Anti-Oxidative and Anti-inflammatory Effect of Combined Extract and Individual Extract of GamiSaengmaeksan (가미생맥산(加味生脈散) 및 개별약재의 항산화 및 항염증 효능에 대한 비교 연구)

  • Objectives : The aim of this study is to investigate the various effects of individual or combined extract of GamiSaengmaeksan (GSS) on cell viability, anti-inflammatory and antioxidant activityMethods : In order to evaluate cytotoxicity, MTT assay was performed. We investigated the levels of proinflammatory cytokines such as tumor necrosis factor (TNF)-α and interleukin (IL)-6 and interleukin (IL)-1β, and nitric oxide(NO) in LPS-induced RAW 264.7 cells to check the effects on anti-inflammatory activity. The level of NO production in RAW 264.7 cells was measured by using Griess reagent. The levels of cytokines and ROS were measured by Luminex and Flow cytometry, respectively.Results : At concentration of 200 ㎍/㎖ GSS, cytotoxicity was observed in RAW 264.7 cells. However, at concentration less than 100 ㎍/㎖ of both combine and individual GSS, cytotoxicity was not observed in Raw 264.7 cells. However, the level of ROS in RAW 264.7 cells were decreased at both extract of 100 ㎍/㎖ GSS. Also, the level of NO in RAW 264.7 cells were decreased from extraction of concentration of 100 ug/ml in GSS and individual-extraction of Liriopis Tuber, White Ginseng and Glycyrrhizae Radix. In addition, productions of pro-inflammatory cytokines (TNF-α) in LPS-induced RAW 264.7 cells were decreased from extraction of concentration of 10 and 100 (㎍/㎖) in GSS and individual-extraction of Liriopis Tuber.Conclusions : It is concluded that combined extract of GSS appears to be more effective in anti-oxidation and anti-inflammatory effect than those in individual-extraction of GSS. These results may be developed as a raw material for new therapeutics to ease the symptoms related with inflammatory and oxidative stress.

Separation of Copper & Cobalt by Solvent Extraction in Organic Acid Leaching Solution (유기산 침출용액에서 용매추출법에 의한 구리 및 코발트 분리)

  • A study has been made on the recovery & separation of cobalt and copper from organic acid leaching solution by solvent extraction. The experimental parameters such as the equilibrium pH, concentration of extractant and phase ratio were observed. Copper was extracted using LIX 84 and Cobalt was extracted using cyanex 272 and versatic acid 10. Experimental results showed that extraction percent of copper was 99% at above eq. pH 2.0 and then more than 90% of cobalt were extracted by cyanex 272 in eq. pH 6.0 and versatic acid 10 in eq. pH 7.5. Stripping of copper and cobalt from the loaded organic phases can be accomplished by sulfuric acid as a stripping reagent and 120 ~ 150 g/L of $H_2SO_4$ was effective for the stripping of copper and cobalt respectively. Finially, the basic optimal process for recovery of copper and cobalt from the bio-leaching solution was proposed.

High-Speed RNA Isolation Using Magnetic Oligo(dT) Beads and Lateral Magnetophoresis (올리고-dT 자성입자와 측면방향 자기영동을 이용한 초고속 RNA 추출 기술)

  • This paper presents a high-speed RNA microextractor for the direct isolation of RNA from blood lysate using magnetic oligo(dT) beads. The extraction is performed through lateral magnetophoresis, which is induced by a ferromagnetic wire array inlaid. With this RNA microextractor, more than 80% of the magnetic beads could be separated at a flow rate up to 20 ml/h, and the overall extraction procedure was completed within 1 min. The absorbance ratio of RNA to protein(A260/A280) was greater than 1.7, indicating that the extraction technique yields pure RNA. The feasibility of using this technique in reverse transcription polymerase chain reaction procedures was investigated by cDNA synthesis and PCR processes. The results confirmed that the RNA microextractor is a practical device for easy, fast, and high-precision RT-PCR using minimal amounts of reagent.

Recovery of Li from the Lithium Containing Waste Solution by D2EHPA (리튬함유 폐액으로부터 D2EHPA에 의한 리튬의 회수)

  • A study on the solvent extraction for the recovery of Li from lithium-containing waste solution was investigated using $D_2EHPA$ as an extractant. The experimental parameters, such as the pH of the aqueous solution, concentration of extractant and phase ratio were observed. Experimental results showed that the extraction percentage of Li was increased with increasing the equilibrium pH. More than 50% of Li was extracted in eq. pH 6.0 by 20% $D_2EHPA$. From the analysis of McCabe-Thiele diagram, 95% of Li was extracted by four extraction stage at phase ratio(O/A) of 3.0. Stripping of Li from the loaded organic phases can be accomplished by sulfuric acid as a stripping reagent and 90 ~ 120 g/L of $H_2SO_4$ was effective for the stripping of Li. Finially, Li was concentrated about 11.85 g/L by continuous stripping process, and then lithium carbonate was prepared by precipitation method.

Effective Extraction of Oligomeric Proanthocyanidin (OPC) from Wild Grape Seeds

  • The Oligomeric proanthocyanidin (OPC) in green and black tea, grape seeds, grapes and wine has raised much attention but that OPC in wild grape seed remains to be intensively investigated. This study investigated the total OPC contents and total antioxidant activity of wild grape seeds and developed an efficient extraction process with various temperatures, solvent compositions and times. Also, a chromatography column packed with the Dia-ion HP-20 resin was used for further purification of the ope. The total OPC contents were determined with the Folin-Ciocalteu reagent, and the antioxidant activity using total antioxidant potential (TAP) and 1,1-dipheny|-2picrylhydrazy| (DPPH). The yield of final purified OPC was 1.78 (+)-catechin equivalent (CE) g/100 g, with $IC_{50}$ activities of TAP and DPPH of 31.60 and $15.70\;{\mu}\;g/mL$. These activi­ties of the final purified OPC were about two times higher than that of the BHA used as a refer­ence sample.

A Study on Leaching and Solvent Extraction for the Recovery of Copper Ore for Small-Scale Mining in Tanzania (탄자니아의 소규모 광산에서 구리광석 정제를 위한 침출 및 용매 추출에 관한 연구)

  • Tanzania has abundant copper deposits, but copper-metal extraction remains low there, owing to the lack of suitable copper recovery processes and insufficient funds for developing mining technologies. Accordingly, leaching and solvent extraction methods for the extraction of copper from copper ore were studied with a particular emphasis on developing a simple processing method for small-scale copper mining. Chrysocolla ore was used as the copper-bearing mineral and sulfuric acid was used as the leaching reagent. A maximum copper recovery of 95.1% was obtained when the particles in the sample were smaller than $53{\mu}m$, the concentration of 98%(w/w) sulfuric acid in the leaching solution was 5.0 g/L and the stirring rate was between 60 and 80 rpm. The highest selectivity of $Cu^2+$ in the solvent extraction was obtained using 15% LIX-70 in kerosene. In the pH range from 0.5 to 3.0, the efficiency of $Cu^2+$ extraction increased with increasing pH. However, at pH values higher than 3.0, other metal ions were extracted into the organic phase more readily than $Cu^2+$. The highest solvent extraction rate obtained was 96.5% at pH values of 2.0 and 3.0 using 15% LIX-70.

Determination of Aloesin in Plasma by High-Performance Liquid Chromatography as Fluorescent 9-Anthroyl Derivative

  • A sensitive high-performance liquid chromatographic (HPLC) method for the determination of aloesin in plasma was developed. After solid-phase extraction from plasma and derivatization of aloesin and compound AD-1, which was prepared from aloesin as a internal standard, with 9-anthroylnitrile in the presence of quinuclidine, the derivatives were separated on a Inertsil ODS-3 column using acetonitrile/methanol/water (3:1:6) as a mobile phase, and detected fluorimetrically at 460nm with excitation at 360nm. The detection limit of aloesin was 3.2ng/ml in plasma (S/N=3).

Resolution of Salbutamol Enantiomers in Human Urine by Reversed-phase High Performance Liquid Chromatography after Derivatization with (S)-(-)-${\alpha}$-methylbenzyl isocyanate

  • A stereospecific HPLC method has been developed for the resolution of the enantiomers of salbutamol in human urine. After solid-phase extraction and derivatization with (S)-(-)-${\alpha}$-methyl-benzyl isocyanate, the diastereomeric derivatives were resolved $(R_s=1.59)$ on $5{\mu}M$ octadecylsilan column using 47% methanol as a mobile phase with fluorescence detection. The detection limit of each enantiomer was 10 ng/ml (S/N=3).

  • A stereospecific HPLC method has been developed for the resolution of the enantiomers of salbutamol in human urine. After solid-phase extraction and derivatization with 2,3,4,6-tetra-O-acetyl-$\beta$-D-glucopyranosyl isothiocyanate, the diastereomeric derivatives were resolved (Rs=1.83) on $5{\mu}m$ octadecylsilan column using 35% acetonitrile in 0.05M ammonium acetate buffer (pH=6) as a mobile phase with electrochemical detection. The diastereomeric derivatives were formed within 30 min. The detection limit of each enantiomer was 20 ng/ml (S/N=3).

Spectrophotometric Determination of Nickel in Steel with Di-2-pyridylmethanone 2-Thiophenecarboxylic Hydrazone

  • Di-2-pyridylmethanon 2-thiophenecarboxylie hydrazone(DPMTCH) was synthesized and used as a chromogenic reagent in the spectrophotometric determination of nickel in steel. The proton disc spectrophotometrically, were $pK_{a1}= 2.66$ and $pK_{a2}= 10.98$ ${\mu}=0.5(NaCl)$. Nickel(II) reacts with DPMTCH to form a 1:2 metal-2-ligand ratio complex, the xylene solution of which has an absorption maximum at 417 nm. Beer's law is obeyed over the range $0-1.17\;mg\;ml^{-1}$ of nickel(II). The molar absorptivity of the nickel(II) complex is $4.17{\times}10^4l\;mol^{-1}\;cm^{-1}$. The precision of the method was estimated for 4.7 mg of nickel(II), the coefficient of variation for 15 replicate determinations being 0.34%.
