Study on White Ginseng Extract Preparation for Cognition Improvement (인지능 개선 효과 증진을 위한 백삼 추출물 조제 연구)

  • Background: The study was conducted to elucidate the extraction conditions under which white ginseng has cognition-improving efficacy. Methods and Results: Extracts from white ginseng under different solvent and temperature conditions were analyzed for ginsenoside content and inhibitory effect on N-methyl-D-aspartate (NMDA) receptor and acetylcholinesterase. The total ginsenoside contents and amounts of ginsenoside Rb1 plus ginsenoside Rg1 from the 1st extracts (prepared with EtOH/$H_2O$ as solvent) were higher than those from the 2nd extracts (extracted with $H_2O$ after the 1st EtOH/$H_2O$ extraction). The contents in the 1st and 2nd extracts produced at $80^{\circ}C$ were also higher than those obtained at $50^{\circ}C$. Samples from the 1st extraction at $80^{\circ}C$ indicated higher inhibitory activities on NMDA receptors-whose excessive activation is thought to mediate the calcium-dependent neurotoxicity associated with several neurodegenerative diseases-than those from the 2nd extraction. Among the samples prepared at varying temperatures, the extract prepared at $50^{\circ}C$ showed the highest suppression activity on NMDA receptors. Note, however, that the extracts from the 2nd extraction at $50^{\circ}C$ inhibited acetylcholinesterase-whose inhibition could be a therapeutic strategy for neurodegenerative diseases with cognitive deficits and memory malfunction-more effectively than those from the 1st extraction. Conclusions: To enhance the cognition-improving activity of white ginseng extract, it is suggested that the extracts be utilized after being combined the 1st extracts (made with EtOH/$H_2O$ solvent) and the 2nd extracts (prepared with $H_2O$) at low temperature.

Separation and Recovery of Indole from Model Coal Tar Fraction by Batch Cocurrent 5 Stages Equilibrium Extraction (회분 병류 5단 평형추출에 의한 모델 콜타르 유분 중에 함유된 Indole의 분리 및 회수)

  • The separation of indole from a model mixture comprising four kinds of nitrogen heterocyclic compounds [indole (In), quinoline (Q), iso-quinoline (iQ), quinaldine (Qu)], three kinds of bicyclic aromatic compounds [1-methylnaphthalene (1MN), 2-methylnaphthalene (2MN), dimethylnaphthalene (DMN)], biphenyl (Bp) and phenyl ether (Pe) was examined by batch cocurrent 4 stages equilibrium extraction. The model mixture used as a raw material in this work was prepared according to the components and compositions contained in coal tar fraction (the temperature ranges of fraction: $240{\sim}265^{\circ}C$). An aqueous solution of formamide was used as a solvent. Indole was recovered more than 99% through 4 stages of the equilibrium extraction. The range of selectivity of indole in reference to DMN obtained through the 5 stages equilibrium extraction was found to be 63~118. The process for separation and recovery of indole contained in coal tar was studied by using the experimental results obtained from this work and the previous work.

Change in Functional Properties by Extraction Condition of Roasted Pleurotus eryngii (볶음 처리 새송이버섯의 추출조건에 따른 생리기능적 특성 변화)

  • This study was performed to investigate the functional properties of roasted Pleurotus eryngii by the extraction conditions. Total phenolic compound content and electron donating ability (EDA) were high at 50% ethanol concentration. Superoxide dismutase (SOD)-like activity was high at 75% ethanol concentration. Nitrite scavenging ability increased as ethanol concentration in extracting solvent decreased. EDA decreased, SOD-like activity and nitrite scavenging ability increased as the extraction time increased. With the increase in extraction temperature, EDA and SOD-like activity decreased. But extraction temperature did not significantly affect the nitrite-scavenging ability With the increase in ratio of sample content to solvent, EDA, SOD-like activity and nitrite scavenging ability decreased. The results would be useful for understanding the extraction condition of roasted Pleurotus eryngii.

  • The extraction methods of PBBs from environmental samples by soxhlet extraction for solid phase and liquid-liquid extraction for liquid phase were compared. After extraction, silicagel and florisil as column clean up methods were used for the comparison of cleanup efficiency with different solvent system. Also, the analytical instruments were used GC/MSD. The 22 kinds of PBBs standards were used to establish the analytical methods to perform the experiments of recoveries and detection limits in water and soil. The detection limits of PBBs represented 5~10 ng/L for water and 0.5~3.5 ng/g for soil samples, respectively. The recoveries of water samples represented 96~107% in n-hexane, dichloromethane and toluene for water, and 60~80% in combined solvent with n-hexane and acetone. The recoveries of soil sample were surveyed 67~97% in soxhlet extraction and 64~76% in ultrasonic extraction with selected solvents.

  • We evaluated the effects of temperature, time, solvent type, and solvent concentration on the phenolic content and antioxidant activity of rosemary extracts. The antioxidant capacity of rosemary extracts was demonstrated by their ability to scavenge stable free radicals, and the phenolic content of the extracts was determined using the Folin Ciocalteu method. The highest values were obtained by water-based extraction at $90^{\circ}C$ for 30 min, a 75% methanol solution at $60^{\circ}C$ for 30 min, and a 50% ethanol solution at $70^{\circ}C$ for 10 min. The aqueous solution of 75% methanol, extraction temperature of $60^{\circ}C$, and extraction time of 30 min were the most efficient parameters for the extraction of polyphenols from dry rosemary.

  • Extraction behaviour of silver from nitric acid solutions by Cyanex 301 as an extractant was examined. Detailed studies have been conducted to evaluate the effect of different variables influencing the extraction such as concentration of nitric acid and extractant, phase ratio(O/A) and synergism of mixed extractant. The extraction behaviour of associated metals namely Ca, Al, Fe, Zn and Sr has been investigated. The extraction results show that Ag can be extracted effectively by Cyanex 301 and selectively extracted from 3.0M $HNO_3$ using 5% Cyanex 301. Impurity metals loaded in organic phase can be effective scrubbed by 4.0M HCl. Finally, pure silver solution can be obtained efficiently by thiourea as a stripping reagent.

Study on Antioxidant Activities of Pleurotus ostreatus (Jacq.) Que Pleurotus eryngii and Flammulina velutipes Extracts by Different Solvent (느타리, 새송이, 팽이버섯의 용매추출에 따른 항산화활성 연구)

  • This study was conducted to find out the optimal solvent extraction method [Distilled water (DW), 70% ethanol, 99% ethanol] of mushrooms, including Pleurotus ostreatus (Jacq.) Que, Pleurotus eryngii and Flammulina velutipes and improve their usability as natural antioxidants. To analyze antioxidant activities in each mushroom, total polyphenol, flavonoid contents, 2,2-Diphenyl-1-picrylhydrazyl (DPPH), 2,2'-azinobis(3-ethylbenzothiazoline-6-sulfonic acid) cation radical (ABTS+) and fluorescence recovery after photobleaching (FRAP) were measured. All mushrooms showed the highest total polyphenol contents in DW mushroom extract (p<0.001). Total flavonoid contents were the highest in P. eryngii and F. velutipes DW and 70% ethanol mushroom extracts (p<0.05). All mushrooms showed the highest activities using DPPH and FRAP assays in the DW extraction method (p<0.001). P. ostreatus (Jacq.) Que and P. eryngii showed the highest ABTS+ radical scavenging activity in the DW extraction method, and F. velutipes showed the highest activity in the 70% ethanol extraction method (p<0.001). As a result of comparing IC50 values of DPPH and ABTS+ radicals and FRAP EC50 values, the DW P. ostreatus (Jacq.) Que extract showed high antioxidant activities (p<0.001). Pearson's correlation between total polyphenol contents and antioxidant activities showed a positive correlation in all mushrooms (p<0.01). Therefore, extraction of the mushrooms with DW can enhance the extraction of effective bioactive substances and antioxidant activity.

Antioxidant Effects of Hermetia illucens Larvae Extracts Using Different Extraction Temperatures and Solvents (추출 온도 및 용매에 따른 아메리카동애등에(Hermetia illucens) 유충 추출물의 항산화 효과)

  • This study investigated the antioxidant effect of Hermetia illucens larvae using different extraction temperatures and solvents. We found significant differences in total phenolic content (TPC), total flavonoid content (TFC), an in three antioxidant indexes 2,2-diphenyl-1-picrylhydrazyl (DPPH), 1,1'-azino-bis-3-ethylbenzothiazoline-6-sulphonic acid (ABTS), and ferric-reducing antioxidant power (FRAP) contents, among the samples depending on the extraction temperature and solvent used. The sample extracted with water at 45℃ (HIW-45℃) showed the highest TPC, DPPH, ABTS, and FRAP contents, while the sample extracted with water at 90℃ (HIW-90℃) showed the highest TFC. These differences can be due to the different chemical structures of the extracted components. Based on these results, HIW-45℃ was the optimal extraction method for Hermetia illucens. We intend to further investigate the availability of functional materials for Hermetia illucens using this method.

Comparative Characterization of AFC Precipitated Using Vacuum Drying, Dilution Precipitation and Spray Drying (감압건조, 희석침전, 분무건조 방식으로 제조된 무회분석탄의 특성)

  • Solid ash-free coal (AFC) samples recovered from solvent-extracted solution by vacuum drying, dilution precipitation and spray drying methods were compared in terms of physical properties and chemical structure. AFC was prepared by using Kideco coal (Indonesian sub-bituminous coal) and polar N-methyl-2-pyrrolidone (NMP) solvent as raw materials. The physical properties of the AFCs were characterized with proximate, ultimate, and calorific value analysis. In analyzing the chemical structure, FTIR and NMR were used. the proximate analysis showed much reduced ash in the AFCs compared to parent raw coal. The FTIR result showed that the extraction solvent was not fully removed from the AFC prepared by vacuum drying. However, the solvent was not detected in the AFC recovered by using dilution precipitation. Dilution precipitation has advantages over the other two methods, since it can be done at relatively low temperature and separate ash-free coal from extraction solvent more effectively.

Comparison Studies between Conventional Hot Water and Cellulase Extraction for Safflower Dyestuff (홍화색소의 일반추출과 셀룰라아제추출의 비교연구)

  • Natural red and yellow dyestuff was extracted from safflower (Carthamus tinctorius Linnaeus) by a new process of cellulase extraction compared with the conventional hot water extraction. Dyestuffs were extracted from safflower easily and repeatedly by means of cellulose as safflower cell wall destroyer. It means that new dyestuff extraction by cellulase improves not only yields of dyestuff from safflower successfully but also the rate of repetition of extraction. From the above experiments, the conclusions of this study were summarized as follows. 1. The optimum conditions of dyestuff extraction from safflower by general extraction method were that the solvent was the water of pH 6.0 on yellow dyestuff and 3% $K_2CO_3$ solution on red dyestuff, extraction temperature was $55^{\circ}C$, and extraction time was 30 min. 2. Among various cellulase, the NOVO cellulase was the best cell wall destroyer of safflower and finally produced the largest amount of dyestuff from safflower by cellulase extraction method. 3. The optimum conditions of dyestuff extraction by cellulase extraction method were conducted on 10 unit of cellulase per gram of safflower at $100m{\ell}$ water of pH 5.0 at $50^{\circ}C$ for 30 min.
