Application of Fast Atom Bombardment Collision-induced Dissociation Tandem Mass Spectrometry for Structural identification of Glycerolipids Isolated From Marine Sponge

  • Two types of glycerolipids [monoacylglycerols (MAG) and cyclitols] were isolated by reversed phase high-performance liquid chromatography from the methanol extracts of a marine sponge, and analyzed by fast atom bombardment mass spectrometry (FAB-MS) in positive-ion mode. FAB mass spectra of these compounds yielded protonated molecules $[M + H]^+$ and abundant sodiated molecules $[M + Na]^+$ from a mixture of 3-nitrobenzyl alcohol and NaI. The structures of these compounds were elucidated by FAB-collisional-induced dissociation (CID)-tandem mass spectrometry. We carried out collision-indused dissociation (CID) of these lipids in B/E-linked scan mode. The CID B/E-linked scan of $[M + H]^+$ and $[M + Na]^+$ precursor ions resulted in the formation of numerous characteristic product ions through a series of dissociative processes. The product ions formed by charge-remote fragmentation (CRF) provided important information for the identification of the acyl chain structure substituted at the glycerol backbone. Some of the product the ions were diagnostic for the presence of a glycerol backbone or acyl chain structure.

  • Fallen needle (8.5 kg) of Larix kaempferi were collected and extracted with 95% EtOH. The EtOH extracts were evaporated under reduced pressure, concentrated, and successively fractionated with a series of hexane, methylene chloride, ethylacetate and water on a separatory funnel to be freeze dried. A portion of ethylacetate and water soluble powder were chromatographed on a Sephadex LH-20 column eluting with aqueous MeOH and EtOH-hexane mixture. Spectrometric analyses such as NMR and FAB-MS, including TLC, were performed on the seven isolated compounds and were elucidated as (+)-catechin, (-)-epicatechin, 2"-O-rhamnosylvitexin, juglanin, afzelin, laricitrin-3-O-${\beta}$-D-glucopyranoside, isoquercitrin and cedrusin.

  • An antifungal mixture of six members (component A to F), KRF-001 produced by Bacillzts subtilis subsp. krictiensis was isolated from the fermentation broth. Molecular weight of component A to F was determined by FAB-MS to be 1042, 1056, 1056, 1070, 1070 and 1084 respectively. Various instrumental analyses (amino acid analysis, GC-MS, $^1H-NMR, ^1HH$ COSY NMR) revealed that the mixture was a homologous cyclic peptide composed of each one mole of glutamine, proline, tyrosine, serine, unusual $\beta$-amino acid and three moles of asparagine. The structural differences of component A to F were found in carbon number and terminal structure of the unusual $\beta$-amino acid. After determination of the sequence and stereochemistry of those amino acids, the tentative structure of KRF-001 was determined.

Spectrometric analyses such as NMR and FAB-MS including TLC were performed to characterize the structures of isolated compounds. 5 compounds were isolated from the fallen needles of Larix kaempferi. The antioxidative activities of each fraction and isolated compounds were done by DPPH radical scavenging test.

  • 5,8-Epidioxycholest-6-en-3-ol (1), cholesterol (2), glycerol 1-palmitate (3) and glycerol 1,3-dioleate-2-stearate (4) were isolated from the methanol extract of the sea urchin Diadema setosum, which was collected from the Halong sea, Vietnam. Chemical structures were established based on extensive 1D, 2D-NMR, FAB-MS, EI-MS spectroscopic data and GC-MS analysis. The NMR spectral data of compound 1 were reassigned by using HMQC and HMBC. Compound 1 was found to have strong cytotoxic effect against various cancer cell lines, such as KB ($IC_{50}$, 2.0 $\mu\textrm{g}$/mL), FL($IC_{50}$, 3.93 $\mu\textrm{g}$/mL), and Hep-2 ($IC_{50}$, 2.4 $\mu\textrm{g}$/mL) by in vitro assay.

Anti-cancer Activity of Styrax japonica Bark Extrats (때죽나무(Styrax japonica) 수피 추출물의 항암 활성)

  • A compound has been isolated from the methanol extract of Styrax japonica bark using conventional chromatographic methods including silica gel chromatography, TLC and HPLC. The molecular formula of Styraxlignolide F analyzed by spectrometric analyses using FAB-MS, NMR was found to be $C_{27}H_{34}O_{11}Na$. The cytotoxicity of the styralignolide F was showed 15.2% in $1.0mg/m{\ell}$ on human kidney cell (HEK 293). As anticancer activity of $CH_2Cl_2$ fraction, over 60% of AGS and MCF-7 cells were inhibited in concentration of $1.0mg/m{\ell}$. In the results of anticancer test using quantification of Bcl-2, $CH_2Cl_2$ fraction showed lower Bcl-2 and p53 expression than those of styraxlignolide F and other fractions. In apoptosis of human lung carcunoma cancer cell (A549), $CH_2Cl_2$ fraction showed the highest inhibition rate (46.9%) and styralignolide F was the next (43.5%). The $CH_2Cl_2$ fraction showed higher anti-cancer activities than isolated substance (styraxlignolide F), probably due to the crude extract showing synergic effects by other components.

  • The fresh ginseng roots were extracted in aqueous methanol (MeOH), and the obtained extracts were partitioned using ethyl acetate (EtOA), n-butanol (n-BuOH), and water, successively. The repeated silica gel column chromatography for n-BuOH fraction afforded a purified ginsenoside $Rg_1$. The physico-chemical, spectroscopic and chromatographic data of ginsenoside $Rg_1$, such as crystallization characteristics, melting point, specific rotation, infrared spectrometry (IR) data, fast atom bombardment/mass spectrometry (FAB/MS) data, nuclear magnetic resonance (NMR) data, retention factor (Rf) in thin layer chromatography (TLC) experiment, and retention time (r.t.) in HPLC analysis, were measured and compared with those reported in literatures. Especially, the previous literatures reported different data for ginsenoside $Rg_1$ in the $^{1}H-$ and $^{13}C$-NMR experiments. This paper gives the exactly assigned NMR data through 2D-NMR experiments, such as $^{1}H-^{1}H$ correlation spectroscopy (COSY), hetero nuclear single quantum correlation (HSQC), and hetero nuclear multiple bond connectivity (HMBC).

  • Investigation of the fruits of Chaenomeles sinensis (Rosaceae) resulted in the isolation of a minor flavonoid. The structure of flavanone glycoside was determined to be as $2-hydroxynaringenin-7-O-{\beta}-glucoside$ on the basis of FAB-MS and spectral evidence, especially by 2D-NMR $(^1H-^1H\;COSY,\;HMQC,\;HMBC\;and\;NOESY)$.

  • Three flavan-3,4-diol derivatives were isolated from the heartwood of Robinia pseudoacacia and characterized by spectroscopic methods including $^1H$ and $^{13}C$ NMR and positive FAB-MS. The structures were identified as 4,4'-dimethoxy-, 4-ethoxy- and 4-ethoxy-4'-methoxy-2,3-trans-3,4-cis-(+)-leucorobinetinidin.

  • Specific molecular recognition of cyclodextrin and aspirin was determined. A stable 1:1 inclusion complex was established in solution. The distinct structure of inclusion complex was elucidated by FT-IR, FAB-MS, UV, 1H NMR, and 13C NMR spectroscopy. Based on the 1H NMR data, a time-averaged conformation of $\alpha$-cyclodextrin exhibited significant catalytic activity toward the hydrolysis of aspirin in alkaline solution.
