Immunogenicity Study of Recombinant Human Basic Fibroblast Growth Factor

  • The immunogenicity of the recombinant human basic fibroblast growth factor (rh-bFGF) was investigated by tests for active systemic anaphylaxis (ASA), passive cutaneous anaphylaxis (PCA), passive hemagglutination (PHA) and guinea pig maximization test (GPMT) in mice or guinea pigs. Guinea pigs were sensitized with rh-bFGF ($100-1000\;\mu\textrm{g}/kg$) or rh-bFGF-CFA mixture ($1000\;\mu\textrm{g}/kg$). All animals sensitized with rh-bFGF alone or mixture with CFA showed symptoms of anaphylactic shock. IgE antibodies to rh-bFGF were detected in sera obtained from rh-bFGF and rh-bFGF-Alum ($1000\;\mu\textrm{g}/kg$) sensitized mice, indicating that rh-bFGF has immunogenicity eliciting potential. IgG and/or IgM antibodies to rh-bFGF were also detected in all the sera obtained from sensitized mice by PHA. In the GPMT for delayed type skin reaction, no skin reaction was observed in sensitized guinea pigs after intradermal injection and dermal application of 0.01% rh- bFGF. However, these positive reactions were consistent with the results of another rh-bFGF, showing that rh- bFGF is a heterogenous protein to rodents. Considering the fact that rh-bFGF is a genuine human protein of which structure is identical to the endogenous human bFGF, it is thought that rh-bFGF is rarely associated with immunological problems in clinical use.

Carex brevispicula (Cyperaceae), a new species from Korea (좀목포사초, 한국에서 발견된 1신종)

  • A new species, Carex brevispicula G. H. Nam & G. Y. Chung (Cyperaceae), was found in Korea. Carex brevispicula is similar to the related species C. chungii Z. P. Wang and C. genkaiensis Ohwi in that its achenes are constricted in the middle part. However, C. brevispicula is distinguished from C. chungii as the plants, staminate, and pistillate spikes are shorter and its pistillate scales are pale green; C. brevispicula is distinguished from C. genkaiensis by its awned staminate and pistillate scales. The scientific name of this new species was based on the fact that its inflorescence is shorter than that of C. chungii. The corresponding Korean name, "Jom-mok-po-sa-cho," means that the plants of this species are smaller than the "Mok-po-sa-cho" types (C. genkaiensis). We hereby provide a description of C. brevispicula, with corresponding illustrations and photographs, a distribution map, and a key of related taxa.

  • For arbitrary random variables {$X_{n},n{\geq}1$}, the order of growth of the series. $S_{n}\;=\;{\sum}_{j=1}^n\;X_{j}$ is studied in this paper. More specifically, when the series S_{n}$ diverges almost surely, the strong law of large numbers $S_{n}/g_{n}^{-1}$($A_{n}{\psi}(A_{n}))\;{\rightarrow}\;0$ a.s. is constructed by extending the results of Petrov (1973). On the other hand, if the series $S_{n}$ converges almost surely to a random variable S, then the tail series $T_{n}\;=\;S\;-\;S_{n-1}\;=\;{\sum}_{j=n}^{\infty}\;X_{j}$ is a well-defined sequence of random variables and converges to 0 almost surely. For the almost surely convergent series $S_{n}$, a tail series strong law of large numbers $T_{n}/g_{n}^{-1}(B_{n}{\psi}^{\ast}(B_{n}^{-1}))\;{\rightarrow}\;0$ a.s., which generalizes the result of Klesov (1984), is also established by investigating the duality between the limiting behavior of partial sums and that of tail series. In particular, an example is provided showing that the current work can prevail despite the fact that previous tail series strong law of large numbers does not work.

  • Economic development is usually identified with the increased of G.N.P and capital formation connected directly wi:11 the net increase of land, structure, commodity, stocks and foreign claims, etc. The increase of G.N.P. is driving at capital formation. The usual concept of capital formation for economic development misses the important productivity factor of human capital. Because it is now increasingly realized that a massive injection of material into production will 11 not necessarily explain a successful productivity unless that country already possesses 1 suitable human capital. Human capital is built of educational investment which is related to the physical capital. Many statistical investigations identified this reality. Of late years, a great crowd of economists studying an the problem of economic development have turned their attention towards human capital. Thereupon, this paper has dealt with the fact that educational investment affects physical capital and employment. The qualitative increment of human capital will increase the productivity but excessive expenditure on human capital will squander a physical capital. In designing a strategy of human capital for economic development, one needs to consider tile level of educational investment because educational investment of developing countries will be accasionaly made or marred by investment level and direction.

Characterization of a Xylanase Produced by Bacillus sp. N-25 (Bacillus sp. N-25가 생산하는 Xylanase의 특성)

  • To increase the efficency of utilizing cellulosic biomass, a potent xylanase producing bacteria was isolated and identified as Bacillus sp. N-25. Extracellular xylanase from Bacillus sp. N-25 was partially purified by ammonium sulfate precipitation, DEAE-Sephadex A-25 and Sephadex G-IOO column chromatographies. The xylanase was single fraction on chromatography and was true xylanase without cellulase activity. The enzyme was stable at pH 6-8 and 80% activity was remained at $50^{\circ}C$ for 30 min, but was inhibited by $Hg^{2+}$, $Ag^{2+}$, and $Mn^{2+}$. From the fact that the major end product was xylose, we suggested that the enzyme is an exo-xylanase which may be a prime candidate for industrial use.

  • The pseudo relevance feedback suggests that frequent words at the top documents are related to initial query. However, the main drawback associated with the term frequency method is the fact that it relies on feature independence, and disregards any dependencies that may exist between words in the text. In this paper, we propose query expansion based on word graph using term proximity. It supplements term frequency method. On TREC WT10g test collection, experimental results in MAP(Mean Average Precision) show that the proposed method achieved 6.4% improvement over language model.

  • The effects of the five ginsenosides on the activities of particulate adenylate cyclase and particulate guanylate cylase of rat brain have been studied. The range of concentrations of ginsenosides were between 10$\mu\textrm{g}$ and 500$\mu\textrm{g}$ per 500${mu}ell$ reaction mixture, Also, the effects of three ginsenosides on the activity of soluble guanylate cylace have been studied in the same range of concentrations as in particulate adenylate cyclase. Only ginsenoside Re has shown the reciprocal feeects when tested with particulated adenylate cyclase and particulate guanylated cyclase. Regulatory action of the several mononucleotides on the activities of adenylate cyclase and guanylate cyclase was examined. Ginsenoside Rd-inhibited adenylate cyclase was activated in great extent by the addition of increasing amount of GMP. On the other hand, ginsenoside Rc-activated guanylate cyclase was inhibited by the addition of increasing amount of AMP and GMP. The fact that the stimulatory action of GMP is observed only with particulated adenylate cyclase but not with soluble suanylate cyclase suggests that the action is membrane-related one. The competitive action was observed between ginsenoside Rb2 and dopamine in their binding to the receptors. This result is clear-cut evidence that the ginsenoside Rb2 binds specifically to $\beta$-adrenergic receptors.

Effect of Varous Physicochemical Factors on the Biodegradation of Explosive 2,4,6-Trinitrotoluene by Stenotropomonas maltophilia (Stenotrophomonas maltophilia에 의한 폭약 2,4,6-Trinitrotoluene의 생분해에 영향을 미치는 물리화학적 요인)

  • The relationships between the explosive 2,4,6-trinitrotoluene (TNT) degradation by Stenotrophomonas maltophilia and several relevant physicochemical environmental parameters were examined. At neutral pH of the cultures, the degradation of TNT proceeded to completion, whereas only about 50% of TNT was utilized when the cultures were adjusted to acidic pH. The effect of various co-substrates (e.g., glucose, fructose, acetate, citrate, succinate) on the degradation of TNT by the test culture of S. maltophilia was evaluated. The results indicated that, among the various co-substrates studies, the test culture that received 2 mM fructose degraded 100 mg/L of TNT completely within 20 days of incubation at ambient temperature, whereas partial degradation of TNT was observed in the test culture with acetate, citrate, or succinate as a co-substrate, respectively. In fact, fructose was the best co-substrate for TNT degradation in this experiment. The effect of supplemented nitrogens [e.g., (NH$_4$)$_2$,SO$_4$, NH$_4$Cl. urea] on the TNT degradation was monitored. All supplemented nitrogens in this study were inhibitory to TNT degradation. Addition of 1% Tween80 accelerated TNT degradation, and showed complete degradation of TNT within 8 days of incubation. Addition of yeast extract resulted higher growth yields, based on turbidity measurement, but it inhibited TNT degradation.

  • Gambierdiscus toxicus, Adachi et. Fukuyo, is a benthic ciguatoxin-producing armored dinoflagellate, often attached to macroalgae. This organism is the primary causative agent of ciguatera fish poisoning which occurs in tropical and subtropical regions. However, regardless of the fact that the population of Gambierdiscus spp. has expanded to such temperate areas from sub-trophic and trophic areas, monitoring of G. toxicus has been lacking in the Korean coastal waters of temperate areas. This study was performed at the uninhabited Baekdo Islands off the southern coast of Korea and at Seopsom Island in the vicinity of Seogwipo, Jeju Province during April and May, 2011. Cell densities of Gambierdiscus spp. on macroalgae at Baekdo and Jeju Island ranged from zero to 56.4 cells $g^{-1}$. Maximum density was recorded on the brown alga Cladophora japonica at St. 3 of Jeju Island. In particular, the cell densities of Gambierdiscus spp. were influenced by the substrate characteristics of macroalgae. In the future, the continuous monitoring of toxic benthic dinoflagellate is necessary to predict and prevent ciguatera poisoning in Korean coastal waters.

Understanding of Protein Adsorption Kinetics to Contact Lens Hydrogels (콘택트렌즈용 하이드로젤로의 단백질 흡착 반응속도 이해)

  • Protein adsorption kinetics was studied with the amount of proteins adsorbed to contact lens hydrogels over time scales. Hydroxyethylmethacrylate (HEMA) and silicone hydrogels were dipped in protein solutions (albumin or IgG) and adsorption amounts were measured over time scales. The amount of protein adsorbed to both hydrogel types increased rapidly in 10 min, and remained consistently in 90 min. Decreasing interfacial energetics was taken slowly up to an hour in spite of rapid diffusion of protein molecules. This is due to the fact that water deprivation from three dimensional interphase initially formed by protein diffusion took over an hour. Interpretation of adsorption kinetics on contact lens hydrogels was discussed with understanding of relationship between surface energy and protein adsorption capacity.
