Cloning, Purification, and Characterization of a New DNA Polymerase from a Hyperthermophilic Archaeon, Thermococcus sp. NA1

  • Genomic analysis of Thermococcus sp. NA1 revealed the presence of a 3,927-base-pair (bp) family B-type DNA polymerase gene, TNA1_pol. TNA1_pol, without its intein, was overexpressed in Escherichia coli, purified using metal affinity chromatography, and characterized. TNA1_pol activity was optimal at pH 7.5 and $75^{\circ}C$. TNA1_pol was highly thermostable, with a half-life of 3.5h at $100^{\circ}C$ and 12.5h at $95^{\circ}C$. Polymerase chain reaction parameters of TNA1_pol such as error-rate, processivity, and extension rate were measured in comparison with rTaq, Pfu, and KOD DNA polymerases. TNA1_pol averaged one incorrect bp every 4.45 kilobases (kb), and had a processivity of 150 nucleotides (nt) and an extension rate of 60 bases/s. Thus, TNA1_pol has a much faster elongation rate than Pfu DNA polymerase with 7-fold higher fidelity than that of rTaq.

인삼 재배 시 토직모 생산에 적합한 유기물 선발 (Selection of Suitable Organic Matter for To-jik Nursery in Panax ginseng C.A. Meyer)

  • This experiment was carried out to select suitable organic matter in To-jik nursery (self soil nursery) for complement To-jik nursery's defects that are deterioration of raw material by poor quality of seed ginseng and reduction of the quantity in seed ginseng production. Organic matter used were Yacto, rice bean, defatted rice bran, soybean cake and their mixture. As follows, bulk density in soil physical property by treating organic composts was the greatest in soybean cake and the next was followed by mix, Yacto, defatted rice bran, and rice bran treatment in order. Soil pore space ratio was totally the opposite; that was rice bran the first and followed by defatted rice bran, Yacto, mix and soybean cake treatment. The incidence rate of damping off by treating organic composts was 1.5% in both soybean cake and mix while the others was 1.0%. Emergence time was the same among treatment on April 16 and Emergence rate was the highest at 73% in Yacto. There was no significant differences among treatment in the growth of aboveground part but it was a little better in defatted rice bran treatment. In Yacto treatment, the growth of underground part, total root number per kan, rate of first grade ginseng seedling, and rate of usable ginseng seedling etc. were entirely higher but there was little differences. Using defatted rice bran was slightly lower in productivity compared to Yacto, but the possibility was high as a alternative for Yacto in a view of managing cost down.

초기 자궁경부암의 수술후 방사선치료 결과 (Therapeutic Results of Postoperative Radiation Therapy for Early Stage Uterine Cervical Cancer)

  • This is a retrospective analysis of 67 patients with histologically proven invasive carcinoma of uterine cervix treated with surgery followed by adjuvant radiotherapy at Inje University Seoul Paik Hospital between october 1983 and september 1991, Postoperative radiotherapy was carried out in patients with high risks of locoregional recurrence such as positive pelvic lymph node (38 pts), large tumor size more than 3 cm (22 pts), cervical stromal invasion more than 2/3 (46 pts), parametrial involvement (9 pts), positive resection margin (14 pts), endo/myometrial extension (10 pts), and angiolymphatic invasion (13 pts). Stage I A, I B, and IIA were 2 $(3\%),$ 39 $(58.2\%),\;and\;26\;(38.8\%),$ respectively. Median follow-up period was 48 months with ranges from 13 to 115 months. All 67 patients were treated externally with standard pelvic field with radiation dose ranging from 4080 to 6120 cGy in 4~6 weeks period of time. Of these, 45 patients received intracavitary radiotherapy. The overall survival rate and disease free survival rate at 5-year were $88.0\%\;and\;82.1\%,$ respectively. The survival rates by stage were $87.1\%$ in IB and $88.4\%$ in IIA. Local control rate was $80.6\%(58\;pts).$ The treatment failure was noted in 12 of 67 patients $(17.9\%):$ locoregional failure in $7(10.4\%),$ distant metastasis in 3 $(4.5\%),$ and locoregional and distant metastasis in $2(3\%),$ The univariate analysis of prognostic factors disclosed endo/myometrial extension as a significant factor of survival and recurrence $(70.0\%\;vs\;91.1\%\;P<0.05\;&\;30.0\%\;vs\;15.8\%,\;respectively).$ The complication of postoperative radiothrapy was not significant and all patient were well tolerated. In conclusion, postoperative radiotherapy in patients with high risks of locoreginal recurrence is relatively well tolerated and it gives significantly improved survival rate especially in patients with positive lymph nodes, bulky tumor size $(\geqq3\;cm),$ parametrial involvement, cervical stromal invasion more than 2/3, positive resection margin and angiolymphatic invasion.

Insect Adaptations to Changing Environments - Temperature and Humidity

  • The most important factors in environment that influence the physiology of insects are temperature and humidity. Insects display a remarkable range of adaptations to changing environments and maintain their internal temperature (thermoregulation) and water content within tolerable limits, despite wide fluctuations in their surroundings. Adaptation is a complex and dynamic state that widely differs in species. Surviving under changing environment in insects depends on dispersal, habitat selection, habitat modification, relationship with ice and water, resistance to cold, diapause and developmental rate, sensitivity to environmental signals and syntheses of variety of cryoprotectant molecules. The mulberry silkworm (Bombyx mori) is very delicate and sensitive to environmental fluctuations and unable to survive naturally because of their domestication since ancient times. Thus, the adaptability to environmental conditions in the silkworm is quite different from those of wild insects. Temperature, humidity, air circulation, gases and photoperiod etc. shows a significant interaction in their effect on the physiology of silkworm depending upon the combination of factors and developmental stage affecting growth, development, productivity and quality of silk. An attempt has been made in this article to briefly discuss adaptation in insects with special emphasis on the role of environmental factors and their fluctuations and its significance in the physiology of mulberry silkworm, B. mori.

스판덱스 혼합 면 편성물과 면 편성물의 세탁에 따른 변형 비교 (The Deformation of Knitted Cotton Fabrics with/without Spandex During Laundering)

  • This study evaluate the effect of laundering on the deformation of knitted spandex/cotton fabrics compared with that of knitted cotton fabrics. Commercial knitted spandex/cotton and knitted cotton fabrics for T-shits were laundered in a drum-type washing machine and dried in a tumble dryer. Wale spirality, shrinkage, elastic recovery and surface contour of knitted fabrics were investigated under different laundering conditions: washing temperature, presoaking time and washing cycles. Knitted spandex/cotton fabrics had a lower angle of spirality than knitted cotton fabrics. After the first washing cycle, the angles of spirality of all the fabrics had decreased greatly. Knitted cotton fabric of low density deformed more than that of higher density. Knitted spandex/cotton. fabric of low density shrank less, because of the greater extension given during heat-set. Permanent elongation length at the 80$\%$ extension was longer than at the 50$\%$ extension, and the knitted spandex/cotton fabric which was expanded greatly during heat-set had a lower elastic recovery rate. The surface appearance of the knitted spandex/cotton fabrics was worsl~ than that of the knitted cotton fabrics before laundering and after repeated laundering, because of the much protruded cotton fibers from the yarns.

Effects of Stubble Height, Irrigation and Nitrogen Fertilization on Rice Ratooning in Korea

  • Rice (Oryza sativa L.) ratooning is the production of a second rice crop from the stubble left behind after the main crop harvest. The objectives of this study were to evaluate the effects of main-crop stubble height, irrigation and fertilization on ratoon grain yield. Main crop 'Jinbuol' rice cultivar was harvested to leave with 10, 20, 30, or 40 cm stubble height. When the main crop stubble was harvested with 10 cm height, ratoon rice grain yield was increased by 2,810 kg/ha. Irrigation on stubbles after main crop harvest did not affect the ratoon crop yield and rice quality. The results showed a large variation in the ratoon performance by fertilizer application methods. Top-dressed nitrogen fertilizer on the stubble after harvest caused increase in panicle production and higher maturity rate. However, there was no significant difference in protein content, amylose content of milled rice and cooked rice characters between plots managed with and without nitrogen fertilizer.

한국표준형원전의 격납건물종합누설률 시험 주기연장에 대한 리스크 평가 (Risk Assessment of Integrated Leak Rate Test(ILRT) Extension for Korea Standard Nuclear Power Plant)

  • An ILRT Interval for a nuclear power plant in Korea was extended from once in five years to once in ten years. Therefore, it is necessary to evaluate risk impact for ILRT interval extensions. In this paper, input data were generated for the reference plants, KSNP, using raw data such as meteorological data, population distribution data and source term data. And, using MACCS II code the risk impact assessment was performed based on the two methodologies of NUREG-1493 and NEI Interim Report. The risk impact derived from an ILRT interval extension was identified not to be significant. It is considered to apply this study and results to making an accident management plan and safety goal, and to the field of public acceptance.

Multi-view Rate Control based on HEVC for 3D Video Services

  • In this paper, we propose two rate control algorithms for multi-view extension of HEVC with two rate control algorithms adopted in HEVC and analyze the multi-view rate control performance. The proposed multi-view rate controls are designed on HEVC-based multi-view video coding (MV-HEVC) platform with consideration of high-level syntax, inter-view prediction, etc. not only for the base view but also for the extended views using the rate control algorithms based on URQ (Unified Rate-Quantization) and R-lambda model adopted in HEVC. The proposed multi-view rate controls also contain view-wise target bit allocation for providing the compatibility to the base view. By allocating the target bitrates for each view, the proposed multi-view rate control based on URQ model achieved about 1.83% of average bitrate accuracy and 1.73dB of average PSNR degradation. In addition, about 2.97% of average bitrate accuracy and 0.31dB of average PSNR degradation are achieved with the proposed multi-view rate control based on R-lambda model.

송풍식 줄기절단기에 의한 적정 양파 잎 절단 시기 및 잔여 엽장에 따른 양파 저장 특성 (Appropriate Working Period and Storage Characteristics Based on Residual Leaf Length of Onion (Allium cepa L.) Harvested with a Blower-type Stem Cutter)

  • 최근 국내에서 개발된 송풍식 양파 줄기절단기를 수확기 양파 엽 절단에 활용할 경우 적정 작업 조건을 구명하기 위해서 이 실험을 수행하였다. 처리구 중 식물체 엽 건조가 가장 많이 진행된(엽 건조 정도 : 66.3%, 엽 수분함량 : 50.5%) 6월 20일 엽 절단 처리구에서 평균 잔여 엽장은 6.7±3.5cm로서 작업 후 적정 잔여 엽장에 해당되는 범위인 4-10cm에 포함되므로 기계 엽 절단 성능이 처리구 중 가장 우수한 것으로 판단된다. 줄기절단기 이용 양파 엽 기계 절단 시 평균 작업 속도는 0.17m·s-1였는데, 이는 인력 엽 절단 처리구의 평균 작업 속도인 0.05m·s-1보다 3.4배 정도 빨랐으며, 이를 통해 해당 기종을 이용하여 10a 면적을 작업할 경우에는 인력 작업(1인 기준)에 비해 2.6시간 정도를 절감할 수 있을 것으로 기대된다. 또한 기계 엽 절단 처리구에서의 손상구 발생률은 1.3%로서 인력 엽 절단 처리구의 0.0%에 비해 높았는데, 이로 인해 기계 엽 절단 처리구가 인력 엽 절단 처리구보다 저장 중 양파 구 부패율이 평균적으로 높았던 것으로 판단된다. 처리별 저장 특성을 살펴보면, 저장 8개월 후 구 부위(기부, 정부)별 부패율은 기계 엽 절단 후 잔여 엽장이 5.0cm 미만인 처리구에서 잔여 엽장이 5.0cm 이상인 처리구보다 높았다. 이는 잔여 엽 길이가 5.0cm 미만인 처리구에서는 5.0cm 이상인 처리구보다 저장 중 구 부패를 유발하는 병원균의 감염이 쉬우므로 장기 저장 시 부패율이 높은 것으로 생각된다. 본 실험 결과와 실험 기종의 성능 목표(작업 후 잔여 엽장 : 5cm) 및 양파 수확 시 적정 잔여 엽장에 관한 기존의 연구결과 등을 종합적으로 고려할 경우, 본 실험에 사용된 줄기절단기 이용 양파 엽 절단 시 양파의 적정 잔여 엽장은 5-10cm 정도일 것으로 판단된다.

베리류 발효톱밥 종류에 따른 흰점박이꽃무지 발육 및 산란 특성 (Development and Oviposition Characteristics of Protaetia brevitarsis (Coleoptera: Cetoniidae) fed with Fermented Sawdust from Different Berries)

  • 뽕나무, 복분자, 블루베리의 전정 가지 대부분은 버려지고 있다. 따라서, 이들 버려지는 전정 가지를 활용하여 흰점박이꽃무지의 사육 가능성을 검토하기 위하여 발육 및 산란 특성을 조사하였다. 베리류 발효톱밥을 먹인 흰점박이꽃무지 유충 발육 기간은 뽕나무 발효톱밥 157.3일, 복분자 발효톱밥 130.3일, 블루베리 발효톱밥은 140일로 참나무 발효톱밥 169.3일에 비하여 짧아지고, 유충 무게가 증가하였으며, 생존율이 높았다. 흰점박이꽃무지 유충 집단 사육시 2.5 g 이상인 유충 비율은 3종류의 발효톱밥 모두 6주에서 8주사이에 60% 이상인 반면 같은 기간에 참나무 발효톱밥은 10% 이내였고, 10주가 지나야 2.5 g 이상인 유충의 비율이 높아졌다. 흰점박이꽃무지의 평균 산란수는 80개 이상으로 평균 산란기간은 9주정도 소요되었지만, 개체당 편차가 커서 유의차는 없었다. 3종류의 베리류 발효톱밥은 서로 혼합하여 사용해도 참나무 발효톱밥에 비해 유충 발육속도가 빠르고, 산란수는 차이가 없었다. 따라서, 흰점박이꽃무지를 사육하는데 있어서 베리류 발효톱밥이 참나무 발효톱밥에 비하여 효과적으로 추정된다.
