Regeneration Studies in Grateloupia filicina (J.V. Lamouroux) C. Agardh - An Important Carrageenophyte and Edible Seaweed

  • Grateloupia filicina (J.V. Lamouroux) C. Agardh (Halymeniaceae, Cryptonemiales, Rhodophyta) is an edible seaweed as well as an important source of carrageenan. In the present study, attempt has been made to develop a suitable protocol for effective regeneration of the seaweed and the rapid multiplication of the desired varieties. The young upright thallus of G. filicina was grown in axenic culture using both solid and liquid media. The various media tested were f/2, Provasoli’s Enriched Seawater (PES) and Enriched Seawater (ESW). The effect of glycerol (as a carbon source) and various plant growth regulators i.e. auxin (NAA) and cytokinins (Kinetin and BA) were tested. Although, regeneration of young thalli was observed from the cut ends in all the media, better growth was found in f/2, PES, f/2 (0.5% Glycerol), f/2 (NAA ${10^{-5}}_M)\;and\;f/2\;(BA\;{10^{-6}}_M$). On the other hand callusing was observed only in solid media supplemented with low concentration of Glycerol (0.5%) in f/2, NAA ${10^{-5}}_M\;in\;f/2,\;PES\;and\;BA\;{10^{-5}}_M$ in f/2. Young thalli were developed from the callus sub culture after 40 days of inoculation.

Growth, Photosynthesis and Chlorophyll Fluorescence of Chinese Cabbage in Response to High Temperature (고온 스트레스에 대한 배추의 생장과 광합성 및 엽록소형광 반응)

  • In order to gain insight into the physiological responses of plants to high temperature stress, the effects of temperature on Chinese cabbage (Brassica campestris subsp. napus var. pekinensis cv. Detong) were investigated through analyses of photosynthesis and chlorophyll fluorescence under 3 different temperatures in the temperature gradient tunnel. Growth (leaf length and number of leaves) during the rosette stage was greater at ambient $+4^{\circ}C$ and ambient $+7^{\circ}C$ temperatures than at ambient temperature. Photosynthetic $CO_2$ fixation rates of Chinese cabbage grown under the different temperatures did not differ significantly. However, dark respiration rate was significantly higher in the cabbage that developed under ambient temperature relative to elevated temperature. Furthermore, elevated growth temperature increased transpiration rate and stomatal conductance resulting in an overall decrease of water use efficiency. The chlorophyll a fluorescence transient was also considerably affected by high temperature stress; the fluorescence yield $F_J$, $F_I$, and $F_P$ decreased considerably at ambient $+4^{\circ}C$ and ambient $+7^{\circ}C$ temperatures, with induction of $F_K$ and decrease of $F_V/F_O$. The values of RC/CS, ABS/CS, TRo/CS, and ETo/CS decreased considerably, while DIo/CS increased with increased growth temperature. The symptoms of soft-rot disease were observed in the inner part of the cabbage heads after 7, 9, and/or 10 weeks of cultivation at ambient $+4^{\circ}C$ and ambient $+7^{\circ}C$ temperatures, but not in the cabbage heads growing at ambient temperature. These results show that Chinese cabbage could be negatively affected by high temperature under a future climate change scenario. Therefore, to maintain the high productivity and quality of Chinese cabbage, it may be necessary to develop new high temperature tolerant cultivars or to markedly improve cropping systems. In addition, it would be possible to use the non-invasive fluorescence parameters $F_O$, $F_V/F_M$, and $F_V/F_O$, as well as $F_K$, $M_O$, $S_M$, RC/CS, ETo/CS, $PI_{abs}$, and $SFI_{abs}$ (which were selected in this study), to quantitatively determine the physiological status of plants in response to high temperature stresses.

  • The films with ferromagnetic fine particles dispersed in nonmagnetic matrix, such as $Fe-Al_2O_3$ and Fe-Cu have been studied for use of magnetic recording medium, optically device and sensor. Their magnetic properties depend strongly on structural parameter such as size and volume fraction of ferromagnetic particles. Fe-Cr-Ni alloy sputtered films also have microstructure with ferromagnetic -- b.c.c phase and nonmagnetic f.c.c phase grains. Magnetic properties of these films depend strongly on such a unique structure. These are depend on the ratio in volume of ferromagnetic particles to nonmagnetic ones $V_F/V_N$, the saturation magnetization Ms increased with increase of $V_F/V_N$. The coercivity Hc of the as-deposited films took maximum value of about 200 Oe at adequate $V_F/V_N$ and then Ms and Squareness S were 500 emu/cc and 0.5, respectively.(omitted)

  • In this article we make a local classification of n-dimensional Riemannian manifolds (M, g) with harmonic curvature and less than four Ricci eigenvalues which admit a smooth non constant solution f to the following equation $$(1)\hspace{20}{\nabla}df=f(r-{\frac{R}{n-1}}g)+x{\cdot} r+y(R)g,$$ where ∇ is the Levi-Civita connection of g, r is the Ricci tensor of g, x is a constant and y(R) a function of the scalar curvature R. Indeed, we showed that, in a neighborhood V of each point in some open dense subset of M, either (i) or (ii) below holds; (i) (V, g, f + x) is a static space and isometric to a domain in the Riemannian product of an Einstein manifold N and a static space (W, gW, f + x), where gW is a warped product metric of an interval and an Einstein manifold. (ii) (V, g) is isometric to a domain in the warped product of an interval and an Einstein manifold. For the proof we use eigenvalue analysis based on the Codazzi tensor properties of the Ricci tensor.

  • Given an $m{\in}\mathbb{N}$ and two vector spaces V and W, a function f : $V^m{\rightarrow}W$ is called multi-Jensen if it satisfies Jensen's equation in each variable separately. In this paper we unify these m Jensen equations to obtain a single functional equation for f and prove its stability in the sense of Hyers-Ulam, using the so-called direct method.

Supercapacitive Properties of Carbon-Nano Fiber/MnO2 Composite Electrode (나노탄소섬유/MnO2 복합전극의 초고용량 캐폐시터 특성)

  • In order to improve the specific capacitance of amorphous hydrous manganese oxide ($MnO_2$) for supercapacitors, it is made into composites with vapour-grown carbon nanofibers (VGCF) having the VGCF ratio as 40 wt% in the composites. The electrochemical properties of these composites are investigated in 1.0 M $Na_2SO_4$ by cyclic voltammetry (CV), impedance measurements and chronopotentiometric charger/discharger. The composite with 40 wt% VGCF shows the superior electrochemical performance, whose specific capacitance (based on the mass of $MnO_2$, $0.8mg/cm^2$) is 380 F/g at 20 mV/s and 230 F/g at 500 mV/s. Also, the cycle-life testing of this electrode carried out for 3,000 charge/discharge cycles at $2.0mA/cm^2$ shows 97% capacitance retention.

Overall Prevalence and Distribution of Knockdown Resistance (kdr) Mutations in Aedes aegypti from Mandalay Region, Myanmar

  • Knockdown resistance (kdr) mutations in the voltage-gated sodium channel (VGSC) of mosquitoes confer resistance to insecticides. Although insecticide resistance has been suspected to be widespread in the natural population of Aedes aegypti in Myanmar, only limited information is currently available. The overall prevalence and distribution of kdr mutations was analyzed in Ae. aegypti from Mandalay areas, Myanmar. Sequence analysis of the VGSC in Ae. aegypti from Myanmar revealed amino acid mutations at 13 and 11 positions in domains II and III of VGSC, respectively. High frequencies of S989P (68.6%), V1016G (73.5%), and F1534C (40.1%) were found in domains II and III. T1520I was also found, but the frequency was low (8.1%). The frequency of S989P/V1016G was high (55.0%), and the frequencies of V1016G/F1534C and S989P/V1016G/F1534C were also high at 30.1% and 23.5%, respectively. Novel mutations in domain II (L963Q, M976I, V977A, M994T, L995F, V996M/A, D998N, V999A, N1013D, and F1020S) and domain III (K1514R, Y1523H, V1529A, F1534L, F1537S, V1546A, F1551S, G1581D, and K1584R) were also identified. These results collectively suggest that high frequencies of kdr mutations were identified in Myanmar Ae. aegypti, indicating a high level of insecticide resistance.

Development and Luminescent Characteristics of $CaSiN_2$ Based Phosphors ($CaSiN_2$를 모체로 하는 형광체의 개발 및 발광 특성)

  • The $CaSiN_2:Eu$ and $CaSiN_2:Tb$ phosphors were synthesized and analyzed to develop new nitride compound phosphors. $Ca_3N_2$, $Si_3N_4$ and $EuF_3$(or $TbF_3$) powders were mixed, cold-pressed, and sintered to synthesize $CaSiN_2:Eu$ and $CaSiN_2:Tb$ phosphors. Photoluminescence(PL) and electroluminescence(EL) characteristics of the synthesized phosphors were measured and found to be similar to general emission spectra of 뗘 and Tb ion, respecticely. Threshold voltage($V_{th)$) and luminance of the $CaSiN_2:Eu$ TFEL device fabricated by sputtering were 90 V and 1.62 $cd/m^2$ at 280 V, respectively. The charge-voltage(Q-V) and transferred charge-phosphor field($Q_t-F_p$) characteristics of the TFEL devices were also measured.

Alteration of 4-Aminopyridine-Sensitive, Voltage-Dependent $K^+-Channel$ in Arterial Smooth Muscle Cells of One-Kidney, One-Clip Goldblatt Hypertensive Rats

  • Using the patch-clamp technique, we investigated the alteration of 4-aminopyridine(4-AP)-sensitive, voltage-dependent $K^+$ channel (Kv) in the mesenteric arterial smooth muscle cell (MASMC) of renovascular hypertensive model, one-kidney one-clip Goldblatt hypertensive rat (GBH). To isolate $K_V$ current, internal pipette solution contained 5 mM ATP and 10 mM EGTA. Under these condition, MASMC was depolarized by 4-AP, but charybdotoxin did not affect membrane potential. Membrane potential of hypertensive cell $(-40.3{\pm}3.2\;mV)$ was reduced when compared to that of normotensive cell $(-59.5{\pm}2.8\;mV).$ Outward $K^+$ current of hypertensive cell was significantly reduced when compared to normotensive cell. At 60 mV, the outward currents were $19.10{\pm}1.91$ and $14.06{\pm}1.05$ pA/pF in normotensive cell and hypertensive cell respectively. 4-AP-sensitive $K^+$ current was also smaller in hypertensive cell $(4.28{\pm}0.38\;pA/pF)$ than in normotensive cell $(7.65{\pm}0.52\;pA/pF).$ The values of half activation voltage $(V_{1/2})$ and slope factor (k1) as well as the values of half inactivation voltage $(V_{1/2})$ and slope factor (k1) were virtually similar between GBH and NTR. These results suggest that the decrease of 4-AP-sensitive $K^+$ current contributes to a depolarization of membrane potential, which leads to development of vascular tone in GBH.
