed with and without animal oil, but the amount of nitrate in kimchi stew did not decrease both treatments, but it increased after the processing since the water in Kimchi stew has got low by boiling.
