Effect of Titanium Coating on Cell Adhesion and Extracellular Matrix Formation in Human Osteoblast-like MG-63 Cells

  • A variety of titanium (Ti) and its alloys are used in the clinical procedures of bone regeneration for periodontal and dental implant therapies. This study was performed to determine the effect of different surface dental implant materials on biologic responses of a MG-63 human osteoblast-like cell line. MG-63 cells were cultured on Ti coated with hydroxyapatite (HA), calcium metaphosphate (CMP), anodized (A), which compared with non-coated Ti (control). The appearances of surface of dental implant materials and the morphology of these cells were assessed by scanning electron microscopy (SEM). The gene expression profiles of MG-63 cells cultured on Ti were examined by human cDNA microarray (1,152 elements). The expression of several genes was up- and down-regulated by different surfaces of dental implant materials. Interesting, the genes correlated with cellular adhesion and extra cellular matrix (ECM) formation were enhanced, in accordance surface morphology of the dental implant materials used.

Endothelial Cell Seeding onto Extracellular Matrix for Development of Polyurethane Vascular Prosthesis (폴리우레탄 인공혈관을 위한 extracellular matrix 기질상의 내피세포이식)

  • Many experiments about endothelial cell seeding on artificial vessels were studied and conducted For this one or a combination of the extramatrix was used for the underlying matrix. But we used the whole ECM(extracellular matrix) that made excreated from flbroblasl. In thls study, we obtained human adult omental microvascular endothelium by collagenase digestion and used polyurthane sheets in order to make a new artificial vessel material. We cultured fibroblast on the polyurethane and gelatin - coated polyurethane. After confluent ingrowth we treated the polyure thane with triton in order to destroy the cytoskeleton and nucleus. We observed the preformed extra cellular matrix on the ployurethane and cultured the isolated microvascular endothelium. We also ok served the growth of microvascular endothelium on the polyurethane and gelatin. We conclude that the use of the whole ECM is promising fair as a new underying substrate for endothelial cell seeding on artificial vessels.

Study on Photo-aging Inhibition Effect of Microalgae-derived Oil for Cosmetic Material Development (화장품 소재 개발을 위한 미세조류 유래 오일의 광노화 억제 효과 연구)

  • Ultraviolet (UV) radiation causes inflammation and matrix metalloproteinase (MMP) over-expression and extra cellular matrix depletion, leading to skin photo-aging such as wrinkle formation, dryness, and sagging. In this study, we demonstrated that pretreatment with the hexane extract of microalgae protects UVB mediated cell damages. The results of clinical study showed that Microalgal Oil treated group reduced wrinkle and improve elasticity. All these results suggest Microalgal Oil may be useful as new photo-aging cosmetics for protection against UV induced activity.

  • Backgrounds and Objectives: Metastasis is a complex multistep process that requires sequential interactions between the invasive cell and the extra-cellular matrix. Transforming growth factor-${\beta}1$ ($TGF-{\beta}1$) is a multifunctional regulator of cellular differentiation, motility and growth. Loss of sensitivity to the growth inhibitory effects by $TGF-{\beta}1$ plays important roles in neoplastic progression. The aim of this study was to investigate the role of $TGF-{\beta}1$ in the neoplastic invasion and metastasis through matrix metalloproteinase (MMP) of laryngeal cancer cell lines. Material and Methods: Two laryngeal cancer cell lines, SNU-899 and SNU-1076 were treated with recombinant $TGF-{\beta}1$, and the expression of MMP-2 and MMP-9 was immunohistochemically evaluated and gelatinase activity was studied by gelatin zymogram. Results: The cell growth inhibition was evident on 4th days after 1ng/ml and 10ng/ml $TGF-{\beta}1$ treatment. The expressions of MMP-2 and MMP-9, and their gelatinase activities were increased in dose-dependent manner. Conclusion: $TGF-{\beta}1$ treatment in laryngeal cancer cell lines induces the expression of MMP-2 and MMP-9, thus playing a role in the digestion of extracellular matrix gelatin.

  • Tissue engineering (TE) has been developed to create functional organs and tissue by combining 3D matrix and cells in vitro. Vascularization and angiogenesis are utmost important for supply of nutrients and oxygen in tissue engineered organs. The present study was performed to isolate and characterize primary endothelial cells (EC) from aorta of alpha 1, 3-enzyme galactosyltransferase knock out (GalT KO) pig, to minimize immune rejection and analyze body immune system for future xenotransplantation studies. Isolation of primary EC from aorta were performed by incubation with dispase for 8-10 min at $37^{\circ}C$. Primary EC were cultured in EC growth medium on different extra cellular matrix (ECM), either collagen or gelation. Primary EC exhibits morphological characteristics and showed positive expressions of EC specific marker proteins i.e. PECAM1, KDR and VWF despite of their ECM surface; however, on collagen based surface they showed increase in mRNA level analyzed by qPCR. Primary EC cultured on collagen were sorted by flow cytometer using KDR marker and cultured as KDR positive cells and KDR negative cells, respectively. KDR positive cells showed dramatically increased in PECAM1 and VWF level as compared to KDR negative cells. Based on the above results, primary EC derived from GalT KO are successfully isolated and survived continuously in culture without becoming overgrown by fibroblast. Therefore, they can be utilize for xeno organ transfer, tissue engineering, and immune rejection study in future.

  • We have shown that it is possible to form a fibrilar network of fibronectin on a polyelectrolyte polymer film whose dimensions are similar to those reported on the extra cellular matrix. The fibronectin network was observed to form only when the charge density of the polymer was in excess of the natural charge density of the cell wall. Furthermore, the self-organized fibronectin layer was much thicker than the polymer film, indicating that long ranged interaction may playa key role in the assembly process. It is therefore important to understand the structure of the polymer layer/protein interface. Here we report on a neutron reflectivity study where we explore the structure of the polyelectrolyte layer, in this case sulfonated polystyrene (PSSx,), with varying degree of sulfonation (x<30%), as a function of sulfur content and counter ion concentration. These results are then correlated with systemic study of the adsorption and the multilayer formation of fibronectin as a function of incubation time for various sulfonation levels of $PSSx.^1$

  • The diseases caused by Xylella fastidiosa are associated with aggregation of the bacteria m xylem vessels, formation of a gummy matrix and subsequent blockage of water uptake. In the closely related pathogen, Xanthomonas campestris, xanthan gum is known to be an important virulence factor, probably contributing to bacterial adhesion, aggregation and plugging of xylem. Xanthan gum, produced by X. campestris, is an extra-cellular polysaccharide consisting of a cellulose backbone ($\bate$-1,4-linked D-glucose) with trisaccharide side chains composed of mannose, glucuronic acid and mannose attached to alternate glucose residues in the backbone. We had constructed a mutant of X. campestris lacking gumI gene that is responsible for adding the terminal mannose for producing modified xanthan gum which is similar to xanthan gum fromX. fastidiosa. The modified xanthan gum degrading endgphytic bacterium Acineto-bacter johnsonii GX123 isolated from the oleander infected with leaf scorch disease.

In vitro Cartilage Regeneration using Primary Chondrocytes Cultured within Porous Poly(lactide-co-glycolide) Scaffolds

  • Cartilage injuries are frequent nowadays. The previous surgical treatment of cartilage defect was limited. Another approach in the treatment of cartilage injuries is the use of reconstitute cartilage consisting of chondrocytes cultured in suitable biodegradable scaffolds. Current studies have demonstrated the compatibility of chondrocytes with different biomaterials and the chondrogenesis in various types of porous scaffolds. The cell ingrowth into the porous scaffolds is modulated by initial cell loading efficiency. Therefore, well-interconnected pore structure and even pore distribution of the scaffolds are essential for efficient cell seeding. According to our previous work, well-interconnected macroporous scaffolds can be prepared by gas-foaming/salt-leaching method using ammonium bicarbonate salt as porogen additives. In this work, primary chondrocytes were cultured in PLGA 65/35 scaffolds fabricated by using our method. Cells seeded in the scaffolds showed well distribution by agitated seeding method. Histochemical staining of proteoglycans present in the scaffolds was used to visualize the chondrocyte ingrowth in the scaffolds. At 3 weeks, the population of chondrocytes was increased for the most part of the scaffolds, and extra cellular matrix (ECM) secretion was increased as culture periods progressed.

Functional Classification of Gene Expression Profiles During Differentiation of Mouse Embryonic Cells on Monolayer Culture

  • Embryonic stem (ES) cells have a capability to generate all types of cells. However, the mechanism by which ES cells differentiate into specific cell is still unclear. Using microarray technology, the differentiation process in mouse embryonic stem cells was characterized by temporal gene expression changes of mouse ES cells during differentiation in a monolayer culture. A large number of genes were differentially regulated from 1 day to 14 days, and less number of genes were differentially expressed from 14 days to 28 days. The number of up-regulated genes was linearly increased throughout the 28 days of in vitro differentiation, while the number of down-regulated genes reached the plateau from 14 days to 28 days. Most differentially expressed genes were functionally classified into transcriptional regulation, development, extra cellular matrix (ECM),cytoskeleton organization, cytokines, receptors, RNA processing, DNA replication, chromatin assembly, proliferation and apoptosis related genes. While genes encoding ECM proteins were up-regulated, most of the genes related to proliferation, chromatin assembly, DNA replication, RNA processing, and cytoskeleton organization were down-regulated at 14 days. Genes known to be associated with embryo development or transcriptional regulation were differentially expressed mostly after 14 days of differentiation. These results indicate that the altered expression of ECM genes constitute an early event during the spontaneous differentiation, followed by the inhibition of proliferation and lineage specification. Our study might identify useful time-points for applying selective treatments for directed differentiation of mouse ES cells.
